Abstract

技术领域
本发明涉及包含粘结剂或粘合剂的材料的领域。更具体地,糖浆作为木质纤维素生物质发酵过程的联产品有效地用作粘结剂或粘合剂。
背景技术
粘结剂和粘合剂在许多应用中特别是在具有期望性能的材料因其颗粒性质而难以处理时是有用的。用作粘结剂的物质(例如具有细煤粉的物质)包括淀粉和糖蜜(US 5,009,671)、沥青粘结剂材料(US 823,025)、沥青粘结剂(US 3,966,427)、或液化生物质(US5,916,826)。另外,将木质素磺酸盐即木材的硫酸盐制浆工艺的废液用作各种材料的粘结剂和粘合剂。
在将木质纤维素生物质用作用于发酵的碳源的纤维素乙醇方法中,通常将来自蒸馏塔(啤酒塔)的全塔馏物分离为固体(湿饼或滤饼)和液体(稀塔馏物)馏分。使稀塔馏物通过蒸发器以产生糖浆。滤饼和糖浆是纤维素乙醇方法的联产品。如在共同拥有的US20120102823中所公开的,可燃烧具有至少约40%固体的糖浆,由此提供能量。也可燃烧滤饼以提供能量。
仍然需要可有效地用作粘结剂和粘合剂的另外材料,以及使用这些材料的方法。
发明内容
在一个方面,提供一种用于粘结颗粒物质的方法,该方法包括:
a)提供木质纤维素糖浆;
b)提供至少一种颗粒物质;
c)使b)的颗粒物质与a)的糖浆接触;
其中所述糖浆粘结所述颗粒物质。
在另一方面,提供了一种用于使两个表面粘合的方法,该方法包括:将包含木质纤维素糖浆的组合物施用于至少一个表面,以及使两个表面彼此接触,使得木质纤维素糖浆置于两个表面之间。

术语“能源甘蔗”是指培育用于产生能量的甘蔗。其因纤维百分比高于糖而被选择。
术语“木质纤维素生物质水解产物”是指木质纤维素生物质糖化产生的产物。生物质也可在糖化前进行预处理或预加工。
术语“木质纤维素生物质水解产物发酵液”是指包含产物的发酵液,所述产物得自包含木质纤维素生物质水解产物的介质中的生物催化剂生长和生产。所述发酵液包含木质纤维素生物质水解产物的成分,所述成分为没有被生物催化剂消耗的,以及生物催化剂本身和生物催化剂产生的产品。

术语“糖浆”或“木质纤维素糖浆”可互换使用并且意指浓缩的产物,其由稀塔馏物去除水而产生,一般来讲为蒸发。
术语“目标产物”是指任何由发酵中的微生物生产宿主细胞生成的产物。目标产物可为宿主细胞中经遗传工程改造的酶途径的产物或可由内源性途径产生。典型的目标产物包括但不限于:酸、醇、烷烃、烯烃、芳烃、醛、酮、生物聚合物、蛋白质、肽、氨基酸、维生素、抗生素和药物。
术语“发酵”是指广义地使用生物催化剂来产生目标产物。通常生物催化剂在发酵液中利用发酵液中的碳源生长,并且通过其代谢产生目标产物。
“固体”是指可溶固体和不可溶固体。来自木质纤维素发酵过程的固体包含用于制备水解产物培养基的木质纤维素生物质的残余物。
本文“挥发物”是指在引入热的过程中将较大程度气化的组分。通过确定严格受控条件下由加热至950℃所引起的重量损失来测量本文挥发物含量(如在ASTM D-3175中)。典型的挥发物包括但不限于氢气、氧气、氮气、乙酸以及一些碳和硫。
本文“固定碳”是指通过将水分百分比、灰分百分比、和挥发物百分比相加,然后用100减去该百分比得到的计算百分比。
“灰分”是根据ASTM D-3174,在受控条件下燃烧后剩余的残余物的重量。
如木质纤维素糖浆组合物中所提及的“糖”意指单糖和可溶性低聚糖的总和。
木质纤维素糖浆的制备和组合物
本发明的木质纤维素糖浆作为来自于使用木质纤维素生物质的过程的联产品而产生,所述木质纤维素生物质作为可用作生物催化剂的碳源的可发酵糖的来源。生物催化剂在发酵过程中利用糖产生目标产物。
