Abstract

本发明提供了一种吴茱萸碱喷雾剂,每1000ml喷雾剂的原料由如下重量配比的组份组成:吴茱萸碱0.1~1.0g,透皮吸收促进剂20~50g,乙醇或含水乙醇加至全量;所述透皮吸收促进剂由如下重量配比的组份组成:薄荷油3~5份、油酸1~2份、冬青油2~4份。本发明喷雾剂将薄荷油、油酸和冬青油联合使用作为透皮吸收促进剂,相比单用冰片、薄荷油、油酸或冬青油而言,吴茱萸碱的渗透量明显增加,说明本发明将三者联合使用后发挥了协同作用,能够更为有效地促进吴茱萸碱这一成分的皮肤渗透性。

Description

一种吴茱萸碱透皮吸收喷雾剂
技术领域
本发明涉及一种吴茱萸碱透皮吸收喷雾剂。
背景技术
吴茱萸功效散寒止痛、降逆止呕、助阳止泻,临床常用于治疗厥阴头痛、寒湿脚气、经行腹痛等,具有保护心脏、抗心律失常、抗炎镇痛等作用。其主要功效成分为吴茱萸碱和吴茱萸次碱。吴茱萸碱为难溶性化合物,几乎不溶于水和多数有机溶剂,口服吸收困难。全身给予吴茱萸碱不是发挥该类化合物镇痛作用的理想给药途径。而局部施用药物可直达病灶,提高局部药物浓度,减少药物在全身循环或其它组织的分布,降低出现潜在不良反应的风险。
外用局部施药的关键是药物的经皮渗透能力,经皮渗透能力可以通过添加促渗剂来改善,如何嘉仑等采用油酸、氮酮或1,2-丙二醇来促进吴茱萸碱的经皮吸收,并发现油酸的促渗效果优于其他两类成分(吴茱萸提取物的透皮吸收促进剂的筛选,中成药,2013年35卷6期)。张广唱等采用冰片作为透皮吸收促进剂,同样能够改善吴茱萸碱的透皮性能(吴茱萸水包油微乳喷雾剂的体外透皮吸收特性,中国实验方剂学杂志,2015年21卷17期)。
发明内容
本发明的目的在于提供一种能够有效地促进吴茱萸碱透皮吸收的喷雾剂。
本发明提供了一种吴茱萸碱喷雾剂,每1000ml喷雾剂的原料由如下重量配比的组份组成:
吴茱萸碱0.1~1.0g,透皮吸收促进剂20~50g,乙醇或含水乙醇加至全量;
所述透皮吸收促进剂由如下重量配比的组份组成:
薄荷油3~5份、油酸1~2份、冬青油2~4份。
进一步地,所述透皮吸收促进剂由如下重量配比的组份组成:
薄荷油4份、油酸1份、冬青油3份。
其中,所述含水乙醇为30~95%v/v乙醇。
更进一步地,所述含水乙醇为70%v/v~95%v/v乙醇。
本发明上述原料亦可制备气雾剂,若制备气雾剂则需要加入相适应的抛射剂。
本发明还提供了上述喷雾剂的制备方法,它包括如下操作步骤:
(1)取吴茱萸碱溶于适量乙醇或含水乙醇中,得吴茱萸碱乙醇溶液;
(2)取透皮吸收促进剂与适量乙醇或含水乙醇混匀,再与吴茱萸碱乙醇溶液混合,加乙醇或含水乙醇至全量,混匀,装瓶即得。
本发明还提供了上述喷雾剂在制备抗炎止痛的药物中的用途。
本发明还提供了上述喷雾剂在制备治疗或辅助治疗痛风的药物中的用途。
本发明的有益效果如下:
(1)张广唱等采用冰片作为透皮吸收促进剂,同样能够改善吴茱萸碱的透皮性能(吴茱萸水包油微乳喷雾剂的体外透皮吸收特性,中国实验方剂学杂志,2015年21卷17期)。
本发明喷雾剂将薄荷油、油酸和冬青油联合使用作为透皮吸收促进剂,相比单用冰片、薄荷油、油酸或冬青油而言,吴茱萸碱的渗透量明显增加,说明本发明将三者联合使用后发挥了协同作用,能够更为有效地促进吴茱萸碱这一成分的皮肤渗透性。
(2)何嘉仑等采用油酸、氮酮或1,2-丙二醇来促进吴茱萸碱的经皮吸收,并发现油酸的促渗效果优于其他两类成分(吴茱萸提取物的透皮吸收促进剂的筛选,中成药,2013年35卷6期)。
相比单用油酸而言,本发明喷雾剂将薄荷油、油酸和冬青油联合使用作为透皮吸收促进剂,吴茱萸碱的渗透量明显更大,说明本发明将三者联合使用后,其促渗作用优于现有技术中记载的经典成分油酸。另外,薄荷油自身具有清凉感,能够降低疼痛,减轻不适;冬青油具有消肿、消炎和镇痛作用。三者合用,不仅可以促进吴茱萸碱透皮吸收,还可协助吴茱萸碱止痛,相较于油酸、氮酮或丙二醇等透皮吸收促进剂而言,更有利于对痛风的治疗或辅助治疗。
具体实施方式
实施例1本发明喷雾剂的制备
成分:
吴茱萸碱0.5g、透皮吸收促进剂30g(薄荷油:油酸:冬青油=4:1:3w/w)、95%v/v乙醇加至1000mL;
制备方法:
(1)取吴茱萸碱溶于适量乙醇中;
(2)取透皮吸收促进剂混合于适量乙醇中;
(3)将吴茱萸碱乙醇溶液与透皮吸收促进剂乙醇溶液混合,加乙醇至全量,混匀,装瓶,即得喷雾剂。
实施例2本发明喷雾剂的制备
成分:
吴茱萸碱1.0g、透皮吸收促进剂50g(薄荷油:油酸:冬青油=4:1:3w/w)、95%v/v乙醇加至1000mL;
制备方法同实施例1。
实施例3本发明喷雾剂的制备
成分:
吴茱萸碱0.1g、透皮吸收促进剂20g(薄荷油:油酸:冬青油=4:1:3w/w)、95%v/v乙醇加至1000mL;
制备方法同实施例1。
实施例4本发明组合物促进吴茱萸碱透皮吸收作用测试
本发明测试实验主要参照“吴茱萸提取物的透皮吸收促进剂的筛选.何嘉仑,等.中成药,2013年35卷6期”。
(1)裸鼠皮肤的处理
取健康裸鼠,处死后小心剥离腹部皮肤,剔除皮下脂肪组织及黏连物,用生理盐水反复冲洗干净,置于-20℃冰箱中保存备用。实验前自然解冻,每次实验前检查皮肤的完整性,不能有任何破损。
(2)体外透皮吸收实验方法
将处理好的裸鼠腹部皮肤固定于扩散池的供给池与接收池之间,角质层面向供给池。供给池中加入1.5mL吴茱萸碱(1mL含吴茱萸碱0.5mg)及相应的促渗剂(3%g/mL),接收池中加入7mL70%v/v乙醇生理盐水作为接收介质,排出气泡使液面恰与皮肤内层接触。调节扩散仪至恒温(32±1)℃,恒速搅拌(约200r/min),分别于搅拌开始后0.5,1,2,4,6,8,10,12,24h从样品接收池中抽取1mL接收液,同时补加同温度同体积接收液。将各个时间点取出的接收液用0.22μm微孔滤膜过滤过,供HPLC进样分析,并以此计算累积渗透量Qn。
(3)实验结果
结果参见下表。

