vacuum drying cabinet, model DZF-6050, purchased from Shanghai Boxun industries, Inc.
The following examples further illustrate embodiments of the present invention:
example 1 preparation of a Chinese medicinal composition of six-in-one decoction of Astragalus
(1) Preparation of astragalus root six-in-one decoction traditional Chinese medicine composition granules
1) Weighing 600g of astragalus mongholicus, 100g of liquorice, 300g of bighead atractylodes rhizome and 300g of white paeony root, adding 8 times of water, heating, refluxing and extracting for 3 times, each time for 2 hours, filtering for several times, combining filtrates, concentrating under reduced pressure to relative density of 1.10(70 ℃), cooling to room temperature, centrifuging, concentrating supernatant under reduced pressure to relative density of 1.20(70 ℃), and obtaining traditional Chinese medicine extract.
2) Taking the Chinese medicinal extract, adding 720g of sucrose powder, vacuum drying at 60 ℃ for 24h, pulverizing, granulating with 90% ethanol as wetting agent, drying at 50 ℃ for 2h, and grading to obtain granule.
(2) Preparation of astragalus root six-one decoction traditional Chinese medicine composition oral liquid
1) Weighing 600g of astragalus mongholicus, 100g of liquorice, 300g of bighead atractylodes rhizome and 300g of white paeony root, adding 8 times of water, heating, refluxing and extracting for 3 times, each time for 2 hours, filtering for several times, combining filtrates, concentrating under reduced pressure to relative density of 1.10(70 ℃), cooling to room temperature, centrifuging, concentrating supernatant under reduced pressure to relative density of 1.20(70 ℃), and obtaining traditional Chinese medicine extract.
2) Adding water into the Chinese medicinal extract to 1200ml, adding 65.0g tween 80 and 13.0g potassium sorbate, stirring for dissolving, diluting with water to 1300ml, filtering with 0.45 μm filter membrane, bottling, plugging, and sterilizing at 100 deg.C for 30min to obtain oral liquid.
(3) Preparation of astragalus root six-one decoction traditional Chinese medicine composition powder
1) Weighing 600g of astragalus mongholicus, 100g of liquorice, 300g of bighead atractylodes rhizome and 300g of white paeony root, adding 8 times of water, heating, refluxing and extracting for 3 times, each time for 2 hours, filtering for several times, combining filtrates, concentrating under reduced pressure to relative density of 1.10(70 ℃), cooling to room temperature, centrifuging, concentrating supernatant under reduced pressure to relative density of 1.20(70 ℃), and obtaining traditional Chinese medicine extract.
2) Taking the Chinese medicinal extract, pouring into a stainless steel tray, vacuum drying at 60 deg.C for 24 hr, taking out, pulverizing, and sieving with 80 mesh sieve to obtain powder.
Example 2 pharmacodynamic test conditions for enhancing immunity
The experiment provides a basis for the clinical application of the astragalus mongholicus six-one decoction traditional Chinese medicine composition in enhancing the immune function by observing the influence of the astragalus mongholicus six-one decoction traditional Chinese medicine composition on T lymphocytes in chicken blood.
Experimental animals: the test chickens are 90 healthy AA broilers of 10 days old, and are managed by a conventional feeding method, and the feeding management conditions are consistent during the test period.
Grouping and processing experimental animals: 90 healthy AA broilers at age of 10 days were randomly divided into A, B, C three groups of 30 broilers each. Group A, the astragalus root six-one decoction traditional Chinese medicine composition granules prepared in the example 1 are given; group B administration of Astragalus polysaccharides; group C was a blank control group, not dosed. The administration and treatment methods are detailed in table 1.
Feeding management: the test chickens were only housed in groups in the isolator in the same animal laboratory. 3 test groups were routinely fed on basal diets. During the whole test period, the patient can eat and drink water freely, and the patient can be cleaned every day.

The experimental method comprises the following steps: each group was administered for 7 days, and 10 chickens per group were randomly sampled after drug withdrawal for blood sampling and E-rosette test.
And (4) judging a result: observing about 200 lymphocytes under a light microscope, wherein the cells with 3 or more sheep red blood cells adhered to the surfaces of the lymphocytes are rosette forming cells, and calculating the percentage of the rosette forming cells in the total number of the lymphocytes. Statistical processing was performed using SPSS analysis software. In particular, see the following table:

The results show that: the difference of the forming rate of lymphocyte rosette in group A and group C is very obvious, P is less than 0.01, which shows that the astragalus root six-one decoction traditional Chinese medicine composition can obviously increase the number of T lymphocytes in chicken blood and improve the immune function of the T lymphocytes; the difference of the lymphocyte rosette formation rate of the group B and the comparison group is obvious, P is less than 0.05, which indicates that astragalus polysaccharide can also obviously increase the number of T lymphocytes in chicken blood and improve the immune function of the T lymphocytes; the difference between the forming rate of lymphocyte rosette in group A and the forming rate of lymphocyte rosette in group B is extremely obvious, and P is less than 0.01, which shows that the astragalus root six-in-one decoction traditional Chinese medicine composition can obviously increase the number of T lymphocytes in chicken blood and improve the immune function of the T lymphocytes compared with astragalus root polysaccharide.
Example 3 Experimental conditions for increasing antibody levels in combination with vaccines
The experiment mainly aims to provide a basis for the traditional Chinese medicine composition of astragalus root six-one decoction to enhance the immune function by comparing the effect of the medicine on enhancing the immunity of the newcastle disease vaccine.
Vaccine: newcastle disease virus and avian influenza virus bivalent inactivated vaccine (La Sota strain + H9 subtype, HL strain), lot No. 150633, manufacturer: praco bioengineering, Inc.
Experimental animals: 1 day old healthy SPF Roman egg male chicks are raised in an isolator, and during the test period, SPF chicken feed is fed, and free feeding and drinking are carried out.
Grouping experimental animals: feeding to 8 days of age, selecting 90 healthy chickens, randomly dividing into A, B, C test groups, each group comprising 30 chickens, each numbered, and feeding in 3 isolators according to the group.
The experimental method comprises the following steps: at 9 days of age, randomly extracting 5 chickens from each group, collecting non-anticoagulated blood, and determining the antibody titer of Newcastle disease by reference to the GB/T16550-2008 standard hemagglutination inhibition method. At 10 days of age, each group was immunized by subcutaneous injection of newcastle disease virus and avian influenza virus bivalent inactivated vaccine (La Sota strain + H9 subtype, HL strain) into the neck and back at 0.1 ml/feather. The next day of immunization, group a was administered the astragalus siyitang chinese medicinal composition granules prepared in example 1; group B administration of Astragalus polysaccharides; group C was a blank control group, not dosed. The administration and treatment methods are detailed in table 3. In each group, 10 chickens were randomly selected 28 days after immunization, each chicken was bled with 1ml of blood, and serum was separated to determine the newcastle disease antibody titer.

The experimental results are as follows:

The results show that: before immunization, the Newcastle disease antibody titer of each group of chickens is 0; at 28 days after immunization, the antibody titers in group A were significantly different from those in group C (P <0.01), those in group B (P <0.05), and those in group A (P > 0.05). The astragalus root six-one decoction traditional Chinese medicine composition has a strong immunity enhancing effect, and can improve the immune response of a vaccine and the antibody level.
Example 4 test conditions for improving broiler productivity
Experimental animals: 10077 new yellow feather broilers are tested in a certain culture farm in Luoyang Ischuan.
The experimental method comprises the following steps: 10077 yellow-feathered broilers of 7 days old are divided into a group A and a group B, 5065 in the group A are given the astragalus six-one decoction traditional Chinese medicine composition granules prepared in the example 1, and 5012 in the group B are blank control groups and are not given. The 2 groups were all managed according to the normal procedure of the farm, and the administration and treatment methods are detailed in table 5. At 7, 24, 45, 58 days of age (slaughter), 500 chickens per group were randomly picked and weighed, and the average weight was calculated. And respectively calculating the daily average weight gain, daily average feed intake, total feed consumption and total weight gain of 3 time periods of 8-24 days old, 25-45 days old and 46-58 days old, and calculating the feed-weight ratio. During the test period, the number of the sick and dead chickens in each group of chickens is observed and recorded, and the morbidity and the mortality in the whole test period are calculated.

As can be seen from Table 6, compared with the blank control group, the daily average weight gain of the administration groups at 3 days of 8-24d, 25-45d and 46-58 d is respectively improved by 4.40%, 2.32% and 5.86%, and the material weight ratio of the administration groups at 3 days is respectively reduced by 2.34%, 4.90% and 8.60%. In the whole test period, compared with a blank control group, the daily gain of the administration group is increased by 4.11%, and the material-weight ratio is reduced by 5.88%. The astragalus root six-in-one decoction traditional Chinese medicine composition has the effects of improving the weight gain rate of chicken flocks, reducing the feed weight ratio and improving the feed reward.

As can be seen from table 7, the morbidity and mortality of the blank control group were 7.88% and 2.89% respectively, while the morbidity and mortality of the administration group under the same feeding and management conditions were 4.78% and 1.50% respectively, which were reduced by 3.10% and 1.39% respectively. The astragalus root-six-one decoction traditional Chinese medicine composition can reduce the disease rate and the death rate of chickens and improve the economic benefit of breeding.

Claims (3)
1. The application of the astragalus mongholicus six-first decoction traditional Chinese medicine composition in preparing a medicine for improving the weight gain rate of chicken flocks and reducing the material weight ratio is characterized in that the astragalus mongholicus six-first decoction traditional Chinese medicine composition is prepared from 4-8 parts of astragalus mongholicus, 1 part of liquorice, 2-4 parts of bighead atractylodes rhizome and 2-4 parts of white paeony root.
2. The use of claim 1, wherein the raw materials of the astragalus root-six-one decoction traditional Chinese medicine composition are 6 parts of astragalus root, 1 part of liquorice, 3 parts of bighead atractylodes rhizome and 3 parts of white peony root.
3. The use of claim 1, wherein the astragalus decoction-six traditional Chinese medicine composition is in the dosage forms of granules, oral liquid and powder.
