Abstract
本发明提供了一种WS2‑PVP纳米片,在WS2纳米片的表面修饰有聚乙烯吡咯烷酮。本发明还提供了上述一种WS2‑PVP纳米片的制备方法,先将硫源和钨源溶解于溶剂中,搅拌使之完全溶解;在搅拌作用下,将聚合物溶解于硫源和钼源的溶液中,得硫源、钼源和聚合物的混合溶液;将所得溶液转移至对位聚苯内衬的不锈钢反应釜中密封反应一段时间,离心分离、清洗所得产物,即得表面修饰有聚合物、具有良好的胶体稳定性的二维WS2‑PVP纳米片。本发明还提供了WS2‑PVP纳米片作为光热转化材料中的应用。本发明的WS2‑PVP纳米片具有优秀的光热转化能力、计算机断层扫描和光声成像能力,可有望应用于肿瘤的高效协同诊断和治疗。

技术领域
本发明属于生物纳米材料领域,具体来说涉及一种简单易行、高效的超小WS2-PVP纳米片及其制备方法和应用。
背景技术
光热转化材料,包括无机材料诸如氧化钨纳米线、硫化铜纳米颗粒、碳纳米材料(如石墨烯)和贵金属纳米材料(如金纳米棒,钯的纳米片),有机材料诸如有机化合物/聚合物(如吲哚菁绿,聚吡咯纳米颗粒)等,已经被用于肿瘤的光热治疗研究。在近红外激光(NIR,波长范围:700-1100 nm)的照射下,富集在肿瘤组织中的上述光热材料可以吸收并将NIR转化为热量,使肿瘤组织局部升温而导致肿瘤消融死亡、进而杀死肿瘤细胞。然而,上述纳米材料作为光热转化材料却存在着诸多不足之处:对于贵金属纳米材料而言,其形貌会随着近红外激光的辐射而发生明显的变化,导致NIR吸收能力减弱,光热性能也随之减弱。而碳材料也存在着吸收系数和光热转化效率低等缺点。在人们日益追求高质量生活的今天,开发新的光热转化材料,对推动肿瘤的光热治疗研究具有重要意义。
过渡金属硫化物(如MoS2,MoSe2,WS2和WSe2)等因具有低成本、低毒等优点成为了光热材料领域的一个重要研究方向。作为过渡金属硫化物一员, WS2纳米片/量子点在肿瘤治疗的研究领域应用得到了研究者的关注。例如,Zhao 等人利用外力研磨和超声双重作用,“自上而下”地制备出了超小WS2量子点(直径3nm),实现了对肿瘤的光热治疗/放疗联合治疗(ACS Nano, 2015,9: 12451-12463)。Liu等人利用溶剂热法,合成出了掺杂有不同金属元素的WS2纳米片,通过后续的聚乙二醇(PEG)修饰,得到了PEG修饰的Fe,Co,Ni,Mn和Gd掺杂的WS2纳米片,实现了肿瘤的诊断和治疗(ACS Nano, 2015,9: 11090-11101)。与MoS2纳米片类似,WS2纳米片本身无法像石墨烯类碳材料那样通过“π-π共轭”的方式与PEG等有机物碳链连接而实现表面修饰,故已有报道的合成和表面PEG修饰的工艺非常复杂,而且也无法实现对MoS2纳米片形貌的有效控制。我们前期的研究表明(Biomaterials,2015,39,206),通过向溶剂热反应溶剂中添加PEG-400,可以一步实现MoS2的可控合成和表面PEG修饰。
基于此,我们设想利用溶剂热法“自下而上”制备WS2纳米片;向溶剂热反应体系中添加聚合物分子,在溶剂热合成的同时,实现对WS2纳米片的表面修饰。截止目前,尚没有文献报道通过添加聚合物分子,利用溶剂热法一步实现WS2纳米片的合成及同步表面修饰的报道。
发明内容
针对现有技术中的上述技术问题,本发明提供了一种WS2-PVP纳米片及其制备方法和应用,所述的这种WS2-PVP纳米片及其制备方法和应用要解决现有技术中WS2-PVP纳米片合成和表面修饰工艺非常复杂,纳米片的形貌不能进行有效控制的技术问题。
