Abstract

本发明提供了一种含有薁磺酸钠的漱口散剂及其制备方法,所述漱口散剂含有茶多酚,以质量百分比计,由包括以下组分的原料制备而成:薁磺酸钠0.1%~1%、茶多酚0.1%~1%、余量为辅料,所述辅料包括填充剂、崩解剂、pH调节剂、粘合剂以及调味剂。本发明所提供的制备方法为将薁磺酸钠和茶多酚原料混合均匀;将所得的混合物再与其他辅料通过等量递加法混合均匀,并过筛;最后分装到铝塑袋内。本发明所提供的漱口散剂具有稳定性好,疗效好,便于携带和使用的优点。

Description

一种含有薁磺酸钠的漱口散剂及其制备方法
技术领域
本发明属于药物制剂技术领域,具体涉及一种含有薁磺酸钠的漱口散剂及其制备方法。
背景技术
薁磺酸钠是洋甘菊花的有效成分,其药理机制是不经过垂体-肾上腺系统,直接抑制白细胞向体外迁移,抑制肥大细胞释放组胺及PGE2合成,抑制多种炎症物质引起炎症,作用直接持久,且能促进肉芽的合成和上皮细胞的修复,因此主要用于治疗咽炎、扁桃体炎、口腔炎、急性牙龈炎、舌炎和口腔溃疡。目前,薁磺酸钠用于治疗口腔疾患的只有水溶液,存在携带不方便、不稳定等缺点。散剂虽然是一种常见的药剂,但目前还没有一种较为稳定的用于口腔治疗和护理的漱口散剂。
发明内容
本发明的目的之一是提供一种含有薁磺酸钠的漱口散剂,以解决现有薁磺酸钠药剂携带不方便、不稳定的问题。
本发明的目的之二是提供上述含有薁磺酸钠的漱口散剂的制备方法。
本发明的目的一是通过以下技术方案实现的:所述漱口散剂含有茶多酚。
以质量百分比计,所述漱口散剂茶多酚含量为0.1%~1%。
以质量百分比计,所述漱口散剂茶多酚含量为0.2%~0.5%。
以质量百分比计,所述漱口散剂由包括以下组分的原料制备而成:薁磺酸钠0.1%~1%、茶多酚0.1%~1%、余量为辅料。
以质量百分比计,所述漱口散剂由包括以下组分的原料制备而成:薁磺酸钠0.1%~1%、茶多酚0.2%~0.5%、余量为辅料,优选薁磺酸钠用量范围为0.4%~0.6%。
以质量百分比计,所述漱口散剂由包括以下组分的原料制备而成:薁磺酸钠0.4%、茶多酚0.2%~0.5%、余量为辅料。
所述辅料包括填充剂、崩解剂、pH调节剂、粘合剂以及调味剂。
所述填充剂选自甘露醇、乳糖、木糖醇或蔗糖中的至少一种;
所述崩解剂选自淀粉、羧甲淀粉钠、预胶化淀粉中的至少一种;
所述pH调节剂选自碳酸氢钠、磷酸氢二钠或磷酸二氢钠中的至少一种;
所述粘合剂选自聚乙烯醇、羟丙基纤维素或聚维酮中的至少一种;
所述调味剂包括薄荷醇和香精。
所述辅料还包括润滑剂,所述润滑剂选自二氧化硅、硬脂酸或硬脂酸盐中的至少一种。
所述辅料不限于上述种类范围,可选用现有技术中的其他任意辅料,辅料用量均为理论上的合理用量范围。
本发明提供了上述漱口散剂的制备方法,包括以下步骤:
(1)将薁磺酸钠和茶多酚原料混合均匀;
(2)将步骤(1)所得的混合物再与其他辅料通过等量递加法混合均匀,并过筛;
(3)将步骤(2)所得产品按剂量分装到铝塑袋内。
本发明所提供的漱口散剂具有稳定性好,疗效好,便于携带和使用的优点,使用时,将散剂用水溶解,直接漱口使用。茶多酚具有良好的抗氧化作用,通过加入茶多酚并配合其他辅料,有效解决了传统散剂易氧化、易吸湿的问题,稳定性优于传统水溶液制剂,同时将薁磺酸钠和茶多酚配合使用使得本发明漱口散剂疗效显著增强。此外,本发明漱口散剂制备工艺简单,质量易于控制,储运方便。
本发明漱口散剂具有口腔消毒的作用,在临床上用于咽炎、扁桃体炎、口腔炎、急性牙龈炎、舌炎、口腔溃疡的治疗,也可用作日常口腔清洁剂,适用于老人、儿童或患口腔疾病不适宜刷牙的人群;此外,本发明漱口散剂还可用于病毒性感冒的预防和辅助治疗;作为日常保健品,也用于严重污染雾霾天气的口腔鼻腔清洗剂。
具体实施方式
下面结合具体实施例进一步说明本发明的技术方案,本发明所用原料均为市售普通产品。
实施例1
一种含有薁磺酸钠的漱口散剂,以质量百分比计,由以下组分的原料制备而成:薁磺酸钠0.4%、茶多酚0.25%、碳酸氢钠2%、甘露醇94.65%、聚维酮K30 1.5%、薄荷醇1%、香精0.2%。将按上述量称取的薁磺酸钠和茶多酚混合均匀,将得到的混合物与其他辅料通过等量递加法混合,然后过100目筛两遍,混合10分钟,最后将制成的散剂按1g/包分装到铝塑袋内。
实施例2~7、对比例1
改变茶多酚的量,同时配合改变甘露醇的量(具体见表1~4),其他原料和制备方法同实施例1。
对上述所有样品进行稳定性测试,测试方法如下。
本发明的稳定性测试分为加速试验和长期试验,加速试验为将样品置于恒温恒湿箱中,在温度40℃±2℃、湿度75%±5%条件下储存6个月,并分别在第0天和第1、2、3、6个月末时取样,观察颜色变化并检测样品中薁磺酸钠的含量,得到不同时间样品中薁磺酸钠含量占初始配方中薁磺酸钠用量的百分比,颜色变化结果如表1所示,薁磺酸钠的检测结果如表2所示。
长期试验为将样品置于恒温恒湿箱中,在温度30℃±2℃、湿度65%±5%条件下储存12个月,并分别在第0天和第3、6、9、12个月末时取样,观察颜色变化并检测样品中薁磺酸钠的含量,得到不同时间样品中薁磺酸钠含量占初始配方中薁磺酸钠用量的百分比,颜色变化结果如表3所示,薁磺酸钠的检测结果如表4所示。

