本发明公开了一种铁皮石斛合剂,铁皮石斛合剂由以下重量份的中药材为原料制成的铁皮石斛10‑20份,生白术15‑25份,玄参10‑20份,麦冬10‑20份,生地10‑20份。本发明具有增液润燥,滋阴清热,健脾益气的功效,用于便秘,症见排便次数减少,粪便量少质硬,便后没有畅快感,排便时间或排便间隔时间延长等。为了更好地证明本发明对便秘的良好效果,我们通过动物实验考察了本合剂对复方地芬诺酯诱导的便秘小鼠的作用,结果显示本合剂可促进便秘小鼠小肠推进,促进排便,保护肠粘膜,提高血清SOD。

技术领域
本发明涉及一种铁皮石斛合剂,具体地说,是一种缓解便秘的铁皮石斛合剂。
背景技术
便秘是一种可由多因素诱导的症状,主要表现为排便次/量减少或排便不畅等,临床发病率较高,也是一种较为常见的临床症状,严重影响人们的生活质量。便秘会引起肠神经节膜面积减少及肠神经递质异常,引发结肠病变,严重便秘可发展为肠梗阻并危及生命。由于便秘易发展为慢性,患者多选择简便的导泄药物疗法,而峻泻剂刺激性大,缓泻剂长期使用亦有副作用。导泄药物长期的攻伐会导致气虚阳衰,气虚则大肠传导无力,反而会加重便秘症状,介时通过灌肠甚至手术治疗,雪上加霜,增加病人痛苦。
本铁皮石斛合剂在古代名方增液汤(玄参、生地、麦冬配伍)的基础上增加了补脾益胃的铁皮石斛及健脾补气的生白术,因此,铁皮石斛合剂除了包含增液汤的增液行舟、润肠通便之效,还具有一定的补虚作用,相对于一般导泻药的治标不治本有一定优势。
发明内容
本发明的目的在于提供一种针对便秘的铁皮石斛合剂,其效果显著,可增加复方地芬诺酯诱导的便秘小鼠的小肠推进,促进排便,保护肠粘膜,提高血清SOD,可起到缓解便秘的效果。
本发明是通过以下技术方案实现的:
本发明公开了一种铁皮石斛合剂,所述的铁皮石斛合剂由以下重量份的中药材为原料制成的:
铁皮石斛10-20份,生白术15-25份,玄参10-20份,麦冬10-20份,生地10-20份。
作为进一步地改进,本发明所述的铁皮石斛合剂由以下重量份的中药材为原料制成的:
铁皮石斛15份,生白术20份,玄参15份,麦冬15份,生地15份。
本发明还公开了一种铁皮石斛合剂的制备方法,具体制备工艺如下:
1)、将铁皮石斛、生白术、玄参、麦冬及生地混合,置于不锈钢锅内,加水800ml浸泡半小时,备用;
2)、沸水煎煮60min后将药液移至烧杯,加水800ml再次煎煮30min;
3)、两次药液合并,用旋转蒸发仪浓缩至100ml,即得铁皮石斛合剂。
作为进一步地改进,本发明所述的旋转蒸发仪的转速70rpm,温度60℃。
本发明的有益效果如下:
本发明的缓解便秘的铁皮石斛合剂,在古代名方增液汤(玄参、生地、麦冬配伍)的基础上增加了铁皮石斛及生白术。铁皮石斛(Dendrobium officinale Kimura et Migo)为兰科石斛属多年生草本植物,是滋阴清热、益胃生津的补阴类药材,具有主伤中、除弊、下气、补五脏虚劳羸弱、强阴、久服厚肠胃等功效,对改善胃肠道运动,缓解便秘症状具有一定作用。生白术较白术而言较为甘润,升清降浊,其主要成分苍术酮可与胆碱能受体相作用而促进平滑肌收缩来加强胃肠道运动,通便作用较强。另外,增液汤中玄参质润多液,滋阴润燥,苦泄滑肠而通便;生地凉血润燥,壮水生津而通便;麦冬,滋养肺胃阴津以润肠燥,养阴增液而通便。
