Abstract
本发明公开了抗菌脂肽bacaucin衍生物及其在抑制细菌感染中的应用。本发明公开的抗菌脂肽bacaucin衍生物为序列表中SEQ ID No.4和SEQ ID No.9‑18所示的多肽或其衍生物。实验证明,本发明中的Bacaucin衍生物对细菌具有抑制作用。尤其是Bacaucin‑1对金黄色葡萄球菌具有特异性的抑制作用,最小抑菌浓度为4μg/mL;且对Vero,HEp‑2和A549无细胞毒性作用,不具有溶血性,具备较好的组织相容性,对斑马鱼胚胎无毒性作用。Bacaucin衍生物对革兰氏阳性菌和革兰氏阴性菌具有抑制作用。表明,Bacaucin衍生物可以用来抑制细菌。
Description
技术领域
本发明涉及生物技术领域中,抗菌脂肽bacaucin衍生物及其在抑制细菌感染中的应用。
背景技术
抗生素的大规模使用始于20世纪40年代,在治疗细菌感染中起到决定性作用,挽救了无数生命。由于低剂量抗生素对动物生长具有促进作用,一些抗生素被添加到饲料中用作抗菌促长剂(antibiotic growth promoter,AGP)在世界范围内广泛应用。在以规模化、集约化为特征的现代化养殖业中,抗生素在动物疫病防控中发挥着无可替代的作用,保障了充足的肉、蛋、奶等食品供应。特别是在我国养殖业管理水平偏低,集约化程度不高的情况下,抗生素在减少动物发病、降低死亡率、促进生产、减少病原菌排放和改善公共卫生等方面发挥了极其重要的作用。但是由于长期以来抗生素的大量使用,细菌耐药性问题日益突出,特别是近年来,面对一波又一波超级耐药菌的侵袭,人类逐渐到了面临细菌感染而无药可用的窘境。例如,2011年南亚发现能产生NDM-1的革兰氏阳性细菌和以前未见报道的致病菌志贺氏菌和霍乱弧菌,2011年北美发现与环境相关的耐甲氧西林金黄色葡萄球菌(CA-MRSA)和2015年中国发现携带MCR-1的大肠杆菌,这些超级耐药菌的不断出现对整个人类疾病的治疗和预防带来了巨大的挑战。正是由于迅速产生的细菌耐药性和新型抗生素研发的巨大投入,减少畜牧生产过程中抗生素使用量的呼声越来越高。为确保养殖业持续发展,新型抗生素或抗生素替代品的开发势在必行。目前用于畜牧业的抗生素替代品主要包括有机酸、酶制剂、低聚糖、中草药和微生态制剂等,但这些替代品不仅生产成本高,而且在实际生产中效果亦不是很理想。从上个世纪70年代开始,新型抗生素的的研发速度不断下降,这主要是相同来源的细菌能产生新型抗生素的比例逐渐减小。目前世界范围内都掀起了寻找新型抗菌物质的热潮,如开放式抗菌药物发现组织(CO-ADD)发起了全球搜寻新抗生素项目,邀请全球化学家提交自己的化合物,进行抗菌活性筛查,旨在与超级细菌的赛跑中早日寻找到下一代抗生素。
小分子多肽类化合物,因其良好的抗菌活性、独特的抗菌机制受到越来越多的关注。然而目前大多数的小分子多肽的抗菌性只局限于体外抗菌活性,对热或酸碱的稳定性较差,对蛋白酶的稳定性较差,大大限制了其在体内的活性和在疾病治疗领域的应用。
发明内容
本发明所要解决的技术问题是如何抑制细菌的生长。
为解决上述技术问题,本发明首先提供了一种具有抑制细菌生长的bacaucin衍生物;bacaucin的化学结构式如式1所示;
所述bacaucin衍生物为名称为bacaucin-1、bacaucin-6、bacaucin-7、bacaucin-8、bacaucin-9、bacaucin-10、bacaucin-11、bacaucin-12、bacaucin-13、bacaucin-14或Bacaucin-15的物质;

所述Bacaucin衍生物可为环状多肽。所述环状多肽可为将相应氨基酸序列的首位相连得到的环状多肽。所述bacaucin-8可为将序列表中SEQ ID No.11的第1位与末位相连得到的环状多肽。
所述Bacaucin衍生物还可为对相应氨基酸序列进行Ac修饰得到的物质。所述Ac修饰可在相应氨基酸序列的N端进行。所述bacaucin-12可为将序列表中SEQ ID No.15的N端进行Ac修饰得到的多肽。
为解决上述技术问题,本发明还提供了与所述Bacaucin衍生物相关的生物材料,所述生物材料为下述B1)至B12)中的任一种:
