Abstract

本发明公开了一种区分不同海参品种的分析鉴定方法,步骤是:1、先进行显微镜骨片鉴定,方法是:(1)将采集的海参个体,用无水乙醇固定;(2)从海参背脊中部,切取1cmZ X 1mm的体壁组织,作为分析样品;(3)每个样品3个重复,双蒸水洗净,用灭菌吸水纸吸干水分,至于5ml小烧杯中(每个样品配有单独的烧杯和吸管,防止相互污染);(4)滴加1ml 100o NaClO溶液消化30s;2、进行扫描电镜的制样;本发明采用了显微镜骨片差异辅助扫描电镜的方法进行鉴定。在显微镜下,可以发现不同形状的骨片,骨片是随海参胚胎发育而逐渐完善稳定的重要特征,形状、大小随种类的不同而差别很大,骨片是进行海参分类及快速鉴别的重要依据。

一种区分不同海参品种的分析鉴定方法
技术领域
本发明涉及的是海参的品种鉴定技术领域,具体的说是一种区分不同海参品种的分析鉴定方法。
背景技术
海参一直是广受人们推崇的海洋珍品,名列海味八珍之首。刺参是目前我国唯一规模化养殖的海参品种。但近年来刺参的养殖相继出现了生长缓慢、死亡率升高、病害频发等一系列问题,刺参种质显著下降,于是,海参的国外引种及相关养殖非常热门,如从日本、俄罗斯等国家引进了红海参,从加拿大、美国引进了加利福尼亚海参等,以期提高海参种质资源的遗传多样性,满足目前日益增长的市场需求及急剧下降的野生资源危机,增加国内海参养殖品种的多样性。如何区分不同的海参品种问题迫在眉睫。
发明内容
本发明的目的在于针对现有技术的缺陷和不足,提供了一种区分不同海参品种的分析鉴定方法。
为实现上述目的,本发明采用的技术方案是:一种区分不同海参品种的分析鉴定方法,步骤是:
一、显微镜骨片鉴定方法是:
(1)将采集的海参个体,用无水乙醇固定;
(2)从海参背脊中部,切取1cmZ X 1mm的体壁组织,作为分析样品;
(3)每个样品3个重复,双蒸水洗净,用灭菌吸水纸吸干水分,至于5ml小烧杯中(每个样品配有单独的烧杯和吸管,防止相互污染);
(4)滴加1ml 100o NaClO溶液消化30s;
(5)挑出未消化的组织块,滴加3ml双蒸水,待组织块完全消融沉淀;
(6)吸去上层溶液,滴加双蒸水,重复漂洗3次;
(7)通过显微镜观察;
二、扫描电镜的制样方法是:
(1)在显微镜下吸取骨片悬液,小心滴加在滤纸上;
(2)用65℃的温度进行烘干4h;
(3)轻柔抖动滤纸,使骨片散落至胶膜上;
(4)HITACHIE-1010型离子溅射仪喷镀金膜;
(5)用HITACHI S-3400N扫描电镜观察并摄影。
本发明的有益效果为:除了从外形、棘刺和凸起等特征方面来区分外,采用了显微镜骨片差异辅助扫描电镜的方法进行鉴定。在显微镜下,可以发现不同形状的骨片,骨片是随海参胚胎发育而逐渐完善稳定的重要特征,形状、大小随种类的不同而差别很大,骨片是进行海参分类及快速鉴别的重要依据。

实施例:
一种区分不同海参品种的分析鉴定方法,步骤是:
一、显微镜骨片鉴定方法是:
(1)将采集的海参个体,用无水乙醇固定;
(2)从海参背脊中部,切取1cmZ X 1mm的体壁组织,作为分析样品;
(3)每个样品3个重复,双蒸水洗净,用灭菌吸水纸吸干水分,至于5ml小烧杯中(每个样品配有单独的烧杯和吸管,防止相互污染);
(4)滴加1ml 100o NaClO溶液消化30s;
(5)挑出未消化的组织块,滴加3ml双蒸水,待组织块完全消融沉淀;
(6)吸去上层溶液,滴加双蒸水,重复漂洗3次;
(7)通过显微镜观察。

本具体实施方式除了从外形、棘刺和凸起等特征方面来区分外,采用了显微镜骨片差异辅助扫描电镜的方法进行鉴定。通过在显微镜下,可以发现不同形状的骨片。骨片是随海参胚胎发育而逐渐完善稳定的重要特征,形状、大小随种类的不同而差别很大,骨片是进行海参分类及快速鉴别的重要依据。骨片分析是海参快速鉴定的理想指标。
虽然发现取海参的不同部位,比如背脊部、腹部触手、口部和肛门部组织,其骨片的特征有所不同,但通过取海参相同部位可以有效规避这个问题。
对于本领域技术人员而言,显然本发明不限于上述示范性实施例的细节,而且在不背离本发明的精神或基本特征的情况下,能够以其他的具体形式实现本发明。因此,无论从哪一点来看,均应将实施例看作是示范性的,而且是非限制性的,本发明的范围由所附权利要求而不是上述说明限定,因此旨在将落在权利要求的等同要件的含义和范围内的所有变化囊括在本发明内。
此外,应当理解,虽然本说明书按照实施方式加以描述,但并非每个实施方式仅包含一个独立的技术方案,说明书的这种叙述方式仅仅是为清楚起见,本领域技术人员应当将说明书作为一个整体,各实施例中的技术方案也可以经适当组合,形成本领域技术人员可以理解的其他实施方式。

Claims (1)

1.一种区分不同海参品种的分析鉴定方法,其特征在于:步骤是:
一、显微镜骨片鉴定方法是:
(1)将采集的海参个体,用无水乙醇固定;
(2)从海参背脊中部,切取1cmZX1mm的体壁组织,作为分析样品;
(3)每个样品3个重复,双蒸水洗净,用灭菌吸水纸吸干水分,至于5ml小烧杯中(每个样品配有单独的烧杯和吸管,防止相互污染);
(4)滴加1ml 100o NaClO溶液消化30s;
(5)挑出未消化的组织块,滴加3ml双蒸水,待组织块完全消融沉淀;
(6)吸去上层溶液,滴加双蒸水,重复漂洗3次;
(7)通过显微镜观察;
