Abstract
本发明公开了一种鹰嘴豆芽多糖及其应用,所述鹰嘴豆芽多糖中总多糖含量为33.85%‑40.38%,蛋白含量为0.60%‑3.25%,硫酸基含量为1.19%‑1.58%,糖醛酸含量为1.02%‑2.01%。本发明的鹰嘴豆芽多糖具有很好的保湿功效,可用于化妆品、保健食品和医药领域,尤其是可应用于化妆品中。

技术领域
本发明属于天然产物化学领域,尤其涉及一种鹰嘴豆芽多糖及其应用。
背景技术
皮肤保护内部组织的屏障功能有赖于皮肤的水合保湿能力,皮肤的储水量占全身的18%-20%。空调、环境污染、季节转换带来的温度变化与压力,以及皮肤衰老、代谢减缓等因素都会造成肌肤水分过度流失,令肌肤粗糙、黯哑、形成干纹、引发诸多肌肤问题。防止皮肤水分过度流失,对于保持良好的皮肤屏障功能和皮肤的健康,乃至皮肤美丽的外观是非常重要的。
为了达到保持皮肤水分,减少水分过度散失的目的,通常在皮肤外用剂中加入保湿成分。保湿剂种类很多,常用的保湿成分可以分为封闭型保湿剂和吸湿性保湿剂两大类。封闭型保湿剂作用是通过在皮肤表面形成膜层,阻挡皮肤水分蒸发,从而使角质层保持一定的含水量,这一类保湿剂包括矿物油和蜡如凡士林,植物油与动物脂肪如羊毛脂,磷脂、胆固醇等。吸湿性保湿剂是一类类似于表皮中天然保湿因子,可以与水强力结合而达到使角质层保湿的效果。此类保湿成分有:天然保湿因子、蛋白质、多糖、神经酰胺、各种维生素等。动物和矿物来源的保湿剂容易引起皮肤过敏、皮肤刺激等问题,从植物中寻找天然保湿剂符合人们对源于自然的化妆品的需求。
鹰嘴豆(Cicer arietinum Linn)为维吾尔族习用药材,维吾尔族名为奴乎特、诺胡提,因种子形如鹰头、鸡头或山状突起而得名,别名称羊头豆、鸡头豆、鸡豌豆、脑豆子和桃豆等。鹰嘴豆盛产于新疆天山北坡海拔1300m的木垒县,已有2500年的生长历史。鹰嘴豆味甘、性平、无毒,有补中益气、温肾壮阳、主消渴、解血毒、润肺止咳等作用。对糖尿病、高血脂、高血压病、心脑血管疾病、肺病、消化不良等均有良好的食疗作用。它的花可以治疗痢疾,效果很好,浸泡水后服用可以治疗中药中毒和白带增多等。籽粒可做利尿剂、催奶剂,可治疗失眠、预防皮肤病和胆病等。研究发现发芽的鹰嘴豆种子胚芽部分含异黄酮类:鹰嘴豆芽素A、鹰嘴豆谷芽素和鹰嘴豆芽素C等,异黄酮类物质具有降血糖、降血脂等多种生物学功能。
豆芽也称芽苗菜,是各种谷类、豆类、树类的种子培育出可以食用的“芽菜”,也称“活体蔬菜”。常见的芽苗菜有:香椿芽苗菜、荞麦芽苗菜、苜蓿芽苗菜、花椒芽苗菜、绿色黑豆芽苗菜、相思豆芽苗菜、葵花籽芽苗菜、萝卜芽苗菜、龙须豆芽苗菜、花生芽苗菜、蚕豆芽苗菜等30多个品种,是常见的蔬菜。鹰嘴豆芽为鹰嘴豆种子培育而出的,鹰嘴豆芽中含丰富异黄酮类:鹰嘴豆芽素A和芒柄花素等。植物异黄酮有弱的雌激素样作用,其雌激素活性仅为天然雌激素的1/100至1/1000,在活体内起雌激素和抗雌激素双重调节作用,可调节乳腺对雌激素的反应,使雌激素不易引起乳腺组织发生异常,植物异黄酮也减少男士发生前列腺癌危险。
发明内容
本发明的目的在于:提供一种鹰嘴豆芽多糖及其应用,旨在将鹰嘴豆芽多糖应用于皮肤外用剂中,以起到美容护肤的作用。