B1)编码所述Bacaucin衍生物的核酸分子;
B2)含有B1)所述核酸分子的表达盒;
B3)含有B1)所述核酸分子的重组载体;
B4)含有B2)所述表达盒的重组载体;

其中,所述核酸分子可以是DNA,如cDNA、基因组DNA或重组DNA;所述核酸分子也可以是RNA,如mRNA或hnRNA等。
本领域普通技术人员可以很容易地采用已知的方法,例如定向进化和点突变的方法,对本发明的编码所述Bacaucin衍生物的核苷酸序列进行突变。那些经过人工修饰的,具有与本发明的所述Bacaucin衍生物的核苷酸序列75%或者更高同一性的核苷酸,只要编码所述Bacaucin衍生物且具有所述Bacaucin衍生物的功能,均是衍生于本发明的核苷酸序列并且等同于本发明的序列。
这里使用的术语“同一性”指与天然核酸序列的序列相似性。“同一性”包括与本发明的编码SEQ ID No.1或SEQ ID No.6-SEQ ID No.18中任一所示的氨基酸序列组成的多肽的核苷酸序列具有75%或更高,或85%或更高,或90%或更高,或95%或更高同一性的核苷酸序列。同一性可以用肉眼或计算机软件进行评价。使用计算机软件,两个或多个序列之间的同一性可以用百分比(%)表示,其可以用来评价相关序列之间的同一性。
上述应用中,B2)所述的含有编码所述Bacaucin衍生物的核酸分子的表达盒(所述Bacaucin衍生物基因表达盒),是指能够在宿主细胞中表达所述Bacaucin衍生物的DNA,该DNA不但可包括启动所述Bacaucin衍生物基因转录的启动子,还可包括终止所述Bacaucin衍生物基因转录的终止子。
可用现有的表达载体构建含有所述Bacaucin衍生物基因表达盒的重组载体。
所述转基因细胞系不包括繁殖材料。
为解决上述技术问题,本发明还提供了所述Bacaucin衍生物或所述生物材料在制备抗菌肽中的应用。
上述应用中,所述抗菌肽抑制的病原菌可为细菌。
为解决上述技术问题,本发明还提供了所述Bacaucin衍生物或所述生物材料在防止细菌引起腐败中的应用。
为解决上述技术问题,本发明还提供了所述Bacaucin衍生物或所述生物材料在制备防止细菌引起腐败的产品中的应用。

为解决上述技术问题,本发明还提供了含有所述Bacaucin衍生物的产品,所述产品为下述任一产品:
Y1)抑制细菌生长产品;

上述产品可以所述Bacaucin衍生物作为活性成分,还可将所述Bacaucin衍生物与其他物质组合在一起作为活性成分。
本发明中,所述细菌可为革兰氏阳性菌或革兰氏阴性菌。所述革兰氏阳性菌可为枯草芽孢杆菌(Bacillus subtilis)、解淀粉芽孢杆菌(Bacillus amyloliquefaciens)、地衣芽孢杆菌(Bacillus licheniformis)、蜡样芽孢杆菌(Bacillus cereus)、金黄色葡萄球菌(Staphylococcus aureus)和/或肠球菌。所述肠球菌可为粪肠球菌(Enterococcusfaecalis)(如粪肠球菌(Enterococcus faecalis)VRE A4、粪肠球菌(Enterococcusfaecalis)1F-1或粪肠球菌(Enterococcus faecalis)2F-7(optrA))或屎肠球菌(Enterococcus faecium)(如屎肠球菌(Enterococcus faecium)5F-10或屎肠球菌(Enterococcus faecium)4W-9(optrA+cfr))。所述金黄色葡萄球菌(Staphylococcusaureus)可为金黄色葡萄球菌(Staphylococcus aureus)MRSA T144、金黄色葡萄球菌(Staphylococcus aureus)MRSA 115、金黄色葡萄球菌(Staphylococcus aureus)MRSA W7、金黄色葡萄球菌(Staphylococcus aureus)MRSA 417(cfr)、金黄色葡萄球菌(Staphylococcus aureus)MRSA 1518(cfr)、金黄色葡萄球菌(Staphylococcus aureus)MRSA 1530(cfr)或金黄色葡萄球菌(Staphylococcus aureus)215(LZDR+cfr)。
