Abstract
本发明公开了一种包含五味子和磷脂酰丝氨酸的药物及其制备方法,包含五味子和磷脂酰丝氨酸的药物按照重量份的原料包括:五味子1?100份、磷脂酰丝氨酸0.001?0.4份,制备时,以五味子为原料,经清洗、回流提取、浓缩,并经过初滤和精滤制成五味子提取物,五味子提取物中加入磷脂酰丝氨酸后,制成饮料、茶或片、颗粒、胶囊等剂型。本发明在五味子提取物中加入了磷脂酰丝氨酸,不仅保留原五味子的用于久嗽虚喘,梦遗滑精,遗尿尿频,久泻不止,自汗,盗汗,津伤口渴,短气脉虚,内热消渴,心悸失眠等功效,另因加入磷脂酰丝氨酸,还可以增强记忆力、预防老年痴呆等功效。上述配方,不仅适用于老年人用,因加入了磷脂酰丝氨酸而变成老少皆宜的大众食品。

技术领域
本发明涉及药物生产技术领域,具体是一种包含五味子和磷脂酰丝氨酸的药物及其制备方法。
背景技术
五味子分北五味子和南五味子,北五味子质比南五味子优良。五味子喜微酸性腐殖土,生长期需要足够的水分和营养;分布于黑龙江、吉林、辽宁、内蒙古、河北等地。古医书称它荎蕏、玄及、会及,最早列于神农本草经上品中药,能滋补强壮之力,药用价值极高,有强身健体之效。2015年8月,中国科学家在五味子中发现抗艾滋病毒成分。五味子为著名中药,其果含有五味子素及维生素C、树脂、鞣质及少量糖类。有敛肺止咳、滋补涩精、止泻止汗之效。其叶、果实可提取芳香油。种仁含有脂肪油,榨油可作工业原料、润滑油。茎皮纤维柔韧,可供绳索。功能主治:敛肺,滋肾,生津,收汗,涩精、治肺虚喘咳,口干作渴,自汗,盗汗,劳伤羸瘦,梦遗滑精,久泻久痢。
磷脂作为一种生物活性物质,有着独特的理化性质和营养价值,在世界范围内的食品、保健品、医药以及饲料行业已被广泛使用,人们通过使用磷脂可以起到调节血脂、改善记忆、保护肝脏、健脑益智、以及延缓衰老等生理功能的作用。磷脂酰丝氨酸是磷脂家族中的一员,磷脂酰丝氨酸,又称复合神经酸,简称PS,由天然大豆榨油剩余物提取,是细胞膜的活性物质,尤其存在于大脑细胞中,其功能主要是改善神经细胞功能,调节神经脉冲的传导,增进大脑记忆功能,由于其具有很强的亲脂性,吸收后能够迅速通过血脑屏障进入大脑,起到舒缓血管平滑肌细胞,增加脑部供血的作用,它是唯一能够调控细胞膜关键蛋白功能状态的磷脂,是人体不可或缺的物质。所以只要正常服用磷脂酰丝氨酸类的膳食剂并不会给身体带来额外的负担,并且通过了许多国家的药监局认证。
磷脂酰丝氨酸被誉为继胆碱和“脑黄金”DHA之后的一大新兴的“智能营养素”。专家认为,这种天然物质能够帮助细胞壁保持柔韧性,并且能够增强传送大脑信号的神经递质的效率,帮助大脑高效运转,激发大脑的活化状态。具体来说,磷脂酰丝氨酸有以下功能:
1.提高大脑机能,集中注意力,改善记忆力;欧美涌现出大量对磷脂酰丝氨酸的荟萃分析(荟萃分析指对某个问题所做的多个独立研究结果进行系统的定量或定性的综合),其主要目的是将以往的研究结果更为客观地综合反映出来。分析9个双盲、安慰剂对照、共1224例患者参与的临床试验,结果显示,补充磷脂酰丝氨酸后,关于认知和记忆的参数得到显著改善。毫无疑问,补充磷脂酰丝氨酸可以提高长期记忆、长期认知以及自由谈吐和逻辑性发言能力;随着年龄的增长,磷脂酰丝氨酸和其他重要的脑内化学物质会逐渐减少,从而导致记忆力、认知力减弱。补充磷脂酰丝氨酸能增加脑突刺数目、脑细胞膜的流动性及促进脑细胞中葡萄糖代谢,从而使脑细胞更活跃,促进注意力集中,提高警觉性和记忆力。意大利、斯堪的纳维亚半岛和欧洲其他国家都广泛应用磷脂酰丝氨酸补充剂来治疗衰老引发的认知障碍症及老年记忆损失。
