Abstract
本发明涉及TLR3抑制剂在制备防治肿瘤转移产品中的应用。本发明具体涉及对模式识别受体TLR3具有抑制作用的物质在制备用于预防和/或治疗对象中肿瘤转移的产品中的应用、包含对模式识别受体TLR3具有抑制作用的物质的用于预防和/或治疗对象中肿瘤转移的产品、以及预防和/或治疗肿瘤转移的方法。本发明为肿瘤及其转移的预防和/或治疗提供了新的治疗靶点和途径。

技术领域
本发明涉及生物技术和医学领域,具体涉及模式识别受体TLR3的抑制剂在制备防治肿瘤转移产品中的应用在制备预防和/或治疗肿瘤转移产品中的应用。
背景技术
恶性肿瘤是严重危害国民健康的重大疾病之一,而且其发病率与死亡率仍持续攀升。目前研究发现,肿瘤的发生发展不仅仅与肿瘤细胞本身相关,肿瘤微环境作为肿瘤生存的重要土壤也发挥着极其重要的功能,成为肿瘤生物学研究以及寻找肿瘤治疗靶点的一大热点。
肿瘤微环境是由肿瘤细胞以及肿瘤相关细胞组成(主要是宿主细胞),包括血管内皮细胞、成纤维细胞、周细胞以及一些炎性细胞,如中性粒细胞、巨噬细胞、单核细胞、T细胞等(Hanahan,D.和Weinberg,R.A.,Hallmarks of cancer:the nextgeneration.Cell.2011;144:646-674)。
肿瘤生长过程中产生大量膜型和分泌型物质,诱导基质和相关炎性细胞功能转化,形成肿瘤的免疫微环境。越来越多的研究证实,肿瘤及远隔转移器官中的炎性细胞能通过与肿瘤细胞相互接触或分泌可溶性的细胞因子(如VEGF-α、IL-10、TGF-β等),改变肿瘤的侵袭及转移的能力,发挥肿瘤的负向免疫调控功能。因此,研究肿瘤免疫微环境的产生机制、诱导肿瘤相关免疫细胞的功能逆转从而打破抑制性肿瘤免疫微环境成为肿瘤免疫治疗学界关注的重点。
目前,针对免疫检查点(PD-1和CTLA4)抗体治疗有效的增强了微环境中T细胞对肿瘤的杀伤功能,成为近年来免疫治疗领域的重大突破(Topalian S.L.等,Immunecheckpoint blockade:a common denominator approach to cancer therapy.CancerCell.2015;27:450)。
肿瘤微环境包括肿瘤原位微环境、侵袭微环境以及转移微环境。肿瘤转移前微环境(Pre-metastatic niche)概念的提出为肿瘤转移尤其是转移器官选择性提供了新的理论解释。转移前微环境是指原发灶的肿瘤能够通过肿瘤来源的分泌物质(Tumor-derivedsecreted factors,TDSFs)动员和募集骨髓来源细胞(Bone marrow-derived cells,BMDCs)到转移靶器官,通过与靶器官中基质细胞及细胞外基质的协同作用,创造了利于肿瘤细胞定居的“土壤”(Kaplan,R.N.等.VEGFR1-positive haematopoietic bone marrowprogenitors initiate the pre-metastatic niche.Nature.2005,438:820-827)。多种BMDCs(VRGFR+骨髓前体细胞、CD11b+髓系细胞等)被证实参与了转移前微环境的形成。因此,研究肿瘤转移前微环境形成的影响因素对于肿瘤转移的预防和治疗至关重要。
目前,中性粒细胞被证实在转移前微环境中发挥了关键的促进作用。中性粒细胞是天然免疫系统中抵御病原体感染的重要成员,当炎症发生时,它们被迅速趋化至炎症部位,吞噬入侵的病原体和组织碎片,发挥抗感染和创伤修复的作用。在肿瘤转移前微环境中,中性粒细胞被大量募集到预转移器官。这些中性粒细胞发挥着抑制抗肿瘤免疫、促进肿瘤细胞存活而促进肿瘤转移前微环境的形成的功能。
目前,中性粒细胞所介导的肿瘤免疫抑制及促肿瘤功能在多种小鼠及临床肿瘤中得到验证(Wculek,S.K.和Malanchi,I.Neutrophils support lung colonization ofmetastasis-initiating breast cancer cells.Nature.2015;528:413-417)。因此,基于中性粒细胞及其影响因素的干预已成为肿瘤治疗新的生长点。
例如,肿瘤学顶级杂志Cancer Cell发表的最新研究显示抑制趋化因子CXCR2的表达能够显著抑制胰腺癌的转移与发展,其作用机制就是抑制了肿瘤微环境中中性粒细胞的募集(Steele C.W.等,CXCR2Inhibition Profoundly Suppresses Metastases andAugments Immunotherapy in Pancreatic Ductal Adenocarcinoma.Cancer Cell.2016;29:832)。
