Abstract
本发明公开了一种NTDs鼠胚动物模型及其构建方法,对孕6~10天的正常孕鼠一次性给予剂量19~23mg/kg的维甲酸进行灌胃干预,能够在孕10.5天后快速诱导构建稳定高效的NTDs鼠胚动物模型,进一步分离鼠胚脑泡组织细胞培养,还可以建立NTDs细胞模型。利用本发明构建的动物模型和细胞模型,可以在体内和体外为研究NTDs的分子和细胞机制提供良好的平台。

技术领域
本发明涉及一种动物模型的构建方法,特别是涉及一种NTDs鼠胚动物模型的构建方法,以及利用该方法构建的NTDs鼠胚动物模型和细胞模型。
背景技术
神经管畸形(neural tube defects, NTDs)是人类最主要的出生缺陷疾病,发病率排在第二位。
NTDs是由于在胚胎发育过程中,神经管闭合不全引起的一类先天畸形,主要表现是脑和脊髓的异常,临床上以先天性无脑畸形和脊柱裂常见,但其发病机制尚不清楚。
全球有每年300000新生儿罹患,发病率约为0.5~2‰。发展中国家发病率更高,在山西省的某些地区高达13.9‰,严重影响人口质量的同时,也给社会和家庭带来了沉重的负担。
神经管发生是胚胎早期一个重要的涉及中枢神经系统原基形成的发育学事件,从单侧神经上皮细胞构成的神经板逐渐演变为长管状神经管的过程称为神经胚形成。神经管闭合包含四个有序而重叠的阶段,即神经板形成、神经板塑形、神经板卷褶和神经褶融合,其中任何一个环节受到干扰,都会导致NTDs的发生。
人NTDs标本来源有限,构建NTDs动物模型有助于深入认识人类NTDs的病因、发病机制和病理特点,对研究防治措施具有重要意义。鼠类与人的神经管发育模式非常相似,所以是目前研究NTDs最常见的模式生物。
NTDs是遗传和环境因素共同作用的结果。现有NTDs动物模型的构建方法主要包括基因敲除和环境诱导两种方式。
目前在小鼠中已经发现了超过200个基因可导致NTDs表型,但这些基因在人群中却没有得到很好的验证。
环境因素对于神经管闭合具有深远的影响,叶酸能有效预防NTDs就是很好的证据。但近年来研究发现,30~50%的NTDs是不能通过叶酸来预防的。因此,其他的环境因素也应该引起重视。目前已构建了妊娠期高温、高糖以及一些化学因素(如顺铂、环磷酰胺、丙戊酸等)诱导的NTDs鼠胚模型,以用于NTDs发病机制的研究。
由于环境因素复杂多样,研究不同环境对于NTDs发生的影响具有重要意义。维生素A在女性生殖和胚胎发育中具有重要作用,其在NTDs发生中的作用及机制值得探讨,建立相应的NTDs模型,将会为相关研究提供良好的平台。
发明内容
本发明的目的是提供一种NTDs鼠胚动物模型及其构建方法,以利用所构建的鼠胚动物及细胞模型深入研究NTDs的发生机制。
本发明具体是提供了一种采用维甲酸干预以构建NTDs鼠胚动物模型的方法。维甲酸是维生素A在体内的活性衍生物,其在体内的分布对胚胎发育具有重要的调节作用。维甲酸代谢紊乱可以引起鼠胚发育异常,本发明发现,维甲酸干预能够快速诱导构建稳定高效的NTDs鼠胚动物模型。
本发明所述NTDs鼠胚动物模型的构建方法是:使用正常孕鼠,在孕6~10天时一次性给予剂量19~23mg/kg的维甲酸进行灌胃干预,孕10.5天后得到NTDs鼠胚动物模型。
进一步地,所述维甲酸的最佳干预剂量为21mg/kg。
孕6~10天是鼠胚神经管发育的关键阶段,在这个阶段,以合理剂量的维甲酸进行干预,能够成功得到NTDs鼠胚动物模型。
优选地,构建NTDs鼠胚动物模型的最佳维甲酸干预时间是孕7.5天。
本发明将小鼠按照一定的雌雄比例合笼得到孕鼠后,采用上述方法进行维甲酸药物干预,并以采用等体积香油干预的孕鼠作为对照。规定孕周时在体视显微镜下观察鼠胚发育情况,对照孕鼠鼠胚发育正常,药物干预孕鼠的鼠胚发育异常,出现明显的发育迟缓、神经管闭合障碍,主要表现为脑部畸形。证明本发明方法能够快速稳定的构建成功NTDs鼠胚动物模型。
进而,本发明还在NTDs鼠胚动物模型的基础上,进一步分离出鼠胚脑泡组织进行细胞培养,建立了NTDs细胞模型,并对其细胞增殖、细胞周期和细胞分化等特性进行了研究。利用本发明所构建的动物模型和细胞模型,可以为研究NTDs的分子和细胞机制提供良好的平台。