由以上结果可知,茶多酚对薁磺酸钠有保护作用,剂量越大,效果越好。在加速实验条件下,茶多酚用量达到0.25%时,能起到很好的保护作用,在长期实验条件下,茶多酚用量达到0.2%时,就能起到很好的保护作用,结合成本及不同地域气候考虑,选取茶多酚用量为0.25%为最佳实施例,茶多酚用量在0.1%~1%时,本发明漱口散剂的稳定性均显著提高。
实施例8
一种含有薁磺酸钠的漱口散剂,以质量百分比计,由以下组分的原料制备而成:薁磺酸钠0.1%、茶多酚0.25%、碳酸氢钠2%、甘露醇94.95%、聚维酮K30 1.5%、薄荷醇1%、香精0.2%。将按上述量称取的薁磺酸钠和茶多酚混合均匀,将得到的混合物与其他辅料通过等量递加法混合,然后过100目筛两遍,混合10分钟,最后将制成的散剂按1g/包分装到铝塑袋内。
实施例9
一种含有薁磺酸钠的漱口散剂,以质量百分比计,由以下组分的原料制备而成:薁磺酸钠1.0%、茶多酚0.25%、碳酸氢钠2%、甘露醇94.05%、聚维酮K30 1.5%、薄荷醇1%、香精0.2%。将按上述量称取的薁磺酸钠和茶多酚混合均匀,将得到的混合物与其他辅料通过等量递加法混合,然后过100目筛两遍,混合10分钟,最后将制成的散剂按1g/包分装到铝塑袋内。
临床实验
1、实验对象:选取各种口腔不适病例160人,分成两组,每组80人;第一组口腔溃疡病例38人,自述牙龈出血病例25人,口腔异味病例17人;第二组口腔溃疡病例36人,自述牙龈出血病例28人,口腔异味病例16人。
2、实验方法:第一组使用实施例1的处方散剂进行治疗,第二组使用对比例1的处方散剂进行治疗,治疗时,取一包散剂,倒入100ml的水中,轻摇,完全溶散后,漱口,一天3-4次,疗程为7天。
3、实验结果:
第一组,口腔溃疡病例中,35人口腔溃疡面不再疼痛,溃疡面不再发红,其余3人疼痛减轻,有效率92.1%;牙龈出血病例中,24人不再出血,1人出血减轻,有效率96%;口腔异味病例中,17人均感觉异味消除,口腔清爽,有效率100%。
第二组,口腔溃疡病例中,31人口腔溃疡面不再疼痛,溃疡面不再发红,其余5人疼痛减轻,有效率86.1%;牙龈出血病例中,23人不再出血,5人出血减轻,有效率82.1%;口腔异味病例中,15人感觉异味消除,口腔清爽,1人口腔异味减轻,有效率93.7%。
抗氧化性对比实验
将薁磺酸钠分别与茶多酚1.5g、BHT 0.25g、维生素C 2.5g混合均匀(抗氧剂皆用其有效剂量的最大剂量),薁磺酸钠的量均为2.0g。
将以上混合物分别放在不同环境下进行测试:一、室温下(温度25℃±5℃,湿度60%±10%)放置30天;二、在4500±500lx强光照射10天;三、在75%±5%高湿条件下放置10天。最后观察样品颜色、薁磺酸钠含量变化,以考察其抗氧化效果。结果见表5,表5中含量为薁磺酸钠的检测含量占初始量的百分比。

