本发明公开了一种辣根过氧化物酶稳定底物AB混合液,高压灭菌水定容至1000ml,包括.1mol/L柠檬酸243ml、0.2mol/L磷酸氢二钠257ml、联苯二胺(OPD)40mg、甘油50ml、焦磷酸钠500mg、五水合硫代硫酸钠(Na2S2O3.5H2O)50mg、30%过氧化脲150ul。这种混合液性能稳定,能避免酶联免疫吸附试验由于A、B液的分别加入而造成的操作误差所造成的风险。

技术领域
本发明涉及医药实验试剂领域,尤其涉及一种辣根过氧化物酶稳定底物AB混合液。
背景技术
酶联免疫吸附试验(Enzyme-Linked Immunosorbent Assays,ELISA)方法的基本原理是酶分子与抗体或抗抗体分子共价结合,这种结合既不会改变抗体的免疫学特性,也不影响酶的生物学活性。这种酶标记抗体可与吸附在固相载体上的抗原或抗体发生特异性结合。滴加底物溶液后,底物可在酶作用下使其所含的供氢体由无色还原型变成有色的氧化型,出现颜色反应。因此,可通过底物的颜色反应来判定有无相应的免疫反应,颜色反应的深浅与标本中相应抗体或抗原的量呈比例。此种显色反应可通过ELISA检测仪进行定量测定,通过结合酶化学反应的敏感性和抗原抗体反应的特异性,使ELISA方法成为一种既特异又敏感的检测方法。
用于标记抗体或抗抗体的酶须具有下列特性:有高度的活性和敏感性;在室温下稳定;反应产物易于显现;能商品化生产。如今应用较多的有辣根过氧化物酶(HorseradishPeroxidase,HRP)、碱性磷酸酶、葡萄糖氧化酶等,其中以HRP应用最广。
辣根过氧化物酶HRP具有分子量小,标记方法简单,性质稳定等优点,是临床检验试剂中的常用酶。HRP广泛用于生化检测项目和免疫类ELISA试剂盒,作为试剂盒显色体系的关键成分,对试剂盒的质量有重要影响。一般以OD403nm /OD275nm的比值RZ(德文Reinheit Zahl,RZ)表示酶的纯度。高纯度的酶RZ值应在3.0左右(最高可达3.4)。RZ值越小,非酶蛋白就越多。
辣根过氧化物酶发色底物为过氧化物和供氢体(DH2),其真正底物是H2O2,但人们习惯把供氢体称为底物或统称供氢体底物。在ELISA中常用的供氢体有以下几种:1.邻苯二胺(o-Phenylenediamine,OPD)是ELISA技术中应用较多的供氢体,酶作用后显黄色(最大吸收波长为492 nm),其灵敏度高,测定方便。可用肉眼观察判别,容易被浓硫酸终止反应,颜色可在数小时内不改变,是目前国内ELISA中最常用的一种; 2.TMB(3,3',5,5'-Tetramethylbenzidine, TMB,3,3',5,5'-四甲基联苯胺)是近年来常用的一种供氢底物,经酶作用后显天蓝色,目测对比度鲜明,加酸终止酶反应后变黄色(最大吸收波长为450nm),易比色定量测定。3.DBA (3,3'-Diaminobenzidine, DAB,3,3'-二氨基联苯胺)供氢底物,其反应产物为不能溶解的棕色吩嗪衍生物,可用不同光镜观察。此种多聚物能被还原和螯合四氯化碳,形成具有电子密度的产物,便于用电镜观察。
辣根过氧化物酶是酶联试剂最常用的工具酶之一,当催化反应时的供氢体为邻苯二胺OPD时,HRP能催化底物H2O2(过氧化氢)反应,生成物能供还原剂联苯二胺发生氧化,而产生较为稳定的显色反应。
过氧化氢有一定的腐蚀性且较不稳定,使用中常采用过氧化氢和尿素的结合物过氧化脲来替代,但过氧化脲亦不是十分稳定的化合物,在放置过程中可分解成脲素和过氧化氢,生成的过氧化氢可使显色剂变色,造成试剂中底物的变质、失效。
联苯二胺是一种强还原剂,可被氧化剂(包括过氧化氢)氧化而变色,因此在使用中常将底物A液(过氧化脲)和B液(联苯二胺)分别配制,在显色时先后加入,以完成显色反应。
由于A、B液的分别加入操作过程繁琐,有一定的时间限制,增加了操作误差所造成的风险,使用时亟待有一种稳定的混合试剂替代目前常用的A、B液。
发明内容
本发明提供一种辣根过氧化物酶稳定底物AB混合液,具有性能稳定的特点,在显色反应中能够避免误差,使得检测结果更精准。
一种辣根过氧化物酶稳定底物AB混合液,高压灭菌水定容至1000ml,包括:0.1mol/L柠檬酸243ml、0.2mol/L磷酸氢二钠257ml、联苯二胺(OPD) 40mg、甘油50ml、焦磷酸钠 500mg、五水合硫代硫酸钠 (Na2S2O3.5H2O)50mg、30%过氧化脲150ul。
本发明的有益效果是:混合液性能稳定,避免了酶联免疫吸附试验中A、B液的分操作误差所造成的风险,使得试验效果大大提高。

一种辣根过氧化物酶稳定底物AB混合液,包括:高压灭菌水定容至1000ml,0.1mol/L柠檬酸243ml、0.2mol/L磷酸氢二钠257ml、联苯二胺(OPD) 40mg、甘油50ml、焦磷酸钠 500mg、五水合硫代硫酸钠 (Na2S2O3.5H2O)50mg、30%过氧化脲150ul。