本发明的目的是这样实现的:
在本发明的鹰嘴豆芽多糖中,所述鹰嘴豆芽多糖中总多糖含量为33.85%-40.38%,蛋白含量为0.60%-3.25%,硫酸基含量为1.19%-1.58%,糖醛酸含量为1.02%-2.01%。
通过以下方法制备:
A、取鹰嘴豆,洗净,于温水中浸泡6-8小时后,取出置于豆芽箱中,豆芽箱的温度30℃,淋水时间5秒,淋水间隔为180分钟。豆芽长至5-6cm左右,将豆芽和豆瓣分开,取豆芽,备用;
B、取新鲜鹰嘴豆芽加70%乙醇超声提取2次,每次1小时,过滤,滤液合并,得豆芽残渣备用;
C、取鹰嘴豆芽醇提后残渣加入沸水煮3次,将每次滤液合并,浓缩,加三氯醋酸在4℃下脱蛋白,离心,上清液加NaOH中和,流水透析2d,浓缩,再加入95%乙醇使体系含醇量达75-85%,沉淀冷冻干燥得鹰嘴豆芽多糖。
进一步的,在本发明的鹰嘴豆芽多糖的制备方法中,C步骤为:
1)、取鹰嘴豆芽醇提后残渣加入沸水煮3次,将每次滤液合并,浓缩,加三氯醋酸在4℃下脱蛋白,离心,上清液加NaOH中和,流水透析2d,浓缩,加入95%乙醇使体系含醇量达75%,沉淀冷冻干燥得鹰嘴豆芽多糖-1;
2)、取鹰嘴豆芽醇提后残渣,加入沸水煮3次,将每次滤液合并,浓缩,加三氯醋酸在4℃下脱蛋白,离心,上清液加NaOH中和,流水透析2d,浓缩,加入95%乙醇使体系含醇量达80%,沉淀冷冻干燥得鹰嘴豆芽多糖-2;
3)、取鹰嘴豆芽醇提后残渣,加入沸水煮3次,将每次滤液合并,浓缩,加三氯醋酸在4℃下脱蛋白,离心,上清液加NaOH中和,流水透析2d,浓缩,加入95%乙醇使体系含醇量达85%,沉淀冷冻干燥得鹰嘴豆芽多糖-3。
本发明的鹰嘴豆芽多糖具有保湿作用,并且可以作为皮肤外用剂组合物中的添加剂。
在本发明的含有鹰嘴豆芽多糖作为有效成分的皮肤外用剂组合物中,其中鹰嘴豆芽多糖通过以下方法制备:
A、取鹰嘴豆,洗净,于温水中浸泡6-8小时后,取出置于豆芽箱中,豆芽箱的温度30℃,淋水时间5秒,淋水间隔为180分钟。豆芽长至5-6cm左右,将豆芽和豆瓣分开,取豆芽,备用;
B、取新鲜鹰嘴豆芽加70%乙醇超声提取2次,每次1小时,过滤,取豆芽残渣备用;
C、取鹰嘴豆芽醇提后残渣加入沸水煮3次,将每次滤液合并,浓缩,加三氯醋酸在4℃下脱蛋白,离心,上清液加NaOH中和,流水透析2d,浓缩,再加入95%乙醇使体系含醇量达75-85%,沉淀冷冻干燥得鹰嘴豆芽多糖。
可以选择的是,在本发明的含有鹰嘴豆芽多糖作为有效成分的皮肤外用剂组合物中,其中鹰嘴豆芽多糖通过以下方法制备:
A、鹰嘴豆芽的培育取鹰嘴豆,洗净,于温水中浸泡6-8小时后,取出置于豆芽箱中,豆芽箱的温度30℃,淋水时间5秒,淋水间隔为180分钟。豆芽长至5-6cm左右,将豆芽和豆瓣分开,取豆芽,备用;
B、取新鲜鹰嘴豆芽150克加70%乙醇1500毫升超声提取2次,每次1小时,过滤,得豆芽残渣备用;
C、取鹰嘴豆芽醇提后残渣30克,加入300毫升沸水煮3次,将每次滤液合并,浓缩到300毫升,加三氯醋酸在4℃下脱蛋白,离心,上清液加NaOH中和,流水透析2d,浓缩到150毫升,加入95%乙醇使体系含醇量达75%,沉淀冷冻干燥得鹰嘴豆芽多糖-1;