2.提高学生成绩,专门测试磷脂酰丝氨酸缓解精神压力的效果,试验采用了双盲、安慰剂对照,连续30天,每天给予健康大学生300毫克磷脂酰丝氨酸。大学生必须在给定时间内完成具有一定难度的数学测试,并记录他们面对压力的反应。结果发现,使用磷脂酰丝氨酸的学生在反应力、自信和表现方面均好于对照组,他们在考试中的成绩也更好。一项研究针对120名重庆巴蜀中学高三年级学生的研究还发现,服用强化磷脂酰丝氨酸的学生,经过40天后,其语言和非语言的记忆力都有显著提高。
3.缓解压力,促进用脑疲劳的恢复、平衡情绪;多项研究表明,磷脂酰丝氨酸能显著降低工作紧张者体内过多的应激激素的水平,减轻压力,缓解脑部疲劳;磷脂酰丝氨酸还可作用于大脑内影响心情的神经递质水平,可以促进注意力集中、提高警觉性和记忆力,帮助缓解不良情绪(如抑郁、沮丧等)。
4.帮助修复大脑损伤;磷脂酰丝氨酸是脑部神经的主要成分之一,可营养和活化脑中各种酶的活性,能延缓神经递质的减少进程,有助于修复、更新大脑受损细胞和清除有害物质。它可以使老年人的记忆力恢复到14年前的水平,研究显示,食用磷脂酰丝氨酸12周后,66岁的人拥有了52岁人的记忆力;除此之外,磷脂酰丝氨酸和DHA可以相互促进吸收,对神经2A细胞起到保护作用。丰富的磷脂酰丝氨酸可以增加细胞膜的流动性,促进智力的发育。磷脂酰丝氨酸和DHA一起可以保护中枢神经系统,促进胎儿智力发育;磷脂酰丝氨酸之所以能够增强人的智力,主要原因在于它能够迅速穿过血脑屏障,进入大脑。在脑部起到舒缓大脑毛细血管平滑肌细胞,增加脑部供血的作用。所以近几年有很多针对脑卒中方面的产品都以PS作为原料。
磷脂酰丝氨酸是一种天然存在于食物中的成分,在母乳中也存在。肉类及鱼类中均含有磷脂酰丝氨酸,脑或内脏(如肝、肾)中的含量较高。奶制品以及蔬菜中(除豆类)磷脂酰丝氨酸的含量非常少。现代人饮食习惯的改变(如不再食用内脏),食用健康食品的意识增强(如吃低脂、低胆固醇食品和避免食用大量肉类食品),以及食品危机(如疯牛病和口蹄疫)导致人们摄入磷脂酰丝氨酸的数量已经越来越少。更严重的是,现代工业化生产出的脂类和油类产品中的天然磷脂含量下降,进一步减少了正常人每日应该摄取磷脂酰丝氨酸的数量。一般来讲,如果饮食中有较多的肉和鱼类,磷脂酰丝氨酸的摄取量大约为180毫克/天;如果食物中脂肪含量低,那么每天磷脂酰丝氨酸的摄取量将仅为100毫克,甚至少于50毫克。据统计,正常人每日磷脂酰丝氨酸的摄取量与正常需求量间差距在70—150毫克之间,素食者尤为缺乏,其差距达到200—250毫克。鉴于以上原因,正常人每日需要补充100—300毫克的纯磷脂酰丝氨酸,对于素食主义者、低脂肪或低胆固醇饮食者以及老年人来说,这点更加重要。此外,其他一些饮食习惯也会降低大脑中磷脂酰丝氨酸的含量。例如,饮食中缺乏欧米伽3脂肪酸会使大脑中磷脂酰丝氨酸的含量降低28%,进而改变大脑海马体的生物化学功能。
发明内容
本发明的目的在于提供一种可以治疗内热消渴、预防老年痴呆的包含五味子和磷脂酰丝氨酸的药物及其制备方法,以解决上述背景技术中提出的问题。
为实现上述目的,本发明提供如下技术方案:
一种包含五味子和磷脂酰丝氨酸的药物,按照重量份的原料包括:按照重量份的原料包括:五味子1-100份、磷脂酰丝氨酸0.001-0.4份。
作为本发明进一步的方案:所述包含五味子和磷脂酰丝氨酸的药物,按照重量份的原料包括:五味子5份、磷脂酰丝氨酸0.1份。
