背景技术
化疗是化学药物治疗的简称,是利用化学药物阻止癌细胞的增殖、浸润、转移,直至最终杀灭癌细胞的一种治疗方式。它是一种全身性治疗手段,和手术、放疗一起,并称为癌症的3大治疗手段。由于化疗药物的选择性不强,在杀灭癌细胞的同时也会不可避免地损伤人体正常的细胞,从而出现药物的不良反应。因此,在接受化疗药物的时候,一方面希望能够达到最佳的抗肿瘤作用,另一方面也要注意预防和识别化疗药物的不良反应。
德国科学家弗里茨·绍丁在1905年发现了引起梅毒(一种经性传播的疾病)的微生物。恩利希就用他的化合物来试验对这种新微生物的治疗作用。他高兴地发现,606号化合物有效果。他把这种化合物称为洒尔氟散,并戏称它为“神奇的子弹”,因为它对梅毒有特效。1911年,它第一次运用于梅毒的治疗。从那以后,科学家们一直在寻找能杀伤肿瘤细胞、不对人体造成严重伤害的化学物质。在发现有效的化学物质之前,科学家得对几千种化学物质进行测试。许多种癌症现在能被治愈。干扰素是人体针对某些病毒发生反应后而自然产生的蛋白质。它们刺激人体自身的防御系统,杀伤一些癌症细胞。它们已经被成功地用于治疗某些类型的白血病,并能延缓一些肿瘤的发展。
对某些肿瘤,手术切除非但不能治愈,反而促进其生长和增殖。特别是对于中晚期肿瘤的治疗,不能一味追求肿瘤的消退,而要控制肿瘤,与其共存。放化疗往往是手术治疗后预防复发与转移的措施之一,但是它在杀伤肿瘤细胞的同时,也同样会杀伤正常的免疫细胞,损害机体的正常机能,破坏内环境的平衡。放化疗造成的毒性反应主要有:骨髓抑制最初多表现为白细胞下降,其中尤以粒细胞更为明显。随着剂量的增加,血小板和红细胞也会受到影响,有的甚至可发生再生障碍性贫血。因而在进行放疗和化疗过程中要定期检查血象。当白细胞降低至2000~3000/mm3,血小板降至5万~8万/mm3时,应暂时中止治疗;消化道反应如口干、食欲减退、恶心、呕吐、腹痛或腹泻等;免疫功能降低,抗感染能力下降;皮肤、粘膜损害。如皮肤干燥、红斑、口腔溃疡、色素沉着、脱发及指甲变形等;有些药物对神经系统、心肌、肝、肾、肺等重要脏器可发生损害放化疗不仅仅存在诸多的不良反应,给人身带来不同程度的损伤,而且随着放化疗疗程的加长放化疗的敏感性越来越差,因此治疗效果也会大打折扣。
化疗是目前治疗肿瘤及某些自身免疫性疾病的主要手段之一,但在治疗中,患者普遍有明显的恶心呕吐等副作用,给患者带来不适感。

(1)将无花果、散血莲、当归、山楂、夏至草和喜树洗净,放入容器内,加入8~10倍量的蒸馏水,浸泡5~6小时后,煮沸3~4小时,提取;再次加入6~8倍量蒸馏水,煮沸2~3小时,提取;最后,加入4~6倍量蒸馏水,加热煮沸1~2小时,提取;合并三次提取液,过滤,得滤液,浓缩为60℃下相对密度为1.10~1.13的稠膏,备用;
(2)将仙茅洗净,用黄酒拌匀,润透后文火炒至微干,取出晾至全干,置于研钵内,研末,得到过200目的细粉,备用;
(3)将大蓟、甘蔗、乌梅和三白草洗净,切碎,放入容器内,绞汁,滤杂质,留汁液备用;
(4)将地锦草和地核桃洗净,浸入5倍量的70%的乙醇溶液浸泡1~2小时,加热提取3小时,回收乙醇,滤过,得滤液,浓缩为65℃下相对密度为1.12~1.16的稠膏,备用;
(5)将阴香皮、谷芽和蛇蜕放入容器内,进行粉碎处理,研末,得到过180目的细粉,备用;