本发明提供了一种WS2-PVP纳米片,在WS2纳米片的表面修饰有聚乙烯吡咯烷酮。
本发明还提供了一种WS2-PVP纳米片的制备方法,包括如下步骤:
1)将硫源和钨源溶解于溶剂中,搅拌使之完全溶解;所述的钨源为钨酸铵、钨酸钠、或者四硫代钨酸铵中的任意一种,所述的硫源为单质硫、二硫化碳、硫化氢、硫脲、四硫代钨酸铵中的任意一种;
2)在搅拌作用下,将聚合物溶解于硫源和钼源的溶液中,得硫源、钼源和聚合物的混合溶液,所述的聚合物为聚乙二醇或聚乙烯吡咯烷酮,聚乙二醇的分子量为400Da-1000Kda,聚乙烯吡咯烷酮的分子量为10kDa-1300kDa;
3)将所得溶液转移至对位聚苯内衬的不锈钢反应釜中密封反应,反应完全后,离心分离、清洗所得产物,即得表面修饰有聚合物的WS2-PVP纳米片。
进一步的,所述的钨源和所述的硫源均为四硫代钨酸铵。
进一步的,步骤(1)中,所述的溶剂为蒸馏水、N,N-二甲基甲酰胺、乙醇、或者聚乙二醇-400中的一种或两种以上的组合。
进一步的,步骤(2)中所述的钨源的浓度为1~50 mg/mL,所述的硫源的浓度为1~50 mg/mL。
进一步的,步骤(1)-(2)中,所述的搅拌为磁力搅拌,其速率为50-200 r/min,搅拌时间为10-90分钟。
进一步的,聚四氟乙烯高压釜中的反应温度为200-220 ℃,反应时间为12-24小时;分离方法为离心分离,转速为5000-10000 r/min。
进一步的,产物洗涤分别使用乙醇胺溶液和蒸馏水;乙醇胺溶液的体积百分比浓度为10-80%,乙醇胺水溶液和蒸馏水的清洗次数均为3-5次。
本发明还提供了WS2-PVP纳米片作为光热转化材料中的应用。
本发明通过溶剂反应法处理聚合物、硫源和钨源材料的混合溶液特定时间,即得具有良好胶体稳定性的WS2-PVP纳米片。一方面,添加的聚合物分子链可以约束纳米片的生长、控制纳米片的形貌;其次,聚合物分子链还可以有效地修饰在纳米片表面,改善纳米材料的胶体稳定性。
本发明和已有技术相比,其技术进步是显著的。本发明工艺简单,产品易得通过本发明的方法制备得到的WS2-PVP纳米片具有良好的胶体稳定性和优异的光热转换效应,具有计算机断层扫描造影和光声造影能力,有望应用于肿瘤的诊断和光热治疗等领域。

具体实施方式
下面结合具体实施例,进一步阐述本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。此外应理解,在阅读了本发明讲授的内容之后,本领域技术人员可以对本发明作各种改动或修改,这些等价形式同样落于本申请所附权利要求书所限定的范围。
实施例1
称取0.15 g 四硫代钨酸铵和0.15g PVP(分子量为360kDa),与30 N,N-二甲基甲酰胺混合,室温下搅拌1小时,得澄清透明溶液。将所得溶液转移至100 mL容积的对位聚苯内衬的不锈钢反应釜中密封。将反应釜置入高温烘箱中220 ℃热处理12 h,待自然冷却至室温后,离心分离反应混合物(10000 r/min,5min),并用50%的乙醇胺溶液和蒸馏水各洗涤3次。通过TEM观察材料的微观形貌:将适量纳米片分散于无水乙醇中,超声分散均匀后,将镀有碳膜的铜网浸入上述无水乙醇中。待样品自然干燥后,通过TEM观察、拍照,TEM操作电压为200 kV。
对比例1