为了从木质纤维素生物质产生可发酵糖,对生物质进行处理以释放糖诸如来自于生物质多糖的葡萄糖、木糖和阿拉伯糖。本领域技术人员可用任何已知的方法处理木质纤维素生物质,以在水解产物中产生可发酵糖。通常,所述生物质使用物理的、热的和/或化学的处理进行预处理,并酶解糖化。热-化学预处理方法包括蒸汽喷发或使生物质溶胀以释放糖的方法(参见例如WO2010113129;WO2010113130)。也可以采用化学糖化。用于预处理生物质的物理处理包括但不限于磨削、研磨和切割。诸如这些的物理处理可用于在进一步化学处理前减小粒度。化学处理包括碱处理诸如用强碱(氨或NaOH)或酸处理(US8545633;WO2012103220)。在一个实施方案中,用氨处理生物质(US 7932063;US 7781191;US7998713;US7915017)。这些处理从生物质释放聚合糖。低氨预处理是特别有用的,其中生物质与包含氨的水溶液接触,形成生物质-氨水混合物,其中氨的浓度足够保持生物质-氨水混合物的碱性pH,但相对于生物质的干重小于约12重量%,并且相对于生物质-氨水混合物的重量,生物质的干重为至少约15重量%的固体,如US 7,932,063所公开的,其以引用方式并入本文。
将聚合糖转化为单体糖的糖化可以通过酶处理或化学处理进行。在一个方面,使经预处理的生物质与糖化酶聚生体在合适的条件下接触,以产生可发酵糖。在糖化之前,可使经预处理的生物质达到期望的水分含量并对其进行处理以改变pH、组合物或温度使得糖化酶聚生体中的酶将有活性。可通过添加固体或液体形式的酸改变pH。另选地,可利用可从发酵中回收的二氧化碳(CO2)来降低pH。例如,如果存在足够的液体,可从发酵罐中收集CO2并将其进料到闪蒸槽中的预处理产物顶部空间或起泡通过经预处理的生物质,同时监控pH,直至达到所需的pH。可使温度达到下文提到的适合糖化酶活性的温度。通常,合适的条件可包括在约40℃至50℃的温度以及在约4.8至5.8的pH。
纤维素或木质纤维素生物质的酶促糖化通常利用酶组合物或共混物来降解纤维素和/或半纤维素并且产生包含糖(诸如例如葡萄糖、木糖和阿拉伯糖)的水解产物。糖化酶参见Lynd,L.R.等人(“Microbiol.Mol.Biol.Rev.,66:506-577,2002)。使用至少一种酶,并且通常使用糖化酶共混物,其包含一种或多种糖苷酶。糖苷酶水解二糖、低聚糖和多糖的醚键,并且存在于广义“水解酶”(EC 3.)的酶分类EC 3.2.1.x(Enzyme Nomenclature 1992,Academic Press,San Diego,CA,以及增补1(1993)、增补2(1994)、增补3(1995)、增补4(1997)和增补5[分别在Eur.J.Biochem.,223:1-5,1994;Eur.J.Biochem.,232:1-6,1995;Eur.J.Biochem.,237:1-5,1996;Eur.J.Biochem.,250:1-6,1997;以及Eur.J.Biochem.”,264:610-6501999中])中。可用于糖化的糖苷酶可通过它们水解的生物质组分进行分类。可用于糖化的糖苷酶可包括纤维素水解糖苷酶(例如,纤维素酶、内切葡聚糖酶、外切葡聚糖酶、纤维二糖水解酶、β-葡糖苷酶)、半纤维素水解糖苷酶(例如,木聚糖酶、内切木聚糖酶、外切木聚糖酶、β-木聚糖苷酶、阿拉伯糖基木聚糖酶、甘露聚糖酶、半乳糖酶、果胶酶、葡糖苷酸酶)和淀粉水解糖苷酶(例如,淀粉酶、α-淀粉酶、β-淀粉酶、葡糖淀粉酶、α-葡糖苷酶、异淀粉酶)。