(6)将上述步骤(1)~(5)所制得的稠膏、细粉以及汁液混合后,制成片剂,即得所述的治疗肿瘤化疗引起的不良反应的药物。
本发明所用中药的药性如下:
无花果:味甘,性凉;归肺、胃、大肠经。健胃清肠,消肿解毒。治肠炎,痢疾,便秘,痔疮,喉痛,痈疮疥癣,利咽喉,开胃驱虫。用于食欲不振,脘腹胀痛,痔疮便秘,消化不良,痔疮,脱肛,腹泻、乳汁不足,咽喉肿痛,热痢,咳嗽多痰等症。《云南中草药》载:“健胃止泻,祛痰理气。治食欲不振,消化不良,肠炎,痢疾,咽喉痛,咳嗽痰多,胸闷。”
仙茅:味辛,性热;归肾、肝、脾经。补肾阳,强筋骨,祛寒湿。用于阳痿精冷,筋骨痿软,腰膝冷痹,阳虚冷泻。《海药本草》载:“主风,补暖腰脚,清安五脏,强筋骨,消食。宣而复补,主丈夫七伤,明耳目,益筋力,填骨髓,益阳。”
大蓟:味甘、苦,性凉,无毒;归心、肝经。主治吐血,衄血,尿血,血淋,血崩,带下,肠风,肠痈,痈疡肿毒,疔疮。现代医学研究证实,大蓟的水溶液具有麻醉作用;大蓟对凝血酶原激活物的生成有促进作用,并能抑制病菌生成。
地锦草:味辛,性平;归肺、肝、胃、大肠、膀胱经。清热解毒,利湿退黄,活血止血。主治痢疾,泄泻,黄疸,咳血,吐血,尿血,便血,崩漏,乳汁不下,跌打肿痛及热毒疮疡。《上海常用中草药》:载:“止血,利尿,健胃,活血解毒。治黄疸,痢疾,腹泻,尿路感染,便血,尿血,子宫出血,痔疮出血,跌打肿痛,女人乳汁不通,蛇咬伤,头疮,皮肤疮毒。”
散血莲:味微辛、微苦,性凉;归肝经。祛风除湿,散血止痛,清热解毒。主治风湿关节痛,瘀血腹痛,闭经,跌打损伤,目赤肿痛,乳痈,各种肿毒初起。
甘蔗:味甘、涩、性平,无毒。治下气和中,助脾气,利大肠,消痰止渴,除心胸烦热,解酒毒;还可治呕吐反胃,宽胸膈。现代医学研究表明,甘蔗中含有丰富的糖分、水分,此外,还含有对人体新陈代谢非常有益的各种维生素、脂肪、蛋白质、有机酸、钙、铁等物质。
阴香皮:味辛、微甘,性温,归脾经。温中止痛,祛风散寒,解毒消肿,止血。主治寒性胃痛,腹痛泄泻,食欲不振,风寒湿痹,腰腿疼痛,跌打损伤,创伤出血,疮疖肿毒。广州部队《常用中草药手册》载:“祛风散寒,温中止痛。治寒性胃痛,腹胀,腹泻。”
乌梅:味酸,性平;入肝、脾、肺、肾、胃、大肠经。敛肺止咳,涩肠止泻,止血生津,安蛔。主治久咳,虚热烦渴,久疟,久泻,痢疾,便血,尿血,血崩,蛔厥腹痛,呕吐,钩虫病。《纲目》载:“敛肺涩肠,治久嗽,泻痢,反胃噎膈,蛔厥吐利,消肿,涌痰,杀虫,解鱼毒、马汗毒、硫黄毒。”现代医学研究证实,乌梅有明显的抗菌作用,对痢疾杆菌、绿脓杆菌、伤寒杆菌、大肠杆菌、结核杆菌、变形杆菌、金黄色葡萄球菌、零乱弧菌和各种真菌均有抑制作用,且能刺激腮腺荷尔蒙的分泌,澄化血液,增强新陈代谢功能。
谷芽:味甘,性平;归脾、胃经。消食和中,健脾开胃。用于食积不消,腹胀口臭,脾胃虚弱,不饥食少,炒谷芽偏于消食,用于不饥食少,焦谷芽善化积滞,用于积滞不消。《本草经疏》载:“蘖米即稻蘖也,具生化之性,故为消食健脾、开胃和中之要药,脾胃和则中自瘟,气自下,热自除也。”
三白草:味甘、辛,性寒;归肺、脾、胃、大肠经。清热利水,解毒消肿。主治热淋,血淋,水肿,脚气,黄疸,痢疾,带下,痈肿疮毒,湿疹,蛇咬伤。现代医学研究证实,三白草所含金丝桃甙具明显的抗炎作用,1.50%煎剂对金黄色葡萄球菌、伤寒杆菌有抑制作用。较强的止咳作用和抑制眼醛糖还原酶的作用,可能对预防糖尿病性白内障有益。
