Description
技术领域

发明背景
现有技术中涉及靶向坏死细胞的系统和方法不是最佳的和/或适用的:
a.坏死性对比凋亡性细胞死亡
坏死的靶向是特异的和独特的,因为坏死只出现在细胞死亡的病理情况下,所述病理情况归因于不充足的血液供应并因此氧气缺乏、创伤或者归因于直接的细胞毒性剂或其它如放射性治疗或光动力疗法的癌症治疗。凋亡性细胞死亡与之相反,其在组织更新过程中连续发生。因此相比于坏死性细胞死亡,凋亡性细胞死亡是不适用于靶向特征为坏死的疾病,所述坏死是例如在肿瘤(快速生长的肿瘤自发性进展坏死核)、创伤、梗塞、骨关节炎、糖尿病、动脉粥样硬化斑块、烧伤、某些细菌感染等的情况下发现的。
b.抗体对比小分子
我们的化合物是小分子、青色素基染料(cyanine based dyes),其深深的渗入到坏死组织中。抗体已经被用于靶向坏死(肿瘤坏死靶向),但是具有抗体太大而不能渗入到坏死组织中的缺点。
c.其它化学试剂对比青色素染料

本发明的一个目的是克服现有技术中组合物的一个或多个缺点,并且提供替代当前组合物而用于诊断和治疗涉及坏死性细胞死亡的癌症和其它疾病,并且不损害功能和优点。
发明简介
本发明涉及根据权利要求1所述的一种包括结合坏死细胞的靶向分子,和一种或多种试剂,载体和在体内的非反应性分子的系统。
本系统包括至少两个实体,所述实体例如通过化学键而连接,每个实体在系统中起到不同功能。应当注意的是,可以存在一个以上的试剂,和一个以上的载体,和一个以上的非反应性分子。因此例如组合疗法对本发明的系统是可行的。
本发明的靶向分子是非常有选择性的和非常特异的结合坏死细胞。作为结果,本系统非常低浓度c.q.数量可以被用于提供其有益效果。应当注意的是,相比于坏死特异性探针,细胞凋亡的特异性探针也将靶向健康组织,因为细胞凋亡参与正常组织更新。
本发明人在早期的研究中已经发现,靶向分子特异性非共价结合细胞内蛋白质,如(仅当细胞的细胞膜完整性丧失时,即在坏死细胞的情况下,可供给靶向分子的)肌动蛋白。就最佳的靶点而言,坏死细胞优选是在坏死的早期阶段,例如细胞已经死亡小于几天,优选小于半天,如几个小时,如2个小时,如刚刚死亡的细胞。负责本发明中靶向分子的有效靶向性和/或安全性的特征,是它们为细胞膜不可渗透的,即,它们不能(显著的)穿透健康细胞的细胞膜;它们是非激活的;它们能够非共价地结合到细胞内蛋白(它们的靶向分子);和,它们没有能力/不显著的结合DNA(或RNA)。
假使该系统包括试剂,该试剂可以涉及成像化合物,例如用于在成像技术中,治疗性化合物,以对坏死细胞或有此治疗需求的身体的一部分(包括坏死细胞)提供治疗作用,如癌症,和释放因子,其用于释放进一步化合物,如作为刺激物。