称取0.15 g 四硫代钨酸铵和0.15g PVP(分子量为30kDa),与30 N,N-二甲基甲酰胺混合,室温下搅拌1小时,得澄清透明溶液。将所得溶液转移至100 mL容积的对位聚苯内衬的不锈钢反应釜中密封。将反应釜置入高温烘箱中220 ℃热处理12 h,待自然冷却至室温后,离心分离反应混合物(10000 r/min,5min),并用50%的乙醇胺溶液和蒸馏水各洗涤3次。通过TEM观察材料的微观形貌:将适量纳米片分散于无水乙醇中,超声分散均匀后,将镀有碳膜的铜网浸入上述无水乙醇中。待样品自然干燥后,通过TEM观察、拍照。
对比例2
称取0.15 g 四硫代钨酸铵和0.15g PVP(分子量为2100kDa),与30 N,N-二甲基甲酰胺混合,室温下搅拌1小时,得澄清透明溶液。将所得溶液转移至100 mL容积的对位聚苯内衬的不锈钢反应釜中密封。将反应釜置入高温烘箱中220 ℃热处理12 h,待自然冷却至室温后,离心分离反应混合物(10000 r/min,5min),并用50%的乙醇胺溶液和蒸馏水各洗涤3次。通过TEM观察材料的微观形貌:将适量纳米片分散于无水乙醇中,超声分散均匀后,将镀有碳膜的铜网浸入上述无水乙醇中。待样品自然干燥后,通过TEM观察、拍照。
用Image J 1.40G软件(http://rsb.info.nih.gov/ij/download.html,NationalInstitutes of Health,美国)测量纳米片的片径(每个样品至少测量50片)约为100 nm。用布鲁克公司的Dimension Icon型原子力显微镜观察样品的形貌像,以统计其厚度。
由图1(a-c)可以看出,本发明合成的WS2-PVP纳米片形貌均匀,片径100nm左右,片层厚度6-7nm(图1(d))。
由图2可以看出,不同分子量的PVP介导合成出的WS2-PVP纳米片形貌不同(30kDa,图2(a,c,d),2100kDa,图2(b,d,e)),在不同环境下,表现出了各异的胶体分散性(分子量为2100kDa的PVP介导的WS2-PVP纳米片较分子量为30kDa和360kDa的胶体稳定性差,如图4(a,b,c))。这可能是因为,在溶剂热反应过程中,不同分子量的PVP对片状材料的生长的约束作用不同,导致产物具有不同的形貌;在液相环境中,不同长度的聚合物空间位阻作用不同,进而导致WS2-PVP纳米片具有不同的分散性。
实施例2
用Thermal Scientific公司的ESCAlab250型X射线光电子能谱仪(XPS)表征纳米片中W和S元素的化合价。激发源为单色器Al Kα X射线(λ = 0.8339 nm),能量为1486 eV,线宽为0.9 eV,功率为150 W。结合能用C的1s峰(284.8 eV)校正。使用XRD(Rigaku D/max-2200PC,日本)研究纳米片的XRD衍射图谱的晶体结构。以Cu2Kα射线为光源,操作电压为40 kV,电流为200 mA,扫描角度(2θ)范围为3° - 70°。使用FTIR(Nicolet Nexus 670红外光谱仪)对WS2-PVP纳米片表征,取少许WS2-PVP纳米片粉体和PVP粉体(对照组),与干燥的KBr粉末混合研磨均匀后压片。置于Nicolet Nexus 670红外光谱仪样品架上进行扫描(扫描范围400-4000cm-1)。采用热重分析(TG,德国NETZSCH)对WS2-PVP表征,通过样品经高温灼烧后的质量残余来证明WS2-PVP表面PVP的存在。测试时,称取5 mg左右的粉末样品,置于氧化铝坩埚中,选择空气为吹扫气体,吹扫速度为20 mL/min,温度范围为室温到800 ℃,升温速度为10℃/min。
