Abstract
本发明公开了一种具有抗白色念珠菌功能的口腔洗护用品,该口腔洗护用品含有以下重量配比的活性成分:藜蒿叶提取物2~15%、薰衣草提取物、黄芩提取物、莪术提取物中的一种或多种1~20%。本发明还公开了一种抗白色念珠菌的药物牙膏。本发明对白色念珠菌有很好的抑制效果,可用于预防或缓解由白色念珠菌引起的口腔念珠菌疾病。
Description
技术领域
本发明涉及药物洗护用品,尤其是一种抗白色念珠菌的口腔洗护用品。
背景技术
白色念珠菌又称白色假丝酵母,是广泛分布于人体体表粘膜的一种条件致病菌。通常菌体以单细胞浮游状态存在,几乎没有致病力,而当细胞形成了利于粘附在宿主细胞粘膜的生物膜时,白色念珠菌可进行大量增殖而侵染宿主。
口腔是白色念珠菌在人体的主要分布区域,通常情况下白色念珠菌是口腔内正常微生物群的组成之一。然而当宿主机体免疫力下降,或者抗生素药物的使用影响到口腔的微生态平衡时,白色念珠菌可过量增殖继而引起疾病。在口腔念珠菌患者的口腔内白色念珠菌属优势菌株,数量约占念珠菌总菌株的61.6%,且78%的白色念珠菌菌株具有形成生物膜的能力,即较强的致病性。
目前,已有很多添加了植物提取物的药物洗护用品的文献报道,如CN102038620A公开了一种添加艾叶提取物和菖蒲提取物的洗护用品及其制备方法;CN104825368A公开了一种天然植物活性牙膏,该牙膏添加了皂角、无患子和玉米等天然植物提取物;CN104688658A公开了一种中草药牙膏,含有牛蒡子、芦根等中药提取物,对牙龈炎、牙周炎等口腔疾病具有治疗功效;CN102846498A公开了一种竹叶提取物漱口水及其制备方法;CN103006513A公开了一种中草药漱口水,其由矿化水、添加剂和中草药提取物组成。这些天然植物提取物洗护用品只有少数具有抗白色念珠菌的作用:CN103536451A公开了一种含丹皮酚和长梗冬青苷的中药牙膏,可抑制口腔中的白色念珠菌;CN102379827A公开了一种含有三七素的牙膏,能杀灭口腔内引起疾病的白色念珠菌。然而,目前未见一种添加藜蒿叶提取物且具有抗白色念珠菌功效的洗护用品。
藜蒿,又名芦蒿、蒌蒿,菊科蒿属,是盛产于湖北、湖南等地的一种多年生草本植物。藜蒿含有丰富的黄酮、绿原酸、挥发油等活性成分,全草具有特殊香味,是一种宝贵的野生蔬菜资源。而藜蒿仅以嫩茎食用,存在着大量藜蒿叶被废弃的资源浪费问题。基于藜蒿资源的综合利用以及天然植物提取物抗菌洗护产品的研制,本发明提出了一种以藜蒿叶提取物为主要活性成分,并与多种中药材提取物配伍,开发一种具有抗白色念珠菌特殊功效的口腔洗护用品,以预防或缓解由白色念珠菌引起的口腔念珠菌疾病,尤其适用于口腔念珠菌病患者。
发明内容
本发明的目的是提供一种以藜蒿叶提取物为主要活性成分,具有抗白色念珠菌特殊功效的口腔洗护用品。
为解决上述问题,本发明通过以下技术方案实现:
一种抗白色念珠菌的口腔洗护用品,它含有以下重量配比的活性成分:
藜蒿叶提取物2~15%
薰衣草提取物、黄芩提取物、莪术提取物中的一种或多种1~20%,
所述藜蒿叶提取物、薰衣草提取物、黄芩提取物、莪术提取物是将藜蒿叶、薰衣草、黄芩、莪术分别用水提取,提取液浓缩后用酒精醇沉,醇沉后的上清液经过浓缩、干燥后得到的。
优选地,所述藜蒿叶提取物的制备方法是:将藜蒿叶用10~20倍重量的水煎煮提取1~3次,合并提取液并浓缩成清膏,将清膏用60~90%的酒精醇沉,醇沉后的上清液经过浓缩、干燥,即得。
进一步优选地,所述口腔洗护用品为药物牙膏、漱口剂、口含片。