当归:味甘、辛,性温;归肝、心、脾经。补血活血,调经止痛,润肠通便。主治血虚萎黄,眩晕心悸,虚寒腹痛,风湿痹痛,痈疽疮疡。《本草正》载:“其味甘而重,故专能补血,其气轻而辛,故又能行血,能养营养血,补气生精,安五脏,强形体,益神志。”现代医学研究证实,当归具有抗菌、消炎作用。
地核桃:味苦、辛,性寒;归肺经。清热解毒,消肿止血。治痈疽疮毒,肺痈,跌打损伤,刀伤出血。《泉州本草》载:“清热解毒,退癀止痛。治痈疽肿毒,瘰疬恶疮,以及各种化脓性疮疡。”
蛇蜕:味咸、甘,性平;归肝经。祛风,定惊,解毒,退翳。用于小儿惊风,抽搐痉挛,翳障,喉痹,疔肿,皮肤瘙痒。《纲目》载:“祛风,杀虫。烧末服,治妇人吹奶,大人喉风,退目翳,消木舌,敷小儿重舌,重腭,唇紧,解颅,面疮,月蚀,天泡疮,大人疔肿,漏疮肿毒,煮汤洗诸恶虫伤。”
山楂:味酸、甘,性微温;归脾、胃、肝经。消食健胃,行气散瘀。用于肉食积滞,胃脘胀满,泻痢腹痛,瘀血经闭,产后瘀阻,心腹刺痛,疝气疼痛;高脂血症。焦山楂消食导滞作用增强。用于肉食积滞,泻痢不爽。《纲目》载:“化饮食,消肉积,癥瘕,痰饮痞满吞酸,滞血痛胀。”
夏至草:味辛、微苦,性寒;、归肝经。养血活血,清热利湿。主月经不调,产后瘀滞腹痛,血虚头昏,半射不遂,跌打损伤,水肿,小便不利,目赤肿痛,疮痈,冻疮,牙痛,皮疹瘙痒。《陕西中草药》载:“活血,调经。治贫血性头昏,半身不遂,月经不调。”
喜树:味苦、辛,性寒,有毒;归脾、胃肝经。抗癌,清热,杀虫。用于胃癌,结肠癌,直肠癌,膀胱癌,慢性粒细胞性白血病,急性淋巴细胞性白血病;外用治牛皮癣。《新编中医学概要》载:“破血化瘀。治慢性淋巴性白血病、慢性髓性白血病有较好疗效;对急性白血病只能暂时缓解症状。”
本发明与现有技术相比,具有以下有益效果:
本发明药物所选药材配伍相宜,符合中医药学和现代医药学理论,并具有清热解毒、补血活血和消食健胃之功效,对于肿瘤化疗引起的恶心、呕吐、腹泻、便秘、食欲不振和脱发等不良反应具有独特疗效,且疗效显著,见效较快,疗程较短,成本较低,安全可靠,无任何毒副作用,奇效实用,值得临床上广泛推广应用。
具体实施方式
下面结合具体实施例对本发明做进一步阐述。
实施例1
本发明药物有效成分的原料组成及重量为:
一种治疗肿瘤化疗引起的不良反应的药物的制备方法,包括以下工艺步骤:
(1)将无花果、散血莲、当归、山楂、夏至草和喜树洗净,放入容器内,加入8~10倍量的蒸馏水,浸泡5~6小时后,煮沸3~4小时,提取;再次加入6~8倍量蒸馏水,煮沸2~3小时,提取;最后,加入4~6倍量蒸馏水,加热煮沸1~2小时,提取;合并三次提取液,过滤,得滤液,浓缩为60℃下相对密度为1.10~1.13的稠膏,备用;
(2)将仙茅洗净,用黄酒拌匀,润透后文火炒至微干,取出晾至全干,置于研钵内,研末,得到过200目的细粉,备用;
(3)将大蓟、甘蔗、乌梅和三白草洗净,切碎,放入容器内,绞汁,滤杂质,留汁液备用;
(4)将地锦草和地核桃洗净,浸入5倍量的70%的乙醇溶液浸泡1~2小时,加热提取3小时,回收乙醇,滤过,得滤液,浓缩为65℃下相对密度为1.12~1.16的稠膏,备用;
(5)将阴香皮、谷芽和蛇蜕放入容器内,进行粉碎处理,研末,得到过180目的细粉,备用;
(6)将上述步骤(1)~(5)所制得的稠膏、细粉以及汁液混合后,制成片剂,即得所述的治疗肿瘤化疗引起的不良反应的药物。