所述包含五味子和磷脂酰丝氨酸的药物的制备方法,具体步骤如下:
(1)按照重量份称取各原料;
(2)将五味子清洗干净后,加入3-15倍的水,温度控制在80-110℃,回流连续提取0.5-4h;
(3)将步骤(2)得到的提取液温度降低至70-90°C,进行抽真空压缩至相对密度为1.0-1.1;
(4)进行初滤后再精滤,得五味子提取物;
(5)五味子提取物中加入磷脂酰丝氨酸,搅拌均匀后得到药物制剂。
作为本发明再进一步的方案:所述药物制剂为饮料、茶、片剂、颗粒剂或胶囊剂。
与现有技术相比,本发明的有益效果是:
本发明在传统的五味子提取物中加入了磷脂酰丝氨酸,不仅保留原五味子的用于久嗽虚喘,梦遗滑精,遗尿尿频,久泻不止,自汗,盗汗,津伤口渴,短气脉虚,内热消渴,心悸失眠等功效,另因加入磷脂酰丝氨酸,还可以增强记忆力、预防老年痴呆等功效。上述配方,不仅适用于老年人用,因加入了磷脂酰丝氨酸而变成老少皆宜的大众食品。
具体实施方式
下面结合具体实施方式对本专利的技术方案作进一步详细地说明。
实施例1
一种包含五味子和磷脂酰丝氨酸的药物,按照重量份的原料包括:五味子1份、磷脂酰丝氨酸0.001份。
所述包含五味子和磷脂酰丝氨酸的药物的制备方法,具体步骤如下:
(1)按照重量份称取各原料;
(2)将五味子清洗干净后,加入3倍的水,温度控制在80℃,回流连续提取0.5h;
(3)将步骤(2)得到的提取液温度降低至70°C,进行抽真空压缩至相对密度为1.0;
(4)进行初滤后再精滤,得五味子提取物;
(5)五味子提取物中加入磷脂酰丝氨酸,搅拌均匀后得到饮料。
实施例2
一种包含五味子和磷脂酰丝氨酸的药物,按照重量份的原料包括:五味子5份、磷脂酰丝氨酸0.1份。
所述包含五味子和磷脂酰丝氨酸的药物的制备方法,具体步骤如下:
(1)按照重量份称取各原料;
(2)将五味子清洗干净后,加入6倍的水,温度控制在87℃,回流连续提取1.3h;
(3)将步骤(2)得到的提取液温度降低至75°C,进行抽真空压缩至相对密度为1.02;
(4)进行初滤后再精滤,得五味子提取物;
(5)五味子提取物中加入磷脂酰丝氨酸,搅拌均匀后得到茶。
实施例3
一种包含五味子和磷脂酰丝氨酸的药物,按照重量份的原料包括:五味子50份、磷脂酰丝氨酸0.2份。
所述包含五味子和磷脂酰丝氨酸的药物的制备方法,具体步骤如下:
(1)按照重量份称取各原料;
(2)将五味子清洗干净后,加入9倍的水,温度控制在95℃,回流连续提取2.1h;
(3)将步骤(2)得到的提取液温度降低至80°C,进行抽真空压缩至相对密度为1.05;
(4)进行初滤后再精滤,得五味子提取物;
(5)五味子提取物中加入磷脂酰丝氨酸,搅拌均匀后得到片剂。
实施例4
一种包含五味子和磷脂酰丝氨酸的药物,按照重量份的原料包括:五味子80份、磷脂酰丝氨酸0.3份。
所述包含五味子和磷脂酰丝氨酸的药物的制备方法,具体步骤如下:
(1)按照重量份称取各原料;
(2)将五味子清洗干净后,加入12倍的水,温度控制在102℃,回流连续提取3h;
(3)将步骤(2)得到的提取液温度降低至85°C,进行抽真空压缩至相对密度为1.08;
(4)进行初滤后再精滤,得五味子提取物;
(5)五味子提取物中加入磷脂酰丝氨酸,搅拌均匀后得到颗粒剂。
实施例5
一种包含五味子和磷脂酰丝氨酸的药物,按照重量份的原料包括:五味子100份、磷脂酰丝氨酸0.4份。