由以上结果可知,茶多酚对薁磺酸钠制剂的保护作用是显而易见的,显著优于BHT和VC。

1.一种含有薁磺酸钠的漱口散剂,其特征在于,以质量百分比计,所述漱口散剂由以下组分的原料制备而成:薁磺酸钠0.1%~1%、茶多酚0.1%~1%、余量为辅料。
2.根据权利要求1所述的含有薁磺酸钠的漱口散剂,其特征在于,以质量百分比计,所述漱口散剂由以下组分的原料制备而成:薁磺酸钠0.1%~1%、茶多酚0.2%~0.5%、余量为辅料。
3.根据权利要求1所述的含有薁磺酸钠的漱口散剂,其特征在于,以质量百分比计,所述漱口散剂由以下组分的原料制备而成:薁磺酸钠0.4%、茶多酚0.2%~0.5%、余量为辅料。
4.根据权利要求1~3任一所述的含有薁磺酸钠的漱口散剂,其特征在于,所述辅料包括填充剂、崩解剂、pH调节剂、粘合剂以及调味剂。
5.根据权利要求4所述的含有薁磺酸钠的漱口散剂,其特征在于,所述填充剂选自甘露醇、乳糖、木糖醇或蔗糖中的至少一种;
所述崩解剂选自淀粉、羧甲淀粉钠、预胶化淀粉中的至少一种;
所述pH调节剂选自碳酸氢钠、磷酸氢二钠或磷酸二氢钠中的至少一种;
所述粘合剂选自聚乙烯醇、羟丙基纤维素或聚维酮中的至少一种;
所述调味剂包括薄荷醇和香精。
6.根据权利要求1~3任一所述的含有薁磺酸钠的漱口散剂,其特征在于,所述辅料包括润滑剂,所述润滑剂选自二氧化硅、硬脂酸或硬脂酸盐中的至少一种。
7.一种权利要求1~6任一所述含有薁磺酸钠的漱口散剂的制备方法,其特征在于,包括以下步骤:
(1)将薁磺酸钠和茶多酚原料混合均匀;
(2)将步骤(1)所得的混合物再与辅料通过等量递加法混合均匀,并过筛;
(3)将步骤(2)所得产品按剂量分装到铝塑袋内。