所述革兰氏阴性菌可为大肠杆菌(Escherichia coli)、沙门氏菌(Salmonellaenterica)、绿脓杆菌(Pseudomonas aeruginosa)(如绿脓杆菌(Pseudomonas aeruginosa)PAO1)或奇异变形杆菌(Proteus mirabilis)。
其中,粪肠球菌(Enterococcus faecalis)2F-7(optrA)携带有耐药基因optrA,屎肠球菌(Enterococcus faecium)4W-9(optrA+cfr)携带有耐药基因optrA和cfr,金黄色葡萄球菌(Staphylococcus aureus)MRSA 417(cfr)、金黄色葡萄球菌(Staphylococcusaureus)MRSA 1518(cfr)和金黄色葡萄球菌(Staphylococcus aureus)MRSA 1530(cfr)均携带有耐药基因cfr,金黄色葡萄球菌(Staphylococcus aureus)215(LZDR+cfr)携带有耐药基因cfr且利奈唑胺耐药,粪肠球菌(Enterococcus faecalis)VRE A4为万古霉素的耐药菌株。
本发明中,所述预防和/或治疗细菌引起的疾病的产品均可为药物或疫苗。所述防止细菌引起腐败的产品均可为防腐剂。
实验证明,本发明中的Bacaucin衍生物对细菌具有抑制作用。尤其是Bacaucin-1对金黄色葡萄球菌具有特异性的抑制作用,最小抑菌浓度为4μg/mL;且对Vero,HEp-2和A549无细胞毒性作用,不具有溶血性,具备较好的组织相容性,对斑马鱼胚胎无毒性作用。此外,本发明中的小分子多肽Bacaucin-1还具有分子量小,氨基酸组成简单,便于大规模合成;耐高温,在pH 4-7范围内稳定,对多种蛋白酶稳定等优点。本发明的小分子多肽Bacaucin-1不仅可以有效地抑制肌肉腐败模型和小鼠大腿感染模型中金葡菌的繁殖,而且大大提高了菌血症模型中小鼠的存活率。Bacaucin衍生物对革兰氏阳性菌和革兰氏阴性菌具有抑制作用。表明,Bacaucin衍生物可以用来抑制细菌。

下面结合具体实施方式对本发明进行进一步的详细描述,给出的实施例仅为了阐明本发明,而不是为了限制本发明的范围。
下述实施例中的实验方法,如无特殊说明,均为常规方法。
下述实施例中所用的材料、试剂、仪器等,如无特殊说明,均可从商业途径得到。
以下实施例中的定量试验,均设置三次重复实验,结果取平均值。
下述实施例中的BALB/c雌性小鼠为北京维通利华实验动物技术有限公司产品。
实施例1、抗菌脂肽bacaucin的制备及其抗菌活性
本发明从土壤中分离得到一株菌,利用在16S rRNA的两端设计的引物对AGAGTTTGATCCTGGCTCAG(序列1)与ACGGCTACCTTGTTACGACTT(序列2))对该菌株的16S rRNA进行PCR扩增,扩增产物的16S rRNA的序列如序列表中序列3所示,该菌株16S rRNA的序列分析结果如图1所示,经序列比对发现该16S rRNA的序列表明该菌株为枯草芽孢杆菌(Bacillus subtilis),将该菌命名为枯草芽孢杆菌(CAU21)。
枯草芽孢杆菌(CAU21),细胞呈杆状,直径0.6μm~1.0μm,长1.5μm~2.0μm,芽孢中生,椭圆,孢囊不膨大,革兰氏反应阳性。在锰营养琼脂平板上37℃培养36h形成的菌落为圆形,直径3.0mm~10.0mm,淡黄色,中间色深,表面平整不光滑,无光泽,边缘不整齐。
枯草芽孢杆菌(CAU21)已于2016年12月16日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏中心登记入册编号:CGMCC No.13463。
一、抗菌脂肽bacaucin的制备
(1)甲醇提取:枯草芽孢杆菌(CAU21)接种于大豆琼脂平板(Sigma公司)上于37℃培养10天后。刮取菌落置于1mL甲醇中,80℃下水浴30min。待甲醇挥发完全后,用0.5mL甲醇复溶,充分涡动混匀后,13000g离心3min,弃沉淀,收集上清液,将上清液利用N2吹干,得到菌体粗提物;将菌体粗提物用500μL甲醇复溶,得到粗提取液,保存于4℃。