然而,转移靶器官如何识别肿瘤来源的刺激信号而募集中性粒细胞形成转移前微环境、中性粒细胞如何迁移到转移的靶器官尤其是肺脏中而促进微环境形成,这些问题的解决有利于寻找新的预测肿瘤转移及肿瘤治疗的靶点。
肺泡的上皮层主要由I型和II型上皮细胞组成,其在气体交换和表面张力维持方面发挥了重要作用。此外,肺上皮细胞也能够识别病原体和损伤相关的刺激信号,发挥免疫调节功能维持肺脏的稳态。肺上皮细胞表达多种模式识别受体(Pattern recognitionreceptors,PRRs),其激活后能够产生细胞因子和趋化因子等诱导免疫细胞在肺部的浸润。同样,肺上皮细胞的长期刺激导致的异常炎症反应也是肺部慢性炎症的产生因素,表明其在肿瘤发生中的潜在功能(Whitsett,J.A.和Alenghat,T.,Respiratory epithelialcells orchestrate pulmonary innate immunity.Nat.Immunol.,2015;16:27-35)。然而,作为肺泡的重要组成成分,目前尚未见报道肺上皮细胞对肿瘤肺转移的调节功能。
模式识别受体(PRRs)是机体识别外源性及内源性危险信号启动免疫应答的主要媒介。目前发现的PRR主要分为三类:TLRs(Toll-like receptors);RLHs(RIG-I-likehelicases);NLRs(Nucleotide-oligomerization domain-like receptors)。其中,Toll样受体(Toll-like receptors,TLRs)是最早发现且研究较为深入的一类模式识别受体。细菌或病毒成分如蛋白质、RNA等能够被免疫细胞的TLRs识别,进而活化下游信号途径,促使免疫细胞活化并表达促炎症细胞因子和干扰素,提高细胞的抗病原体能力,发挥清除病原体感染的作用。
TLRs是一种进化上高度保守的胚系编码的I型跨膜蛋白,由胞外区、跨膜区和胞内区组成。目前发现并克隆了十多种哺乳动物的TLRs分子,分别选择识别不同的病原体相关分子模式(Cao,X.,Self-regulation and cross-regulation of pattern-recognitionreceptor signalling in health and disease.Nat.Rev.Immunol.2016;16:35-50)。
TLR3是机体识别双链RNA(double-stranded RNA,dsRNA)的一种Toll样受体,其在抗病毒免疫反应中发挥着很重要的作用。TLR3的特异性配体是双链RNA,病毒在复制和转录过程中可产生大量dsRNA。TLR3和dsRNA结合后,可经由MyD88依赖和非依赖两条途径来诱导NF-κB的激活,促进其转位进入细胞核内和DNA片段结合,启动相应的炎性因子和细胞因子的转录和翻译,如IL-8、IL-12、TNFα、IFNα等,从而引起抗病毒免疫反应,抵抗病毒的感染。
除了感知双链RNA病毒感染,TLR还被报道参与识别了内源性的来自坏死或凋亡细胞的RNA,提示TLR3也参与了坏死诱导的炎症的触发(Cavassani,K.A.等,TLR3is anendogenous sensor of tissue necrosis during acute inflammatoryevents.J.Exp.Med.2008;205:2609-2621)。
在感染过程中,TLRs促发的炎症信号对于中性粒细胞向感染部位的迁移及趋化至关重要,但目前尚未见报道TLR3影响中性粒细胞在肿瘤内或转移前微环境中的浸润及迁移。
在肿瘤研究领域,Toll样受体的功能日益受到关注。肿瘤细胞高表达多种TLRs,其在肿瘤发生发展中发挥着双向调控功能。本领域中已知某些模式识别受体与肿瘤具有一定的相关性,如TLR4信号的活化促使胃癌恶化(X,Yuan等,Activation of TLR4signalingpromotes gastric cancer progression by inducing mitochondrial ROSproduction.Cell Death and Disease.2013;4,e794),帮助肺癌细胞实现免疫逃逸(Weigang,H.等,TLR4signaling promotes immune escape of human lung cancer cellsby inducing immunosuppressive cytokines and apoptosis resistance.MolecularImmunology.2007;44(11):2850-2859);TLR3在乳腺癌细胞中发挥促凋亡作用(Salaun B.等,TLR3can directly trigger apoptosis in human cancer cells.J Immunol.2006;176:4894-4901)。