如图3(a)和(b)所示,对比文献可知产物中W和S的价态均为W+4和S-2,可分别归属为WS2和WOX中W的4f7/2、4f5/2电子轨道。S-2的电子能谱谱图则归因于S2p1/2、和2p3/2轨道。(c)为WS2-PVP纳米片的X射线衍射图谱,材料的X射线衍射图谱和WS2标准图谱(JCPD 35-0651)各晶面位置并不吻合(主要衍射峰出现了一定程度的右移),这主要是因为PVP的氧原子链接在所得WS2-PVP纳米片表面,替代了原子半径交大的硫原子所致;另外可能也可PVP高分子约束下纳米片的定向生长有关,这证明了本发明制备的WS2-PVP的形成本质为:在溶剂热反应过程中,部分PVP分子链连接在WS2表面,并取代其中1个S原子。PVP和WS2-PVP纳米片的(d)傅里叶变换红外光谱图和(e-f)PVP(分子量360kDa)和WS2-PVP纳米片的TG曲线;从傅里叶变换红外光谱图中可以清晰地看出PVP的骨架振动吸收峰,说明PVP分子链已经成功地连接在r/min纳米片的表面。TG表征结果表明,在空气气氛下升温至800℃后,PVP已经完全燃烧,而WS2-PVP纳米片的质量残余约为67.5%,进一步证明了WS2-PVP纳米片表面包裹一层PVP,其质量约占WS2-PVP纳米片的32.5%。
实施例3
使用英国马尔文公司的Zetasizer Nanoseries(Nano ZS90)测量表征纳米片在不同分散介质中的粒径,及其水中的zeta电位。图4(a)、(b)和(c)可以看出,分散于去离子水、生理盐水和1640细胞培养基中72h后,分子量为2100kDa的PVP介导的WS2-PVP纳米片较分子量为30kDa和360kDa的胶体稳定性差,分子量为30kDa和360kDa的PVP介导合成的WS2-PVP纳米片的水合动力学直径并没有发生明显的变化,具有良好的胶体稳定性(如明显的丁达尔效应(图4(d-f)),且仍为稳定的胶体(图4g(左:PVP30kDa,中:PVP360kDa,右:PVP2100kDa),为在活体水平评价它们的光热转换、杀伤肿瘤能力奠定了基础。不同分子量PVP介导合成的WS2-PVP纳米片在水中的均显示为负的表面Zeta电势(图4(d-h))。
实施例4
用UV-Vis-NIR(Lambda 25,Perkin Elmer公司,美国)分析其光吸收性质(波长范围350-1100 nm)。将不同浓度的WS2-PVP纳米片分散在96孔细胞培养板的培养孔中,取蒸馏水作为对照,用如图5(e)所列出的预先设定功率的808 nm波长的近红外激光照射不同浓度的WS2-PVP纳米片(图5(b,c和e))或蒸馏水,通过FLIR E60红外热像仪记录材料分散液或蒸馏水温度随时间的变化情况及对应的红外热成像照片。
从图5(a)可以看出,不同浓度的WS2-PVP均可以吸收一定强度的波长为808 nm的近红外激光。可以预计,上述不同材料吸收的这部分近红外激光将转化为热量,因而具备近红外光吸收-热转化能力。从图5(b)可以看出,与其红外光吸收特性类似,WS2-PVP纳米片对808nm近红外激光的吸收和热转化能力较980nm激光更强。从图5(c)可以看出,高浓度的WS2-PVP纳米片分能更加有效地进行光热转换而升高水温。在同一浓度下(W的浓度为25ppm),从图5(e)可以看出,激光密度越高,能量越大,进而水温升高越高;从图5(d)和5(f)为与图5(c)和5(e)可以看出,红外热成像照片进一步验证了WS2-PVP纳米片的光热转化与浓度或激光密度的关系。总之,在实验条件下,本发明制备的WS2-PVP纳米片表现出了良好的光热转换能力。
实施例5