(2)纯化:将步骤(1)得到的粗提取液上样于Sephadex LH-20凝胶过滤层析柱(该层析柱的填料为Sephadex LH-20,Sephadex LH-20为GE公司产品)(1.6×80cm(直径×高度)),采用甲醇以0.8mL/min的流速对层析住进行等度洗脱,收集6h内的洗脱液。利用多功能酶标仪SpectraMax M5测定洗脱液的吸光值(214nm),绘制对应的吸收曲线,得到三个特征吸收峰(图2中A),将三个吸收峰对应的洗脱液分别称为洗脱液1、洗脱液2和洗脱液3。对洗脱液1、洗脱液2和洗脱液3利用肉汤稀释法(CLSI(Clinical and Laboratory StandardsInstitute)2015)进行金黄色葡萄球菌(ATCC29213)抗菌活性测试,发现对应于2号峰的洗脱液2具有最高的抗菌活性。
对洗脱液2进行富集浓缩后,进一步采用反相高效液相色谱(RP-HPLC)对其进行分离,固定相为C18(Waters公司),流动相为乙腈和水,流速为1mL/min,采用梯度洗脱的方法对其进行进一步的分离和纯化,具体方法如下:

结果得到四个色谱峰(图2中B),1号峰、2号峰、3号峰和4号峰,对应的洗脱液分别为洗脱液A、洗脱液B、洗脱液C和洗脱液D,将这四个洗脱液利用肉汤稀释法(CLSI(Clinicaland Laboratory Standards Institute)2015)进行金黄色葡萄球菌(ATCC29213)进行抗菌活性测定,发现保留时间为7.88min的1号峰具有最高的抗菌活性,收集1号峰的洗脱液(洗脱液A),N2吹干,得到精提物,-20℃保存。将精提物以相同的色谱条件进行纯度鉴定发现,其纯度大于98%,如图2中C所示,结果显示精提物为单一组分,纯度达到化学结构测定的要求。
(3)结构鉴定:采用紫外色谱(UV)、红外光谱(FTIR)、超高效液相色谱串联质谱(UPLC-MS/MS)和核磁共振氢谱(1H NMR)对其分子结构进行鉴定。
紫外吸收光谱(UV):将上述步骤(2)得到的精提物极微量溶于超纯水中,用PDA检测器(waters)在200nm~400nm波长范围内以超纯水为空白对照进行全波长扫描,自动形成紫外吸收图谱(图3)。紫外光谱结果表明该精提物的最大吸收波长为219.2nm。
红外光谱(FTIR):用Spotlight 200Micro-Infrared spectrometer(PE)进行400cm-1-4000cm-1区域内扫描。结果显示,该精提物具有酰胺键特征吸收峰(图3)。图3中,N-H伸缩振动(3217cm-1)和N-H弯曲振动(1545cm-1),伸缩振动of C=O and C-N chemicalbonds(1639cm-1and 1397cm-1),说明bacaucin中含有酰胺键,即肽键。
紫外吸收光谱与红外光谱的结果表明该精提物是由氨基酸组成的多肽类化合物。
超高效液相色谱串联质谱(UPLC-MS/MS):采用Waters;条件:雾化器,28;毛细管电压,3200V;脱溶剂温度,350℃;,源温:100℃,scan range:50~1037Da,syringe pump:0.3ml/min,数据分析软件为masslynx v4.1。一级质谱表明该精提物的分子量为1035.8Da。
核磁共振氢谱(1H NMR):以D2O为溶剂,室温下测定。采用Bruker AvanceIII400MHzNMR spectrometer(Bruker),进行氢谱(1HNMR)的测定,共振频率为400MHZ。
二级质谱和核磁共振氢谱结果(图3)表明该精提物是由7个氨基酸残基和β-羟基脂肪酸链组成的环肽类化合物,其化学结构式如式1所示,将其命名为抗菌脂肽bacaucin。
二、抗菌脂肽bacaucin的抗菌活性
试验菌株包括革兰氏阳性菌(枯草芽孢杆菌、解淀粉芽孢杆菌、蜡样芽孢杆菌、金黄色葡萄球菌、粪肠球菌和屎肠球菌)和革兰氏阴性菌(大肠杆菌、绿脓杆菌、沙门氏菌和奇异变形杆菌),菌株信息如表1所示,采取肉汤稀释法(CLSI(Clinical and LaboratoryStandards Institute)2015)对其抗菌谱及最小抑菌浓度进行测定,测定温度为37℃,pH为7.0。