此外,将其它活性物质加入糖化酶聚生体(诸如肽酶(EC 3.4.x.y)、脂肪酶(EC3.1.1.x和3.1.4.x)、木质素酶(EC 1.11.1.x)或阿魏酸酯酶(EC3.1.1.73))中以促进从生物质的其它组分中释放多糖可为有用的。本领域熟知生产多糖水解酶的微生物常常表现出某种活性,诸如降解纤维素的能力,该活性由具有不同底物特异性的若干种酶或一组酶催化。因此,来自微生物的“纤维素酶”可包括一组酶、可有助于纤维素降解活性的一种或多种酶或所有酶。取决于获取酶时利用的纯化方案,商业或非商业酶制剂(诸如纤维素酶)可包括多种酶。对于糖化有用的许多糖基水解酶和它们的组合物公开于WO 2011/038019中。用于糖化的另外的酶包括例如糖基水解酶,其水解两个或更多个碳水化合物之间的糖苷键,或碳水化合物与非碳水化合物部分之间的糖苷键。
糖化酶可商购获得。此类酶包括例如CP纤维素酶、木聚糖酶、1500、DUET、以及TrioTM(DupontTM/Wilmington,DE),以及Novozyme-188(Novozymes,2880 Bagsvaerd,Denmark)。此外,糖化酶可以是未经纯化的并以细胞提取物或全细胞制剂的形式提供。可使用已经工程化以表达一种或多种糖化酶的重组微生物来制备所述酶。例如,可用于使经预处理纤维素类生物质糖化的H3A蛋白质制备物是由里氏木霉(Trichoderma reesei)的遗传工程菌株生产的酶的未纯化的制备物,其包括纤维素酶和半纤维素酶的组合,并且描述于WO 2011/038019中,该文献以引用方式并入本文。
可使用化学糖化处理并且其对于本领域的技术人员是已知的,诸如用无机酸包括HCl和H2SO4处理(US5580389;WO2011002660)。
通过木质纤维素生物质的糖化来释放糖诸如葡萄糖、木糖和阿拉伯糖,并且这些单体糖提供用于生物催化剂(用于发酵过程)的碳水化合物源。糖存在于用作发酵培养基的生物质水解产物中。发酵培养基可仅由水解产物构成,或可包含水解产物之外的组分,诸如山梨醇或甘露糖醇,最终浓度为约5mM,如US 7,629,156中所述,其以引用方式并入本文。生物质水解产物通常占发酵培养基的至少约50%。通常发酵液终体积的约10%是包含生物催化剂的种菌。
在发酵罐中发酵包含水解产物的培养基,所述发酵罐为任何容纳水解产物发酵培养基和至少一种生物催化剂并且具有阀门、通气孔、和/或用于管理发酵过程的端口的容器。
天然地或通过基因工程利用葡萄糖且优选地还利用木糖生产目标产物的任何生物催化剂可用于木质纤维素生物质制成的生物质水解产物中可发酵糖的发酵。可通过发酵生产的目标产物包括例如酸、醇、烷烃、烯烃、芳烃、醛、酮、生物聚合物、蛋白质、肽、氨基酸、维生素、抗生素和药物。醇包括但不限于甲醇、乙醇、丙醇、异丙醇、丁醇、乙二醇、丙二醇、丁二醇、甘油、赤藓醇、木糖醇、甘露糖醇和山梨醇。酸可包括乙酸、甲酸、乳酸、丙酸、3-羟基丙酸、丁酸、葡萄糖酸、衣康酸、柠檬酸、琥珀酸、3-羟基丙酸、富马酸、马来酸和乙酰丙酸。氨基酸可包括谷氨酸、天冬氨酸、甲硫氨酸、赖氨酸、甘氨酸、精氨酸、苏氨酸、苯丙氨酸和酪氨酸。附加的目标产物包括甲烷、乙烯、丙酮和工业酶。