用法用量:口服,每日2次,早晚各1次,每次2片(每片0.5g),饭后温开水送服;10天为1个疗程,2~3个疗程即可痊愈,病情严重者遵医嘱酌增用量。

临床资料:
1、病例选择:
作为治疗肿瘤化疗引起的不良反应的药物,选择确诊的肿瘤化疗过后出现不良反应的患者78例临床观察,病人随机分成治疗组和对照组两组,治疗组39例,其中男性19例,女性20例,年龄50~71岁,平均年龄60.5岁,平均病程2年;对照组39例,其中男性20例,女性19例,年龄49~70岁,平均年龄59.5岁,平均病程2年。
治疗组和对照组两组病例的病程、症状轻重程度基本一致,无显著差异,具有可比性。
2、药物选择:
治疗组服用本发明药物,口服,每日2次,早晚各1次,每次2片(每片0.5g),饭后温开水送服;10天为1个疗程,2~3个疗程即可痊愈,病情严重者遵医嘱酌增用量。
对照组服用柔红霉素,口服,成人1日1~2g,分3~4次服用,整片吞服;静滴1日1~2g,分3~4次滴注。
3、疗效判定:
(1)治愈:恶心、呕吐、腹泻、便秘、食欲不振和脱发等不良反应全部消失,身体恢复正常。
(2)显效:恶心、呕吐、腹泻、便秘、食欲不振和脱发等不良反应明显减轻,身体有所好转。
(3)有效:恶心、呕吐、腹泻、便秘、食欲不振和脱发等不良反应有所减轻,身体明显好转。
(4)无效:通过治疗后恶心、呕吐、腹泻、便秘、食欲不振和脱发等不良反应无减轻或加重。
4、结果:
选用本发明药物的治疗组中,治愈21例,显效11例,有效5例,无效2例,治愈率53.8%,总有效率94.9%。
对照组中治愈18例,显效10例,有效4例,无效7例,治愈率46.2%,总有效率82.1%。
以上结果显示,治疗组的治愈率和总有效率明显高于对照组。
典型病例举例:
1、袁某,女,50岁,重庆沙坪坝区人。患者查出患乳腺癌,接受了乳房切除手术以及整形再造手术,手术之后经过化疗,当时身体已恢复,化疗后身体出现不适现象,经医生给予本发明药物治疗,服用1个疗程后,症状明显减轻,服用3个疗程后,症状全部消失,化疗期间未出现身体不适现象。
2、谭某,男,69岁,河北秦皇岛人。患者因持续发热就医,检查双颈部进行性、无疼痛淋巴结肿大伴发热,右侧最大,约3cm*2cm,均活动,无压痛。辅助检查发现右颈部肿大淋巴结活检见淋巴结结构完全破坏,诊断为淋巴瘤。患者因害怕手术引起全身淋巴转移,选择化疗。化疗期间出现恶心、呕吐、脱发、体恤气短、精神萎靡等症状,在医生的建议下,患者从化疗开始服用本发明药物治疗,服用1个疗程后,化疗期间出现恶心、呕吐、脱发、体恤气短、精神萎靡等症状有所减轻,继续服用2个疗程后,化疗期间没有出现恶心、呕吐、脱发、体恤气短、精神萎靡等症状。目前患者已出院,精神乐观,每天饭量都能达到正常。
3、曹某,女,61岁,江苏无锡人。患者因腹痛、乏力、食欲下降,体重下降就医,做胃镜检查后发现胃窦小弯侧直径为2.5cm的壁内龛影,周围粘膜僵硬有中断,诊断为胃癌。医生建议行胃大部切除术,切除2/3。术后进行化疗,化疗过程中出现恶心、呕吐等症状,连头发都出现脱落现象。化疗期间给予本发明药物治疗,服用2个疗程后,化疗过程中未出现胃肠道症状如恶心、呕吐等,连头发都没出现脱落现象。