假使该系统包括在体内的非反应性分子,它旨在使该分子与配对物相互作用。相互作用可以(进一步)涉及经由激活剂的一个或多个激活作用,由催化剂支持,基于相互作用产生药物,并基于相互作用释放药物。应当注意的是,从功能点的角度,分子和配对物可以互换。
本系统涉及无毒的小分子,其具有结合坏死细胞和组织的能力。这些分子不与DNA相互作用,即这些分子不是诱变剂。通常的,本系统具有广泛的生物分布,穿透血-脑屏障,不结合到细胞表面蛋白并且是足够稳定的。
在称为的一个实例中,在癌症治疗期间的关键时间被保全。即时提供所给予的化学治疗药物效力的相关数据。本发明系统的一个优点是,在化疗开始之后的24-36小时之内提供该治疗是或否为有效的信息。的使用将防止病人经受没有临床益处的过度治疗,这被认为是在癌症治疗中的一个重要改进。的使用将导致成本节约。
应当注意到,不幸的是对化疗的(正)反应率被限制为20-35%。此外,由于人体的局限,治疗周期的数量通常被限制为最多4次。换而言之,在开始周期之前,从开始就确定合适的治疗方法是至关重要的。通过在治疗的早期阶段的识别,如果治疗被认为是有效的,治疗本身可以进行下去,并且如果治疗被认为是无效的,可以被跳过。在非有效的治疗的情况下,第二种治疗可以类似地启动。这种方法可以被重复额外数量的次数。一旦治疗被认为是有效的,化疗的周期可以启动。应当注意的是,该识别可以被重复用于每个和任何化疗。
使用本系统没有必要培养收集的肿瘤细胞,例如为了进行测试,这可能需要相对较长的时间,例如一周。该坏死细胞在它们的天然环境中被标记,这例如减少了人为错误的风险。
提供在上述方面的实时数据,而不需要进行穿刺来收集肿瘤细胞。在这方面应当注意的是通过穿刺仅仅获得肿瘤的一个样品。所获得的样品不一定是肿瘤的真实再现。还应当注意的是肿瘤是形态学上的非均质的。甚至进一步的,肿瘤可能随着时间而在形态特征上进展。其后果是,穿刺的样品至多给出了肿瘤特征的指示。本发明给出了实时和高度可靠的特征。
除了癌症之外,目前的疾病涉及例如心脏和大脑的梗死组织,骨关节炎,神经系统疾病如阿尔茨海默氏病。
本发明还涉及该组合物作为药物的应用;该系统在诊断方法中的应用,该系统在用于涉及坏死性细胞死亡的疾病治疗中的应用,该组合物在用于在体内和/或体外检测和/或靶向坏死细胞中的应用;涉及包括所述系统的剂量;涉及使用所述组合物确定样品内组合物的定位的方法;和,涉及使用所述组合物用于治疗涉及坏死性细胞死亡的癌症和/或疾病的方法。
本发明的靶向分子已经被发现非常有选择性的结合死亡(坏死)细胞。
如先前所述,本发明人已经发现坏死细胞和/或坏死通常是有吸引力的靶点。这样涉及坏死细胞区域的观测通常是存在于癌症(肿瘤)中,例如由于不充分的血液供应和因此缺乏氧气,并且在(或导致)涉及坏死性细胞死亡的疾病中。应当注意到坏死细胞区域典型地不会在健康组织内发现。涉及坏死性细胞死亡的疾病的实例包括脑梗塞或缺血性损伤(即心肌梗死,中风,肾,肝等),创伤(如脑,肌肉,骨)感染或炎症(即感染性休克,类风湿性关节炎,骨坏死),退行性疾病(即阿尔茨海默病,帕金森氏痴呆症),斑块(动脉粥样硬化,淀粉样蛋白)烧伤,辐射诱导的坏死和糖尿病。
至于癌症,一旦肿瘤已经被识别,进一步的坏死性细胞死亡可以在部分肿瘤上被有意诱导(例如通过局部辐照,光动力或局部热疗法和/或聚焦的超声波),以提供所述组合物的一个较大靶点。此外,本组合物被用于治疗目的,坏死细胞的数量将随着治疗的不断进展而增加从而导致因时间作用而剂量扩增。
对细胞死亡分类的一个有益讨论可以在Kroemer等人,细胞死亡和差异(CellDeath and Differentiation),2005年,12,1463中发现。在本申请中,坏死细胞被认为是已经失去细胞膜完整性的细胞。本领域技术人员能够确定细胞膜是否完全,即完整性,如通过使用荧光染料,如使用商业化的胺反应性染料。本发明的组合物对于死亡细胞的选择性,可以在体外试验中被证明,这将在下面的实施例中显示,所述死亡细胞即已经失去细胞膜完整性的细胞。重要的是注意结合于死亡细胞的胺活性染料仅在体外是有用的。在体内,它们会与例如血液中蛋白质或其它化合物的胺反应,并且将不能够用于靶向坏死细胞。

