1、提取液液的制备
1)取赤芍30~50g,马钱子40~60g,红花30~40g,透骨草40~50g,淫羊藿40~50g,三七20~30g,黄芪30~40g,防风20~40g,泽兰20~30g,杜仲20~30g,当归20~30g,骨碎补10~20g,血竭10~20g,混匀粉碎后,加入成分重量5~10倍的50~80%乙醇溶液在常温下浸泡3~5天,过滤取液,再将提取液旋转蒸发浓缩至含水量为5%~10%的浓缩液。
2)称取薄荷40~60g,放入三口烧瓶中以水蒸气蒸馏法提取1h,对馏出液离心,取上层油液,得薄荷挥发油。
2、复方中药固体脂质纳米粒的制备:
1)取浓缩液,以200~1000ml纯化水及10~50ml聚山梨酯-80溶解,超声分散30分钟,加热至65~75℃保温备用,制得水相。
2)按1:1~3:1的比例称取大豆磷脂和单硬脂酸甘油酯混合物10~30克,以200~500ml比例为4:1(体积比)的乙醇/二氯甲烷混合物为溶剂溶解,加入中药薄荷挥发油继续搅拌,超声溶解分散15分钟,制得油相。
3)将油相注入正在搅拌且水浴保温的水相中,搅拌速度1500~2000rpm,形成乳白色混悬液。继续保温搅拌蒸发有机溶剂后,超声分散30分钟,得脂质纳米粒混悬液粗品。
4)将上述脂质纳米粒混悬液粗品注入高压均质机,在冷水(2~6℃)循环水浴中对脂质纳米粒混悬液粗品进行高压均质,设定工作压力为2000Bar,均质乳匀浓缩后,得复方中药固体脂质纳米粒。
3、复方中药微针透皮给药贴的制备:
采用激光-蚀刻法将多孔硅、多孔钛或多孔聚苯乙烯基板材料的一面切削成微针阵列,以0.5*0.5cm大小制成多孔微针单元;将所述微针单元放置在所述复方中药固体脂质纳米粒中,经过包被、填充,取出晾干后,将所述微针单元固定于药膏空白贴的内表面,再覆上药膏背贴即得复方中药微针透皮给药贴。

其中,基板的正面经过激光-蚀刻法后在其表面上形成许多微针,这些微针在基板上形成一组矩形状微针阵列即微针单元,微针单元为多个,微针单元的长和宽都为0.5cm,相邻微针阵列之间的间距为0.1cm。
其中,基板材料可选用多孔硅、多孔钛或多孔聚苯乙烯中的一种或几种的复合材料。
本发明将固体脂质纳米粒技术和微针透皮技术结合运用于经皮给药贴剂,通过将难溶性成分制成固体脂质纳米粒,提高载药效率,多孔微针还具备类似多孔微球的缓释效果,可以避免患者重复给药,延长药效。与现有技术相比具有如下优点,以中药为基本原料,应用现代技术开发出新一代外用皮下给药贴,将药物有效组分直接作用于皮下,通过微针透皮给药,具有治疗效果好、其见效快,对人体皮肤几乎无损伤,无毒副作用特点,是中医药发展的方向。

具体实施方式
下面对本发明的实施例作详细说明,本实施例在以本发明技术方案为前提下进行实施,给出了详细的实施方式和具体的操作过程,但本发明的保护范围不限于下述的实施例。本发明包括:药膏空白贴内表面1、基板2、药膏背贴3、药剂、微针阵列间隙4、尖刺部5、微针6和微针间隙7;
实施例1
如图1~4所示,基板2位于药膏空白贴与药膏背贴的中间,基板的正面经过激光-蚀刻法后在其表面上形成许多微针6,这些微针6在基板2上形成一组矩形状微针阵列。基板2采用具有多孔结构的多孔硅材料,,微针6的顶端设有尖刺部5,微针6垂直固定在基板2上,微针6之间具有微针间隙7,药剂存储在微针间隙7和多孔结构之中,基板2的背面为平面,基板2的背面粘贴在药膏空白贴的内表面1上,药膏背贴3的内表面覆盖在基板2的正面上,药膏空白贴的内表面1与药膏背贴的内表面通过黏贴的方式进行固定连接,微针阵列之间设有微针阵列间隙4,相邻微针阵列的间距为0.1cm,微针阵列的大小为0.5*0.5cm,布阵范围2*2cm。