D、取鹰嘴豆芽醇提后残渣30克,加入300毫升沸水煮3次,将每次滤液合并,浓缩到300毫升,加三氯醋酸在4℃下脱蛋白,离心,上清液加NaOH中和,流水透析2d,浓缩到150毫升,加入95%乙醇使体系含醇量达80%,沉淀冷冻干燥得鹰嘴豆芽多糖-2;
E、取鹰嘴豆芽醇提后残渣30克,加入300毫升沸水煮3次,将每次滤液合并,浓缩到300毫升,加三氯醋酸在4℃下脱蛋白,离心,上清液加NaOH中和,流水透析2d,浓缩到150毫升,加入95%乙醇使体系含醇量达85%,沉淀冷冻干燥得鹰嘴豆芽多糖-3;
在本发明的皮肤外用剂组合物中,所述鹰嘴豆芽多糖在皮肤外用剂组合物中的重量百分比是0.001-20,较好的是5-20。
根据试验可知,鹰嘴豆芽多糖在保湿方面具有很好的功效,可以作为活性成分添加到化妆品配方中,这里所指的活性成分在某些情况下可以提供一种或一种以上的功效。
本发明中所述的活性成分还可以为泛醇、甘草酸二钾、尿囊素、烟酰胺、抗坏血酸磷酸酯钠、三色堇提取物、透明质酸钠、生育酚乙酸酯、十一碳烯酰基苯并氨酸等。此外,此类描述是为了便于理解,并不是使成分限于具体指出的应用或者所列出的应用。
鹰嘴豆芽多糖可应用于皮肤外用剂、保健食品和医药,尤其是可应用于化妆品中,根据制剂的不同类型添加不同的用量。
所述皮肤外用剂组合物是通常用于皮肤外部的所有成分的统称概念,例如可以是化妆料组合物或药学组合物。所述化妆料组合物中可以是基础化妆料、面部妆容化妆料、身体用化妆料、头发护理用化妆料等,对其剂型无特殊限制,根据不同目的可合理选择。
所述化妆料组合物中根据剂型和目的的不同还含有不同的化妆品学层面允许的介质或基质赋形剂。
可以用于本发明皮肤外用剂组合物的化妆品、皮肤病学或药学上可接受的赋形剂可以为水相、油相、凝胶、水包蜡型乳液、水包油型乳液或油包水型乳液的形式。水相为一种或多种水溶性或分散性组分的混合物,其在室温下可以为液体、半固体或固体。赋形剂包括或可以为在水或水-醇赋形剂中的混悬液、分散液或溶液的形式,其可以含有增稠剂或凝胶剂。本领域技术人员可以基于本领域技术人员掌握的知识选择合适的产品形式,其中包含的组分。
所述的组合物可以包括水相,该水相可以含有水或水与至少一种亲水性有机溶剂的混合物,所述的亲水性有机溶剂诸如醇,尤其是含有2-5个碳原子的直链或支链低级一元醇,如乙醇或丙醇;多元醇,如丙二醇、山梨醇、甘油、泛醇或聚乙二醇及其混合物。
当发明的组合物为乳液形式时,该组合物还可以任选包含表面活性剂。
所述的组合物还可以包含成膜聚合物,如聚氨基甲酸酯、聚丙烯酸均聚物或共聚物、聚酯、基于烃的树脂和/或硅氧烷树脂。可以将聚合物溶于或分散于化妆品可接受的赋形剂中并且任选与增塑剂合并。
本发明的组合物还可以包含油相,所述的油相含有在室温下为液体的油溶性或油分散性组分和/或在室温下为油状或蜡状的物质,如蜡、半固体、树胶及其混合物。该油相还可以含有有机溶剂。