本发明进一步提供一种抗白色念珠菌的药物牙膏,它由以下重量配比的成分组成:
藜蒿叶提取物2~15%
薰衣草提取物、黄芩提取物、莪术提取物中的一种或多种1~20%
藜蒿叶超微粉末0.05~1%
余量为洗护基质,
所述藜蒿叶提取物、薰衣草提取物、黄芩提取物、莪术提取物是将藜蒿叶、薰衣草、黄芩、莪术分别用水提取,提取液浓缩后用酒精醇沉,醇沉后的上清液经过浓缩、干燥后得到的。
优选地,所述抗白色念珠菌的药物牙膏由以下重量配比的成分组成:
藜蒿叶提取物6%
薰衣草提取物8%
黄芩提取物5%
莪术提取物1%
藜蒿叶超微粉末0.6%
余量为牙膏基质。
进一步优选地,所述牙膏基质由磷酸氢钙、木糖醇、甘油、二氧化硅、丙二醇、十二烷基硫酸钠、黄原胶、尼泊金乙酯、甜菊苷、去离子水组成。
本发明的有益效果是:
1.本发明提供的药物洗护用品对白色念珠菌有很好的抑制效果,可用于预防或缓解由白色念珠菌引起的口腔念珠菌疾病。
2.本发明提供了一种新的以天然植物提取物为活性成分的抗菌洗护产品,有效地减少了藜蒿的资源浪费。
3.本发明提供的药物牙膏质地细腻,味甘甜,色泽翠绿,具有藜蒿特有的清香,非常受消费者欢迎。
具体实施方式
以下结合具体实施例对本发明进行详细地说明,但本发明并不为实施例所限制。
实施例1
一种抗白色念珠菌的药物牙膏,它由以下重量配比的成分组成:
藜蒿叶提取物6%
薰衣草提取物8%
黄芩提取物5%
莪术提取物1%
藜蒿叶超微粉末0.6%
余量为牙膏基质。
所述牙膏基质由以下成分组成,各成分占牙膏总重量的百分比分别为:磷酸氢钙30%、木糖醇8%、甘油6%、二氧化硅5%、丙二醇5%、十二烷基硫酸钠1%、黄原胶0.3%、尼泊金乙酯0.05%、甜菊苷0.03%、去离子水24%。
制备方法如下:
1)藜蒿叶提取物的制备:将藜蒿叶用15倍重量的水煎煮提取3次,合并提取液并浓缩成清膏,向清膏中加入酒精并使混合溶液中的含醇量达90%(体积含量),搅拌后静置12小时,醇沉,醇沉后的上清液经过真空浓缩、干燥,即得。
2)其它提取物的制备:将薰衣草、黄芩、莪术药材分别粉碎成粗粉,加水煎煮提取,提取液浓缩后加入酒精并使混合溶液中的含醇量达70%(体积含量),搅拌后静置12小时,醇沉,醇沉后取上清液经过真空浓缩、干燥,分别得到薰衣草提取物、黄芩提取物、莪术提取物。
3)藜蒿叶超微粉末的制备:摘取藜蒿叶清洗干净后,50℃干燥12h,粉碎并过60目筛得到藜蒿叶粗粉,将藜蒿叶粗粉用超离心研磨仪(16000r/min,10min)进行研磨,过250目筛网,得到藜蒿叶超微粉末。
4)将藜蒿叶提取物、薰衣草提取物、黄芩提取物、莪术提取物用去离子水溶解,加入木糖醇、甜菊苷、尼泊金乙酯,搅拌,制得水溶液,备用。
5)将磷酸氢钙、二氧化硅、十二烷基硫酸钠、黄原胶和藜蒿叶超微粉末搅拌均匀,然后分散于甘油和丙二醇混合溶液中,搅拌,形成浆状溶液,备用。
6)将上述水溶液和浆状溶液混匀,均质并真空脱气,制得膏体灌装。
实施例2
一种抗白色念珠菌的药物牙膏,它由以下重量配比的成分组成:
藜蒿叶提取物5%
薰衣草提取物8%
黄芩提取物8%
莪术提取物4%
藜蒿叶超微粉末0.5%
余量为牙膏基质。
所述牙膏基质由以下成分组成,各成分占牙膏总重量的百分比分别为:磷酸氢钙25%、木糖醇6%、甘油5%、二氧化硅8%、丙二醇8%、十二烷基硫酸钠0.8%、黄原胶0.5%、尼泊金乙酯0.08%、甜菊苷0.02%、去离子水21.1%。
制备方法如下:
1)藜蒿叶提取物的制备:将藜蒿叶用20倍重量的水煎煮提取2次,合并提取液并浓缩成清膏,向清膏中加入酒精并使混合溶液中的含醇量达80%(体积含量),搅拌后静置6小时,醇沉,醇沉后的上清液经过真空浓缩、干燥,即得。
2)薰衣草、黄芩、莪术都是通过市售途径购买得到,均是将原料药材采用水提醇沉的方法制备而得。
3)藜蒿叶超微粉末的制备:将藜蒿叶清洗干净后,干燥,粉碎得藜蒿叶粗粉,将藜蒿叶粗粉进行研磨,过300目筛网,得到藜蒿叶超微粉末。
4)将藜蒿叶提取物、薰衣草提取物、黄芩提取物、莪术提取物用去离子水溶解,加入木糖醇、甜菊苷、尼泊金乙酯,搅拌,制得水溶液,备用。
5)将磷酸氢钙、二氧化硅、十二烷基硫酸钠、黄原胶和藜蒿叶超微粉末搅拌均匀,然后分散于甘油和丙二醇混合溶液中,搅拌,形成浆状溶液,备用。
6)将上述水溶液和浆状溶液混匀,均质并真空脱气,制得膏体灌装。
实施例3
一种抗白色念珠菌的药物牙膏,它由以下重量配比的成分组成:
藜蒿叶提取物15%
薰衣草提取物2%
黄芩提取物5%
莪术提取物3%
藜蒿叶超微粉末1%
余量为牙膏基质。
所述牙膏基质由以下成分组成,各成分占牙膏总重量的百分比分别为:磷酸氢钙20%、木糖醇5%、甘油10%、二氧化硅6%、丙二醇4%、十二烷基硫酸钠1.5%、黄原胶0.6%、尼泊金乙酯0.1%、甜菊苷0.06%、去离子水26.7%。
制备方法如下:
1)藜蒿叶提取物的制备:将藜蒿叶用10倍重量的水煎煮提取3次,合并提取液并浓缩成清膏,向清膏中加入酒精并使混合溶液中的含醇量达60%(体积含量),搅拌后静置24小时,醇沉,醇沉后的上清液经过真空浓缩、干燥,即得。
2)薰衣草提取物、黄芩提取物、莪术提取物都是通过市售途径购买得到。
3)藜蒿叶超微粉末的制备:将藜蒿叶清洗干净后,干燥,粉碎得藜蒿叶粗粉,将藜蒿叶粗粉进行研磨,过200目筛网,得到藜蒿叶超微粉末。
4)将藜蒿叶提取物、薰衣草提取物、黄芩提取物、莪术提取物用去离子水溶解,加入木糖醇、甜菊苷、尼泊金乙酯,搅拌,制得水溶液,备用。
5)将磷酸氢钙、二氧化硅、十二烷基硫酸钠、黄原胶和藜蒿叶超微粉末搅拌均匀,然后分散于甘油和丙二醇混合溶液中,搅拌,形成浆状溶液,备用。
6)将上述水溶液和浆状溶液混匀,均质并真空脱气,制得膏体灌装。
实施例4
一种抗白色念珠菌的药物漱口水,它由以下重量配比的成分组成:
藜蒿叶提取物2%
薰衣草提取物3%
黄芩提取物1%
莪术提取物1%
甘油8%、食用酒精6%、木糖醇1%、食盐0.5%、尼泊金乙酯0.1%、甜菊苷0.06%、黄原胶0.04%、水77.3%。
制备方法如下:
将以上成分混合,按药物漱口水的常规工艺制备。
试验例1醇沉浓度对藜蒿叶提取物抑菌效果的影响
1.将藜蒿叶用15倍水煎煮提取3次,每次2h,合并滤液并浓缩成清膏;向清膏中加入酒精使混合溶液中的乙醇体积含量分别达到60%、70%、80%、90%,搅拌后静置12小时,醇沉,取醇沉后的上清液,经过真空浓缩、干燥,得到藜蒿叶提取物。
2.以微量平皿稀释法分别测定60%、70%、80%、90%乙醇沉淀处理后藜蒿叶提取物对白色念珠菌的最小抑菌浓度、最小杀菌浓度和最小生物膜抑制浓度,确定最适乙醇处理浓度。具体实验方法为:
(1)将活化后的白色念珠菌以RPMI1640培养液配制并稀释为含菌量1×103-5×103CFU/mL的菌悬液;无菌96孔板1至11号孔分别加入100μL菌悬液和100μL藜蒿提取物药液(以无菌培养液配制)等度稀释至药物浓度为0-250mg·mL-1,12号孔以无菌培养液代替药液作阳性对照,同时以无菌培养液代替菌悬液作对应的阴性对照;采用酶标仪测定各孔在600nm处吸光值,与阳性对照相比下降80%的药物处理浓度为最小抑菌浓度;