宿主细胞表达的TLRs也日益受到关注,TLRs是宿主促肿瘤发生的慢性炎症的启动因素。宿主TLRs也影响了肿瘤的转移,肿瘤产生的物质能够激活宿主髓系细胞的TLR2而促进肿瘤的肺转移(Kim,S.等,Carcinoma-produced factors activate myeloid cellsthrough TLR2to stimulate metastasis.Nature.2009;457:102-106);肿瘤分泌的小RNA(microRNAs)能够结合TLR7/8促进肿瘤诱导的相关炎症而促进肿瘤转移(Fabbri,M.等,MicroRNAs bind to Toll-like receptors to induce prometastatic inflammatory response.Proc.Natl.Acad.Sci.USA.2012,109:E2110-2116)。
然而,由于肿瘤转移模型以及研究的靶细胞不同,宿主TLRs在肿瘤转移中功能报道往往不一致,这也提示着不同细胞以及不同肿瘤阶段TLRs功能的多样性。深入研究TLRs在宿主肿瘤微环境中发挥的功能有利于肿瘤治疗靶点的寻找。
由于癌症是危害人类健康的主要疾病之一,为了有效地治疗和预防肿瘤(如肺癌),目前人们已经越来越关注肿瘤转移及患者预后预判以指导肿瘤的治疗。用临床指标(如TNM分级)和单个分子指标(如NSE)预测肺癌进展、复发转移的研究已经有较长的历史。但是,临床指标或者病理类型相似的患者却有截然不同的临床结局,所以用临床指标或者单独的分子标志来预测或评价患者难以取得满意的效果。对肿瘤实行个体化、预见性的治疗有助于更加深入理解临床病理学特征,改善对患者的临床处理,从而提高肿瘤患者的无瘤生存期及绝对生存期。确定肺癌发展过程中的肿瘤细胞以及微环境中相关基因及其参与肺癌发病机理,可为肺癌个体化预见性治疗提供基础,也为新的治疗方案提供靶点,从而有利于提高肺癌的治愈率。
另一方面,肿瘤的发展包括转移不仅与肿瘤细胞本身有关,还与肿瘤所处的微环境(如上述的转移前微环境)密切相关。目前,针对肿瘤微环境中的免疫检查点(PD-1和CTLA4)抗体治疗有效的增强了微环境中T细胞对肿瘤的杀伤功能,成为近年来免疫治疗领域的重大突破(Topalian S.L.等,Immune checkpoint blockade:a common denominatorapproach to cancer therapy.Cancer Cell.2015;27:450)。
此外,针对中性粒细胞及其影响因素的干预已日益受到肿瘤治疗领域研究者的关注,并取得了良好的效果(Steele C.W.等,CXCR2Inhibition Profoundly SuppressesMetastases and Augments Immunotherapy in Pancreatic DuctalAdenocarcinoma.Cancer Cell.2016;29:832)。以上进展提示除了肿瘤细胞本身针对肿瘤微环境重要靶点的检测及干预治疗方法成为肿瘤诊断及治疗新的增长点。
目前,国内外尚无有关宿主模式识别受体TLR3与中性粒细胞迁移及肿瘤转移相关性的研究报道。
而本领域迫切需要寻找到可有效用于肿瘤诊断、肿瘤治疗方案选择、肿瘤预后评估的PRR分子,并将其用于这些用途中。
发明内容
本发明中发现了模式识别受体在预防和/或治疗肿瘤转移中的作用,并由此提供了可用于预防和/或治疗肿瘤转移的新途径、方法和产品。
本发明发现了对肿瘤转移前微环境中中性粒细胞迁移及趋化具有调控作用的肺上皮细胞表达的分子——模式识别受体TLR3,由此本发明进一步提供了对模式识别受体TLR3具有抑制作用的物质在预防和/或治疗肿瘤转移中的新用途,有由此提供了可用于预防和/或治疗肿瘤转移的新途径、方法和产品。
在本发明的一个方面中,提供了对模式识别受体TLR3具有抑制作用的物质在制备用于预防和/或治疗对象中肿瘤转移的产品中的应用。在与此相应的本发明的另一方面中,提供了一种预防和/或治疗对象中肿瘤转移的方法,所述方法包括给予所述对象对模式识别受体TLR3具有抑制作用的物质。
在一些实施方式中,所述物质选自:抑制TLR3表达和/或功能的物质,例如抗TLR3的抗体(优选单克隆抗体)、针对TLR3的干扰RNA(siRNA)、针对TLR3的反义寡核苷酸、TLR3表达或功能抑制化合物、用于敲除或敲减TLR3表达的物质。