通常在室温下为液体,合适的油性物质包括:来源于动物的基于烃的油,如全氢化角鲨烯;基于烃的植物油,如液体的C4-10脂肪酸的甘油三酯类,例如庚酸或辛酸甘油三酯类,或油,例如向日葵油、玉米油、大豆油、葡萄籽油、蓖麻油、鳄梨油、辛酸/癸酸甘油三酯类、霍霍巴油;矿物或合成来源的直链或支链烃类,例如液体石蜡及其衍生物、凡士林;合成酯类和醚类,特别是脂肪醇的酯类,例如肉豆蔻酸异丙酯、棕榈酸2-乙基己酯、硬脂酸2-辛基十二烷基酯、异硬脂酸异硬脂醇酯;羟基化酯类,例如乳酸异硬脂醇酯、羟基硬脂酸辛酯、羟基硬脂酸辛酯、羟基硬脂酸辛基十二烷基酯、脂肪醇的庚酸酯类、辛酸酯类和癸酸脂类;多元醇酯类,例如丙二醇二辛酸酯、新戊二醇二庚酸酯、二甘醇二异壬酸酯和季戊四醇酯类;含有C12-26的脂肪醇类,例如辛基十二烷醇、2-丁基辛醇、2-己基癸醇、2-十一烷基十五烷醇、油醇;基于部分烃的氟油和/或氟硅油,硅油,在室温下为液体或半固体的挥发性或非挥发性的直链或环状聚甲基硅氧烷,例如环状聚二甲基硅氧烷和聚二甲基硅氧烷,其任选包含苯基,例如苯基三甲基硅氧烷、硅氧烷及其混合物。
本发明的组合物可以进一步包含常用于化妆品领域中的任何组分。这些组分包括防腐剂、水相增稠剂(多糖生物聚合物、合成聚合物)和脂肪相增稠剂、芳香剂、亲水性和亲脂性活性剂及其混合物。
本发明的组合物还可以包含另外的颗粒相,所述的颗粒相可以为化妆品组合物中使用的颜料和/或珠光剂和/或填充剂。
颜料可以存在于组合物中,合适的无机颜料包括氧化钛、氧化锆和氧化铈以及氧化锌、氧化铁和铁蓝;合适的有机颜料包括钡、锶、钙和铝色淀和碳黑。
珠光剂可以存在于组合物中,合适的珠光剂包括涂覆了氧化钛、氧化铁或天然颜料的云母。
填充剂可以存在于组合物中,合适的填充剂包括滑石粉、二氧化硅、硬脂酸锌、云母、高岭土、尼龙粉末、聚乙烯粉末、特氟龙、淀粉、一氮化硼、共聚物微球,例如硅氧烷树脂微珠。
本发明组合物的油相可以包含一种或多种蜡、树胶或其混合物。蜡包括基于烃的蜡、氟蜡和/或硅氧烷蜡,并且可以来源于植物、矿物、动物和/或合成来源。合适的蜡包括蜂蜡、巴西棕榈蜡、小烛树蜡、石蜡、微晶蜡、地蜡;合成蜡包括聚乙烯蜡、含有C16-45的硅氧烷蜡。树胶一般为聚二甲基硅氧烷或羧甲基纤维素钠或多糖类,并且半固体物质一般为基于烃的化合物,如羊毛脂及其衍生物。
可以将本发明的组合物配制成任何合适的产品形式。这类产品形式包括,但不限于气溶胶型喷雾剂、霜剂、乳液、固体、液体、分散体、泡沫、凝胶、化妆水、摩丝、软膏、粉剂、贴剂、润发油、溶液、手按泵型喷雾剂、棒状物、面膜和湿纸巾。可以将本发明的组合物通过本领域众所周知的各种方法便利地用于制备或作为化妆品、皮肤病学或药物局部施用产品。
本发明的皮肤外用剂组合物可以包括一种或多种下列成分:抗过敏剂、抗微生物剂、抗氧化剂、螯合剂、着色剂去色素剂、润肤剂、乳化剂、表皮脱落剂、成膜剂、香料、保湿剂、昆虫驱避剂、润滑剂、药物活性剂、增湿剂、耐光剂、防腐剂、护肤剂、皮肤渗透增强剂、防晒剂、稳定剂、表面活性剂、增稠剂、粘度调节剂、维生素或其任意组合。

具体实施方式
为了使本发明的目的、技术方案及优点更加清楚明白,以下结合实施例,对本发明进行进一步详细说明。应当理解,此处所描述的具体实施例仅用以解释本发明,并不用于限定本发明。
实施例1:鹰嘴豆芽的培育
取鹰嘴豆(经石河子大学王琪教授鉴定为迪西类型鹰嘴豆),洗净,于温水中浸泡至表面起皱后,取出置于豆芽箱中,豆芽箱的温度30℃,淋水时间5s,淋水间隔为180min。豆芽长至5-6cm左右,将豆芽和豆瓣分开,取豆芽,备用。
