Description
Technical Field
The invention belongs to the technical field of aquaculture, and relates to an anti-stress method for temporarily breeding coilia ectenes in a small water body.
Background
Yangtze river Coilia ectenes (Coilia nasus), commonly called saury, is rare and famous reproductive migratory fish in Yangtze river, is famous for delicacy, tenderness and beauty, enjoys the first reputation of three delicacies in Yangtze river, and is an important famous and special economic species in China. However, the coilia ectenes has strong stress response and dies after water discharge, namely, temporary culture in a small water body is needed in a plurality of links in the culture process, and the stress death is easily caused in the small water body, so that serious economic loss is caused. Therefore, there is a need to develop a method for improving the survival rate of the small water temporary culture of the Yangtze river coilia ectenes, so as to reduce the mortality rate and the economic loss in the process.
In other aquatic animals, compound anti-stress agents are usually prepared from compound vitamins, Chinese herbal medicines, glucose and the like, and have a certain anti-stress effect. According to the test of the inventor, the effect of the aquatic anti-stress agent on the current market in the Yangtze river coilia ectenes is not obvious.
disclosure of Invention
The invention aims to provide an anti-stress method for temporarily culturing coilia ectenes in a small water body, which starts from a physiological mechanism of stress regulation and control of coilia ectenes and effectively improves the survival rate of temporarily culturing coilia ectenes in the small water body for 6 hours.
In order to achieve the purpose, the invention adopts the following technical scheme:
The method for temporarily culturing coilia ectenes in a small water body to resist stress is characterized by comprising the following steps:
(1) Temporary culture tool: an open container is adopted;
(2) Denoising: adding elastic sound insulation material at the contact position of the open container and the ground;
(3) Water body regulation and control: adding water for culturing coilia ectenes into an open container, ensuring that the dissolved oxygen is higher than 7.0mg/L, the pH value is 7.0-7.6, the water temperature is lower than 25 ℃, adding 0.6-1.2% of sorbitol by mass ratio, and fully dissolving by stirring;
(4) Transporting the coilia ectenes with water to an open container for temporary culture, wherein the mouth part of the open container is provided with a shading device, so that light is reduced, and stress is reduced.
Preferably: the volume of the open container is 1L-1000L.
The elastic sound insulation material is sponge with the thickness larger than 10 cm.
the sunshade device is a double-layer sunshade net.
The water amount in the step (3) is 2/3 of the open container.
The temporary culture density of the coilia ectenes in the step (4) is 0.5-2.5 g/L.
compared with the prior art, the invention has the following beneficial effects:
1. The invention has simple using tools, is a conventional appliance in aquaculture and is convenient to obtain.
2. The anti-stress agent is a single component, is non-toxic and harmless, has low cost and obvious anti-stress effect, and can improve the temporary culture survival rate of a small water body for 6 hours to over 96 percent.

Detailed Description
The present invention will be described in further detail below with reference to specific examples, but the embodiments of the present invention are not limited thereto. Reagents, equipment and methods adopted by the invention are all reagents, equipment and methods which are conventionally and commercially available in the technical field and are conventionally used in the technical field.
1. An open plastic water tank aquarium with length, width and height (100cm, 60cm and 70cm) is selected as a temporary rearing tool.
2. And a sponge with the thickness of 20cm is additionally arranged on the part of the temporary rearing tool, which is contacted with the ground, so that the noise and the vibration are reduced to be transmitted through the ground.
3. Adding culture water of coilia ectenes to 2/3 of temporary culture tool, detecting dissolved oxygen of 9.0mg/L, pH 7.4, water temperature below 20 deg.C, adding 1.0% sorbitol, and stirring to dissolve completely.
4. Taking 1-year-old cultured coilia ectenes, putting the coilia ectenes with the average weight (20.5 +/-2.3) g according to the density of 1.5g/L, and covering two layers of sunshade nets.
5. counting the survival rate of the coilia ectenes for 6 hours, and detecting stress regulation and control indexes of the surviving coilia ectenes, including the transcription expression quantity and the protein expression quantity of blood sugar, cortisol and glyoxalase 1, wherein the detection results are as follows:
As shown in Table 1, the survival rate of the temporary culture is increased by 97.0% in 6 hours and 20.0% in the control group when the temporary culture is carried out by the method of the invention.
TABLE 1 temporary rearing survival rate
plasma cortisol and blood sugar are important indexes of the stress process, and as shown in fig. 1 and fig. 2, the method of the invention can obviously inhibit the severe rise of blood sugar, thereby being beneficial to the survival of the coilia ectenes.
The glyoxalase 1 is an important index of stress injury, the improvement of the activity of the glyoxalase can obviously reduce the stress injury, and through the formula and the operation of the invention, as shown in figure 3, the expression of the glyoxalase 1 gene is obviously improved, and the anti-stress capability of the glyoxalase is obviously improved.
As shown in FIG. 4, the protein expression level of glyoxalase 1 is also significantly increased, and the protein level shows that the anti-stress capability of the glyoxalase 1 is significantly improved.
