技术领域
本发明属于水产养殖饲料领域,具体涉及一种提高鲤鱼酸性磷酸酶活力的饲料及其制备方法。
背景技术
鲤鱼属底栖杂食性鱼类,身体侧扁而腹部圆,口呈马蹄形,须2对。鲤鱼免疫系统分为固有免疫和适应性免疫两部分,其相互协作共同实现机体的防御功能,识别并清除来自外界及自身的威胁。硬骨鱼是目前已知的最低等的兼具固有与适应性免疫系统的脊椎动物。
随着水产集约化程度的不断发展,水产动物疾病逐步呈现多发性、复杂化、高危害的特点,养殖水环境的恶化也造成鱼类免疫力下降。为应对鱼类疾病蔓延趋势,施用大量渔用杀虫药和外用杀菌消毒药物,导致多种抗生素的滥用,这些措施虽然对疾病有一定的控制作用,但往往造成致病微生物的耐药性、药物残留,甚至造成整个水体生态环境平衡破坏。
中草药具有残留少、致病菌不易产生耐药性的特点,其作用不仅对病原微生物有直接的抑制和杀灭作用,更重要的是对鱼类的免疫系统进行调整,提高鱼类的抗应激能力和免疫机能。中草药作为疾病防治药物或饵料添加剂,尤其适用于当前水产养殖业的集约化、规模化生产的需要,便于进行养殖鱼类病害的群体防治。而且应用中草药防治鱼病符合发展无公害水产品、生产绿色水产品的病害防治准则。通过中草药的施用提高水产动物的免疫力和抗病力,降低水产动物的发病率是近几年研究的热点。
随着中药化学和中药免疫药理学研究的进展,现已证明200余种中草药有多方面的免疫活性,能影响和调节动物机体的免疫功能。鱼的免疫应答类型与人类似,人及哺乳动物免疫所用的中草药大多数也适用于鱼类。中草药免疫活性成分主要有多糖、苷类、生物碱、有机酸、挥发油等,中药免疫增强剂可对水产动物免疫器官产生影响,从而激活机体免疫力。中药免疫增强剂对水产动物免疫能力的作用主要体现在增强机体的细胞免疫与体液免疫上。
鱼类的免疫应答器官与组织主要包括前肾、脾脏、胸腺以及消化道淋巴组织与血液淋巴等。中草药免疫增强剂能促进动物免疫功能,促进动物免疫器官发育,提供更多更好的免疫活性细胞,活化的免疫细胞包括T细胞、B细胞、K细胞、NK细胞等。同时也促进了细胞各种免疫有关受体的表达;激活巨噬细胞功能,加强其吞噬、处理、传递抗原的作用;促进细胞因子的分泌及活性,促进和调节补体、抗体的产生;提高酸性磷酸酶、碱性磷酸酶和溶菌酶活力;影响核酸和核苷酸的合成;改善骨髓造血功能。
酸性磷酸酶是一类在酸性条件下催化磷酸酯水解的酶,包括溶酶体性和非溶酶体性两大类。溶酶体性酸性磷酸酶主要存在于巨噬细胞,定位于溶酶体内,是溶酶体的特征性酶,主要参与磷酸酯的代谢,与细胞的消化、自溶和免疫有关。非溶酶体性酸性磷酸酶与细胞增殖、能量转化、质膜的吸收与运输功能等一些重要的生物学功能有关。由于酸性磷酸酶参与了免疫反应,可作为免疫活性的标志物之一。
发明内容
针对现有技术的不足,本发明的目的是提供一种提高鲤鱼酸性磷酸酶活力的饲料,能够提高鲤鱼的免疫力和抗病力,降低发病率,符合发展无公害水产品、生产绿色水产品的病害防治准则,尤其适用于当前水产养殖业的集约化、规模化生产的需要,便于进行养殖鱼类病害的群体防治;本发明同时提供其科学合理、简单易行的制备方法。
为达到上述目的,本发明的具体技术方案为:
本发明所述的提高鲤鱼酸性磷酸酶活力的饲料,由以下重量百分比的原料混合组成:
鱼粉 45~55%;
豆粕 8~12%;
玉米蛋白粉 25~35%;