其中,粪肠球菌(Enterococcus faecalis)2F-7(optrA)携带有耐药基因optrA,屎肠球菌(Enterococcus faecium)4W-9(optrA+cfr)携带有耐药基因optrA和cfr,金黄色葡萄球菌(Staphylococcus aureus)MRSA 417(cfr)、金黄色葡萄球菌(Staphylococcusaureus)MRSA 1518(cfr)和金黄色葡萄球菌(Staphylococcus aureus)MRSA 1530(cfr)均携带有耐药基因cfr,金黄色葡萄球菌(Staphylococcus aureus)215(LZDR+cfr)携带有耐药基因cfr且利奈唑胺耐药,粪肠球菌(Enterococcus faecalis)VRE A4为万古霉素的耐药菌株。

结果显示,本发明中的抗菌脂肽bacaucin对革兰氏阳性菌具有很好的抗菌作用,最小抑菌浓度(minimal inhibitory concentration,MIC)为2-16μg/mL,且对包括粪肠球菌(E.faecalis)VRE A4、粪肠球菌(E.faecalis)2F-7(optrA)、屎肠球菌(E.faecium)4W-9(optrA+cfr)、金黄色葡萄球菌MRSA 1518(cfr)和金黄色葡萄球菌(S.aureus)215(LZDR+cfr)在内的耐药菌仍然具有很好的抗菌活性,抗菌脂肽bacaucin的抗菌谱见表2和图5。

实施例2、抗菌脂肽bacaucin衍生物的制备及其抗菌活性
对实施例1的抗菌脂肽bacaucin进行优化,得到多种bacaucin衍生物(bacaucinDerivatives),即Bacaucin-1~Bacaucin-16,Bacaucin-1为小分子多肽,Bacaucin其他衍生物Bacaucin-2~Bacaucin-16均为多肽,其中Bacaucin-8为环状多肽,Bacaucin-10的第5位为鸟氨酸(Orn)残基,Bacaucin-12的N端由Ac修饰,Bacaucin-1~Bacaucin-16的氨基酸序列分别为序列表中序列4-19,具体信息如图4所示,其中1至16分别表示Bacaucin-1~Bacaucin-16。
由上海吉尔生化有限公司合成Bacaucin-1~Bacaucin-16,合成后利用高效液相色谱法(HPLC)和超高效液相-串联质谱对这几种bacaucin衍生物的纯度和分子量进行测定,结果表明Bacaucin-1~Bacaucin-16的纯度均大于95%,具体信息如表3所示。

1、抗菌脂肽bacaucin衍生物的抗菌谱及最小抑菌浓度的测定
试验菌株包括革兰氏阳性菌(枯草芽孢杆菌、解淀粉芽孢杆菌、蜡样芽孢杆菌、金黄色葡萄球菌、粪肠球菌和屎肠球菌)和革兰氏阴性菌(大肠杆菌、绿脓杆菌、沙门氏菌和奇异变形杆菌),采取肉汤稀释法(CLSI(Clinical and Laboratory Standards Institute)2015)对抗菌脂肽bacaucin衍生物抗菌谱及最小抑菌浓度进行测定,测定温度为37℃,pH为7.0。将实施例1的bacaucin作为对照。
结果表明,小分子多肽Bacaucin-1对金黄色葡萄球菌,包括金黄色葡萄球菌(MRSA)T144,金黄色葡萄球菌MRSA 1518(cfr),金黄色葡萄球菌(S.aureus)215(LZDR+cfr)携带耐药基因的金黄色葡萄球菌具有特异性的抑制作用,其最小抑菌浓度为2-4μg/mL,其他Bacaucin衍生物的抑菌作用能力均小于Bacaucin-1,结果如表4和图5所示,图5中1至16分别表示Bacaucin-1~Bacaucin-16,金黄色葡萄球菌29213表示金黄色葡萄球菌(Staphylococcus aureus)ATCC 29213。

2、小分子多肽Bacaucin-1的热稳定性和酸碱稳定性
利用金黄色葡萄球菌(S.aureus,ATCC 29213)在Bacaucin-1的最小抑菌浓度下测定其热稳定性和酸碱稳定性。
