Abstract

本发明提供了一种防治骨质疏松症的中药制剂及其制备方法,所述中药制剂由淫羊藿、桑寄生、怀牛膝、延胡索、女贞子、木香、当归、苍术、海螵蛸经水提醇沉制成,其中淫羊藿、桑寄生补肝肾、强筋骨,为君药,怀牛膝、延胡索活血通经、行气止痛,为臣药,辅以女贞子、木香、当归、苍术、海螵蛸滋阴健脾、补血理气,对骨质疏松症有显著疗效,而且毒副作用小,安全系数高,长期服用能明显提高患者的骨密度和骨强度。

一种防治骨质疏松症的中药制剂及其制备方法
技术领域
本发明涉及一种防治骨质疏松症的中药制剂及其制备方法,属中药应用领域。
背景技术
骨质疏松症是一种老年人中常见的退行性病变,也是绝经后女性中的多发病。导致该病症的原因复杂多样,在病理学上,以骨量减少、骨组织的显微结构发生改变,以松质骨骨小梁变细、断裂、数量减少,皮质骨多孔和变薄为特征。在临床上,主要表现为全身骨痛、脊柱畸形甚至骨折。目前,防止骨质疏松症的西药主要有雌激素、降钙素、钙制剂及维生素D类药物等。虽然疗效确切,但是长期服用易于引起各种副作用,给患者的生活带来极大的负面影响。
中医学根据骨质疏松症的症状将其分为“骨痿”、“骨痹”、“骨枯”、“骨极”、“骨折”、“绝经前后诸”、“虚劳”或“腰背痛”等。在中医理论中,认为骨质疏松症主要与肾虚、脾虚、气血亏虚和肝血不足有关。“肾藏精、主骨,藏真经而寓元阳,为先天之本”,肾虚是其发病的重要病理基础。肝脾也是骨痿发病的影响因素之一,血瘀则能加重病情,因此补肾壮骨、调理肝脾、活血化瘀为其治疗原则。例如,中国专利CN 201110084454.8公开了一种由地黄、怀牛膝、杜仲、肉苁蓉、淫羊藿、当归、鸡血藤、木香、龟甲、牡蛎、泽泻制成的治疗骨质疏松症的中药组合物。又如中国专利CN 201210079204.X公开了一种由补骨脂、淫羊藿、黄精、骨碎补、枸杞子、怀牛膝制成的具有治疗骨质疏松功能的中药组合物。再如,中国专利CN201310176471.3公开了一种以淫羊藿、骨碎补、怀牛膝制成的增加骨密度的中药组合物。
发明内容
为解决现有技术中的不足,本发明提供了一种防治骨质疏松症的中药制剂及其制备方法。
为解决以上技术问题,一方面,本发明提供了一种防治骨质疏松症的中药制剂。所述中药制剂由以下重量份数的原料药制成:
淫羊藿25-35份,桑寄生15-30份,怀牛膝15-25份,延胡索15-25份,女贞子10-20份,木香10-20份,当归10-20份,苍术10-20份,海螵蛸10-20份。
在本发明的一个优选的实施方案中,所述中药制剂由以下重量份数的原料药制成:
淫羊藿30-35份,桑寄生20-25份,怀牛膝15-20份,延胡索20-25份,女贞子15-20份,木香10-15份,当归10-15份,苍术15-20份,海螵蛸15-20份。
在本发明的另一个优选的实施方案中,所述中药制剂由以下重量份数的原料药制成:
淫羊藿32份,桑寄生24份,怀牛膝18份,延胡索20份,女贞子15份,木香12份,当归12份,苍术15份,海螵蛸15份。
在本发明的另一个优选的实施方案中,所述中药制剂是胶囊剂、片剂或粉剂。
另一方面,本发明提供了所述中药制剂的制备方法,包括以下步骤:
(1)按照原料药重量份数称取中药材,净选;
(2)海螵蛸粉碎成细粉,过200-300目筛,备用;
(3)淫羊藿、桑寄生、怀牛膝、延胡索、女贞子、木香、当归、苍术,加9-12倍量水,加热煎煮2-4次,每次2-4小时,过滤,合并滤液,浓缩至滤液体积的二分之一,加入95%的乙醇直至含醇量为60%-75%为止,静置24-36小时,离心,取上清液,过滤,将滤液减压浓缩至稠浸膏,置真空干燥箱50-60℃充分干燥,得干浸膏,将干浸膏粉碎成细粉,过200-300目筛,备用;
(4)将步骤(2)所得细粉与步骤(3)所得的干膏粉合并,即为所述中药制剂的活性成分;