在一些实施方式中,所述物质为针对TLR3的干扰RNA,例如针对TLR3基因mRNA不同位点选取的siRNA序列,优选为选自如下序列的siRNA:SEQ ID No.:1、SEQ ID No.:2、SEQID No.:3和SEQ ID No.:4。
在本发明的另一个方面中,提供了一种用于预防和/或治疗对象中肿瘤转移的产品,其包含对模式识别受体TLR3具有抑制作用的物质,所述物质为针对模式识别受体TLR3的干扰RNA,例如针对TLR3基因mRNA不同位点选取的siRNA序列(优选所述siRNA的长度为18~25bp,更优选21~23bp),优选为选自如下序列的siRNA:SEQ ID No.:1、SEQ ID No.:2、SEQ ID No.:3和SEQ ID No.:4。
在本发明中各方面的一些实施方式中,所述模式识别受体TLR3表达于所述对象的非肿瘤细胞中,优选肿瘤微环境中的非肿瘤细胞(或称宿主细胞),所述细胞选自:怀疑发生肿瘤转移组织中的细胞、肿瘤转移高风险组织中的细胞、已发生肿瘤转移组织中的细胞、已建立转移灶中的细胞或它们的组合,或者所述宿主细胞选自:肿瘤侵袭微环境中的细胞、转移微环境中的细胞、肿瘤转移前微环境中的细胞或它们的组合,优选表达于肺上皮细胞、内皮细胞、巨噬细胞、树突状细胞。
在本发明的一些实施方式中,所述对象为哺乳动物,例如灵长类动物、啮齿类动物、家畜、宠物等,优选人、大鼠、小鼠、犬、马、牛、兔或猴。
在本发明的一些实施方式中,所述模式识别受体TLR3具有Gene ID:7098,或为其同源序列。
在一些实施方式中,所述产品的形式适于选自下组的给予途径:经胃肠给予、非经胃肠给予,例如经静脉、黏膜、鼻腔、腹腔内、颅内、瘤内、舌下、颊部、透皮(如离子导入)等方式给予。例如可通过选自下组的途径给予:脂质体包裹DNA直接注射法、金包被DNA基因枪轰击法、繁殖缺陷细菌携带质粒DNA法以及复制缺陷腺病毒携带目的DNA法。

具体实施方式
本申请的发明人经长期而深入的研究发现模式识别受体TLR3能够启动肿瘤诱导的炎症反应,诱导中性粒细胞向肿瘤转移部位募集,促进了肿瘤转移前微环境的形成。在此基础上,发明人通过进一步的研究完成了本发明。
具体而言,通过细胞实验和动物实验,发明人发现:
1)利用肺癌LLC及黑色素瘤B16/F10自发肺转移的模型,在TLR3基因缺陷的小鼠中,小鼠肿瘤肺转移明显减少,生存期延长;
2)中性粒细胞是肺脏中促进肿瘤肺转移的重要因素,在TLR3基因缺陷的小鼠中,中性粒细胞向肺脏的迁移及趋化明显减少,血清及肺泡灌洗液中的趋化因子显著减低;
3)分析肺脏中TLR3表达发现,TLR3主要表达于肺上皮细胞中,肺上皮细胞TLR3缺陷后,其趋化因子的表达明显减少;
4)体外细胞实验证实,分别用si-TLR3 1、si-TLR3 2及si-TLR3 3、si-TLR34干扰人和小鼠肺上皮细胞系,能够明显抑制肿瘤诱导的肺上皮细胞趋化因子的表达;
5)本发明经临床样本研究发现,在非小细胞肺癌患者的癌旁组织中,TLR3的高表达与中性粒细胞的浸润增加显著相关,两者均与肺癌患者生存期缩短、预后差相关。
由此,本发明提供了TLR3分子在肿瘤转移的判断和抗肿瘤转移药物制备中的新用途,为模式识别受体的研究和开发利用提供了新的思路和途径;并且,本发明的TLR3分子可有效用于肿瘤转移的预判及治疗,从而为本领域提供一种新颖的肿瘤转移的诊断剂和/或治疗剂,具有一定的临床应用前景。
此外,针对促癌基因进行特定基因沉默是肿瘤治疗的热点之一,RNA干扰技术是目前高特异性的有效技术,因此无论是在功能基因组研究,还是肿瘤的基因治疗方面均具有广阔地应用前景。本发明分别针对人和小鼠的TLR3基因不同靶位点设计了2条干扰siRNA序列,用以抑制肺上皮细胞模式识别受体TLR3的表达。这些发明证实TLR3的siRNA可望成为预防和治疗肿瘤转移的有效手段。本发明的siRNA可以单独使用或几条siRNA联合,还可以与其它药物和治疗手段联合,用于恶性肿瘤转移的治疗。
如本文所用,术语“模式识别受体TLR3”或“Toll样受体3”或“TLR3”可互换使用,且具有本领域中已知的含义。例如,TLR3可为Gene ID:7098所示的序列,或可为其同源序列。TLR3可表达于对象宿主细胞中,例如表达于肺上皮细胞、内皮细胞、巨噬细胞、树突状细胞。