所述包含五味子和磷脂酰丝氨酸的药物的制备方法,具体步骤如下:
(1)按照重量份称取各原料;
(2)将五味子清洗干净后,加入15倍的水,温度控制在110℃,回流连续提取4h;
(3)将步骤(2)得到的提取液温度降低至90°C,进行抽真空压缩至相对密度为1.1;
(4)进行初滤后再精滤,得五味子提取物;
(5)五味子提取物中加入磷脂酰丝氨酸,搅拌均匀后得到胶囊剂。
药理作用
五味子:
《本草求原》:五味子,为咳嗽要药,凡风寒咳嗽,伤暑咳嗽,伤燥咳嗽,劳伤咳嗽,肾水虚嗽,肾火虚嗽,久嗽喘促,脉浮虚,按之弱如葱叶者,天水不交也,皆用之。先贤多疑外感用早,恐其收气太骤,不知仲景伤寒咳喘,小青龙汤亦用之,然必合细辛、干姜以升发风寒,用此以敛之,则升降灵而咳嗽自止,从无舍干姜而单取五味以治咳嗽者。丹溪又谓其收肺气之耗散,即能除热;潜江亦谓其滋肺以除热,补肾以暖水,而联属心肾;凡嗽在黄昏,是虚火浮入肺中,忌用寒凉,止宜重用五味以敛降,此则不合干姜,而合炒麦冬者也。总之,肺气随阴以下降,则气化精而精盈,肾水从阳以上布,则精化气而气盛,阴阳二气,实一气之变动,以肝为关捩子,五味专精于肝,而交合肺肾,故其效如此,有不同于他味之酸敛者。肺气阳中有阴,故能降,治肺气以阴降为主。然元气之降,先本于升,五味升降咸备,所以阳邪伤阴,固宜清阳,以之收阳;阴邪伤阳,亦宜此辛温畅阳,而寓收阴。东垣谓寒喘热喘,不能舍五味者此也,惟外邪杂病不关肺气者忌。
《本草正义》:阴火上冲激肺之嗽,阳虚火浮,故当黄昏阴盛之时,虚焰发动,乃始作嗽,宜以收摄肺肾为治。然惟脉虚、舌红、无痰者乃合,若舌腻有痰,亦当知所顾忌。
磷脂酰丝氨酸通常位于细胞膜的内层,磷酯化合物中的磷酸甘油酯类,是细胞膜组分之一,与一系列的膜功能有关。尤其在人体的神经系统,是大脑的细胞膜的重要,组成成分之一,同时对大脑的各种功能(尤其是对大脑的记忆力和情绪的稳定)起到重要的调节作用,如它能影响着细胞膜的流动性、通透性,并且能激活多种酶类的代谢和合成;磷脂酰丝氨酸可由人体利用丝氨酸合成产生,它可影响脑内化学讯息的传递,并帮助脑细胞储存和读取资料,是维持大脑正常记忆力、反应和健康情绪的重要营养元素。
本发明在传统的五味子提取物中加入了磷脂酰丝氨酸,不仅保留原五味子的用于久嗽虚喘,梦遗滑精,遗尿尿频,久泻不止,自汗,盗汗,津伤口渴,短气脉虚,内热消渴,心悸失眠等功效,另因加入磷脂酰丝氨酸,还可以增强记忆力、预防老年痴呆等功效。上述配方,不仅适用于老年人用,因加入了磷脂酰丝氨酸而变成老少皆宜的大众食品。

1.一种包含五味子和磷脂酰丝氨酸的药物,其特征在于,按照重量份的原料包括:五味子1-100份、磷脂酰丝氨酸0.001-0.4份。
2.根据权利要求1所述的包含五味子和磷脂酰丝氨酸的药物,其特征在于,按照重量份的原料包括:五味子5份、磷脂酰丝氨酸0.1份。
3.一种如权利要求1-2任一所述的包含五味子和磷脂酰丝氨酸的药物的制备方法,其特征在于,具体步骤如下:
(1)按照重量份称取各原料;
(2)将五味子清洗干净后,加入3-15倍的水,温度控制在80-110℃,回流连续提取0.5-4h;
(3)将步骤(2)得到的提取液温度降低至70-90°C,进行抽真空压缩至相对密度为1.0-1.1;
(4)进行初滤后再精滤,得五味子提取物;
(5)五味子提取物中加入磷脂酰丝氨酸,搅拌均匀后得到药物制剂。
4.根据权利要求3所述的包含五味子和磷脂酰丝氨酸的药物的制备方法,其特征在于,所述药物制剂为饮料、茶、片剂、颗粒剂或胶囊剂。