神经干细胞(neural stem cells, NSCs)具有增殖和自我更新能力,在特定条件诱因下,能够向各种类型神经元或神经胶质细胞分化,是神经系统形成和发育的基础,其增殖、凋亡和分化对于神经管的发育非常重要。本发明发现,维甲酸干预孕鼠的鼠胚发育过程中,可以引起鼠胚神经干细胞增殖及向神经元和神经胶质细胞的分化能力受损。
本发明进一步发现,Nr2e1基因是孤儿核受体家族成员,主要在脑中表达,其异常表达可以导致小鼠视网膜和脑部发生异常。基于本发明建立的NTDs鼠胚动物模型和细胞模型,对Nr2e1基因在NTDs发生中的作用机制进行了研究,表明Nr2e1可通过影响NSCs的增殖和分化参与维甲酸诱导的NTDs发生过程,为探讨NTDs的发病机制提出了新的思路。

下面结合具体实施例对本发明技术方案进行进一步的说明。本发明所述实施例仅用于解释本发明,而不应理解为对本发明范围的限制。在不背离本发明技术方案的前提下,对本发明所作出的本领域技术人员容易实现的任何改动,都应认为是本发明的内容。
本发明所述实施例中的主要试剂和材料如下所述,未注明具体条件的实验方法,通常按照常规条件,或按照制造厂商所建议的条件进行。
实验动物:C57BL/6小鼠,9~10周龄,体重18~23g,SPF级,由广东省医学实验动物中心提供,涉及到小鼠的相关操作已通过广东省医学实验动物中心动物研究伦理委员会审查。
NTDs造模试剂:全反式维甲酸,购自美国Sigma公司。
细胞培养试剂:DMEM/F12培养基,购自美国HyClone公司;B27因子(不添加维生素A)、青链霉素和FBS,购自美国Gibco公司;bFGF和EGF,购自美国PeproTech公司;CCK-8试剂盒,购自东仁化学科技(上海)有限公司;LV3-Nr2e1 shRNA,由上海生工生物工程公司合成。
PCR试剂:Trizol,购自美国Ambion公司;反转录和PCR试剂,购自美国Thermo公司;所有引物由上海生工生物工程公司合成。
Western blot试剂:Nestin、Gfap、NF-H、Map2和Nr2e1抗体,均购自英国Abcam公司,beta actin抗体、鼠源和兔源二抗,购自北京中杉金桥生物技术有限公司。
原位杂交试剂:核糖核酸酶A(RNase A)、蛋白酶K、左旋咪唑、圆酵母RNA(tRNA),购自美国Sigma公司;骨髓细胞紫色指示剂AP底物(BM Purple AP substrateprecipitation)和抗地高辛抗体,购自瑞士Roche公司;小鼠Nr2e1探针,由南京尧舜禹生物公司合成。
实施例1:维甲酸干预构建C57BL/6小鼠胚胎NTDs模型。
于下午18时,将C57BL/6小鼠按雌雄比2∶1合笼交配,次日晨8时取出检查阴栓,将发现阴栓者的当天中午12时定为孕0.5天(E0.5d)。
将得到的孕鼠随机分为对照组(正常组)和实验组(NTDs组),在E7.5d,实验组孕鼠给予19~23mg/kg剂量范围的以香油配制的维甲酸溶液灌胃造模;对照组孕鼠给予等体积香油。
E10.5d,将孕鼠颈椎脱臼处死,75%乙醇擦拭腹部,捏起一小块皮肤,沿腹部中线剪至乳头线水平,再沿两侧剪开,打开腹腔。移开肠管和脂肪垫,分离出子宫,置于有冰PBS的平皿里。用冰PBS将子宫清洗3次,去除周围的血管和脂肪组织。
体视显微镜下,依次剥离子宫壁、蜕膜、Reichert’s膜及卵黄囊,分离出胚胎,观察对照组和实验组胎鼠形态及神经管发育情况,记录活胎鼠数、死胎鼠数、吸收胎数、畸形情况及其数量,计算致畸率。
如图1所示,正常组鼠胚发育良好,NTDs组鼠胚发育迟缓,神经管闭合障碍,呈现明显的脑部畸形,并伴随心脏、鳃弓及前、后肢芽等多部位畸形。
表1给出的不同剂量维甲酸诱导产生神经管畸形鼠胚统计结果显示,21mg/kg剂量组神经管畸形比例最高,吸收胎比例相对较低,为最佳造模剂量。
实施例2:NTDs鼠胚NSCs培养。