未活化的青色素是对如胺和硫醇非反应性的青色素。未活化的青色素不能通过共价键连接到呈现在细胞内的分子上的胺、硫醇或其它活性官能团而显著的(反应不是热力学有利的)结合死细胞(其分子的官能团)。也就是说,在本发明上下文中的选择性结合坏死细胞是不通过共价键,而是经由青色素核心结构通过非共价键结合,并且不是通过活性基团所连接的侧链而结合。术语活化的青色素是本领域技术人员所公知的,并且包括例如酯类,N-羟基琥珀酰亚胺酯,马来酰亚胺,酰基氯,SDS酯类等激活的羧酸。未活化的青色素包括如包含羧酸功能的青色素,即羧酸没有被激活。
应当注意的是活化的青色素可以在组合物的形成中使用,例如对于连接成像的和/或治疗的化合物至青色素的目的;然而在该组合物中,所述青色素是未活化的,并且通过青色素核心结构仍将结合坏死细胞。

靶向分子优选选自:HQ4、HQ5、HQ6、HQ7、ICG、CW 800、ZW800、L4、L7、L11、CY3、CY3b、CY3.5、CY5、CY5.5、CY7、Dy 676、Dy 681、Dy 731、Dy 751和Dy 776;而且,最优选选自HQ4,HQ5,CW800和ZW800。这些化合物在图2中显示。一些这些化合物具有例如非常好的荧光特性。这是相当出乎意料的,因化合物本身可能具有良好的荧光特性,但是其在结合到另一个分子时可能发生变化,所述另一个分子如所存在的蛋白质。

可以考虑的载体是用作提高药物的递送和效力的机制的物质。载体可以用于控释以降低药物的代谢、在体内延长药物的作用、和降低药物的毒性。本载体还用于提高药物递送到靶点位置效力,如药理作用的靶点。
在一个实例中,在体内的非反应性分子,其可以被称作点击A,是能够与配对物相互作用的,所述配对物可以被称作点击B。该非反应性分子优选是一种或多种第一点击化学组分,第二点击化学组分(互补于第一点击化学组分),和自标记蛋白质标签的组分,如Snap-Tag。优选的,点击化学组分“点击”而不需要如铜(I)的催化剂。

测量技术
在本次调查中,应用了一些现有的和/或内部开发的技术:
荧光
[维基百科:]“荧光是由已经吸收光或特定波长的其它电磁辐射的物质发射的光。在大多数情况下,所发射的光具有更长的波长,因此比所吸收的辐射具有较低能量。”
用荧光测量,所以也是体内荧光,很难被量化。在活的生物或细胞组织中所测量的强度是-除了荧光化合物本身的固有亮度和其浓度-也依赖该化合物的物理性质,即与它周围蛋白质的结合比和与它们相互作用的类型和贯穿组织的该物质的分布。
最后,测量的灵敏度和特异性是非常强烈的依赖于实验方案的。
干冰死细胞分析
在乙醇中的固态二氧化碳(-80℃)被用于冻结具有活细胞培养基(和可能还有其它组分)的24孔板的孔中心。已经通过使用具有针脚的冷却块而优化该步骤,所述冷却块被保持至24孔板的底部一定时间(大多约15秒)。其结果是冷冻和随之发生被仍然存活的细胞包围的孔中心的细胞死亡,从而能进行死细胞和活细胞之间的比较研究。