4、程某,男,58岁,黑龙江大庆人。患者体检发现右肺占位,后行开胸探查诊断为肺腺癌。因距主动脉较近未行手术.随后接受化疗1周期,因心肺功能较差停用。三个月后患者出现右胁部及后背疼痛,胸闷,憋气、睡眠差,家人带其去医院检查结果为:CT示:胸部右侧大量胸水,并有纵隔淋巴结转移。经服用本发明药物治疗,服用1个疗程后,胸痛减轻,仍有气短的现象,继续服用2个疗程后,气短的现象消失,痊愈,随访至今未复发。
5、萧某,女,67岁,广东珠海人。患者自诉1年前无明显诱因出现上胸部痛,间歇性无规律,为隐痛可耐受,不向它处放射,胸骨有压痛,咳嗽,深呼吸时不加重,胸闷不伴呼吸困难,当地市医院就诊,查胸片示慢性血行播散性肺结核,肺内转移性病变不排除;CT示:双肺广泛结节性病灶,入院体查:T36.7℃左腹股沟用扪及淋巴结一个,花生米大小,质韧,可活动,有压痛,边清,局部皮肤无红肿,双侧语颤正常,呼吸活动对称双肺叩诊清音,CT扫描报告单:左肺下叶背段见一约1.76×2.25cm2大小软组织肿块,边缘不规整见“毛刺征”及“空泡征”其内密度欠均匀,平扫CT值约为12~46Hu,增强扫描明显强化,CT值约为49~92Hu,左肺下叶背段支气管远端闭塞。双肺余部见弥漫性大小不等结节灶,边缘光整。经化疗治疗,治疗期间出现不良反应,医生推荐服用本发明药物治疗,服用1个疗程,不良反应明显减轻,继续服用2个疗程后,症状消失,患者化疗后未出现不良反应。
6、董某,男,53岁,安徽蚌埠人。患者因刺激性咳嗽伴痰中带血就医,X片可见右肺门上方直径约3cm的团块状阴影,后经确诊为小细胞肺癌。期间经过4次化疗,化疗期间,出现身体无力,食欲不振,精神状态不佳,经服用本发明药物治疗,服用1个疗程后,患者自觉身体明显好转,食欲有所恢复,服用3个疗程后,身体恢复如常,食欲恢复,精神状态也较好。

散血莲26~44份甘蔗26~44份阴香皮25~43份乌梅24~42份
谷芽24~42份三白草23~41份当归23~41份地核桃21~39份
蛇蜕19~37份山楂17~36份夏至草17~36份喜树16~34份。
2.根据权利要求1所述的一种治疗肿瘤化疗引起的不良反应的药物,其特征在于:制成所述药物有效成分的原料组成及重量份数为:
无花果32~44份仙茅31~43份大蓟31~43份地锦草30~42份
散血莲29~41份甘蔗29~41份阴香皮28~40份乌梅27~39份
谷芽27~39份三白草26~38份当归26~38份地核桃24~36份

3.根据权利要求1所述的一种治疗肿瘤化疗引起的不良反应的药物,其特征在于:制成所述药物有效成分的原料组成及重量份数为:

4.根据权利要求1所述的一种治疗肿瘤化疗引起的不良反应的药物,其特征在于:制成所述药物有效成分的原料组成及重量份数为:

散血莲35份甘蔗35份阴香皮34份乌梅33份
谷芽33份三白草32份当归32份地核桃30份
蛇蜕28份山楂27份夏至草27份喜树25份。
5.一种如权利要求1~4任一项所述治疗肿瘤化疗引起的不良反应的药物的制备方法,其特征在于:包括以下工艺步骤:
(1)将无花果、散血莲、当归、山楂、夏至草和喜树洗净,放入容器内,加入8~10倍量的蒸馏水,浸泡5~6小时后,煮沸3~4小时,提取;再次加入6~8倍量蒸馏水,煮沸2~3小时,提取;最后,加入4~6倍量蒸馏水,加热煮沸1~2小时,提取;合并三次提取液,过滤,得滤液,浓缩为60℃下相对密度为1.10~1.13的稠膏,备用;