实施例2
本实施例中,基板2采用的材料为多孔聚苯乙烯,基板2具有多孔结构,基板的正面经过激光-蚀刻法后在其表面上形成许多微针6,这些微针6在基板2上形成一组矩形状微针阵列,微针阵列是由多个微针6按顺序排列而成,微针6的顶端设有尖刺部5,微针阵列垂直固定在基板2上,微针6之间具有微针间隙7,药剂存储在微针间隙7和多孔结构之中,基板2的背面为平面,基板2的背面粘贴在药膏空白贴内表面1上,药膏背贴3覆盖在基板2的正面上,药膏空白贴内表面1和药膏背贴3的大小和形状相适应并且相对平行设置,微针阵列之间设有微针阵列间隙4,相邻微针阵列的间距为0.1cm,微针阵列的大小为0.5*0.5cm,布阵范围为4*4cm,为了将药贴粘贴紧密,可在药膏空白贴内表面1的四周留有1cm大小的粘贴区,这样能够提高药贴的密封效果,防止因为四周沾染灰尘导致粘贴性不强。
实施例3
实施例3中,基板2采用的材料为多孔钛材料,基板2具有多孔结构,基板2的正面设有微针阵列,微针阵列是由多个微针6按顺序排列而成,微针6的顶端设有尖刺部5,尖刺部5的大小可根据实际需要调整激光-蚀刻的精度所实现,微针阵列垂直固定在基板2上,微针6之间具有微针间隙7,药剂存储在微针间隙7和多孔结构之中,基板2的背面为平面,基板2的背面粘贴在药膏空白贴的内表面1上,药膏背贴3的内表面覆盖在基板2的正面上,药膏空白贴的内表面1与药膏背贴的内表面通过黏贴的方式进行固定连接,微针阵列之间设有微针阵列间隙4,相邻微针阵列的间距为0.1cm,微针阵列的大小为0.5*0.5cm,布阵范围为4*4cm。
实施例4
1)称取赤芍30g,马钱子40g,红花30g,透骨草40g,淫羊藿40g,三七20g,黄芪30g,防风20g,泽兰20g,杜仲20g,当归20g,骨碎补10g,血竭10g,混匀粉碎后,加入成分重量5倍的50%乙醇溶液在常温浸泡5天,过滤取液,再将提取液旋转蒸发浓缩至含水量为5%的浓缩液.
2)称取薄荷40g放入三口烧瓶中,以水蒸气蒸馏法提取1h,对馏出液离心,取上层油液得薄荷挥发油。
3)取浓缩液,以500ml纯化水及20ml聚山梨酯-80溶解,超声分散30分钟,加热至65℃保温备用,制得水相。
4)按1:1的比例称取大豆磷脂和单硬脂酸甘油酯混合物20克,以300ml比例为4:1(体积比)的乙醇/二氯甲烷混合物为溶剂溶解,加入中药薄荷挥发油继续搅拌,超声溶解分散15分钟,制得油相。
5)将油相注入正在搅拌且水浴保温的水相中,搅拌速度1500rpm,形成乳白色混悬液。继续保温搅拌蒸发有机溶剂后,超声分散30分钟,得脂质纳米粒混悬液粗品。
6)将上述脂质纳米粒混悬液粗品注入高压均质机,在2℃冷水循环水浴中对脂质纳米粒混悬液粗品进行高压均质,设定工作压力为2000Bar,均质乳匀浓缩后,得复方中药固体脂质纳米粒。
7)采用激光-蚀刻法将多孔硅基板材料的一面切削成微针阵列,以0.5*0.5cm大小制成多孔微针单元;将所述微针单元放置在所述复方中药固体脂质纳米粒中,经过包被、填充,取出晾干后,将所述微针单元固定于药膏空白贴的内表面,再覆上药膏背贴即得治疗骨折软组织损伤中药微针透皮给药贴。
实施例5
1)取赤芍50g,马钱子60g,红花40g,透骨草50g,淫羊藿50g,三七30g,黄芪40g,防风40g,泽兰30g,杜仲30g,当归30g,骨碎补20g,血竭20g,混匀粉碎后,加入成分重量10倍的80%乙醇溶液在常温浸泡3天,过滤取液,再将提取液旋转蒸发浓缩至含水量为10%的浓缩液.