分离出正常组和NTDs组E10.5d鼠胚的脑泡组织,转移至15ml离心管中。加入少量DMEM/F12培养基,用1ml移液器反复吹吸,直至肉眼看不到组织块,采用200目滤网进行过滤,1000rpm离心5min,弃上清。加入NSCs增殖培养基(DMEM/F12培养基添加2%B27,20ng/mlEGF和20ng/ml bFGF),吹打混匀,调整细胞浓度为1×106/ml,接种于培养瓶,置于37℃/5%CO2培养箱悬浮培养。
图2显示经过培养后,正常组与NTDs组NSCs的增殖能力有显著差别。正常组NSCs在原代培养的第4天就能形成细胞集落,在第6天时集落显著增大增多,而NTDs组NSCs增殖能力明显低于正常组NSCs,在第6天时也几乎见不到细胞集落形成。
实施例3:NTDs鼠胚NSCs鉴定。
将正常组和NTDs组的NSCs调整浓度为1×105/ml,接种于包被多聚赖氨酸的24孔板中,每孔1ml,置于37℃/5% CO2培养箱中增殖24h。采用常规免疫荧光操作方法,选用Nestin抗体进行NSCs鉴定(anti-Nestin:1∶500)。
从图3结果可以看出,与正常鼠胚的NSCs相同,NTDs鼠胚脑泡分离培养的细胞也表达Nestin蛋白,证明其为NSCs。
实施例4:NTDs鼠胚NSCs增殖能力分析。
通过集落计数法和CCK-8法比较正常组和NTDs组鼠胚NSCs的增殖能力。
集落计数法为初级NSCs培养至第6天,计数NSCs集落形成数(直径>50μm)。
CCK-8法为调整NSCs浓度为2×105/ml,接种至96孔板中,每孔100μl,置于37℃/5%CO2培养箱,分别于0、2、4和6天时加入CCK-8,每孔10μl,37℃孵育4h,立即采用Biotek Eon微孔分光光度计测定450nm处吸光度,设定5个平行孔。
结果如图4所示,对两组NSCs在第6天形成的细胞集落进行计数,正常组NSCs集落数显著多于NTDs组,差异具有统计学意义(P<0.001)。CCK-8结果显示在0天时,正常组与NTDs组的OD值差异不明显(P>0.05),在随后的2、4和6天时,NTDs组的OD值都显著低于正常组,差异具有统计学意义(P<0.001)。
上述结果说明,NTDs鼠胚的NSCs增殖能力显著低于正常组NSCs。
实施例5:NTDs鼠胚NSCs细胞周期分析。
调整NSCs浓度为1×106/ml,移去上清液,加入PI染料(含有50μg/ml PI、10μg/mlRnase、1‰ Triton X-100),室温避光30min。采用流式细胞技术对G1、S和G2期的细胞数量进行检测。
由图5可知,NTDs组NSCs处于G1期的细胞数显著高于正常组,而处于S和G2期的细胞数显著低于正常组,差异具有统计学意义(P<0.05)。FCM结果表明,NTDs组NSCs被捕获在了G1期,无法进入S期(即DNA合成期)。
实施例6:NTDs鼠胚NSCs分化能力分析。
将NSCs调整浓度为1×105/ml,接种于包被多聚赖氨酸的24孔板中,每孔1ml,培养基中再添加5% FBS,37℃/5% CO2培养箱诱导分化7天。采用免疫荧光常规操作方法,选用Gfap用于检测星形胶质细胞,NF-H用于检测神经元(anti-Gfap:1∶500,anti-NF-H:1∶500)。
如图6结果显示,NTDs组NSCs向神经元和星形胶质细胞分化的能力都显著低于正常组,进一步说明NTDs组的NSCs是受损的细胞。
实施例7:Nr2e1基因在维甲酸诱导NTDs鼠胚模型中的应用研究。
1. 全胚胎原位杂交检测Nr2e1在不同时期鼠胚的表达。
预杂交和杂交:将分离鼠胚在PBST配制的75%、50%、25%甲醇溶液中进行梯度复水(冰上操作),每次5min。PBST洗2次,每次10min。6%H2O2漂白,冰上至少1h(E8.5~9.5d:1h;E10.5d:2h)。PBST冰上洗3次,每次5min。蛋白酶K处理,常温10min。2mg/ml甘氨酸溶液常温处理2次,每次5min。PBST洗2次,常温5min。4%多聚甲醛和0.2%戊二醛常温重新固定20min。PBST常温洗3次,每次5min。1ml杂交液洗10min。70℃预热杂交液中温育2h。加探针的杂交液中70℃温育过夜。