(5)在步骤(4)所得的活性成分中加入药学上可接受的辅料,采用常规的制剂方法,制成药剂学上可接受的剂型。
为使上述剂型能够实现,需在制备这些剂型时加入药学可接受的辅料,例如:填充剂、崩解剂、润滑剂、助悬剂、粘合剂、甜味剂、矫味剂、防腐剂、基质等。填充剂包括:淀粉、预胶化淀粉、乳糖、甘露醇、甲壳素、微晶纤维素、蔗糖等;崩解剂包括:淀粉、预胶化淀粉、微晶纤维素、羧甲基淀粉钠、交联聚乙烯吡咯烷酮、低取代羟丙纤维素、交联羧甲基纤维素钠等;润滑剂包括:硬脂酸镁、十二烷基硫酸钠、滑石粉、二氧化硅等;助悬剂包括:聚乙烯吡咯烷酮、微晶纤维素、蔗糖、琼脂、羟丙基甲基纤维素等;粘合剂包括,淀粉浆、聚乙烯吡咯烷酮、羟丙基甲基纤维素等;甜味剂包括:糖精钠、阿斯帕坦、蔗糖、甜蜜素、甘草次酸等;矫味剂包括:甜味剂及各种香精;防腐剂包括:尼泊金类、苯甲酸、苯甲酸钠、山梨酸及其盐类、苯扎溴铵、醋酸氯乙定、桉叶油等;基质包括:PEG6000,PEG4000,虫蜡等。为使上述剂型能够实现中药药剂学,需在制备这些剂型时加入药学可接受的其它辅料(范碧亭《中药药剂学》,上海科学出版社1997年12月第1版中各剂型记载的辅料)。
另一方面,本发明提供了所述中药制剂在制备治疗骨质疏松症的药物中的应用。
本发明的中药制剂中,淫羊藿、桑寄生补肝肾、强筋骨,为君药,怀牛膝、延胡索活血通经、行气止痛,为臣药,辅以女贞子、木香、当归、苍术、海螵蛸滋阴健脾、补血理气,对骨质疏松症有显著疗效,而且毒副作用小,安全系数高,长期服用能明显提高患者的骨密度和骨强度。
具体实施方式
通过以下实施方式进一步详细说明本发明的技术方案。需要指出的是,以下说明仅仅是对本发明要求保护的技术方案的举例说明,并非对这些技术方案的任何限制。本发明的保护范围以所附权利要求书记载的内容为准。
实施例1:
原料药:
淫羊藿32份,桑寄生24份,怀牛膝18份,延胡索20份,女贞子15份,木香12份,当归12份,苍术15份,海螵蛸15份;
制备方法为:
(1)按照原料药重量份数称取中药材,净选;
(2)海螵蛸粉碎成细粉,过200目筛,备用;
(3)淫羊藿、桑寄生、怀牛膝、延胡索、女贞子、木香、当归、苍术,加10倍量水,加热煎煮3次,每次2.5小时,过滤,合并滤液,浓缩至滤液体积的二分之一,加入95%的乙醇直至含醇量为75%为止,静置24小时,离心,取上清液,过滤,将滤液减压浓缩至稠浸膏,置真空干燥箱55℃充分干燥,得干浸膏,将干浸膏粉碎成细粉,过200目筛,备用;
(4)将步骤(2)所得细粉与步骤(3)所得的干膏粉合并,充分混合,加入适量淀粉浆制软材,过20目筛制粒,将湿颗粒于55℃干燥,整粒,装入胶囊壳中,即得胶囊剂。
实施例2:
原料药:
淫羊藿30份,桑寄生20份,怀牛膝15份,延胡索18份,女贞子18份,木香10份,当归18份,苍术20份,海螵蛸10份;
制备方法为:
(1)按照原料药重量份数称取中药材,净选;
(2)海螵蛸粉碎成细粉,过300目筛,备用;
(3)淫羊藿、桑寄生、怀牛膝、延胡索、女贞子、木香、当归、苍术,加11倍量水,加热煎煮3次,每次3小时,过滤,合并滤液,浓缩至滤液体积的二分之一,加入95%的乙醇直至含醇量为70%为止,静置30小时,离心,取上清液,过滤,将滤液减压浓缩至稠浸膏,置真空干燥箱60℃充分干燥,得干浸膏,将干浸膏粉碎成细粉,过300目筛,备用;
(4)将步骤(2)所得细粉与步骤(3)所得的干膏粉合并,充分混合,加入适量淀粉浆制软材,过16目筛制粒,将湿颗粒于60℃干燥,整粒,装入胶囊壳中,即得胶囊剂。
实施例3:
原料药:
淫羊藿25份,桑寄生18份,怀牛膝15份,延胡索15份,女贞子10份,木香10份,当归15份,苍术10份,海螵蛸12份;
制备方法为:
(1)按照原料药重量份数称取中药材,净选;
(2)海螵蛸粉碎成细粉,过200目筛,备用;