将Bacaucin-1分别在20-121℃的温度范围内以及pH2-pH12的pH范围分别孵育1h,检测Bacaucin-1的MIC值,计算剩余抗菌活性,剩余抗菌活性=37℃、pH为7.0下的MIC值/待测条件下的MIC值×100%。热稳定性和酸碱稳定性,温度取值分别为25℃、60℃、65℃、70℃、75℃、80℃、85℃、90℃、100℃和121℃,pH的取值分别为2、3、4、5、6、7、8、9、10和12。在测定Bacaucin-1的热稳定性时,pH为7.0;在测定Bacaucin-1的酸碱稳定性时,先将Bacaucin-1溶于PBS溶液中,调节pH至相应的pH值,孵育1h后,调节pH回至7.0,测定温度为37℃。
如图6所示,80℃及以下,pH(4-7)之间,bacaucin-1可以100%的保持其抗菌活性,有较好的热稳定性和酸碱稳定性。
3、小分子多肽Bacaucin-1的毒性评价
对步骤1的各抗菌脂肽bacaucin衍生物的细胞毒性、溶血性和斑马鱼胚胎毒性进行评价,细胞毒性试验将选择Vero,HEp-2和A549作为受试细胞;利用羊血红细胞研究其溶血性,以判断其与组织的相容性;以斑马鱼作为模式动物,研究其对斑马鱼胚胎的毒性。
3.1细胞毒性
采用WST-1测定bacaucin和bacaucin-1细胞毒性:
96孔板先加入100μL含1%的青霉素和链霉素的MEM培养基,加入bacaucin,bacaucin的浓度如下1024μg/mL、512μg/mL、256μg/mL、128μg/mL、64μg/mL、32μg/mL、16μg/mL、8μg/mL、4μg/mL和2μg/mL,每种浓度三个复孔;然后向每孔中加入一层Vero细胞,每孔细胞数为104;37℃孵育24h后,弃去100μL液体,每孔加入10μL的WST-1,孵育1h后测定其450nm。
bacaucin-1细胞毒性的测定同上述方法。
3.2溶血性
96孔板先加入100μL PBS,加入bacaucin,bacaucin的浓度如下1024μg/mL、512μg/mL、256μg/mL、128μg/mL、64μg/mL、32μg/mL、16μg/mL、8μg/mL、4μg/mL和2μg/mL,每种浓度三个复孔;然后向每孔中加入一层100μL 8%红细胞悬浮液(脱纤维羊血3000g,离心10min后去除血清,PBS清洗两次后制备成100%红细胞悬浮液,将8mL该悬浮液与92mL的PBS混合得到8%红细胞悬浮液),37℃孵育1h后,离心,吸取上清液100μL,测定其OD576nm。利用PBS和Triton X-100分别作为阴性对照(N)和阳性对照(P)。溶血率=(待测液体OD值-阴性对照OD值)/(阳性对照OD值-阴性对照OD值)×100%。
bacaucin-1溶血性的测定同上述方法。
3.3斑马鱼胚胎毒性
向胚胎培养液(称取0.8g NaCl、0.04 KCl、0.00358g Na2HPO4、0.006g KH2PO4、0.144g CaCl2、0.246g MgSO4·7H2O、0.35g NaHCO3、0.06g青霉素、0.1g链霉素加入含有800mL灭菌水的1L烧杯中,搅拌至完全溶解,再用灭菌水定容至1L得到胚胎培养液。)中加入Bacaucin-1,得到Bacaucin-1分别为10μg/mL、20μg/mL和40μg/mL的Bacaucin-1溶液;向细胞培养液中加入替米考星得到替米考星浓度为10μg/mL的替米考星溶液。利用Bacaucin-1溶液处理斑马鱼胚胎,并将细胞培养液作为阴性对照,替米考星溶液作为阳性对照,具体步骤如下:受精后6h的胚胎中加入不同浓度的bacaucin-1溶液或替米考星溶液,置于28℃培养箱中培养,96h后取出利用SZM 76立体显微镜(重庆澳浦光电技术有限公司)观察斑马鱼的形态学变化。
结果(图7)表明,小分子多肽Bacaucin-1对受试细胞Vero,HEp-2和A549无任何细胞毒性作用,对羊血红细胞没有溶血性,对模式动物斑马鱼的胚胎也无毒性作用。而Bacaucin不仅对Vero、HEp-2细胞表现出一定的细胞毒性,而且具有溶血性(图8)。
