现有技术中通气鼻贴中中药提取方法,工艺粗糙、落后,杂质多,有效成分含量低,导致患者用量过大,不方便服用,严重影响了本品在临床上应用。
发明内容
发明目的:为了解决上述问题,本发明的目的在于提供一种通气鼻贴。
技术方案:本发明的目的是通过如下的方案实现的:
一种通气鼻贴,由布条和药液组成,药液浸泡在布条中,药液的制备方法为:取灵香蒿20g、鸡心七18g、大尾摇25g、牛舌头10g、陈皮20g、艾叶15g、甘松10g、玫瑰花20g、化橘红18g、灯心草15g、板蓝根20g、夏枯草18g、白花蛇舌草20g,加饮片10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置24小时,过滤,滤液浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,使用超临界萃取,得超临界萃取物,继续浓缩,制备成药液。
所述通气鼻贴,制备方法中将浓缩液加入到CO2超临界萃取器中,乙醇作为夹带剂,夹带剂占总萃取溶剂的体积百分比为4-6%,萃取压力15-30MPa,温度30-50℃,CO2流量1-3m1/g生药·min,萃取时间150-180min,得超临界萃取物,优选条件为:所述CO2超临界萃取夹带剂占总萃取溶剂的体积百分比为5%,所述CO2超临界萃取的萃取压力20MPa,温度40℃,CO2流量2ml/g生药·min,萃取时间160min。
所述通气鼻贴,上述提取方法中浓缩使用装置,包括浓缩筒体,设置在所述浓缩筒体底部的蒸发器,在所述浓缩筒体的顶部设有出气孔和凝结面板,在所述凝结面板内设有降温液存储腔,在所述浓缩筒体的顶部设有与所述降温液存储腔相连通的进液管和排出管,其特征在于:在所述浓缩筒体内的顶壁设有向下延伸的旋转杆,所述旋转杆上设有刮垢刀盘,所述刮垢刀盘的盘边与浓缩筒体的内壁相接触,在所述浓缩筒体的顶壁上设有升降气缸,所述旋转杆的顶部穿过所述浓缩筒体的顶壁且顶端与所述升降气缸相连接,在所述旋转杆内设有旋转电机,所述旋转电机的主轴与所述刮垢刀盘的中心相连接,在所述浓缩筒体的顶部还设有穿过顶壁的升降杆,在所述升降杆的底部设有喇叭状吸泡嘴,在所述升降杆内设有与所述喇叭状吸泡嘴相连通的吸泡通道,在所述升降杆的顶部设有吸附泵体,在所述浓缩筒体的顶壁与所述升降杆之间设有调节气缸,所述升降杆在所述调节气缸的作用下在所述浓缩筒体内做升降移动。
所述通气鼻贴,上述提取方法中浓缩装置所述浓缩筒体的侧壁上设有观察窗,在所述观察窗内设有密封玻璃,所述刮垢刀盘为圆环状,所述刮垢刀盘的外径与浓缩筒体的内径相同且刮垢刀盘的内环面通过连接筋杆与旋转电机的主轴相连接,所述蒸发器为圆筒形结构,所述蒸发器通过连接法兰连接在浓缩筒体的底壁,在所述蒸发器内设有蒸发管,在所述蒸发器的侧壁上设有与所述蒸发管相连通的蒸汽进口和蒸汽出口。
灵香蒿为菊科香青属植物稀毛香青Anaphalis hancockii Maxim.,以全草入药。夏季采收,阴干用。清热解毒,杀虫。用于子宫颈炎、阴道滴虫。
鸡心七为蛇菰科植物多药蛇菰(Balanophora polyandra)的全株。因其形态状如鸡心,故得此名。清热解毒,温经通络,活血止血,接骨。用于跌打损伤,咳血;外用治乳腺炎,刀伤,开发性骨折,疔疮肿毒。本发明以全株入药。
