本发明涉及一种中药材有效成分的提取方法,包括以下步骤:1)将中药材加入足量水进行浸泡,浸泡时同时加入麦芽糖和糖苷合成酶,并保持温度在40~50℃;所述中药材包括山楂、莱菔子、决明子、陈皮、泽兰;2)浸泡后的中药材经过煎煮法得到水溶液;3)水溶液冷却后加入糖苷水解酶,在40~50℃条件下进行水解反应得到水解溶液;4)将水解溶液经过过滤、浓缩和减压干燥后得到中药提取物。本发明的方法可以提高水提法对中药材中黄酮的提取率。

一种中药材有效成分的提取方法
技术领域
本发明涉及一种中药材有效成分的提取方法,属于保健品技术领域。
背景技术
黄酮类化合物是一大类天然化合物物,广泛存在于植物界,是许多中草药的有效成分,它在植物体内大部分与糖结合成黄酮苷,一部分以游离状态存在,黄酮类化合物的生理活性强而广,有的可降低血脂,有的具有很强的保肝作用,有的还具有抗炎和扩冠作用。黄酮苷易溶于热水,而游离黄酮一般难溶或不溶于水。现有技术对中药材中有效成分的提取方法多采用水煎煮法提取,对黄酮苷的提取效果较好,但对于游离黄酮则提取率很低,而采用有机溶剂提取法虽然提取率高,但安全性低。另一方面,人体对黄酮苷的吸收效果较差,对游离黄酮可以直接吸收。
发明内容
本发明的目的在于提供一种持效性好的中药材有效成分的提取方法,具体方案为:
一种中药材有效成分的提取方法,其特征在于,包括以下步骤:
1)将中药材加入足量水进行浸泡,浸泡时同时加入麦芽糖和糖苷合成酶,并保持温度在40~50℃;
2)浸泡后的中药材经过煎煮法得到水溶液;
3)水溶液冷却后加入糖苷水解酶,在40~50℃条件下进行水解反应得到水解溶液;
4)将水解溶液经过过滤、浓缩和减压干燥后得到中药提取物。
进一步的,步骤1)中所述的糖苷合成酶为糖苷水解酶经过非亲核体氨基酸取代亲核体氨基酸得到。
进一步的,步骤3)中所述的糖苷水解酶为纤维素酶和果胶酶的混合物。
进一步的,步骤1)中所述中药材包括山楂、莱菔子、决明子、陈皮、泽兰
本发明的有益点在于:在对中药材有效成分提取时,先加入水、麦芽糖和糖苷合成酶进行浸泡,浸泡时在糖苷合成酶的作用下,游离态的黄酮转化为糖苷,可以提高后续水提取时的提取率。既保留了水提取法的安全性,同时提高了水提取法的提取率。
对提取后的水溶液加入糖苷酶,对黄酮苷进行水解,生成游离态黄酮,有利于在制成产品后提高人体对黄酮类的吸收效率,
本发明中,采用果胶酶能够很好地水解黄酮苷,纤维素酶单独存在时对黄酮苷没有脱糖基作用,但将纤维素酶与果胶酶复合使用时,高达90%的黄酮苷被水解成游离黄酮和苷元。
具体实施方式
实施例1

步骤1)中所述的糖苷合成酶为糖苷水解酶经过非亲核体氨基酸取代亲核体氨基酸得到。

2)浸泡后的中药材经过煎煮法得到水溶液;
3)水溶液冷却后加入糖苷水解酶,在40~50℃条件下进行水解反应得到水解溶液;
4)将水解溶液经过过滤、浓缩和减压干燥后得到中药提取物。
2.根据权利要求1所述的中药材有效成分的提取方法的制备方法,其特征在于:步骤1)中所述的糖苷合成酶为糖苷水解酶经过非亲核体氨基酸取代亲核体氨基酸得到。
3.根据权利要求1所述的中药材有效成分的提取方法的制备方法,其特征在于:步骤3)中所述的糖苷水解酶为纤维素酶和果胶酶的混合物。