如本文所用,术语“宿主细胞”或“非肿瘤细胞”可互换使用,可包括肿瘤微环境中的非肿瘤细胞,例如包括但不限于:怀疑发生肿瘤转移组织中的细胞、肿瘤转移高风险组织中的细胞、已发生肿瘤转移组织中的细胞、已建立转移灶中的细胞或它们的组合,如肺上皮细胞、内皮细胞、巨噬细胞、树突状细胞。从另一角度而言,所述非肿瘤细胞选自:肿瘤侵袭微环境中的细胞、转移微环境中的细胞、肿瘤转移前微环境中的细胞或它们的组合。
“肿瘤微环境(tumor microenvironment,TME)”是指肿瘤存在的细胞环境,主要由肿瘤细胞以及肿瘤相关细胞(非肿瘤细胞)组成,其可包括周围血管、血管内皮细胞、成纤维细胞、上皮细胞、炎性细胞、免疫细胞等,肿瘤微环境包括肿瘤原位微环境、侵袭微环境以及转移微环境(可例如参见Hanahan,D.和Weinberg,R.A.,Hallmarks of cancer:the nextgeneration.Cell.2011;144:646-674)。肿瘤与其微环境密切相关并始终存在相互作用。
如本文所用,术语“对模式识别受体TLR3具有抑制作用的物质”或“TLR3抑制剂”或“本发明的活性物质”可互换使用,均是指可抑制TLR3水平和/或功能的物质。所述抑制可为部分抑制,即降低TLR3水平和/或功能,也可为完全抑制,即完全消除TLR3和/或其功能,所述抑制可为mRNA水平、DNA水平和/或蛋白质水平上的抑制。可用于本发明中的抑制剂包括但不限于,针对TLR3或其编码序列的:抗体(优选单克隆抗体)、siRNA、miRNA、反义寡核苷酸、拮抗剂、阻断剂、化学物质等。可采用本领域中已知的方法获得所述抑制剂或通过市售获得市售抑制剂,例如本领域普通技术人员可通过本领域已知杂交瘤法制备针对TLR3的单克隆抗体,并将其用于本发明中。
如本文所用,术语“产品”或“本发明的产品”可互换使用,是指包含对模式识别受体TLR3具有抑制作用的物质且用于预防和/或治疗对象中肿瘤转移的产品。所述产品包括但不限于:药物、保健品、试剂盒、医疗设备、或它们的组合。所述产品可选的还包含:药学上或保健品学上可接受的载体、辅料、容器、包装物、给予装置,指示给予所含对模式识别受体TLR3具有抑制作用的物质以预防和/或治疗对象中肿瘤转移的指示物(如说明书或使用手册等)。
本发明的产品中可含有有效量的本发明的TLR3抑制剂,以及可选的药学上、保健品学上或免疫学上可接受的载体。如本文所用,术语“可接受的”成分是适用于人和/或动物而无过度不良副反应(如毒性、刺激和变态反应)的,即有合理的效益/风险比的物质。如本文所用,术语“有效量”是指可对人和/或动物产生功能或活性的且可被人和/或动物所接受的量。
如本文所用,术语“载体”指用于向对象递送TLR3抑制剂的载体,包括各种赋形剂和稀释剂。该术语指这样一些药剂载体:它们本身并不是必要的活性成分,且施用后没有过分的毒性。合适的载体是本领域普通技术人员所熟知的。例如,在《雷明顿药物科学》(Remington’s Pharmaceutical Sciences,Mack Pub.Co.,N.J.1991)中可找到关于药学上可接受的赋形剂的充分讨论。
在组合物中可接受的载体可含有液体,如水、盐水、甘油和乙醇。另外,这些载体中还可能存在辅助性的物质,如填充剂、崩解剂、润滑剂、助流剂、泡腾剂、润湿剂或乳化剂、矫味剂、pH缓冲物质等。通常,可将这些物质配制于无毒的、惰性的和药学上可接受的水性载体介质中,其中pH通常约为5-8,较佳地,pH约为6-8。
应理解,所用本发明活性物质的有效剂量可随待施用或治疗的对象的严重程度而变化。具体情况根据对象的个体情况(例如对象体重、年龄、身体状况、所需达到的效果)来决定,这在熟练医师可以判断的范围内。

实施例
下面结合具体实施例,进一步阐述本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。本领域技术人员可对本发明做出适当的修改、变动,这些修改和变动都在本发明的范围之内。
下列实施例中未注明具体条件的实验方法,可采用本领域中的常规方法,例如参考《分子克隆实验指南》(第三版,纽约,冷泉港实验室出版社,New York:Cold SpringHarbor Laboratory Press,1989)或按照供应商所建议的条件。DNA的测序方法为本领域常规的方法,也可由商业公司提供测试。
除非另外说明,否则百分比和份数按重量计算。除非另行定义,文中所使用的所有专业与科学用语与本领域熟练人员所熟悉的意义相同。此外,任何与所记载内容相似或均等的方法及材料皆可应用于本发明方法中。文中所述的较佳实施方法与材料仅作示范之用。
实施例1:肺癌癌旁组织中TLR3的表达与中性粒细胞浸润相关
所用样品为2004年90例肺癌患者的组织切片(获自上海芯超生物科技有限公司),其肺癌组织类型均为非小细胞肺癌(NSCLC),患者在手术前未接受化疗及放疗。患者基本情况如表1所示。