杂交后清洗、封闭和抗体孵育:用在70℃预热的溶液Ⅰ(50ml中含25ml甲酰胺,12.5ml 20×柠檬酸缓冲液,5ml 10%SDS,7.5ml DEPC水)清洗胚胎2~3次,每次30min(E8.5d:2次;E9.5~E10.5d:3次)。70℃预热的溶液Ⅰ与溶液Ⅱ(50ml中含5ml NaCl(5M),0.5ml Tris-HCl(1M,pH7.5),50μl Tween-20,44.5ml DEPC水)以1∶1混合洗10min。常温溶液Ⅱ洗3次,每次5min。加RNaseA的溶液Ⅱ(50ml中含5ml NaCl(5M),0.5ml Tris-HCl(1M,pH7.5),50μl Tween-20,0.25ml RNase A(20mg/ml),44.2ml DEPC水)37℃洗2次,每次30min。65℃预热的溶液Ⅲ(50ml中含25ml甲酰胺,5ml 20×柠檬酸缓冲液,20ml DEPC水)洗2~3次,每次30min(E8.5d:2次;E9.5~E10.5d:3次)。TBST洗3次,每次5min。加入封闭液,20℃孵育2.5h。移去封闭剂,加入1∶4000稀释的抗地高辛抗体,4℃过夜。
抗体后清洗及显色:TBST常温洗3次,每次5min。TBST常温洗5次,每次1~1.5h。TBST中4℃慢摇过夜。常温在NTMT(50ml中含1ml 5MNaCl,2.5ml 2M Tris-HCl(pH9.5),1.25ml MgCl2·6H2O(2M),50μl Tween-20,25mg左旋咪唑,45.2ml DEPC水)中洗3次,每次至少10min。移去NTMT,加入显色剂BM Purple AP substrate,常温避光缓慢摇动,直到显出合适颜色为止。PBST/5mM EDTA清洗几次,终止显色反应。4%多聚甲醛/PBST中固定几小时至过夜,蓝染为阳性结果。
图7A的全胚胎原位杂交结果显示,Nr2e1主要表达在鼠胚的前脑,但在E8.5d的鼠胚中几乎都检测不到蓝染的阳性结果,正常鼠胚脑中的蓝染面积随着发育逐渐增大,说明Nr2e1基因表达逐渐增多,而NTDs组胚胎几乎没有变化。
2. RT-PCR检测Nr2e1在不同时期鼠胚的表达。
采用维甲酸建立NTDs鼠胚模型,分别于E8.5d、E9.5d和E10.5d分离胎鼠脑泡,采用Trizol法提取RNA。
反转录(RT)反应体系:5×Reaction Mix,4μl;RiboLock RNase Inhibitor,1μl;10mM dNTP Mix,2μl;RevertAid M-MuLV RT,1μl;Oligo(dT) primer,1μl;Template RNA,1μg; RNase-Free Water,补足20μl。
反应条件:42℃,60min;70℃加热5min终止反应。
PCR反应体系:PCR Master Mix(2×),5μl;上游引物与下游引物,各0.5μl;基因组DNA,2μl;RNase-Free Water,2μl。
反应条件:(94℃,5min)×1cycle;(94℃,30s;55℃,30s;72℃,1min)×35cycles;(72℃,10min)×1cycle。
配制2%琼脂糖凝胶电泳,采用1×TAE电泳液,电压120V,电泳40min。
图7B结果可以看出,在E8.5d的鼠胚脑中检测到了Nr2e1基因的表达,但其表达量较低,在正常组和NTDs组无明显差异(P>0.05),在E9.5d和E10.5d时,NTDs组Nr2e1基因的表达显著低于正常组,差异具有统计学意义(E9.5d:P<0.05;E10.5d:P<0.01),其变化趋势与全胚胎原位杂交结果一致,即在正常发育过程中Nr2e1基因表达逐渐增高,但在发生NTDs时,其表达一直处于低水平。
3. Nr2e1基因在两种NSCs中的表达。