大尾摇为紫草科天芥菜属植物大尾摇Heliotropium indicum L.,以全草入药。夏秋采收,晒干或鲜用。清热解毒。用于肺炎,肺脓肿,脓胸,腹泻,痢疾,睾丸炎,白喉,口腔糜烂,痈疖。
牛舌头为菊科植物匍茎苦菜的全草,清热解毒。治阑尾炎,痢疾,痔疮,遗精,白浊,乳腺炎,疮疖种毒,汤火伤。其余为药典品种。
现有技术中,人参原来提取方法,工艺粗糙、落后,杂质多,导致患者用量过大,不方便服用,本发明制备的靛玉红含量增加,含量高。本发明中的浓缩设备结构简单、操作便捷,其不但可以根据需要启动刮垢刀盘并根据需要做升降调节从而可以实现对浓缩筒体的内壁进行刮垢处理,同时还可以根据需要调节升降杆的升降操作使喇叭状吸泡嘴处于药物液面的稍上方通过吸附泵体将表面的泡沫吸除,解决了传统技术药物浓缩装置中管壁药垢清除困难的技术不足,使用稳定性好且适用性强。本发明灵香蒿、鸡心七、大尾摇、牛舌头清热解毒为君药,陈皮、艾叶、甘松、玫瑰花、化橘红、灯心草理气止咳为臣药,板蓝根、夏枯草、白花蛇舌草为佐使药,全方理气,治疗鼻塞感冒效果良好。

具体实施方式
以下通过实施例形式,对本发明的上述内容再作进一步的详细说明,但不应将此理解为本发明上述主题的范围仅限于以下的实例,凡基于本发明上述内容所实现的技术均属于本发明的范围。
实施例1
一种通气鼻贴,由布条和药液组成,药液浸泡在布条中,药液的制备方法为:取灵香蒿20g、鸡心七18g、大尾摇25g、牛舌头10g、陈皮20g、艾叶15g、甘松10g、玫瑰花20g、化橘红18g、灯心草15g、板蓝根20g、夏枯草18g、白花蛇舌草20g,加饮片10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置24小时,过滤,滤液浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,使用超临界萃取,得超临界萃取物,继续浓缩,制备成药液。
所述超临界萃取方法为:将浓缩液加入到CO2超临界萃取器中,乙醇作为夹带剂,夹带剂占总萃取溶剂的体积百分比为4%,萃取压力30MPa,温度30℃,CO2流量3m1/g生药·min,萃取时间150min,得超临界萃取物。
见图1:所述通气鼻贴,上述浓缩使用装置,包括浓缩筒体1,设置在所述浓缩筒体1底部的蒸发器2,在所述浓缩筒体1的顶部设有出气孔3和凝结面板4,在所述凝结面板4内设有降温液存储腔5,在所述浓缩筒体1的顶部设有与所述降温液存储腔5相连通的进液管6和排出管7,设有凝结面板,可以将药夜蒸发的气体部分凝结成水分,可以防止药物挥发掉,在所述浓缩筒体1内的顶壁设有向下延伸的旋转杆8,所述旋转杆8上设有刮垢刀盘9,所述刮垢刀盘9的盘边与浓缩筒体1的内壁相接触,在所述浓缩筒体1的顶壁上设有升降气缸10,所述旋转杆8的顶部穿过所述浓缩筒体1的顶壁且顶端与所述升降气缸10相连接,在所述旋转杆8内设有旋转电机11,所述旋转电机11的主轴与所述刮垢刀盘9的中心相连接,在所述浓缩筒体1的顶部还设有穿过顶壁的升降杆12,在所述升降杆12的底部设有喇叭状吸泡嘴13,在所述升降杆12内设有与所述喇叭状吸泡嘴13相连通的吸泡通道14,在所述升降杆12的顶部设有吸附泵体15,在所述浓缩筒体1的顶壁与所述升降杆12之间设有调节气缸16,所述升降杆在所述调节气缸的作用下在所述浓缩筒体内做升降移动。
在所述浓缩筒体1的侧壁上设有观察窗17,在所述观察窗内设有密封玻璃18。