生物质水解产物中的糖转化为目标产物的发酵可通过一种或多种能够在包含生物质水解产物的培养基中生长的合适的生物催化剂在一步或多步发酵过程中进行。生物催化剂可以是选自细菌、丝状真菌和酵母的微生物。生物催化剂可为野生型微生物或重组微生物,并且可包括例如属于埃希氏菌属(Escherichia)、发酵单胞菌属(Zymomonas)、酵母属(Saccharomyces)、假丝酵母属(Candida)、毕赤酵母属(Pichia)、链霉菌属(Streptomyces)、芽胞杆菌属(Bacillus)、乳酸杆菌属(Lactobacillus)和梭菌属(Clostridiuma)的生物体。通常,生物催化剂的示例包括重组大肠杆菌(Escherichiacoli)、运动发酵单胞菌(Zymomonas mobilis)、嗜热脂肪芽胞杆菌(Bacillusstearothermophilus)、酿酒酵母(Saccharomyces cerevisiae)、嗜热梭菌(Clostridiathermocellum)、高温产氢菌(Thermoanaerobacterium saccharolyticum)、以及树干毕赤酵母(Pichia stipitis)。为生长良好和在木质纤维素生物质水解产物发酵液中有高的产物产量,可选择或工程化生物催化剂以对生物质水解产物中存在的抑制剂(诸如乙酸盐)有更高的耐受性。例如,生物催化剂可产生作为目标产物的乙醇,诸如通过运动发酵单胞菌产生乙醇,如在US 8,247,208中所描述,其以引用方式并入本文。
发酵在适用于所用特定生物催化剂的条件下进行。可对条件诸如pH、温度、氧含量和混合进行调节。用于酵母和细菌生物催化剂的发酵的条件在本领域中是已知的。
此外,糖化和发酵可在同一容器中同时发生,这称为同步糖化和发酵(SSF)。此外,在称为HSF(混合糖化和发酵)的过程中,在糖化和发酵并发的时间段之前可发生局部糖化。
对于较大规模的发酵,通常较少的生物催化剂培养物首先生长,其称为种子培养物。将种子培养物作为种菌添加至发酵培养基中,通常在约2%至约20%的终体积的范围内。
通常通过生物催化剂发酵产生包含由生物催化剂制得的目标产物的发酵液。例如,在乙醇方法中,发酵液可为包含约6%至约10%的乙醇的发酵醪。除目标产物之外,发酵液还包含水、溶质和固体,它们得自水解产物培养基以及得自水解产物培养基中糖的生物催化剂代谢。通常使目标产物与发酵液分离,产生枯竭的发酵液(可称为全塔馏物)。例如,当乙醇为产物时,通常使用啤酒塔蒸馏发酵液,以产生乙醇产物流和全塔馏物。蒸馏可使用本领域技术人员已知的任何条件,包括在常压或减压下。使馏出的乙醇进一步通过精馏塔和分子筛以回收乙醇产物。可另选地在随后步骤中去除目标产物,诸如在分离发酵液之后从固体或液体馏分中去除。
木质纤维素生物质发酵过程的糖浆联产品由发酵液或枯竭的发酵液制得。制备糖浆的示例公开于共同拥有的US20120102823,其以引用方式并入本文。发酵液或枯竭的发酵液,诸如全塔馏物,分离为固体和液体流,其中液体流被称为稀塔馏物。可使用多种过滤设备,诸如带束过滤器、压带机、螺杆式压缩机、转筒过滤器、碟式过滤器、Nutsche过滤器、压力过滤机和离心过滤器。可通过诸如施用真空、压力或离心力来辅助过滤。为了提高过滤的效率可使用热处理,如公开在共同拥有的和共同未决的US20120178976中,其以引用方式并入本文。
在对木质纤维素生物质水解产物发酵液或枯竭的发酵液进行液体/固体分离后,固体馏分或滤饼(也称为湿饼)可以燃烧为生产过程提供能量。所述滤饼可在燃烧前干燥,诸如通过空气干燥,以降低水分。
在对木质纤维素生物质水解产物发酵液进行液体/固体分离后,去除产物流。例如,液体流可萃取或蒸馏以产生产物流,诸如蒸馏产生乙醇产物流和剩余液体。
液体馏分通过蒸发而进一步纯化以产生能回收的水和糖浆。在蒸发之前,液体馏分的一部分可回收作为涡流使用,其可添加到过程中需要水的任何一点,诸如在预处理、糖化或生物催化剂种子制备中。蒸发可在任何蒸发系统中进行,诸如降膜、升膜、压力环流、板式或机械和热蒸汽再压缩系统。蒸发可为连续的或分批的,并可用多效蒸发器。蒸发的水可再循环用于木质纤维素生物质水解产物发酵总体过程。