WS2-PVP纳米片的光声成像性能:将1%的琼脂水溶液置于微波炉中加热3 min,然后倒入模具中。制备孔径大小为3 mm的小孔,于室温下冷却凝结、脱模。将15μL的不同浓度的WS2-PVP纳米片注射入凝胶小孔中,使用Vevo LAZR小动物光声成像系统进行光声成像实验。得到图像后,选择成像的区域计算光声信号值,得材料的光声信号值。
WS2-PVP纳米片的CT成像性能:将2mL不同浓度的WS2-PVP纳米片注射入2mL离心管中。以临床用 CT 造影剂碘海醇作为对照样品,其中碘浓度与样品溶液中的W浓度相同。用医用CT测试评价WS2-PVP纳米片的CT造影性能。
如附图6所示,随着W元素浓度的升高,光声造影值(图6(a,b))和CT造影值(图6c,d)均呈近似线性上升趋势。如图6(a),当W的浓度在200ppm时,材料的在808nm处PA值为0.92;在W浓度为50ppm时,其光声值仍可以达到0.42左右。CT造影性能灵敏度相对较低,但仍然优于商品化的造影剂碘海醇(如图6(c-d))。在Bi浓度为2mM时,HU值为22,具有一定的CT造影性能。总之,本发明制备的WS2-PVP纳米片显示出了良好的光声和一定的CT造影性能,因而可以很好地监控其在肿瘤内的分布情况,有望实现肿瘤诊断同步高效治疗。
Claims (9)
1.一种WS2-PVP纳米片,其特征在于:在WS2纳米片的表面修饰有聚乙烯吡咯烷酮。
2.权利要求1所述的一种WS2-PVP纳米片的制备方法,其特征在于包括如下步骤:
1)将硫源和钨源溶解于溶剂中,搅拌使之完全溶解;所述的钨源为钨酸铵、钨酸钠、或者四硫代钨酸铵中的任意一种,所述的硫源为单质硫、二硫化碳、硫化氢、硫脲、四硫代钨酸铵中的任意一种;
2)在搅拌作用下,将聚合物溶解于硫源和钼源的溶液中,得硫源、钼源和聚合物的混合溶液,所述的聚合物为聚乙二醇或聚乙烯吡咯烷酮,聚乙二醇的分子量为400Da-1000Kda,聚乙烯吡咯烷酮的分子量为10kDa-1300kDa;
3)将所得溶液转移至对位聚苯内衬的不锈钢反应釜中密封反应,反应完全后,离心分离、清洗所得产物,即得表面修饰有聚合物的WS2-PVP纳米片。
3.根据权利要求2所述的一种WS2-PVP纳米片的制备方法,其特征在于:所述的钨源和所述的硫源均为四硫代钨酸铵。
4.根据权利要求2所述的一种WS2-PVP纳米片的制备方法,其特征在于:步骤(1)中,所述的溶剂为蒸馏水、N,N-二甲基甲酰胺、乙醇、或者聚乙二醇-400中的一种或两种以上的组合。
5.根据权利要求2所述的一种WS2-PVP纳米片的制备方法,其特征在于:步骤(2)中所述的钨源的浓度为1~50 mg/mL,所述的硫源的浓度为1~50 mg/mL。
6.根据权利要求1所述的一种WS2-PVP纳米片的制备方法,其特征在于:步骤(1)-(2)中,所述的搅拌为磁力搅拌,其速率为50-200 r/min,搅拌时间为10-90分钟。
7.根据权利要求2所述的一种WS2-PVP纳米片的制备方法,其特征在于:聚四氟乙烯高压釜中的反应温度为200-220 ℃,反应时间为12-24小时;分离方法为离心分离,转速为5000-10000 r/min。
8.根据权利要求2所述的一种WS2-PVP纳米片的制备方法,其特征在于:产物洗涤分别使用乙醇胺溶液和蒸馏水;乙醇胺溶液的体积百分比浓度为10-80%,乙醇胺水溶液和蒸馏水的清洗次数均为3-5次;WS2-PVP纳米片作为光热转化材料中的应用。
9.权利要求1所述的WS2-PVP纳米片作为光热转化材料中的应用。