所述刮垢刀盘为圆环状,所述刮垢刀盘的外径与浓缩筒体的内径相同且刮垢刀盘的内环面通过连接筋杆与旋转电机的主轴相连接。由于刮垢与吸泡并不是同时进行,故而两者并不会造成相互限制。
所述蒸发器2为圆筒形结构,所述蒸发器2通过连接法兰19连接在浓缩筒体1的底壁,在所述蒸发器内设有蒸发管20,在所述蒸发器2的侧壁上设有与所述蒸发管20相连通的蒸汽进口21和蒸汽出口22。
实施例2
一种通气鼻贴,由布条和药液组成,药液浸泡在布条中,药液的制备方法为:取灵香蒿20g、鸡心七18g、大尾摇25g、牛舌头10g、陈皮20g、艾叶15g、甘松10g、玫瑰花20g、化橘红18g、灯心草15g、板蓝根20g、夏枯草18g、白花蛇舌草20g,加饮片10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置24小时,过滤,滤液浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,使用超临界萃取,得超临界萃取物,继续浓缩,制备成药液。
所述超临界萃取方法为:将浓缩液加入到CO2超临界萃取器中,乙醇作为夹带剂,夹带剂占总萃取溶剂的体积百分比为6%,萃取压力15MPa,温度50℃,CO2流量1m1/g生药·min,萃取时间180min,得超临界萃取物。
浓缩和干燥设备同上。
实施例3
一种通气鼻贴,由布条和药液组成,药液浸泡在布条中,药液的制备方法为:取灵香蒿20g、鸡心七18g、大尾摇25g、牛舌头10g、陈皮20g、艾叶15g、甘松10g、玫瑰花20g、化橘红18g、灯心草15g、板蓝根20g、夏枯草18g、白花蛇舌草20g,加饮片10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置24小时,过滤,滤液浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,使用超临界萃取,得超临界萃取物,继续浓缩,制备成药液。

浓缩和干燥设备同上。
实施例4:本发明通气鼻贴中靛玉红含量测定实验研究资料
1.1实验药物:本发明通气鼻贴:按实施例1-3方法制备。对照组为按照实施例1采用普通浓缩和干燥方法制备成通气鼻贴。
1.2仪器:高效液相色谱仪系统包括高效液相色谱仪(Waters 515);P200高压泵;Waters2487紫外检测器及GJ605型高压六通进样阀;数据采集及处理采用HS色谱数据工作站V4.0+(杭州英谱科技)。
1.3测定条件:照高效液相色谱法(通则0512)测定。色谱条件与系统适用性试验:色谱柱为MUCLEODUR C18,4.6×250mm,5μm,柱温35℃;以甲醇-水(60:40)为流动相;检测波长为250nm;理论板数按靛玉红峰计算应不低于2000。取靛玉红对照品适量,精密称定,加甲醇制成每lm l含5ug的溶液,即得。供试品溶液的制备取装量差异项下的本品,研细,取约0.5g,精密称定,置25ml量瓶中,加甲醇20ml,超声处理(功率250W,频率40kHz)30分钟,放冷,加甲醇至刻度,摇匀,滤过,取续滤液,即得。分别精密吸取对照品溶液与供试品溶液各20ul注入液相色谱仪,测定,即得。
1.4实验结果

根据上表的试验结果可知,本发明实施例1-3制备的通气鼻贴中靛玉红含量明显高于对照组。
实施例5:本发明的抗病毒作用