4、小分子多肽Bacaucin-1在食品防腐中的应用
将市场上买来的鸡脯肉平均分成24份,每份约5g,随机分成6组,每组4份。各组分别为:PBS组、金黄色葡萄球菌污染组、污染防腐组1、污染防腐组2、污染防腐组3和污染防腐组4。
将金黄色葡萄球菌(S.aureus,ATCC 29213)悬浮于PBS中,得到浓度为6.3×104CFU/mL的菌悬液;将Bacaucin-1溶于PBS中,分别得到浓度为200、500、1000和2000μg/mL的Bacaucin-1溶液。
PBS组鸡脯肉按照如下方式利用PBS进行处理:向每份肉块表面滴加100μL的PBS,随后滴加100μL的PBS缓冲液,作为对照。
金黄色葡萄球菌污染组鸡脯肉按照如下方式处理:向每份肉块表面滴加100μL的菌悬液,随后滴加100μL的PBS缓冲液,得到0mg/kg Bacaucin-1处理的肉块。
污染防腐组1鸡脯肉按照如下方式处理:向每份肉块表面滴加100μL的菌悬液,随后滴加100μL的200μg/mL的bacaucin-1溶液,得到4mg/kg Bacaucin-1处理的肉块。
污染防腐组2鸡脯肉按照如下方式处理:向肉块表面滴加100μL的菌悬液,随后滴加100μL的500μg/mL的bacaucin-1溶液,得到10mg/kg Bacaucin-1处理的肉块。
污染防腐组3鸡脯肉按照如下方式处理:向肉块表面滴加100μL的菌悬液,随后滴加100μL的1000μg/mL的bacaucin-1溶液,得到20mg/kg Bacaucin-1处理的肉块。
污染防腐组4鸡脯肉按照如下方式处理:向肉块表面滴加100μL的菌悬液,随后滴加100μL的2000μg/mL的bacaucin-1溶液,得到40mg/kg Bacaucin-1处理的肉块。
将各组鸡脯肉按照相应的方式处理后,于37度培养24h,然后鉴定各组的肉块菌落总数,并用形态学和HE染色观察肉块组织结构。
将肉块匀浆、混匀,使用金黄色葡萄球菌显色培养基测定细菌数:将肉块研磨液倍比稀释多个梯度后,分别加入9cm平皿中,倾注15mL的金黄色葡萄球菌显色培养基,于37℃下培养24h,选择平板上红色的菌为30-300个之间的进行计数。
结果(图9中A)表明,加入bacaucin-1后的肉块中的细菌数显著降低,并且细菌数随bacaucin-1浓度的增加而降低。形态学和HE染色的结果表明,加入bacaucin-1后,肉块的组织结构和风味得到了较好的保持。表明,bacaucin-1具有很好的防腐作用。
5、小分子多肽Bacaucin-1在治疗小鼠大腿细菌感染和菌血症中的应用
5.1治疗小鼠大腿感染
将30只BALB/c雌性小鼠(18g),平均分成5组,每组6只小鼠。各组分别为:PBS组,不同浓度bacaucin-1治疗组。细菌感染前1天腹腔注射200mg/kg环磷酰胺以构建小鼠的中性粒细胞缺失模型,防止体内中性粒细胞影响后续试验。
将金黄色葡萄球菌(S.aureus,ATCC 29213)悬浮于PBS中,得到浓度为1.5×106CFU/mL的金黄色葡萄球菌悬浮液;将Bacaucin-1溶于PBS中,分别得到浓度为360,900,1800,3600μg/mL的Bacaucin-1溶液。
环磷酰胺注射1天后于小鼠右侧大腿肌肉注射0.1mL的金黄色葡萄球菌悬浮液后,2h后腹腔注射分别给予PBS(即Bacaucin-1浓度为0),360,900,1800和3600μg/mL的Bacaucin-1溶液,注射体积均为0.2mL,即给药剂量分别为0,4,10,20和40mg/kg,每组注射一种浓度的Bacaucin-1溶液或注射PBS。24h后,将所有小鼠安乐死,取右侧大腿肌肉,将肉块匀浆、混匀,按照步骤4中的金黄色葡萄球菌鉴别培养基测定菌含量方法测定细菌数。
结果(图9中B)表明,Bacaucin-1能有效的降低小鼠大腿感染模型中大腿细菌数,并且细菌数随Bacaucin-1浓度的增加而降低。
5.2治疗菌血症
将48只BALB/c雌性小鼠,平均分成8组,每组6只小鼠,平均体重为18g。各组分别为:PBS组、MRSA感染组、不同浓度bacaucin-1治疗组(4组)及不同浓度万古霉素治疗组(2组,作为阳性对照)。
