4. The method adds the distilled water purification and the ethanol dissolution on the basis of the common alcohol extraction process, and the steps can improve the purity of the mango peel bacteriostatic agent and can obviously improve the bacteriostatic effective components.
Detailed Description
The present invention is further illustrated by the following specific examples.
Example 1
A preparation method of a mango peel bacteriostatic agent comprises the following steps:
(1) sample processing
10 jin of mangoes which are free from diseases and insect pests, have green and relatively smooth skins are purchased from the market, and are Yu mango in Guangxi Bai-color Right river district. Cleaning mango, air-drying or airing, cutting mango peel with the thickness of 1-2 mm by using a fruit knife, then putting the fresh mango peel into a dryer, adjusting the temperature to 60 ℃, drying for 24 hours, then crushing, and crushing to 40 meshes to obtain mango peel powder.
(2) Degreasing
Putting 15g of mango peel powder into a conical flask, pouring 30g of petroleum ether with the specification of 60-90 ℃ to enable the petroleum ether to completely immerse the mango peel powder, standing for 25min, oscillating for 8min by using ultrasonic waves (the power is 50%, the temperature is 20 ℃, and the water level is 80), completing degreasing, repeatedly degreasing twice by using the petroleum ether with the temperature of 60-90 ℃ according to the method, pouring out the upper-layer solution, taking out the precipitate, and drying to obtain the degreased mango peel powder.
(3) Ethanol extraction
Taking 10g of degreased mango peel powder, pouring 20g of 85% ethanol in parts by volume, completely immersing the degreased mango peel powder in the 85% ethanol, standing for 20min, oscillating for 8min by using ultrasonic waves (power is 50%, temperature is 20 ℃, and water level is 80), pouring out an upper layer solution, centrifuging for 10min at 2000rpm, centrifuging, taking a supernatant, namely a centrifugal extract, completing ethanol extraction, repeatedly extracting for three times by using the ethanol with the volume fraction of 85% according to the method, collecting the centrifugal extracts of the three times, and uniformly mixing to obtain an ethanol extract.
(4) Purification of distilled water
And (4) taking 10mL of the ethanol extract prepared in the step (3), adding 30mL of distilled water, uniformly mixing, standing for 10min, and after centrifugal separation, taking water-insoluble component substances, namely the water extract.
(5) Dissolving with ethanol
And (3) taking 2g of the water extract prepared in the step (4), adding 10g of ethanol with the volume fraction of 85% for dissolving, uniformly dissolving the water extract to obtain an ethanol solution, adding 440mL of distilled water into the ethanol solution, and uniformly mixing to obtain the mango peel bacteriostatic agent. The preservation temperature of the prepared mango peel bacteriostatic agent is 4 ℃.
The preparation method of the ethanol with the volume parts of 85 percent comprises the following steps: 150mL of distilled water is poured into 850mL of AR-grade ethanol and stirred uniformly.
Example 2
A preparation method of a mango peel bacteriostatic agent comprises the following steps:
(1) mango peel treatment
Selecting mangoes without plant diseases and insect pests and with smooth skins, cleaning and airing the mangoes to prepare mango skins with the thickness of 1-2 mm, then drying the mango skins at the temperature of 65 ℃ for 22h, and crushing the mango skins to 40 meshes to obtain mango skin powder.
(2) Degreasing
Putting the mango peel powder prepared in the step (1) into a container, pouring 60-90 ℃ petroleum ether until the mango peel powder is completely immersed, standing for 30min, oscillating for 5min by ultrasonic waves (power is 50%, temperature is 20 ℃, and water level is 80) to finish degreasing, degreasing twice by the degreasing method with the 60-90 ℃ petroleum ether, pouring out an upper layer solution, taking out a precipitate, and drying to obtain degreased mango peel powder;
(3) ethanol extraction
Putting the degreased mango peel powder into a container, pouring 85% ethanol by volume fraction until the degreased mango peel powder is completely immersed, standing for 15min, oscillating for 10min by ultrasonic waves (power is 50%, temperature is 20 ℃, water level is 80), pouring out an upper layer solution, centrifuging, taking a supernatant, namely a centrifugal extract, completing ethanol extraction, extracting for three times by using 85% ethanol by volume fraction, collecting the centrifugal extracts of the three times, and uniformly mixing to obtain an ethanol extract;
(4) purification of distilled water
Taking the ethanol extract prepared in the step (3), adding distilled water with 4 times of volume, uniformly mixing, standing for 10min, and taking water insoluble component substances after centrifugal separation to obtain a water purified substance;
(5) dissolving with ethanol
And (3) taking the water extract prepared in the step (4), and adding 85% ethanol in volume fraction for dissolving, wherein the mass ratio of the water extract to the 85% ethanol is 1: 6, uniformly dissolving the water extract to obtain an ethanol solution, and adding distilled water into the ethanol solution, wherein the volume ratio of the ethanol solution to the distilled water is 1: 35, and mixing uniformly to obtain the mango peel bacteriostatic agent.
The mango peel bacteriostatic agent prepared in example 1 is used for bacteriostatic tests, and the bacteriostatic test method comprises the following specific steps:
(1) sample liquid preparation
The petri dish, the tip, the 0.22 μm filter, the syringe and the inoculating loop were sterilized (121 ℃, 30 min). Sterilizing the sterile inoculation room and the ultra-clean workbench for 30min under an ultraviolet lamp. After sterilization, the mango peel bacteriostat prepared in example 1 is shaken and evenly shaken in an aseptic inoculation room, 25mL of the mango peel bacteriostat is respectively absorbed by a syringe, filtered by a filter membrane (sterilized) with the diameter of 0.22 μm and placed in a conical flask for standby. (Note: operation under sterile conditions)
(2) Preparation of culture plate
Sucking 1mL of filtered mango peel bacteriostatic agent by using a pipette gun respectively, placing the filtered mango peel bacteriostatic agent into four culture dishes, pouring 14mL of nutrient agar culture medium into three culture dishes, pouring 14mL of PDA nutrient medium into the other culture dish, shaking up the culture dishes while the culture dishes are hot, and standing the culture dishes in a clean bench for cooling. (Note: operating under aseptic conditions, the culture medium must be poured at a temperature below 60 ℃, the culture medium cannot be overheated, otherwise the effective components in the mango peel bacteriostatic agent can be damaged.)