蒸发后剩余的物质是糖浆,其为本发明的木质纤维素糖浆。木质纤维素糖浆组合物包含约40%至约70%的固体或约40%至约60%的固体(可具有约40%、45%、50%、55%、60%、65%或70%的固体)、约10g/l至30g/l的乙酰胺、至少约40g/l的糖、约1g/cm3至约2g/cm3的密度和在100°F(38℃)下小于500 SSU的粘度。“SSU”是以秒计的赛波特通用粘度(Burger VL.,Encycl.Ind.Chem.Anal.(1966),第3卷,768-74)。可调制蒸发程度来实现期望的固体含量。当用于制备生物质(用于糖化)的预处理过程是采用氨的过程时,木质纤维素糖浆包含至少约5g/l的氨。

使木质纤维素糖浆以任意量与颗粒物质混合,其中糖浆起到颗粒物质的粘结剂的作用。在各种实施方案中,糖浆按糖浆颗粒物质的最终组合的重量计为约0.1%至约20%。糖浆可为总合并重量的约0.1%、0.5%、1%、2%、4%、6%、8%、10%、12%、14%、16%、18%或20%。在各种实施方案中,糖浆为糖浆和颗粒物质的最终组合物的约0.1%至20%、0.5%至20%、0.5%至10%、0.5%至5%、2%至20%、或2%至10%。
在各种实施方案中,颗粒物质为炭黑粉末、粉煤灰、或动物饲料成分。炭黑为通常由重质石油产品诸如各种类型的焦油的不完全燃烧所产生的粉末形式的碳。其为具有高表面积与体积比的次晶碳。炭黑被用作轮胎及其它橡胶产品的增强填料和着色剂,以及塑料、油漆、色淀、墨和皮革的彩色颜料,和电气设备的绝缘材料。将炭黑与木质纤维素糖浆粘结产生更易于处理和运输的组合物。该组合物可用于制备轮胎以及用于以上所列的其它应用。
粉煤灰是发电厂中燃烧煤粉的联产品。其由燃烧期间所熔合的煤中的矿物质杂质诸如粘土、长石、石英和页岩构成。该物质存在于废气中。其冷却并硬化,并且通过静电除尘器或袋式过滤器收集。粉煤灰具有水泥质性质并且用作水泥中的添加剂。将粉煤灰与木质纤维素糖浆粘结产生更易于处理和运输的组合物。该组合物可用于制备混凝土用的水泥。
饲料丸粒是动物饲料组分与将组分保持在一起的粘结剂的组合物。饲料组分可包括此类成分如谷物、大豆、磨粉联产品、添加的维生素、矿物质、脂肪/油。谷物及其它组分通常锤磨为颗粒形式以用于饲料丸粒。将动物饲料组分与木质纤维素糖浆粘结产生更易于处理和运输的组合物。
可对木质纤维素糖浆与颗粒物质的混合物进行处理以形成便于处理的固体物质。处理可包括诸如加热、压缩、挤出、粒化、模塑和/或干燥这些处理。可形成各种形状诸如压块、丸粒、不规则形状等等。例如,可在高温下使混合物通过挤出机,然后通过制粒机后干燥,以形成硬化的燃料丸粒。可使用通过模头推动或抽吸半软质固体的任何类型的如本领域技术人员所知的挤出机。挤出可在高温和/或高压下进行。制粒机可用于减小木质纤维素糖浆和颗粒物质组合物的体积并增大其密度,其可单独使用或与挤出结合使用,并且从而使材料成形为丸粒。压块可采用螺旋压机或其它压缩设备形成。在成形后,使组合物干燥,以形成固体木质纤维素糖浆组合物。根据用于制备本发明的固体组合物的起始物和方法,水分可小于约30%、25%、20%、15%、10%或更小。