药品及试剂:按上述实施例1制备本发明。双黄连口服液,每支10ml,中国哈尔滨制药三厂,批号:20130425。RPMI 1640培养基,Life Technologies Inc.U.S.A。胰蛋白酶,上海化学试剂站分装厂,批号:941117。
细胞:Hep—2细胞,由江苏省生物细胞研究中心提供。
菌株及病毒:腺病毒3型,由南京医科大学微生物教研室提供;流感病毒鼠肺适应株FMI,由中国预防医学科学院病毒研究所提供。甲型流感病毒A/PR/8/34(H1N1),由中国预防医学科学院病毒研究所提供。
培养基:营养琼脂,上海市医学化验所,批号:20120101。营养肉汤,江苏省卫生防疫站微生物试剂厂,批号:20131004。
实验方法和结果:对病毒致细胞病变作用(CPE)的影响:取10日龄菜亨种鸡胚,每枚尿囊腔接种病毒0.2ml,37℃培养36h,4℃过夜,用无菌吸管进入尿囊腔吸取尿液,低温保存并作无菌培养。挑选生长良好的Hep-2细胞培养物,按传代细胞培养法,将细胞单层洗涤,消化,计数,细胞数为约28万/mL。将细胞悬液加入微孔塑料面板中,每孔0.1ml,微孔板置37℃CO2培养箱培养18h。将病毒鸡胚尿囊液用1640培养液作连续十倍稀释,弃培养液,将10-1-10-10稀释之病毒液依次加入微孔,每孔0.1mL,细胞对照不加病毒,每个病毒稀释度加4个孔。置CO2培养箱培养,按Reed—Muench法计算TCID50。
将本发明药液(1g/ml)用1640培养液配制成不同稀释度(1:20-1:160)之药液,不同浓度药物与30TCID50作用1h,然后加入Hep-2细胞培养物,37℃CO2培养箱培养,24h后观察细胞病理性改变,实验重复3次。每次设细胞对照、病毒对照、药物对照,待药物和细胞对照仍为正常,而病毒对照出现++-+++病变时判断结果。
本发明对病毒致细胞病变作用(CPE)的影响
注:细胞病变程度:“-”无细胞病变;“+”25%以下的细胞有病变;“++”25-50%的细胞有病变;“+++”50-75%的细胞有病变;“++++”75-100%的细胞有病变。
实验结果表明;本发明在l:20-l:160的药物浓度下,对腺病毒3型的CPE无明显影响,而对流感病毒鼠肺适应袜(FMI)的CPE有抑制作用,其最低抑制浓度为0.0075g/ml。

1.一种通气鼻贴,其特征在于由布条和药液组成,药液浸泡在布条中,药液的制备方法为:取灵香蒿20g、鸡心七18g、大尾摇25g、牛舌头10g、陈皮20g、艾叶15g、甘松10g、玫瑰花20g、化橘红18g、灯心草15g、板蓝根20g、夏枯草18g、白花蛇舌草20g,加饮片10倍重量的水,浸泡0.5h,煎煮1小时,过滤,药渣再加入药材8倍重量的水,煎煮1小时,药液滤出,合并两次煎煮液,浓缩至60℃相对密度为1.05,加乙醇使含醇量的体积百分比达75%,搅拌,静置24小时,过滤,滤液浓缩至65℃时相对密度为1.25,并回收乙醇,得浓缩液,使用超临界萃取,得超临界萃取物,继续浓缩,制备成药液。

4.根据权利要求3所述通气鼻贴,其特征在于上述提取方法中浓缩装置所述浓缩筒体的侧壁上设有观察窗,在所述观察窗内设有密封玻璃,所述刮垢刀盘为圆环状,所述刮垢刀盘的外径与浓缩筒体的内径相同且刮垢刀盘的内环面通过连接筋杆与旋转电机的主轴相连接,所述蒸发器为圆筒形结构,所述蒸发器通过连接法兰连接在浓缩筒体的底壁,在所述蒸发器内设有蒸发管,在所述蒸发器的侧壁上设有与所述蒸发管相连通的蒸汽进口和蒸汽出口。