2)称取薄荷60g放入三口烧瓶中,以水蒸气蒸馏法提取1h,对馏出液离心,取上层油液得薄荷挥发油。
3)取浓缩液,以1000ml纯化水及50ml聚山梨酯-80溶解,超声分散30分钟,加热至75℃保温备用,制得水相。
4)按3:1的比例称取大豆磷脂和单硬脂酸甘油酯混合物30克,以500ml比例为4:1(体积比)的乙醇/二氯甲烷混合物为溶剂溶解,加入中药薄荷挥发油继续搅拌,超声溶解分散15分钟,制得油相。
5)将油相注入正在搅拌且水浴保温的水相中,搅拌速度2000rpm,形成乳白色混悬液。继续保温搅拌蒸发有机溶剂后,超声分散30分钟,得脂质纳米粒混悬液粗品。
6)将上述脂质纳米粒混悬液粗品注入高压均质机,在6℃冷水循环水浴中对脂质纳米粒混悬液粗品进行高压均质,设定工作压力为2000Bar,均质乳匀浓缩后,得复方中药固体脂质纳米粒。
7)采用激光-蚀刻法将多孔聚苯乙烯的一面切削成微针阵列,以0.5*0.5cm大小制成多孔微针单元;将所述微针单元放置在所述复方中药固体脂质纳米粒中,经过包被、填充,取出晾干后,将所述微针单元固定于药膏空白贴的内表面,再覆上药膏背贴即得治疗骨折软组织损伤中药微针透皮给药贴。
实施例6
1)称取赤芍40g、马钱子50g、红花30g、透骨草45g、淫羊藿45g、三七20g、黄芪40g、防风30g、泽兰25g、杜仲20g、当归20g、骨碎补15g、血竭15g,粉碎混合后,以浓度为75%乙醇溶液对混合物浸泡3~5天,过滤,收集滤液并对所述滤液蒸发溶剂后,得到复方中药提取物Ⅰ;
2)称取薄荷50g放入三口烧瓶中,蒸馏,对馏出液离心处理,离心后静置,取上层油液,即得中药活性物薄荷挥发油Ⅱ;
3)将复方中药提取物Ⅰ用500ml纯化水和40ml聚山梨酯-80的混合液进行溶解,超声分散后,加热至65~75℃保温,制得水相A;
4)将质量比为2:1的大豆磷脂和单硬脂酸甘油酯混合物20g混合后,用300ml体积比为4:1的乙醇/二氯甲烷混合液对混合物B进行溶解,得到溶液C,将步骤2)中所得的薄荷挥发油Ⅱ与溶液C混合,超声分散15分钟后,制得油相D;
5)将油相D注入正在搅拌且水浴保温的所述水相A中,搅拌,形成乳白色混悬液E,保温搅拌蒸发有机溶剂后,超声分散,得脂质纳米粒混悬液粗品F;
6)将所述脂质纳米粒混悬液粗品F注入高压均质机中进行高压均质,浓缩后,得复方中药固体脂质纳米粒G;
7)采用激光-蚀刻法将多孔硅基板的正面切削成0.5*0.5cm大小的微针单元,将所述微针单元放置在所述复方中药固体脂质纳米粒中,经过包被、填充和取出晾干后,将蚀刻处理后的基板的背面粘贴于药膏空白贴具有粘性的那一面,所述微针单元用药膏背贴进行覆盖,所述基板位于所述药膏空白贴与所述药膏背贴中间,三者通过黏贴的方式进行固定连接,即得治疗骨折软组织损伤中药微针透皮给药贴。

Claims (7)

1.一种治疗骨折软组织损伤中药微针透皮给药贴的制备方法,其特征在于,包括以下制备步骤:
1)称取赤芍、马钱子、红花、透骨草、淫羊藿、三七、黄芪、防风、泽兰、杜仲、当归、骨碎补和血竭,粉碎混合后,以浓度为50~80%乙醇溶液对混合物浸泡3~5天,过滤,收集滤液并对所述滤液蒸发溶剂后,得到复方中药提取物Ⅰ;
2)称取薄荷放入三口烧瓶中,蒸馏,对馏出液离心处理,离心后静置,取上层油液,即得中药活性物薄荷挥发油Ⅱ;
3)将复方中药提取物Ⅰ用纯化水和聚山梨酯-80的混合液进行溶解,超声分散后,加热至65~75℃保温,制得水相A;