(3)淫羊藿、桑寄生、怀牛膝、延胡索、女贞子、木香、当归、苍术,加9倍量水,加热煎煮3次,每次3小时,过滤,合并滤液,浓缩至滤液体积的二分之一,加入95%的乙醇直至含醇量为65%为止,静置36小时,离心,取上清液,过滤,将滤液减压浓缩至稠浸膏,置真空干燥箱55℃充分干燥,得干浸膏,将干浸膏粉碎成细粉,过200目筛,备用;
(4)将步骤(2)所得细粉与步骤(3)所得的干膏粉合并,充分混合,加入适量淀粉浆制软材,过20目筛制粒,将湿颗粒于60℃干燥,整粒,装入胶囊壳中,即得胶囊剂。
动物试验研究
1.1实验性骨质疏松症小鼠模型的建立
参照文献方法[沈薇等.实验性骨质疏松症小鼠模型的建立[J].沈阳医学院学报,2006,8(4):245-247],取4月龄同批成年雌性Balb/c小鼠72只,体重20-30g,随机分成2组:去卵巢骨质疏松模型组60只,假手术对照组12只。清洁环境饲养,自由进食水,定期环境消毒。用20mg/kg体重的戊巴比妥腹腔注射麻醉,腹部常规消毒,在无菌条件下,取腹侧入路正中手术切口打开腹腔,切除两侧卵巢。彻底止血后,分两层依次缝合腹膜、肌肉和皮肤,术后给予青霉素腹腔注射,8万U/kg。假手术对照组开腹后切除少许脂肪组织后缝合,切除部分的重量基本与两卵巢重量相同,其余操作同上。两组小鼠饲养条件一致。
1.2分组及给药方法
饲养一周后,将60只去卵巢模型小鼠随机分为5组,即模型组,阳性对照组,中药制剂(实施例1制得的胶囊剂)高、中、低剂量组,每组12只,高、中、低剂量组均以0.5%CMC-Na配制为混悬剂灌胃给药,剂量分别为18、9、4.5g生药/kg·d-1,每周6次给药。阳性对照组,戊酸雌二醇片以0.5%CMC-Na配制为混悬剂,灌胃给药1.5mg/kg,每周1次给药。同时将钙尔奇D片剂研磨,以0.5%CMC-Na配制为混悬剂,灌胃给药20mg/kg,每周6次给药;模型组和假手术对照组给以同体积0.5%CMC-Na(1ml/100g)。
1.3骨密度测定
6组动物于0、8、10、12周经戊巴比妥钠腹腔麻醉后称重。四肢展平置于骨密度仪上测量每只小鼠全身BMD。
1.4统计学分析
小鼠全身BMD用x±s表示,用t检验进行组间差异比较。
1.5实验结果

与模型组比较,*:p<0.05,**:p<0.01。
骨密度测定结果表明:与正常对照组比较,模型组骨密度明显降低,表明骨质疏松模型形成。与模型组比较,阳性对照组骨密度有显著升高(p<0.01),中药组合物高、中、低剂量组的骨密度也均有显著性升高(p<0.05~0.01)。
以上所述仅为本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原料的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。

1.一种防治骨质疏松症的中药制剂,其特征在于,所述中药制剂由以下重量份数的原料药制成:
淫羊藿32份,桑寄生24份,怀牛膝18份,延胡索20份,女贞子15份,木香12份,当归12份,苍术15份,海螵蛸15份;
所述制备方法包括以下步骤:
(1)按照原料药重量份数称取中药材,净选;
(2)海螵蛸粉碎成细粉,过200目筛,备用;
(3)淫羊藿、桑寄生、怀牛膝、延胡索、女贞子、木香、当归、苍术,加10倍量水,加热煎煮3次,每次2.5小时,过滤,合并滤液,浓缩至滤液体积的二分之一,加入95%的乙醇直至含醇量为75%为止,静置24小时,离心,取上清液,过滤,将滤液减压浓缩至稠浸膏,置真空干燥箱55℃充分干燥,得干浸膏,将干浸膏粉碎成细粉,过200目筛,备用;
(4)将步骤(2)所得细粉与步骤(3)所得的干膏粉合并,充分混合,加入适量淀粉浆制软材,过20目筛制粒,将湿颗粒于55℃干燥,整粒,装入胶囊壳中,即得胶囊剂。