总的来说,增液汤增液润燥作用较为单一,由于便秘患者长期使用导泻药易形成医源性的习惯性便秘,患者大多气虚津亏血燥,中医辨证属“虚秘”范畴,因此,增液汤加入铁皮石斛及生白术可补脾益气,调理脾胃,加强增液汤增液行舟的通便疗效,同时气阴双补,且生白术可防止全方苦泄寒滑过甚,起到平衡全方药性寒温的作用,本方适用于久病体虚的慢性便秘患者,长期服用安全性较高。
经过常规的水煎液浓缩工艺制成的,具有增液润燥,滋阴清热,健脾益气的功效,用于便秘,症见排便次数减少,粪便量少质硬,便后没有畅快感,排便时间或排便间隔时间延长等。为了更好地证明本发明对便秘的良好效果,我们通过动物实验考察了本合剂对复方地芬诺酯诱导的便秘小鼠的作用,结果显示本合剂可促进便秘小鼠小肠推进,促进排便,保护肠粘膜,提高血清SOD。
具体实施方式
下面结合具体实施例对本发明的技术方案作进一步地说明。
1方法
1.1药液的配制
铁皮石斛合剂:将铁皮石斛、生白术、玄参、麦冬及生地混合,置于不锈钢锅内,加水800ml浸泡半小时,沸水煎煮60min后将药液移至烧杯,加水800ml再次煎煮30min,两次药液合并,用旋转蒸发仪(转速70rpm,温度60℃)浓缩至100ml,即得铁皮石斛合剂。
生白术+增液汤:生白术20g,玄参15g,麦冬15g,生地15g,加水650mL,浸泡半小时,如上煎煮两次合并,浓缩,定容至100mL,冷藏备用。
增液汤:玄参15g,麦冬15g,生地15g,加水450mL,浸泡半小时,如上煎煮两次合并,浓缩,定容至100mL,冷藏备用。
麻仁胶囊去包衣,加水溶解,定容至500mL,超声溶解45min,4℃保存。
复方地芬诺酯片,研钵研磨成极细粉末,蒸馏水配制后浓缩为2.5g/L的悬浊液,置冰箱备用。
1.2对抗药物性便秘实验
将昆明种小白鼠60只单色涂染法编号,通过随机数字法分成6组,每组10只,设空白对照组、便秘模型组(即复方地芬诺酯组)、阳性组(麻仁胶囊组)、铁皮石斛合剂组、生白术+增液汤组及增液汤组,采取灌胃法进行研究。随机将同组同性小鼠5只并入1笼(所有观测指标均按照笼中小鼠只数进行平均处理)。自由饮水进食,小鼠自由饮食连续饲养7天后,各组小鼠禁食24h,称质量,除空白对照组外,其余各组灌胃复方地芬诺酯50mg/kg,建立小鼠便秘模型。1h后各组小鼠称量体重并计算给药量,灌胃给药,麻仁丸组给药0.40mL/只,铁皮石斛合剂组给药0.37mL/只,生白术+增液汤组给药0.37mL/只,增液汤组给药0.40mL/只,空白对照组及模型组给生理盐水0.40mL/只,20min后给各组小鼠灌胃墨汁0.30mL/只,给药后的小鼠置于笼架上,笼内铺滤纸,进行观察,记录小鼠首次排便时间、6h内排便总质量和排便粒数,并观察粪便的形状和质地。
1.2小肠推进率影响实验
各组小鼠称量体重并计算给药量,灌服相应受试物,麻仁丸组给药0.40mL/只,铁皮石斛合剂组给药0.37mL/只,生白术+增液汤组给药0.37mL/只,增液汤组给药0.40mL/只空白组、模型组等体积生理盐水灌胃,15min后灌服墨汁0.30mL/只,给药20min后,脱颈椎处死小鼠,打开腹腔,剪取幽门至盲段的小肠,置于水平光滑面上,轻轻将小肠拉成直线,测量自幽门端至炭沫最前端和盲肠端的长度,以二者之百分比为小肠推进率。墨汁推进率=墨汁推进长度/小肠总长度*100%
