Description

Preparation method of mango peel bacteriostatic agent
Technical Field
The invention relates to the technical field of preparation of bacteriostats, and in particular relates to a preparation method of a mango peel bacteriostat.
Background
Except fresh mango, most of mangoes are used for processing dried mangoes, mango wines, mango vinegar, mango cans and the like. The problem of intolerance of storage of mangoes can be solved by processing fresh mangoes into various mango products, but a large amount of mango peels are generated at the same time. At present, the mango peel is generally discarded in a treatment mode, so that the waste of raw materials is caused, and the environment is polluted. The mango peel contains natural chemical components such as flavonoid, mangiferin and the like, and has obvious inhibition effect on a plurality of microorganisms. According to estimation, the mango peel accounts for 7-24% of the weight of fresh fruits, if the mango peel waste can be utilized, the mango peel waste has an immeasurable effect on increasing the added value of mango products, and a new idea can be provided for developing novel plant bacteriostatic agents.
Bacteriostatic agents are substances that inhibit the growth of bacteria. The bacteriostatic agent mainly achieves the purpose of inhibiting the growth and the reproduction of bacteria by inhibiting the synthesis of substances required in the growth and the reproduction processes of the bacteria, and the bacteriostatic agent can not kill the bacteria but can inhibit the growth of the bacteria and prevent the excessive breeding of the bacteria from harming the health of human bodies. At present, preservatives are usually added in the food production process, and are used for inhibiting the propagation of microorganisms in food and preventing the food from being rotted and deteriorated so as to prolong the storage time of the food. However, the food preservative is also a 'double-edged sword', which may bring certain harm to the health of people. In China, most of the preservatives used in the current food production are artificially synthesized, and certain side effects can be caused when the preservatives are not used properly; some preservatives even contain trace toxins, and long-term excessive intake of the preservatives can cause certain damage to human health. The use of benzoic acid, an example of the food preservative which is widely used at present, has been controversial internationally. For example, since the poisoning by benzoic acid and its sodium salt has been reported, the european union group for child protection has considered that it is not suitable for use in children's food, and japan has made strict restrictions on its use. Even though sorbic acid and potassium sorbate are internationally recognized as safe preservatives, excessive intake of sorbic acid and potassium sorbate can affect the balance of metabolism in the human body.
Disclosure of Invention
The invention aims to solve the problems in the prior art and provides a preparation method of a mango peel bacteriostatic agent, which can well utilize the discarded mango peel of people, thereby reducing the discharge of garbage and beautifying the environment; meanwhile, the bacteriostatic action of natural chemical components such as flavonoid, mangiferin and the like in the mango peel can be fully exerted, the prepared mango peel bacteriostatic agent has the characteristic of good bactericidal effect, the raw materials required for preparation are few, the preparation method is simple, the cost is low, and the mango peel bacteriostatic agent is safe and nontoxic and is a novel plant bacteriostatic agent. The bacteriostatic agent is especially suitable for food, medicine and daily necessities.
In order to achieve the purpose, the technical scheme adopted by the invention is as follows:

(3) ethanol extraction
Pouring 85% ethanol into degreased mango peel powder until the mango peel powder is completely immersed, standing for 15-25min, performing ultrasonic oscillation for 5-10min, pouring out an upper layer solution, centrifuging, taking supernatant, namely centrifugal extract liquid, completing ethanol extraction, extracting for three times by using 85% ethanol according to the method, collecting the centrifugal extract liquid for three times, and uniformly mixing to obtain ethanol extract liquid;
(4) purification of distilled water
Taking the ethanol extract obtained in the step (3), adding distilled water with the volume of 3-4 times of that of the ethanol extract, uniformly mixing, standing for 10min, and after centrifugal separation, taking water insoluble component substances to obtain a water purified product;
(5) dissolving with ethanol
And (3) taking the water extract prepared in the step (4), and adding 85% ethanol in volume fraction for dissolving, wherein the mass ratio of the water extract to the 85% ethanol is 1: 5-6, uniformly dissolving the water extract to obtain an ethanol solution, and adding distilled water into the ethanol solution, wherein the volume ratio of the ethanol solution to the distilled water is 1: 35-40, and uniformly mixing to obtain the mango peel bacteriostatic agent.
According to the technical scheme, preferably, in the step (1), the drying temperature is 55-65 ℃, and the drying time is 22-25 h.
Preferably, in the step (1), the mango peel is crushed to 40 meshes.
In the above technical solution, preferably, in the step (2), the standing time is 25min, and the ultrasonic oscillation time is 8 min.
In the above technical solution, preferably, in the step (2) and the step (3), the power of the ultrasonic wave is 50%, the temperature is 20 ℃, and the water level is 80.
In the above technical scheme, preferably, in the step (2), the specification of the petroleum ether is 60-90 ℃.
According to the technical scheme, preferably, in the step (2), the mass ratio of the mango peel powder to the petroleum ether is 1: 2.

has the advantages that:
1. the preparation method of the mango peel bacteriostatic agent can well utilize wastes, utilizes the mango peel discarded by people, fully exerts the functions of the mango peel, reduces the discharge of garbage and beautifies the environment.
2. The raw material for making is mango peel waste, and the preparation process is simple, easy to operate, low in cost and easy for mass production.
3. The existing plant bacteriostatic agent is generally divided into two types, one type is bacteriostatic agent taking a plurality of plants as raw materials, and the preparation process of the bacteriostatic agent can increase the cost of the raw materials and the cost of the preparation process; the other is a bacteriostatic agent which takes single plants as raw materials, and the bacteriostatic agent generally has good bacteriostatic effect only on certain harmful bacteria, but has weak broad-spectrum bacteriostatic effect. The mango peel bacteriostatic agent prepared by the invention can inhibit staphylococcus aureus, pseudomonas aeruginosa, candida albicans, saccharomyces cerevisiae and other bacteria, not only expands the bacteriostatic spectrum of the bacteriostatic agent taking a single plant as a raw material and has a wide bacteriostatic range, but also can greatly reduce the raw material cost and the process cost because the mango peel bacteriostatic agent is prepared by only utilizing waste mango peel.
4. The method adds the distilled water purification and the ethanol dissolution on the basis of the common alcohol extraction process, and the steps can improve the purity of the mango peel bacteriostatic agent and can obviously improve the bacteriostatic effective components.