本发明的示例性系统
实施例1-参考图12
PLGA纳米微粒的制备。使用O/W乳液和溶剂蒸发-提取法制备具有包埋近红外荧光标记的PLGA纳米微粒。简而言之,在3mL DCM中的90mg PLGA含有近红外(NIR)700nm(680R从LI-COR)(3mg)被逐滴加入到25mL含2%(w/v)PVA的蒸馏水中,并且使用超声波仪乳化90秒(Branson,sonofier 250)。脂类(DSPE-PEG(2000)胺(8mg)和mPEG 2000PE(8mg))的组合溶解在DCM中,并且加入到小瓶中。经由氮气流除去DCM。随后,该乳液被迅速加入到含有脂类的小瓶中,并且使用超声波仪将溶液均质30秒。在4℃下溶剂蒸发过夜之后,经由在60000g下超速离心30分钟收集所述PLGA纳米微粒,用蒸馏水洗涤三次,并且冻干。
共轭CW800-NHS至PLGA纳米微粒。CW800-NHS共轭到含DSPE-PEG(2000)-胺的PLGA纳米微粒制剂上。随后,微粒(50mg)溶解在0.5mL的碳酸缓冲液(pH 8.5)中,并且该激活的CW800-NHS(1mg)在室温下1小时期间内加入到颗粒中。通过离心(10000g,在10分钟内)除去未结合的CW800-NHS并且用PBS洗涤PLGA纳米微粒-CW800四次。通过Odyssey扫描确定在微粒表面的CW800的数量(表1)。
动态光散射和Z电位。在所有纳米微粒上的动态光散射(DLS)测量是在一个ALV光散射仪器上进行的,该光散射仪器配备有ALV5000/60X0多头相关器和在532nm的波长下操作的Oxxius SLIM-532 150mW DPSS激光器。折射率与包围圆柱形散射池的过滤的顺式萘烷浴相匹配,并且使用Haake F3-K恒温器控制温度在21.5±0.3℃。对于各样品的自相关函数,g2(τ),以90°检测角记录10次。对于每个测量的扩散系数,D,使用2阶累积而测量,并且假定粒子是球形的而计算出相应的粒径(参见表1)。在Malvern ZetaSizer 2000(英国)上测定所有纳米微粒的Z电位。

DTPA-PEG-NH2的合成
在Cl-TrtCl树脂上合成含有PEG胺连接的DTPA(4,7,10-三氧杂-1,13-十三烷二胺,在式(DTPA-PEG-NH2)中表示为PEG-NH2)。因此,在含6当量DIEA的DCM存在下,通过在室温下3当量Fmoc-PEG胺反应过夜,Fmoc-PEG胺被并入到Cl-TrtCl树脂(CTC树脂)上。通过Fmoc定量而测定最终负荷:所得的值是约0.8毫摩尔/克。用哌啶-DMF(1∶5)(1×1分钟,2×10分钟)除去Fmoc基。接着,使用DIPCDI(2当量)和HOBt(2当量)使DTPA-(四-叔丁基酯)-COOH(2当量)在DMF中过夜偶联。在偶联过夜之后,茚三酮测试呈阴性,随后,在两个步骤中实施DTPA-(四-叔丁基酯)-CO-NH-PEG-CTC-树脂裂解和脱保护。用含1%三氟乙酸(TFA)的DCM处理DTPA-(四-叔丁基酯)-CO-NH-PEG-CTC-树脂10次,每次1分钟。使用真空除去过量的DCM,并且使用95%TFA,2.5%三异丙基硅烷(TIS)和2.5%的水除去侧链保护基团。除去TFA后,在N2气流中用冷的MTBE沉淀DTPA-CO-NH-PEG-NH2。在水中溶解DTPA-CO-NH-PEG-NH2并且冻干以获得最终产品。通过HPLC(保留时间(tR)是2.28分钟)进行分析,所期望的DTPA-CO-NH-PEG-NH2产率为85.0%,纯度为90.6%。HPLC-MS,m/z计算值:对于C20H37N5O11为523.25,实测值:524.5.28[M+1]+和MALDI-TOF实测值524.2[M+1]+546.3[M+Na]。
实施例3-参考图14
DTPA-CO-NH-PEG-NH2-HQ4的合成。
将溶解于50μL二甲基亚砜(DMSO)的HQ4-NHS(8.3x10-4毫摩尔,1毫克)添加到溶解于含有5μL DIEA的200μL DMSO中的DTPA-CO-NH-PEG-NH2(2.8×10-3毫摩尔,2毫克),并且在室温下搅拌过夜。随后,通过RP-HPLC纯化该复合的DTPA-CO-NH-PEG-HQ4。通过HPLC(tR5.34)进行分析,所期望的Dota-PEG-HQ5的产率为50.5%,纯度为98%。HPLC-MS,m/z计算值:对于C66H90N8O17S2为1331.59,实测值:1332.0[M+1]+和MALDI-TOF实测值1331.9[M+1]+1353.9[M+Na]。

8.根据权利要求1的用途,其中该药物组合物的剂量包括1-250纳摩尔系统/kg体重。
9.根据权利要求1的用途,其中该药物组合物的剂量包括2-100纳摩尔系统/kg体重。