鱼油 5~10%;
酵母粉 1~5%;
甜菜碱 0.3~0.5%;
维生素预混料 0.5~1%;
矿物质预混料 0.5~1%;
中草药 1~2%;
以上原料的重量百分比之和为100%。
优选地,所述的提高鲤鱼酸性磷酸酶活力的饲料,由以下重量百分比的原料混合组成:
鱼粉 50%;
豆粕 10%;
玉米蛋白粉 29~31%;
鱼油 5%;
酵母粉 1.6%;
甜菜碱 0.4%;
维生素预混料 1%;
矿物质预混料 1%;
中草药 1~2%;
以上原料的重量百分比之和为100%。
其中:
所述的中草药为绿豆、山药或黑芝麻中的一种或几种。
优选地,所述的中草药为绿豆、山药和黑芝麻的混合物。
进一步优选地,所述的中草药由如下重量百分比的原料制成:
绿豆 70~80%;
山药 10~20%;
黑芝麻 10~20%。
更进一步优选地,所述的中草药由如下重量百分比的原料制成:
绿豆 70%;
山药 20%;
黑芝麻 10%。
绿豆含蛋白质、脂肪、碳水化物、钙、磷、铁、胡萝卜素、硫胺素、核黄素、尼克酸等。蛋白质主要为球蛋白类,含有蛋氨酸、色氨酸、酪氨酸等多种氨基酸。绿豆的磷脂成分中有磷脂酰胆碱、磷脂酰乙醇胺、磷脂酰肌醇、磷脂酰甘油、磷脂酰丝氨酸、磷脂酸等。
山药含有大量的淀粉及蛋白质、B族维生素、维生素C、维生素E、葡萄糖等营养成分。微量元素尤其是钾含量较高,还含淀粉糖化酶、淀粉酶等多种消化酶。
黑芝麻中脂肪油可达55%,脂肪油含油酸(约48%)、亚油酸(约37%)、棕榈酸、硬脂酸、花生油酸、廿四烷酸的甘油脂,并含芝麻素、芝麻林素、芝麻酚、维生素E、植物甾醇、卵磷脂等成分。另外,黑芝麻中还含有胡麻甙、蛋白质及寡糖类、车前糖、芝麻糖,以及少量磷、钾及细胞色素C、叶酸、烟酸、蔗糖、戊聚糖和多量的钙等。
综上,绿豆、山药、黑芝麻中含有丰富的蛋白质、糖类、脂类和维生素类物质,添加到饲料中可以补充饲料营养成分,显著提高鱼体免疫力。
本发明所述的提高鲤鱼酸性磷酸酶活力的饲料的制备方法,步骤如下:
(1)采用粉碎机将鱼粉、豆粕、中草药分别粉碎,过筛,得粉碎料;
(2)将粉碎料、玉米蛋白粉、酵母粉、甜菜碱、维生素预混料、矿物质预混料依据从小到大的原则,采用高效混合机逐级混合均匀;
(3)将鱼油倒入步骤(2)得到的混合均匀的物料中,过筛,混匀,上机制粒,得粒状物料;
(4)将步骤(3)得到的粒状物料烘干,得饲料产品。
其中:
步骤(1)所述的过筛为过150 μm筛网。
步骤(3)所述的过筛为过40 μm筛网。
步骤(4)所述的烘干为于烘箱中42℃烘干24小时。
本发明采用的鱼粉、豆粕、玉米蛋白粉、鱼油、酵母粉、甜菜碱、维生素预混料、矿物质预混料、中草药等均为市售产品。
与现有技术相比,本发明的有益效果是:
1、本发明所述的提高鲤鱼酸性磷酸酶活力的饲料,根据鲤鱼的营养需求,以及中草药提高免疫力的特性,配方中加入绿豆、山药和黑芝麻,三者协同作用,能够提高鲤鱼的免疫力和抗病力,降低发病率,符合发展无公害水产品、生产绿色水产品的病害防治准则,尤其适用于当前水产养殖业的集约化、规模化生产的需要,便于进行养殖鱼类病害的群体防治。本发明所述的饲料能够为其他养殖鱼类饲料配制提供参考。

具体实施方式
下面结合实施例对本发明做进一步说明,但不限定本发明。
实施例1
含单一中草药绿豆的饲料:
以制备100 kg饲料为例,各原料及用量为:鱼粉50 kg;豆粕10 kg;玉米蛋白粉30kg;鱼油5 kg;酵母粉1.6 kg;甜菜碱0.4 kg;维生素预混料1 kg、矿物质预混料1 kg;绿豆1kg。
采用粉碎机将鱼粉、豆粕、中草药分别粉碎,过150μm筛网,得粉碎料。将粉碎料、玉米蛋白粉、酵母粉、甜菜碱、维生素预混料、矿物质预混料依据含量从小到大的原则,采用高效混合机逐级混合均匀。将鱼油倒入上述混匀的饲料粉末,过40μm筛网,混匀,上机制粒;于烘箱中42℃烘干24小时,即得饲料产品。
实施例2
含单一中草药山药的饲料:

其中,复合中草药含有以下比例的3种中草药,复合中草药制备方法如下:
绿豆,800 g;
山药,100 g;
黑芝麻,100 g。
采用粉碎机将鱼粉、豆粕、中草药分别粉碎,过150μm筛网,得粉碎料。将粉碎料、玉米蛋白粉、酵母粉、甜菜碱、维生素预混料、矿物质预混料依据含量从小到大的原则,采用高效混合机逐级混合均匀。将鱼油倒入上述混匀的饲料粉末,过40μm筛网,混匀,上机制粒;于烘箱中42℃烘干24小时,即得饲料产品。
对比例1
不含中草药:

将鲤鱼养殖桶分为5组,即4个实验组和1个对照组,每组3个重复,每个重复20尾鱼。4个实验组分别采用实施例1-4饲料喂养,对照组采用对比例1饲料喂养,投喂4周。4周后取样,采用酸性磷酸酶活力测定试剂盒(南京建成生物研究所)测定鲤鱼血清中酸性磷酸酶活力,分析各组饲料对酸性磷酸酶活力的影响,结果见表2。与对照组饲料相比,饲料中添加1.0 kg的绿豆、山药、黑芝麻以及复合中草药均可以提高鲤鱼血清酸性磷酸酶活力;与单一添加绿豆、山药或黑芝麻的饲料相比,添加复合中草药组的饲料显著提高鲤鱼血清酸性磷酸酶活力。

Claims (7)
1.一种提高鲤鱼酸性磷酸酶活力的饲料,其特征在于:由以下重量百分比的原料混合组成:
以上原料的重量百分比之和为100%;
所述的中草药由如下重量百分比的原料制成:
绿豆 70~80%;
山药 10~20%;
黑芝麻 10~20%。
2.根据权利要求1所述的提高鲤鱼酸性磷酸酶活力的饲料,其特征在于:由以下重量百分比的原料混合组成:
以上原料的重量百分比之和为100%。
3.根据权利要求1所述的提高鲤鱼酸性磷酸酶活力的饲料,其特征在于:所述的中草药由如下重量百分比的原料制成:
绿豆 70%;
山药 20%;
黑芝麻 10%。
4.一种权利要求1-3任一所述的提高鲤鱼酸性磷酸酶活力的饲料的制备方法,其特征在于:步骤如下:
(1)采用粉碎机将鱼粉、豆粕、中草药分别粉碎,过筛,得粉碎料;
(2)将粉碎料、玉米蛋白粉、酵母粉、甜菜碱、维生素预混料、矿物质预混料依据含量从小到大的原则,采用高效混合机逐级混合均匀;
(3)将鱼油倒入步骤(2)得到的混合均匀的物料中,过筛,混匀,上机制粒,得粒状物料;
(4)将步骤(3)得到的粒状物料烘干,得饲料产品。
5.根据权利要求4所述的提高鲤鱼酸性磷酸酶活力的饲料的制备方法,其特征在于:步骤(1)所述的过筛为过150μm筛网。
6.根据权利要求4所述的提高鲤鱼酸性磷酸酶活力的饲料的制备方法,其特征在于:步骤(3)所述的过筛为过40μm筛网。
7.根据权利要求4所述的提高鲤鱼酸性磷酸酶活力的饲料的制备方法,其特征在于:步骤(4)所述的烘干为于烘箱中42℃烘干24小时。