(2)从高于最小抑菌浓度的培养孔中接种100μL培养液于沙堡琼脂平板,未见明显菌体生长(CFU<5)的最低浓度为藜蒿叶提取物的最低杀菌浓度。
(3)采用结晶紫染色法对以藜蒿叶提取物共同培养72h后的菌体形成的生物膜进行染色,测定不同浓度乙醇处理后的藜蒿叶提取物对菌体生物膜的形成产生80%抑制的最小浓度,即最小生物膜抑制浓度。
实验结果表明,90%乙醇处理浓度所制备藜蒿叶提取物的抑菌效果最强(如表1)。

试验例2不同药材配伍对藜蒿叶提取物抑菌效果的影响
1.选用18种已报道的对白色念珠菌有抑制作用的药材提取物。
2.以微量平皿稀释法(同试验例1)分别测定各提取物和藜蒿叶提取物单独给药时的最小抑菌浓度,即单独用药MICA和单独用药MICB。
3.采用棋盘微量稀释法测定各提取物和藜蒿叶提取物对白色念珠菌产生抑制作用的相互作用,具体实验方法为:将上述藜蒿叶提取物和18种药材提取物分别在96孔板上纵向和横向倍比稀释,两药物以一系列浓度交叉配伍,测定复合给药时各提取物和藜蒿叶提取物对白色念珠菌的最小抑菌浓度,即联合用药MICA和联合用药MICB。
4.以部分抑制浓度指数FICI(factionalinhibitoryconcentrationindex)评价联合用药的相互作用,计算公式为:FICI=联合用药MICA/单独用药MICA+联合用药MICB/单独用药MICB(FICI<0.5即可确定为协同作用);计算各提取物和藜蒿叶提取物对抗白色念珠菌作用的相互作用指数FICI,结果如表2所示。通过实验结果可以筛选出三种对藜蒿叶提取物抗白色念珠菌作用具有极强增效作用的中药提取物:薰衣草、黄芩、莪术,其FICI指数均小于0.5。

1.一种抗白色念珠菌的口腔洗护用品,其特征在于含有以下重量配比的活性成分:
藜蒿叶提取物2~15%
薰衣草提取物、黄芩提取物、莪术提取物中的一种或多种1~20%,
所述藜蒿叶提取物、薰衣草提取物、黄芩提取物、莪术提取物是将藜蒿叶、薰衣草、黄芩、莪术分别用水提取,提取液浓缩后用酒精醇沉,醇沉后的上清液经过浓缩、干燥后得到的。
2.如权利要求1所述的抗白色念珠菌的口腔洗护用品,其特征在于所述藜蒿叶提取物的制备方法是:将藜蒿叶用10~20倍重量的水煎煮提取1~3次,合并提取液并浓缩成清膏,将清膏用60~90%的酒精醇沉,醇沉后的上清液经过浓缩、干燥,即得。
3.如权利要求1或2所述的抗白色念珠菌的口腔洗护用品,其特征在于所述口腔洗护用品为药物牙膏、漱口剂、口含片。
4.一种抗白色念珠菌的药物牙膏,其特征在于由以下重量配比的成分组成:
藜蒿叶提取物2~15%
薰衣草提取物、黄芩提取物、莪术提取物中的一种或多种1~20%
藜蒿叶超微粉末0.05~1%
余量为牙膏基质,
所述藜蒿叶提取物、薰衣草提取物、黄芩提取物、莪术提取物是将藜蒿叶、薰衣草、黄芩、莪术分别用水提取,提取液浓缩后用酒精醇沉,醇沉后的上清液经过浓缩、干燥后得到的。
5.如权利要求4所述的抗白色念珠菌的药物牙膏,其特征在于由以下重量配比的成分组成:
藜蒿叶提取物6%
薰衣草提取物8%
黄芩提取物5%
莪术提取物1%
藜蒿叶超微粉末0.6%
余量为牙膏基质。
6.如权利要求4或5所述的抗白色念珠菌的药物牙膏,其特征在于所述牙膏基质由磷酸氢钙、木糖醇、甘油、二氧化硅、丙二醇、十二烷基硫酸钠、黄原胶、尼泊金乙酯、甜菊苷、去离子水组成。