将金黄色葡萄球菌(S.aureus,ATCC 29213)悬浮于PBS中,得到浓度为1.5×109CFU/mL的MRSA悬浮液;将Bacaucin-1溶于PBS中,分别得到浓度为360,900,1800,3600μg/mL的Bacaucin-1溶液;将万古霉素溶于PBS中,分别得到浓度为180和1800μg/mL的万古霉素溶液。
PBS组:腹腔注射0.5mL PBS 1h后,腹腔注射0.2mL PBS。
MRSA感染组:腹腔注射0.5mL MRSA悬浮液1h后,腹腔注射0.2mL PBS,给药剂量为0mg/kg。
不同浓度bacaucin-1治疗组:腹腔注射0.5mL MRSA悬浮液1h后,腹腔注射0.2mLBacaucin-1溶液进行治疗,每组一种Bacaucin-1溶液,给药剂量分别为0,4,10,20和40mg/kg。
不同浓度万古霉素治疗组:腹腔注射0.5mL MRSA悬浮液1h后,腹腔注射0.2mL万古霉素溶液进行治疗,每组一种万古霉素溶液,给药剂量分别为2和20mg/kg。
48h后观察每组小鼠的存活率,检测小鼠的心、肝、脾、肺和肾中致病菌的细菌数,按照步骤4中的金黄色葡萄球菌鉴别培养基测定菌含量方法进行,对药效进行评价。
结果(图9中C和D)表明,48小时后与MRSA感染组相比,使用bacaucin-1和万古霉素进行治疗后,小鼠存活率得到显著提升,并且使用bacaucin-1进行治疗的小鼠存活率与万古霉素相当,bacaucin-1的治疗效果随着浓度的升高而增加。其次,治疗组小鼠的心、肝、脾、肺和肾中致病菌的菌落总数也显著降低。

1.bacaucin衍生物,bacaucin的化学结构式如式1所示;
所述bacaucin衍生物为名称为bacaucin-1、bacaucin-6、bacaucin-7、bacaucin-8、bacaucin-9、bacaucin-10、bacaucin-11、bacaucin-12、bacaucin-13、bacaucin-14或Bacaucin-15的物质;
所述bacaucin-1为a1)或a2);
a1)氨基酸序列是SEQ ID No.4的多肽;
a2)将SEQ ID No.4的氨基酸序列经过一个或几个氨基酸残基的取代和/或缺失和/或添加得到的多肽;
所述bacaucin-6为b1)或b2);
b1)氨基酸序列是SEQ ID No.9的多肽;
b2)将SEQ ID No.9的氨基酸序列经过一个或几个氨基酸残基的取代和/或缺失和/或添加得到的多肽;

B2)含有B1)所述核酸分子的表达盒;
B3)含有B1)所述核酸分子的重组载体;
B4)含有B2)所述表达盒的重组载体;
B5)含有B1)所述核酸分子的重组微生物;
B6)含有B2)所述表达盒的重组微生物;
B7)含有B3)所述重组载体的重组微生物;
B8)含有B4)所述重组载体的重组微生物;
B9)含有B1)所述核酸分子的转基因动物细胞系;
B10)含有B2)所述表达盒的转基因细胞系;
B11)含有B3)所述重组载体的转基因细胞系;
B12)含有B4)所述重组载体的转基因细胞系。
3.权利要求1所述bacaucin衍生物或权利要求2所述生物材料在制备抗菌肽中的应用。
4.根据权利要求3所述的应用,其特征在于:所述抗菌肽抑制的病原菌是细菌。
5.权利要求1所述bacaucin衍生物或权利要求2所述生物材料在防止细菌引起腐败中的应用。
6.权利要求1所述bacaucin衍生物或权利要求2所述生物材料在制备防止细菌引起腐败的产品中的应用。
7.权利要求1所述bacaucin衍生物或权利要求2所述生物材料在制备预防和/或治疗细菌引起的疾病的产品中的应用。
8.权利要求1所述bacaucin衍生物或权利要求2所述生物材料在预防和/或治疗细菌引起的疾病中的应用。
9.含有权利要求1所述bacaucin衍生物的产品,所述产品为下述任一产品:
Y1)抑制细菌生长产品;
Y2)治疗和/或预防细菌所致疾病产品;
Y3)防止细菌引起腐败的产品。
10.根据权利要求4-8中任一所述的应用,或权利要求9所述的产品,其特征在于:所述细菌为革兰氏阳性菌或革兰氏阴性菌。