The components of the culture medium: a nutrient agar culture medium (3.0 g of beef extract, 10.0g of peptone, 5.0g of NaCl, 20g of agar, 1000mL of distilled water and pH 7.4-7.6); PDA medium (potato 200g, glucose 20g, agar 20g, water 1000mL, natural pH).
(3) Inoculation and culture
0.1mL of staphylococcus aureus, candida albicans and pseudomonas aeruginosa bacterial liquid is respectively inoculated on a nutrient agar culture medium containing a mango peel bacteriostatic agent by using a liquid transfer gun, and the saccharomyces cerevisiae bacterial liquid is inoculated on a PDA nutrient medium containing the mango peel bacteriostatic agent and is uniformly coated by using a coating rod. Labeling, writing date and strain name. The staphylococcus aureus, the candida albicans and the pseudomonas aeruginosa are inversely placed in a constant temperature incubator at 37 +/-1 ℃ for culture, and the saccharomyces cerevisiae is inversely placed in a constant temperature incubator at 27 +/-1 ℃ for culture. (Note: three plates were inoculated per strain to form a parallel control, and the inoculated medium was covered with a freshness-retaining bag to prevent infection with foreign bacteria.)
The bacteriostasis test is also provided with a control group, wherein no mango peel bacteriostat is added into the control group, and other conditions are consistent with the test of the embodiment 1.
Through observation and recording of the tests, the influence of the mango peel bacteriostatic agent prepared in example 1 on four experimental bacteria is shown in table 1.

Note: "-" indicates no growth of the cells; "+" indicates that there is growth of bacteria. "A" represents Staphylococcus aureus; "B" represents Pseudomonas aeruginosa; "C" means Candida albicans; "D" represents Saccharomyces cerevisiae.
As can be seen from the table 1, the mango peel bacteriostatic agent prepared in example 1 has a remarkable bacteriostatic effect on saccharomyces cerevisiae, and for other three experimental bacteria, no bacterial colony exists in the first day, staphylococcus aureus is small and gathered in the second day, and pseudomonas aeruginosa and candida albicans have growth trends; on the third day, the growth vigor of staphylococcus aureus and pseudomonas aeruginosa becomes slow, and the candida albicans is small and dense; the growth of Candida albicans became slow on the fourth and fifth days. By combining the above, the mango peel bacteriostatic agent has a good bacteriostatic effect on staphylococcus aureus, pseudomonas aeruginosa, candida albicans and saccharomyces cerevisiae, and is wide in bacteriostatic range and good in bacteriostatic effect.
Although the present invention has been described with respect to the preferred embodiments, it will be understood by those skilled in the art that various changes in form and details may be made therein without departing from the spirit and scope of the invention as defined by the appended claims.
Claims (8)
1. A preparation method of a mango peel bacteriostatic agent is characterized by comprising the following steps:
(1) mango peel treatment
Selecting mangoes without plant diseases and insect pests and with smooth skins, cleaning and airing the mangoes to prepare mango skins with the thickness of 1-2 mm, and then drying and crushing the mango skins to obtain mango skin powder; the drying temperature is 55-65 ℃, and the drying time is 22-25 h;
(2) degreasing
Putting the mango peel powder prepared in the step (1) into a container, pouring petroleum ether until the mango peel powder is completely immersed, standing for 20-30min, performing ultrasonic oscillation for 5-10min to complete degreasing, degreasing twice with the petroleum ether according to the degreasing method, pouring an upper layer solution, taking out a precipitate, and drying to obtain degreased mango peel powder; the specification of the petroleum ether is 60-90 ℃, and the mass ratio of the mango peel powder to the petroleum ether is 1: 2;
(3) ethanol extraction
Putting the degreased mango peel powder into a container, pouring 85% ethanol by volume fraction until the degreased mango peel powder is completely immersed, standing for 15-25min, performing ultrasonic oscillation for 5-10min, pouring out an upper-layer solution, centrifuging, taking a supernatant, namely a centrifugal extract, completing ethanol extraction, extracting for three times by using 85% ethanol by volume fraction, collecting the centrifugal extracts of the three times, and uniformly mixing to obtain an ethanol extract;
(4) purification of distilled water
Taking the ethanol extract obtained in the step (3), adding distilled water with the volume of 3-4 times of that of the ethanol extract, uniformly mixing, standing for 10min, and after centrifugal separation, taking water insoluble component substances to obtain a water purified product;
(5) dissolving with ethanol
And (3) taking the water extract prepared in the step (4), and adding 85% ethanol in volume fraction for dissolving, wherein the mass ratio of the water extract to the 85% ethanol is 1: 5-6, uniformly dissolving the water extract to obtain an ethanol solution, and adding distilled water into the ethanol solution, wherein the volume ratio of the ethanol solution to the distilled water is 1: 35-40, and uniformly mixing to obtain the mango peel bacteriostatic agent.
2. The method for preparing the mango peel bacteriostatic agent according to claim 1, which is characterized in that: in the step (1), the mango peel is crushed to 40 meshes.
3. The method for preparing the mango peel bacteriostatic agent according to claim 1, which is characterized in that: in the step (2), the standing time is 25min, and the ultrasonic oscillation time is 8 min.