4)将质量比为1:1~3:1的大豆磷脂和单硬脂酸甘油酯混合后,得到混合物B,用体积比为4:1的乙醇/二氯甲烷混合液对所述混合物B进行溶解,得到溶液C,将步骤2)中所得的薄荷挥发油Ⅱ与所述溶液C混合,超声分散后,制得油相D;
5)将油相D注入正在搅拌且水浴保温的所述水相A中,搅拌,形成乳白色混悬液E,保温搅拌蒸发有机溶剂后,超声分散,得脂质纳米粒混悬液粗品F;
6)将所述脂质纳米粒混悬液粗品F注入高压均质机中进行高压均质,浓缩后,得复方中药固体脂质纳米粒G;
7)采用激光-蚀刻法将基板的正面切削成0.5*0.5cm大小的微针单元,将所述微针单元放置在所述复方中药固体脂质纳米粒G中,经过包被、填充和取出晾干后,将蚀刻处理后的基板的背面粘贴于药膏空白贴具有粘性的那一面,所述微针单元用药膏背贴进行覆盖,所述基板位于所述药膏空白贴与所述药膏背贴中间,三者通过黏贴的方式进行固定连接,即得治疗骨折软组织损伤中药微针透皮给药贴。
2.如权利要求1所述的一种治疗骨折软组织损伤中药微针透皮给药贴的制备方法,其特征在于,所述赤芍、马钱子、红花、透骨草、淫羊藿、三七、黄芪、防风、泽兰、杜仲、当归、骨碎补、血竭、薄荷、大豆磷脂/单硬脂酸甘油酯混合物和聚山梨酯-80的组份含量是:
赤芍30~50g
马钱子40~60g
红花30~40g
透骨草40~50g
淫羊藿40~50g
三七20~30g
黄芪30~40g
防风20~30g
泽兰20~30g
杜仲20~30g
当归20~30g
骨碎补10~20g
血竭10~20g
薄荷40~60g
大豆磷脂/单硬脂酸甘油酯混合物10~30g
聚山梨酯-8010~50ml。
3.如权利要求1所述的一种治疗骨折软组织损伤中药微针透皮给药贴的制备方法,其特征在于,所述赤芍、马钱子、红花、透骨草、淫羊藿、三七、黄芪、防风、泽兰、杜仲、当归、骨碎补、血竭、薄荷、大豆磷脂/单硬脂酸甘油酯混合物和聚山梨酯-80的组份含量是:
赤芍40g
马钱子50g
红花30g
透骨草40g
淫羊藿40g
三七20g
黄芪30g
防风20g
泽兰20g
杜仲20g
当归20g
骨碎补10g
血竭10g
薄荷40g
大豆磷脂/单硬脂酸甘油酯混合物10g
聚山梨酯-8010ml。
4.一种治疗骨折软组织损伤中药微针透皮给药贴,其特征在于,所述治疗骨折软组织损伤中药微针透皮给药贴是通过权利要求1~3所述的一种治疗骨折软组织损伤中药微针透皮 给药贴的制备方法中任一方法制作的。
5.如权利要求4所述的一种治疗骨折软组织损伤中药微针透皮给药贴,其特征在于,包括基板、药剂、药膏空白贴和药膏背贴,所述基板位于所述药膏空白贴与所述药膏背贴的中间,所述基板采用具有多孔结构的材料,所述基板的正面设有多于一个的微针,所述微针的顶端设有尖刺部,所述微针垂直固定在所述基板上,所述微针之间具有微针间隙,所述药剂存储在所述微针间隙和所述多孔结构之中,所述基板的背面为平面,所述基板的背面粘贴在所述药膏空白贴的内表面上,所述药膏背贴的内表面覆盖在所述基板的正面上,所述药膏空白贴的内表面与所述药膏背贴的内表面通过黏贴的方式进行固定连接。
6.如权利要求4所述的一种治疗骨折软组织损伤中药微针透皮给药贴,其特征在于,所述基板选用的材料为多孔硅、多孔钛或多孔聚苯乙烯中的一种。
7.如权利要求5所述的一种治疗骨折软组织损伤中药微针透皮给药贴,其特征在于,所述微针排列形成一个矩形状微针阵列,所述微针阵列多于一个,所述微针阵列的长和宽都为0.5cm,相邻所述微针阵列之间的间距为0.1cm。