免疫组化法:取小鼠肺组织,4%多聚甲醛固定过夜。石蜡包埋后切片。70℃烤片3小时。脱蜡置水后,利用枸橼酸进行抗原修复。血清37℃封闭40分钟。PBS洗3次。一抗孵育4℃孵育过夜。PBS洗3次。二抗孵育37℃孵育30分钟。PBS洗3次。辣根过氧化酶孵育37℃孵育20分钟。PBS洗3次。经DAB染色、苏木素染核后脱水、风干,中性树脂封片。送显微镜下观察。所用TLR3抗体,目录号ab13915;CD66b抗体,目录号ab197678,均购自美国abcam公司。
TLR3免疫组化评分:采用quick score(QS)的方法对TLR3蛋白的表达进行评分。首先,对阳性细胞的比例按1-6分进行评分(1=1~4%;2=5~19%;3=20~39%;4=40~59%;5=60~79%;和6=80~100%);其次,对阳性染色细胞的平均强度评分(0=无染色;1=弱,2=中间,和3=强染色);之后,用阳性细胞数比例与阳性平均强度相乘,得到0-18的数值。0-9的数值表示TLR3低表达,10-18数值表示TLR3高表达。
采用免疫组化方法,对该90例肺癌癌旁组织中TLR3及CD66b(反映中性粒细胞数量)蛋白的表达进行了分析,研究TLR3的表达与中性粒细胞浸润的相关性。P值使用SPSS17.0中的Unpaired Student’s t-tests(图1B)或Pearson correlation(图1C)计算。结果分别如图1A至图1C所示。
结果表明:肺癌癌旁组织中TLR3的高表达与中性粒细胞浸润数量多具有相关性。
实施例2:肺癌癌旁组织中TLR3的表达及中性粒细胞浸润与患者生存时间的相关性。
采用免疫组化方法,对该90例肺癌癌旁组织中TLR3及CD66b(反映中性粒细胞数量)蛋白的表达进行了分析,比较肺癌癌旁组织中TLR3的表达及中性粒细胞浸润与患者生存时间的相关性,P值使用SPSS 17.0中的log-rank test计算,结果分别如图2A和图2B所示。
图2A的结果表明:TLR3的高表达与患者更低的总体生存时间显著相关。图2B的结果表明:中性粒细胞浸润多与患者更低的总体生存时间显著相关。
上述结果表明:癌旁组织中TLR3的高表达与中性粒细胞浸润数量多相关,且两者均与患者较差的预后显著相关。该结果提示了癌旁组织中TLR3可能参与了中性粒细胞迁移及趋化的调控,并最终影响了肿瘤患者的预后,由此可作为肿瘤发展判断、抗肿瘤药物制备和/或预后评估的标志物。
实施例3:小鼠肺癌及黑色素瘤自发性肺转移模型中TLR3对肿瘤肺转移的调控。
TLR3缺陷小鼠购自美国Jackson实验室,在SPF级环境中饲养,通过杂交获得TLR3基因缺陷型小鼠及同窝野生型对照小鼠。本研究所使用的实验及对照小鼠均为C57BL/6背景、周龄为8-10周的雌性小鼠。
小鼠肺癌及黑色素瘤自发性肺转移模型的建立:每只小鼠背部皮下注射约1×106个肺癌细胞系LLC或黑色素瘤B16/F10细胞(细胞均购自美国PerkinElmer公司)。待肿瘤长至1cm2大小(18-20天)时,切除背部肿瘤。接种后40-45天时检测小鼠肺部转移情况。
体外肺组织活体成像:利用荧光素酶慢病毒载体FLuc-Puromycin(购自PerkinElmer公司,目录号CLS960002)感染LLC及B16/F10细胞,经嘌呤霉素筛选获得稳定表达荧光素酶的细胞克隆后进行小鼠皮下接种(接种细胞数为1×106/只),建立肿瘤自发转移模型。上机检测前给予小鼠D-Luciferin(15μg/g体重)腹腔注射,10分钟后取小鼠肺脏,置于小动物活体成像仪(PerkinElmer公司)中检测。
生存曲线分析采用Kaplan and Meier分析方法作图,计算P值,P<0.05时,认为两组之间的差别具有统计学意义。
该结果表明:TLR3缺陷后小鼠自发性肺转移极显著减少(图3A),小鼠生存时间明显延长(图3B),提示宿主细胞表达TLR3具有促进肿瘤肺转移的功能。
实施例4:宿主的TLR3影响转移前微环境中中性粒细胞向肺组织的迁移及趋化。
肺组织单细胞悬液的制备:取小鼠肺组织(同实施例3处理后取组织),用剪刀将其剪碎,在含有Ⅳ型胶原酶(1mg/ml)、DNA酶I(40mg/ml)的RPMI 1640培养基中消化60分钟,每10分钟用巴氏吸管轻轻吹打悬液,直到没有可见的团块。然后通过40μm的网格,洗涤两次,并再悬浮于RPMI 1640培养基中。