在一个实施方案中,木质纤维素糖浆用于接触颗粒物质。所述物质可包括任何粒状的物质,诸如为粉末状、尘状或颗粒状。在一个实施方案中,颗粒物质不是可燃物质或土壤调理剂。在一个实施方案中,将木质纤维素糖浆施用于覆盖表面诸如未铺设的道路、土路、苗圃、果园、饲养场、或施工区的颗粒物质。木质纤维素糖浆可施用于任何有灰尘的表面,使得糖浆用于控制粉尘。用于木质纤维素糖浆应用的表面可在与糖浆接触之前首先分级并任选地湿润。可在应用前使糖浆与土壤或其它可铺展物质混合。通常以0.3-0.5加仑每平方码的速度施用糖浆。糖浆的作用在于使路面颗粒粘结在一起,并且随着水蒸发,粉尘颗粒仍存在由粉尘化所引起的团聚最小化问题。
作为粘合剂的木质纤维素糖浆
在各种实施例中,木质纤维素糖浆可用作粘合剂,或粘合剂的组分。糖浆可部分地替换苯酚-甲醛粘合剂中的苯酚。可以干燥的、潮湿的、或液体形式使用包含木质纤维素糖浆的粘合剂。包含木质纤维素糖浆的粘合剂可为增塑剂。木质纤维素糖浆粘合剂可用于一些应用,诸如刨花板、胶合板、将托盘化袋点状胶结在一起以防止它们在储存、处理和装运期间移动、油毡水泥(inoleum cements)、可再润湿的胶带和托盘化容器。
Claims (16)
1.一种用于粘结颗粒物质的方法,所述方法包括:
a)提供木质纤维素糖浆;
b)提供至少一种颗粒物质,使所述(b)的颗粒物质与所述(a)的糖浆接触;
其中所述糖浆粘结所述颗粒物质。
2.根据权利要求1所述的方法,其中所述(c)的接触是使所述(a)的糖浆与所述(b)的颗粒物质混合。
3.根据权利要求2所述的方法,其中所述颗粒物质选自动物饲料成分、炭黑、和粉煤灰。
4.根据权利要求1所述的方法,其中所述(c)的接触是将所述(a)的糖浆施用于所述(b)的颗粒物质。
5.根据权利要求4所述的方法,其中所述(b)的颗粒物质覆盖道路、公路、苗圃、果园、或施工区。
6.一种用于使两个表面粘合的方法,所述方法包括:a)将包含木质纤维素糖浆的组合物施用于至少一个表面;以及b)使所述两个表面彼此接触,使得所述木质纤维素糖浆置于所述两个表面之间。
7.根据权利要求6所述的方法,其中所述组合物选自水泥和可再润湿的胶带。
8.根据权利要求1或6所述的方法,其中所述木质纤维素糖浆是由木质纤维素生物质制备醇的过程的联产品。
9.根据权利要求8所述的方法,其中所述木质纤维素生物质选自玉米棒、玉米秸秆、草、小麦秸秆、大麦秸秆、干草、稻秆、柳枝稷、废纸、甘蔗渣、高粱植物材料、大豆植物材料、木本植物、蔬菜、水果和花。
10.根据权利要求1或6所述的方法,其中所述木质纤维素糖浆是采用氨来预处理木质纤维素生物质的过程的联产品。
11.根据权利要求10所述的方法,其中所述木质纤维素糖浆包含:
a)约40%至约70%的固体;
b)约10g/l至约30g/l的乙酰胺;和
c)至少约40g/l的糖;
其中所述纤维素糖浆具有约1g/cm3至约2g/cm3的密度和在100°F(38℃)下小于500SSU的粘度。
12.一种组合物,所述组合物包含木质纤维素糖浆和至少一种附加颗粒物质。
13.根据权利要求12所述的组合物,其中所述木质纤维素糖浆是由木质纤维素生物质制备醇的过程的联产品。
14.根据权利要求13所述的组合物,其中所述木质纤维素生物质选自玉米棒、玉米秸秆、草、小麦秸秆、大麦秸秆、干草、稻秆、柳枝稷、废纸、甘蔗渣、高粱植物材料、大豆植物材料、木本植物、蔬菜、水果、花、棕榈空果串、和能源甘蔗。
15.根据权利要求12所述的组合物,其中所述木质纤维素糖浆是采用氨来预处理木质纤维素生物质的过程的联产品。
16.根据权利要求15所述的组合物,其中所述糖浆包含:
a)约40%至约70%的固体;
b)约10g/l至约30g/l的乙酰胺;和
c)至少约40g/l的糖;
其中所述纤维素糖浆具有约1g/cm3至约2g/cm3的密度和在100°F(38℃)下小于500SSU的粘度。
