本发明公开了一种神经干细胞添加剂;所述添加剂其组成成分及浓度为:重组人碱性成纤维生长因子1ug/ml,L-丙氨酸2mg/ml,生物素0.1mg/ml,左旋肉碱2mg/ml,2-羟基乙胺1mg/ml,D-半乳糖15mg/ml,L-脯氨酸7.76mg/ml,丁二胺10mM,丙酮酸钠25mg/ml,亚硒酸钠0.5ug/ml,维生素B12?0.35mg/ml,硫酸锌0.2mg/ml,过氧化氢酶16mg/ml,谷胱甘肽1mg/ml,超氧化物歧化酶2.5mg/ml,亚油酸100mg/ml,黄体酮0.6ug/ml,维生素A醋酸酯10mg/ml,醋酸生育酚100mg/ml,β巯基乙醇5mM。本发明的神经干细胞添加剂,配合基础培养基可以在无血清的环境下为神经元干细胞提供生存的营养,也可以维持神经干细胞未分化的状态,配合其他生长因子,可以对神经干细胞进行定向诱导。

技术领域
本发明涉及一种神经细胞培养添加剂,特别涉及一种神经干细胞添加剂,属于神经细胞培养技术领域。
背景技术
干细胞是身体内的原始未分化细胞,而神经干细胞和神经前体(母)细胞是神经系统的干细胞,因为其在治疗神经系统疾病和损伤中的广阔使用前景,近年来的研究和临床应用突飞猛进地发展,而神经干系统应用最首要的是能够在体外成功地培养并获得增殖神经干细胞,因为神经是机体中最复杂和最高度分化的组织,神经干细胞的培养难度极高,在培养过程中极其容易自发分化,从而很快就失去干细胞的特性和功能,现有的基础培养基在无血清环境下,容易失去生存营养;并很难对神经干细胞进行定向诱导。
发明内容
(一)要解决的技术问题
为解决上述问题,本发明提出了一种神经干细胞添加剂,能够配合基础培养基可以在无血清的环境下为神经元干细胞提供生存的营养,也可以维持神经干细胞未分化的状态,配合其他生长因子,可以对神经干细胞进行定向诱导。
(二)技术方案
本发明的神经干细胞添加剂,所述添加剂其组成成分及浓度为:重组人碱性成纤维生长因子1ug/ml,L-丙氨酸2mg/ml,生物素0.1mg/ml,左旋肉碱2mg/ml,2-羟基乙胺1mg/ml,D-半乳糖15mg/ml,L-脯氨酸7.76mg/ml,丁二胺10mM,丙酮酸钠25mg/ml,亚硒酸钠0.5ug/ml,维生素B120.35mg/ml,硫酸锌0.2mg/ml,过氧化氢酶16mg/ml,谷胱甘肽1mg/ml,超氧化物歧化酶2.5mg/ml,亚油酸100mg/ml,黄体酮0.6ug/ml,维生素A醋酸酯10mg/ml,醋酸生育酚100mg/ml,β巯基乙醇5mM。
有益效果
与现有技术相比,本发明的神经干细胞添加剂,配合基础培养基可以在无血清的环境下为神经元干细胞提供生存的营养,也可以维持神经干细胞未分化的状态,配合其他生长因子,可以对神经干细胞进行定向诱导;本发明添加剂设计科学、配方简单、成本低廉、经济合理,能够配合基础培养基培养,使用范围极为广泛。

本发明神经干细胞添加剂,所述添加剂其组成成分及浓度为:重组人碱性成纤维生长因子1ug/ml,L-丙氨酸2mg/ml,生物素0.1mg/ml,左旋肉碱2mg/ml,2-羟基乙胺1mg/ml,D-半乳糖15mg/ml,L-脯氨酸7.76mg/ml,丁二胺10mM,丙酮酸钠25mg/ml,亚硒酸钠0.5ug/ml,维生素B120.35mg/ml,硫酸锌0.2mg/ml,过氧化氢酶16mg/ml,谷胱甘肽1mg/ml,超氧化物歧化酶2.5mg/ml,亚油酸100mg/ml,黄体酮0.6ug/ml,维生素A醋酸酯10mg/ml,醋酸生育酚100mg/ml,β巯基乙醇5mM。
其中,所述维生素A醋酸酯和醋酸生育酚为脂溶性维生素。所述亚油酸由乙醇溶解。
本发明的神经干细胞添加剂,配合基础培养基可以在无血清的环境下为神经元干细胞提供生存的营养,也可以维持神经干细胞未分化的状态,配合其他生长因子,可以对神经干细胞进行定向诱导;本发明添加剂设计科学、配方简单、成本低廉、经济合理,能够配合基础培养基培养,使用范围极为广泛。