流式细胞术:收集细胞,用预冷的流式专用PBS(0.1%NaN3+0.5%BSA+2mM EDTA,pH7.2)洗1遍(500g离心5分钟),再用PBS 100μl重悬,加入抗CD16/CD32抗体(1μg/ml,购自美国BD Pharmingen公司)以消除非特异性结合,分别加入各种荧光素标记的抗体以及相应的同型对照抗体等(Ly6G抗体,目录号12-5931;Ly6C抗体,目录号53-5932,均购自美国BDPharmingen公司),终浓度均为1μg/ml,混匀后置4℃,30分钟,加入500μl PBS洗2遍,加入200μl PBS。
利用ELISA试剂盒检测小鼠血清及肺泡灌洗液中趋化因子的表达(详见趋化因子ELISA说明书,所有趋化因子ELISA检测试剂盒均购自美国R&D公司)。
结果显示:流式细胞术检测小鼠肺部浸润的中性粒细胞显示,随着荷瘤时间的延长,肺组织中的中性粒细胞数明显增多,但TLR3缺陷小鼠荷瘤后肺部中性粒细胞的浸润较正常对照荷瘤小鼠显著减少(图4A)。
ELISA检测小鼠血清及肺泡灌洗液中中性粒细胞相关趋化因子发现,TLR3缺陷小鼠趋化因子的分泌较正常对照小鼠明显减少(图4B)。
上述结果提示:宿主细胞TLR3在小鼠荷瘤后通过促进趋化因子分泌诱导中性粒细胞在预转移的肺组织中聚集。
实施例5:TLR3在肺组织中的定位及肿瘤诱导肺上皮细胞TLR3的表达
组织免疫荧光法:取小鼠肺组织(同实施例3处理后取组织),用OCT胶包埋于塑料小盒中,-80℃冷冻保存。冰冻切片机6微米切片,室温放置20分钟。选择一抗种属来源的血清室温封闭1小时。一抗孵育12小时。PBS洗3次。孵育第二个一抗,室温孵育1~2小时。PBS洗3次。二抗孵育2小时,室温。PBS洗3次。DAPI染色15分钟。PBS洗3次。盖上盖玻片,送激光共聚焦显微镜下观察。所用抗体如下文中所述。
利用免疫荧光的方法对TLR3在肺组织中的定位进行研究,对肺组织切片分别标记TLR3、Sftpd(肺上皮标记)、Ly6G(中性粒细胞标记)(TLR3抗体,目录号ab13915;Sftpd抗体,目录号ab17781;Ly6G抗体,目录号ab25377,均购自美国abcam公司),观察TLR3与Sftpd及Ly6G的共定位情况。
结果显示:TLR3与Sftpd具有较好的共定位,而与Ly6G无共定位,提示TLR3主要在肺组织的肺上皮细胞中表达,而在中性粒细胞中不表达(图5A)。
采用TRIzol(Invitrogen公司)抽提肺组织总RNA。qRT-PCR检测TLR3基因的相对定量使用2-ΔΔCt法计算(β-actin为内参)(Livak,KJ.等,Analysis of relative geneexpression data using real-time quantitative PCR and the 2-ΔΔCtmethod.Methods.2001;25:402-408)。
结果显示:肿瘤能够极显著上调肺上皮细胞中TLR3的表达以及趋化因子的表达(图5B)。
该结果表明:肿瘤诱导的趋化因子分泌及中性粒细胞的浸润可能是通过上调肺上皮细胞TLR3表达,提示肺上皮细胞TLR3的表达参与了中性粒细胞募集及肿瘤肺转移的调控。
实施例6:肺上皮细胞TLR3缺陷或干扰RNA干扰TLR3表达后中性粒细胞相关趋化因子的表达。
分选TLR3缺陷小鼠以及正常对照小鼠荷瘤14天肺上皮细胞(如实施例3处理),采用TRIzol(Invitrogen公司)抽提细胞总RNA,qRT-PCR法检测细胞趋化因子(CXCL1、CXCL2、CXCL5、CXCL12)的基因表达。
体内结果(图6A)显示:TLR3缺陷后肺上皮细胞表达趋化因子的能力下降。
培养小鼠肺上皮细胞细胞系MLE-12及人肺上皮细胞细胞系A549(购自ATCC),细胞接种于含10%的小牛血清的RPMI 1640(Gibco BRL公司产品)培养液,置于37摄氏度、体积分数为5%的CO2培养箱中常规培养。
优化转染条件,利用si-Tlr3l(SEQ ID No.:1)、si-Tlr3 2(SEQ ID No.:2)干扰人肺上皮细胞细胞系A549;利用si-Tlr3 3(SEQ ID No.:3)、si-Tlr3 4(SEQ ID No.:4)干扰小鼠肺上皮细胞细胞系MLE-12的TLR3的表达,并以si-Tlr3neg1(SEQ ID No.:5)和si-Tlr3neg2(SEQ ID No.:6)为阴性对照,各siRNA以50nmol的终浓度加入细胞培养液,于孵育后24~48h利用肿瘤培养上清刺激以后,采用TRIzol(Invitrogen公司)抽提细胞总RNA,qRT-PCR法检测细胞趋化因子(CXCL1、CXCL2、CXCL5、CXCL12)的基因表达。
各siRNA的序列如下所示:
针对人TLR3基因mRNA不同位点选取的2段21个碱基的siRNA序列:
SEQ ID No.1:5’-GAGGUCUUCAAGGAUUUAUTT-3’
SEQ ID No.2:5’-GUCCCAUUUAUUUCCUAAATT-3’
针对小鼠TLR3基因mRNA不同位点选取的2段21个碱基的siRNA序列:
SEQ ID No.3:5’-GGAGAGGUCUGAGAAAUAUTT-3’
SEQ ID No.4:5’-CGGCCUUAAUGAAAUUGAATT-3’
作为阴性对照的随机序列:
SEQ ID No.5:5’-UUCUCCGAACGUGUCACGUdTdT-3’
SEQ ID No.6:5’-ACGUGACACGUUCGGAGAAdTdT-3’。
体外RNA干扰结果(图6B与6C)显示:干扰RNA干扰Tlr3表达后能明显减低肿瘤诱导的肺上皮细胞趋化因子的表达。
该结果表明:肺上皮细胞的TLR3能够调控其趋化因子的分泌,结合实施例3-5的结果,提示肺上皮细胞的TLR3可以作为肿瘤治疗的靶点,利用干扰RNA技术干扰其表达后能够抑制中性粒细胞在转移部位迁移及趋化,从而抑制肿瘤的转移,达到预防和治疗肿瘤转移的目的。

Claims (31)
1.对模式识别受体TLR3具有抑制作用的物质在制备用于预防和/或治疗对象中肿瘤转移的产品中的应用,其中所述肿瘤为肺癌。
2.如权利要求1所述的应用,其中,所述物质为抑制TLR3表达和/或功能的物质。
3.如权利要求1所述的应用,其中,所述物质为用于敲除或敲减TLR3表达的物质。
4.如权利要求1所述的应用,其中,所述物质选自:抗TLR3的抗体、针对TLR3的干扰RNA(siRNA)、针对TLR3的反义寡核苷酸。
5.如权利要求1所述的应用,其中,所述物质为抗TLR3的单克隆抗体。
6.如权利要求1所述的应用,其中,所述物质为选自如下序列的siRNA:SEQ ID No.:1、SEQ ID No.:2、SEQ ID No.:3和SEQ ID No.:4。
7.如权利要求1所述的应用,其中,所述模式识别受体TLR3表达于对象的非肿瘤细胞。
8.如权利要求1所述的应用,其中,所述模式识别受体TLR3表达于肿瘤微环境中的非肿瘤细胞。
9.如权利要求1所述的应用,其中,所述模式识别受体TLR3表达于选自下组的细胞:怀疑发生肿瘤转移组织中的细胞、肿瘤转移高风险组织中的细胞、已发生肿瘤转移组织中的细胞、已建立转移灶中的细胞或它们的组合。
10.如权利要求1所述的应用,其中,所述模式识别受体TLR3表达于肺上皮细胞、内皮细胞、巨噬细胞和/或树突状细胞。
11.如权利要求1所述的应用,其中,所述对象为哺乳动物。
12.如权利要求1所述的应用,其中,所述对象选自:灵长类动物、啮齿类动物、家畜、宠物。
13.如权利要求1所述的应用,其中,所述对象选自:人、大鼠、小鼠、犬、马、牛、兔或猴。
14.如权利要求1所述的应用,其中,所述物质递送后靶向包含模式识别受体TLR3的宿主细胞。
15.如权利要求1所述的应用,其中,所述产品选自:药物、保健品、试剂盒、医疗设备、或它们的组合。
16.一种用于预防和/或治疗对象中肿瘤转移的产品,其包含对模式识别受体TLR3具有抑制作用的物质,其中所述肿瘤为肺癌。
17.如权利要求16所述的产品,其中,所述物质为抑制TLR3表达和/或功能的物质。
18.如权利要求16所述的产品,其中,所述物质为用于敲除或敲减TLR3表达的物质。
19.如权利要求16所述的产品,其中,所述物质选自:抗TLR3的抗体、针对TLR3的干扰RNA(siRNA)、针对TLR3的反义寡核苷酸。
20.如权利要求16所述的产品,其中,所述物质为针对模式识别受体TLR3的干扰RNA。
21.如权利要求16所述的产品,其中,所述物质为选自如下序列的siRNA:SEQ ID No.:1、SEQ ID No.:2、SEQ ID No.:3和SEQ ID No.:4。
22.如权利要求16所述的产品,其中,所述产品的形式适于选自下组的给予途径:脂质体包裹DNA直接注射法、金包被DNA基因枪轰击法、繁殖缺陷细菌携带质粒DNA法以及复制缺陷腺病毒携带目的DNA法。
23.如权利要求16所述的产品,其中,所述模式识别受体TLR3表达于对象的非肿瘤细胞。
24.如权利要求16所述的产品,其中,所述模式识别受体TLR3表达于肿瘤微环境中的非肿瘤细胞。
25.如权利要求16所述的产品,其中,所述模式识别受体TLR3表达于选自下组的细胞:怀疑发生肿瘤转移组织中的细胞、肿瘤转移高风险组织中的细胞、已发生肿瘤转移组织中的细胞、已建立转移灶中的细胞或它们的组合。
26.如权利要求16所述的产品,其中,所述模式识别受体TLR3表达于肺上皮细胞、内皮细胞、巨噬细胞和/或树突状细胞。
