Abstract
本发明公开了一种用于修补腹膜缺损的生物材料,由以下方法制备得到:先按重量份称取各组分;然后将上述原料加入反应釜中,在真空条件下加热至100‑120℃,搅拌10‑20分钟,降温至30‑40℃,搅拌5‑10分钟,得到混合溶液,然后进行湿法纺丝,得到合成纤维,再经过牵伸、水洗、卷绕、后进行织网,最后在100‑120℃的温度下定型。本发明的一种用于修补腹膜缺损的生物材料可有效减少腹腔内脏器的粘连,防止伤口的感染,生物相容性好,对组织无刺激,对人体无毒无害,可以广泛应用与医疗领域。
Description
技术领域
本发明属于生物医用材料领域,具体涉及一种用于修补腹膜缺损的生物材料。
背景技术
腹壁缺损也称腹外疝,是指腹壁在某个部位出现缺损或者薄弱,使腹腔脏器连同腹膜经腹壁薄弱处或孔隙向体表突出而形成局部肿块,可引起疼痛、肠梗阻等不适。腹外疝必须位于有腹膜壁层所组成的囊袋内,以别于内脏脱出,后者无腹膜壁层复盖。腹外疝是腹部外科最常见的疾病之一,并以突出的解剖部位命名,其中以腹股沟疝发生率最高,占90%以上,股疝次之,占5%左右,较常见的腹外疝还有切口疝、脐疝和白线疝。在外科治疗方法中,依赖植入补片的无张力修补术已成为主流术式,补片大多选用以聚丙烯为代表的人工高分子材料,但是传统的聚丙烯补片不能应用于腹壁缺损的腹膜内修补;若用于腹膜内修补则会出现一系列并发症,包括肠瘘、肠梗阻、窦道形成、感染及明显疼痛感等并发症,不利于临床的广泛应用。寻找一种适用于腹腔内腹壁缺损修补的补片,减少传统补片相关并发症是目前腹壁缺损修补研究的难点与热点。因此,需要研发出一种具有优良生物相容性和抗粘连的用于修补腹膜缺损的生物材料。
发明内容
本发明的目的在于提供一种用于修补腹膜缺损的生物材料。
为了实现上述目的,本发明提供了一种用于修补腹膜缺损的生物材料,所述生物材料由以下方法制备得到:先按重量份称取聚乙烯80-90份、海蜇胶原12-18份、硅凝胶14-18份、壳聚糖乳酸盐12-20份、聚酯纤维5-10份、纤维蛋白胶10-15份、聚乙烯醇50-60份、盐酸曲马多5-8份、卵磷脂3-5份、剑麻纤维10-20份、聚丙烯酰胺4-8份、纳米银10-15份和水40-50份;然后将上述原料加入反应釜中,在真空条件下加热至100-120℃,搅拌10-20分钟,降温至30-40℃,搅拌5-10分钟,得到混合溶液,然后进行湿法纺丝,得到合成纤维,再经过牵伸、水洗、卷绕、后进行织网,最后在100-120℃的温度下定型。
所述聚乙烯与硅凝胶重量比为5:1。
所述水为去离子水或蒸馏水。
所述定型温度为110℃。
所述纳米银粒径为40-100nm。
有益效果:本发明的一种用于修补腹膜缺损的生物材料可有效减少腹腔内脏器的粘连,防止伤口的感染,生物相容性好,对组织无刺激,对人体无毒无害,可以广泛应用与医疗领域。
具体实施方式
实施例1
一种用于修补腹膜缺损的生物材料,所述生物材料由以下方法制备得到:先按重量份称取聚乙烯80份、海蜇胶原12份、硅凝胶14份、壳聚糖乳酸盐12份、聚酯纤维5份、纤维蛋白胶10份、聚乙烯醇50份、盐酸曲马多5份、卵磷脂3份、剑麻纤维10份、聚丙烯酰胺4份、纳米银10份和去离子水40份;然后将上述原料加入反应釜中,在真空条件下加热至100-120℃,搅拌10-20分钟,降温至30-40℃,搅拌5-10分钟,得到混合溶液,然后进行湿法纺丝,得到合成纤维,再经过牵伸、水洗、卷绕、后进行织网,最后在100℃的温度下定型。纳米银粒径为40-100nm。
实施例2
一种用于修补腹膜缺损的生物材料,所述生物材料由以下方法制备得到:先按重量份称取聚乙烯90份、海蜇胶原18份、硅凝胶18份、壳聚糖乳酸盐20份、聚酯纤维10份、纤维蛋白胶15份、聚乙烯醇60份、盐酸曲马多8份、卵磷脂5份、剑麻纤维20份、聚丙烯酰胺8份、纳米银15份和水50份;然后将上述原料加入反应釜中,在真空条件下加热至100-120℃,搅拌10-20分钟,降温至30-40℃,搅拌5-10分钟,得到混合溶液,然后进行湿法纺丝,得到合成纤维,再经过牵伸、水洗、卷绕、后进行织网,最后在120℃的温度下定型。纳米银粒径为40-100nm。
实施例3
一种用于修补腹膜缺损的生物材料,所述生物材料由以下方法制备得到:先按重量份称取聚乙烯85份、海蜇胶原15份、硅凝胶17份、壳聚糖乳酸盐16份、聚酯纤维7份、纤维蛋白胶12份、聚乙烯醇55份、盐酸曲马多6份、卵磷脂4份、剑麻纤维15份、聚丙烯酰胺6份、纳米银13份和蒸馏水45份;然后将上述原料加入反应釜中,在真空条件下加热至100-120℃,搅拌10-20分钟,降温至30-40℃,搅拌5-10分钟,得到混合溶液,然后进行湿法纺丝,得到合成纤维,再经过牵伸、水洗、卷绕、后进行织网,最后在110℃的温度下定型。纳米银粒径为40-100nm。
实施例4
动物应用:取10只重约280g-300g雄性大鼠,对大鼠进行腹腔注射麻醉。随即暴露大鼠腹部,用剪刀在腹中央清理出一块无毛区域。常规消毒铺巾,用手术刀在大鼠腹部切开两条长约3cm皮肤切口,切口间隔3cm,切口平行大鼠躯干纵轴,随后沿切口逐层分离并切开腹部肌肉各层,进入腹腔。
准备好实施例3制得生物材料,大小为3*3cm,轻柔的放入腹腔内,良好放置。随即用丝线将补片四角良好的缝合固定在腹壁,逐层关闭腹腔。随后观察大鼠精神、饮食情况,7-10天后打开腹腔,观察补片有无粘连及粘连严重程度。
7-10天后,打开10只大鼠腹腔,腹腔均未见感染,补片均未见明显粘连。

1.一种用于修补腹膜缺损的生物材料,其特征在于所述生物材料由以下方法制备得到:先按重量份称取聚乙烯80-90份、海蜇胶原12-18份、硅凝胶14-18份、壳聚糖乳酸盐12-20份、聚酯纤维5-10份、纤维蛋白胶10-15份、聚乙烯醇50-60份、盐酸曲马多5-8份、卵磷脂3-5份、剑麻纤维10-20份、聚丙烯酰胺4-8份、纳米银10-15份和水40-50份;然后将上述原料加入反应釜中,在真空条件下加热至100-120℃,搅拌10-20分钟,降温至30-40℃,搅拌5-10分钟,得到混合溶液,然后进行湿法纺丝,得到合成纤维,再经过牵伸、水洗、卷绕、后进行织网,最后在100-120℃的温度下定型。