RT-PCR检测Nr2e1在不同NSCs的mRNA表达(方法同前)。Western blot检测Nr2e1在不同NSCs的蛋白水平表达,将收集的NSCs加入蛋白裂解液(含1%PMSF),冰上裂解30min,采用超声粉碎法破碎细胞,10000rpm离心10min,取上清,即蛋白提取物。按常规方法进行SDS-PAGE电泳,上样量50μg,采用半干式转膜2h,5%蛋白封闭液室温封闭1h,加入一抗(Nr2e1:1∶500,β-actin:1∶1000),4℃孵育过夜。二抗分别采用(HRP-羊抗兔IgG:1∶5000,HRP-羊抗小鼠IgG:1∶5000),室温孵育1h。
图8A中的RT-PCR结果显示,Nr2e1基因在NTDs组NSCs中mRNA表达显著低于正常组,差异具有统计学意义(P<0.05),图8B中Western blot结果显示Nr2e1在NTDs组NSCs中蛋白水平显著低于正常组,差异具有统计学意义(P<0.05)。说明Nr2e1基因在NSCs中的变化趋势与NTDs鼠胚中的结果一致。
4. Nr2e1基因慢病毒转染NSCs。
细胞调至5×105/ml,每孔加2ml,37℃/5%CO2培养过夜。NSCs维持液培养基+Polybrene(终浓度5μg/ml),慢病毒原液(MOI=100)加入到稀释液中。移去细胞培养液,加入稀释后的病毒液,37℃/5%CO2过夜。24h后移去细胞侵染后的病毒液,加入2ml完全培养基,37℃/5%CO2过夜。分出1/2,加入1ml完全培养基,继续培养24h。荧光显微镜下观察转染效果,并采用RT-PCR和Western blot检测Nr2e1表达情况。
图9A显示LV3-NC和LV3-Nr2e1 shRNA均成功转染NSCs。图9B的RT-PCR结果显示,转染LV3-Nr2e1 shRNA组细胞中的Nr2e1的mRNA的表达水平较LV3-NC shRNA呈现显著下降(P<0.05)。图9C的Western blot结果显示蛋白表达水平改变与mRNA水平变化趋势一致(P<0.05)。
5. Nr2e1基因在对NSCs增殖和分化的影响。
通过集落计数法和CCK-8法检测转染了LV3-Nr2e1的 NSCs的增殖能力。采用免疫荧光方法,选用Gfap检测星形胶质细胞,Map2检测神经元(anti-Gfap:1∶500,anti-Map2:1∶500)。
图10A和B结果显示,转染LV3-Nr2e1 shRNA组NSCs的集落形成能力显著低于LV3-NC shRNA对照组(P<0.05)。CCK-8法结果显示转染LV3-Nr2e1 shRNA组NSCs的0h吸光度值(OD)与LV3-NC shRNA对照组无明显差异(P>0.05),而在24h、48h和72h的OD值则显著低于LV3-NC shRNA对照组,差异具有统计学意义(24h:P<0.01;48h:P<0.05;72h:P=0.05),表明抑制Nr2e1表达显著降低了NSCs的增殖能力。
图10C和D结果表明,通过7天的诱导分化,与LV3-NC shRNA对照组NSCs相比,LV3-Nr2e1 shRNA组NSCs的Map2阳性的细胞显著下降(P<0.001),而Gfap阳性细胞显著升高(P<0.001),提示其向神经元分化能力减弱,而向星形胶质细胞分化能力增强,即Nr2e1下调引起了NSCs分化的紊乱。
Claims (7)
1.一种NTDs鼠胚动物模型构建方法,是对孕6~10天的正常孕鼠一次性给予剂量19~23mg/kg的维甲酸进行灌胃干预,在孕10.5天后得到NTDs鼠胚动物模型。
2.根据权利要求1所述的NTDs鼠胚动物模型构建方法,其特征是所述维甲酸的干预剂量为21mg/kg。
3.根据权利要求1所述的NTDs鼠胚动物模型构建方法,其特征是所述维甲酸干预时间为孕7.5天。
4.以权利要求1所述构建方法得到的NTDs鼠胚动物模型,所述鼠胚动物模型表现为脑部畸形。
5.利用权利要求4所述NTDs鼠胚动物模型建立NTDs细胞模型的方法,其特征是分离出NTDs鼠胚动物模型的脑泡组织进行细胞培养。
6.权利要求4所述NTDs鼠胚动物模型在NTDs发病机制研究方面的应用。
7.权利要求5所述NTDs细胞模型在NTDs发病机制研究方面的应用。