由上表可知:
(1)本发明喷雾剂将薄荷油、油酸和冬青油联合使用作为透皮吸收促进剂,相比单用冰片、薄荷油、油酸或冬青油而言,吴茱萸碱的渗透量明显增加,说明本发明将三者联合使用后发挥了协同作用,能够更为有效地促进吴茱萸碱这一成分的皮肤渗透性。
(2)相比单用油酸而言,本发明喷雾剂将薄荷油、油酸和冬青油联合使用作为透皮吸收促进剂,吴茱萸碱的渗透量明显更大,说明本发明将三者联合使用后,其促渗作用优于现有技术中记载的经典成分油酸。
另外,薄荷油自身具有清凉感,能够降低疼痛,减轻不适;冬青油具有消肿、消炎和镇痛作用。两者与油酸合用,不仅可以促进吴茱萸碱透皮吸收,还可协助吴茱萸碱止痛,相较于油酸、氮酮或丙二醇等透皮吸收促进剂而言,更有利于对痛风的治疗或辅助治疗。

Claims (7)

1.一种吴茱萸碱喷雾剂,其特征在于:每1000ml喷雾剂的原料由如下重量配比的组份组成:
吴茱萸碱0.1~1.0g,透皮吸收促进剂20~50g,乙醇或含水乙醇加至全量;
所述透皮吸收促进剂由如下重量配比的组份组成:
薄荷油3~5份、油酸1~2份、冬青油2~4份。
2.根据权利要求1所述的喷雾剂,其特征在于:所述透皮吸收促进剂由如下重量配比的组份组成:
薄荷油4份、油酸1份、冬青油3份。
3.根据权利要求1所述的喷雾剂,其特征在于:所述含水乙醇为30~95%v/v乙醇。
4.根据权利要求3所述的喷雾剂,其特征在于:所述含水乙醇为70%v/v~95%v/v乙醇。
5.权利要求1~4任意一项所述喷雾剂的制备方法,其特征在于:它包括如下操作步骤:
(1)取吴茱萸碱溶于适量乙醇或含水乙醇中,得吴茱萸碱乙醇溶液;
(2)取透皮吸收促进剂与适量乙醇或含水乙醇混匀,再与吴茱萸碱乙醇溶液混合,加乙醇或含水乙醇至全量,混匀,装瓶即得。
6.权利要求1~4所述喷雾剂在制备抗炎止痛的药物中的用途。
7.权利要求1~4所述喷雾剂在制备治疗或辅助治疗痛风的药物中的用途。