1.3血清SOD测量
各组小鼠称量体重并计算给药量,灌服相应受试物,麻仁丸组给药0.40mL/只,铁皮石斛合剂组给药0.37mL/只,生白术+增液汤组给药0.37mL/只,增液汤组给药0.40mL/只空白组、模型组等体积生理盐水灌胃,24后再次给药。15min后眼眶静脉丛取血,离心15min后分离血清,根据试剂盒说明书检测血清SOD活力。
2实验结果
2.1铁皮石斛合剂对便秘小鼠排便的影响
结果见表1。比较模型组与空白组,模型组首次排便时间更长,6h内排便粒数以及含水量亦有较大差异,小鼠便秘模型建立成功。阳性对照组及水煎液实验组与模型组相比,首次排便时间、排便粒数和排便质量均有较大差异,含铁皮石斛的铁皮石斛合剂组首次排黑便所需时间明显减少,作用优于麻仁胶囊。

2.2铁皮石斛合剂对便秘小鼠小肠蠕动的影响
结果见表2。相较模型组与空白组的墨汁推进率,差异显著(P<0.01),模型建立成功。结果显示含有铁皮石斛的水煎液组小肠推进率明显高于其他组别,经数据统计分析,与模型组相较有显著差异(P<0.01),此外,生白术+增液汤组与麻仁胶囊组亦与模型组相较均有显著差异(P<0.01),表现出对小鼠小肠蠕动的良好的促进作用。

2.3铁皮石斛合剂对便秘小鼠血清SOD活性的影响
结果见表3。与模型组相较,麻仁胶囊组、铁皮石斛合剂组及增液汤组对便秘小鼠血清的SOD均具有明显的增强作用,而铁皮石斛合剂组作用尤为显著,结果表明铁皮石斛合剂对便秘小鼠的肠粘膜具有良好的保护作用。

3.结论
实验证明,铁皮石斛合剂利于复方地芬诺酯诱导的便秘小鼠的胃肠蠕动,促进排便,疗效显著,亦可提高血清SOD,且相对于临床常用的泻剂,该复方对于小肠粘膜和胃肠道的刺激性小,兼有补虚作用,由此,该方在针对便秘方面有良好的应用前景,有利于提高患者的顺应性。
最后,还需要注意的是,以上列举的仅是本发明的具体实施例。显然,本发明不限于以上实施例,还可以有许多变形。本领域的普通技术人员能从本发明公开的内容直接导出或联想到的所有变形,均应认为是本发明的保护范围。
Claims (4)
1.一种铁皮石斛合剂,其特征在于,所述的铁皮石斛合剂由以下重量份的中药材为原料制成的:
铁皮石斛10-20份,生白术15-25份,玄参10-20份,麦冬10-20份,生地10-20份。
2.根据权利要求1所述的铁皮石斛合剂,其特征在于,所述的铁皮石斛合剂由以下重量份的中药材为原料制成的:
铁皮石斛15份,生白术20份,玄参15份,麦冬15份,生地15份。
3.根据权利要求1或2所述的铁皮石斛合剂的制备方法,其特征在于,具体制备工艺如下:
1)、将铁皮石斛、生白术、玄参、麦冬及生地混合,置于不锈钢锅内,加水800ml浸泡半小时,备用;
2)、沸水煎煮60min后将药液移至烧杯,加水800ml再次煎煮30min;
3)、两次药液合并,用旋转蒸发仪浓缩至100ml,即得铁皮石斛合剂。
4.根据权利要求3所述的铁皮石斛合剂的制备方法,其特征在于,所述的旋转蒸发仪的转速70rpm,温度60℃。