实施例2 鹰嘴豆芽多糖提物制备
①、取新鲜鹰嘴豆芽150g,加70%乙醇1500ml,超声提取2次,每次1h,过滤,滤液合并,豆芽残渣备用。
②、取鹰嘴豆芽醇提后残渣约30g,加入300ml沸水煮3次,将每次滤液合并,浓缩至约300ml,加三氯醋酸在4℃下脱蛋白;离心,上清液NaOH中和,流水透析2d;浓缩至约150ml,加入95%乙醇使体系含醇量达75%;沉淀冷冻干燥得鹰嘴豆芽多糖-1。
③、取鹰嘴豆芽醇提后残渣约30g,加入300ml沸水煮3次,将每次滤液合并,浓缩至约300ml,加三氯醋酸在4℃下脱蛋白;离心,上清液NaOH中和,流水透析2d;浓缩至约150ml,加入95%乙醇使体系含醇量达80%;沉淀冷冻干燥得鹰嘴豆芽多糖-2。
④、取鹰嘴豆芽醇提后残渣约30g,加入300ml沸水煮3次,将每次滤液合并,浓缩至约300ml,加三氯醋酸在4℃下脱蛋白;离心,上清液NaOH中和,流水透析2d;浓缩至约150ml,加入95%乙醇使体系含醇量达85%;沉淀冷冻干燥得鹰嘴豆芽多糖-3。
实施例3 糖含量测定
采用硫酸-苯酚法,显色后用分光光度计测定490nm处吸收度,以葡萄糖为对照,绘制标准曲线,计算多糖中糖含量。
①、制作标准曲线:准确称取标准葡聚糖(或葡萄糖)20mg于500ml容量瓶中,加水至刻度,分别吸取0.4ml、0.6ml、0.8ml、1.0ml、1.2ml、1.4ml、1.6ml及1.8ml,各以蒸馏水补至2.0ml,然后加入6%苯酚1.0ml及浓硫酸5.0ml,摇匀冷却,室温放置20分钟以后于490nm测光密度,以2.0ml水按同样显色操作为空白,横坐标为多糖微克数,纵坐标为光密度值,得标准曲线。
②、取样品1克加1ml 15%三氯乙酸溶液研磨,再加少许5%三氯乙酸溶液研磨,倒上清液于10毫升离心管中,再加少许5%三氯乙酸溶液研磨,倒上清液,重复3次。最后一次将残渣一起到入离心管。注意:总的溶液不要超出10毫升。
③、离心,转速3000转/分钟,共三次。第一次15分钟,取上清液。后两次各5分钟取上清液到25毫升锥形比色管中。最后滤液保持18毫升左右。
④、水浴,在向比色管中加入2毫升6mol/L盐酸之后摇匀,在96℃水浴锅中水浴2小时。
⑤、定容取样。水浴后,用流水冷却后加入2毫升6mol/L氢氧化钠摇匀。定容至25毫升的容量瓶中。吸取0.2ml的样品液,以蒸馏补至2.0ml,然后加入6%苯酚1.0ml及浓硫酸5.0ml,摇匀冷却室温放置20分钟以后于490nm测光密度。每次测定取双样对照。以标准曲线计算多糖含量。
⑥、结果:经计算回归方程为Y=7.4898X-0.00442,R2=0.9996,葡聚糖浓度在0-105.0μg/mL范围内与吸光度呈良好的线性(如图1所示)。同法测定样品吸光度,计算得到鹰嘴豆芽多糖-1中总糖含量为33.85%,鹰嘴豆芽多糖-2中总糖含量为40.38%,鹰嘴豆芽多糖-3中总糖含量为38.67%。
实施例4 蛋白质含量
运用考马斯亮兰G250显色剂测定蛋白含量,于595nm处测定吸光度,以不加白蛋白溶液为空白,绘制标准曲线,计算多糖中蛋白质含量。
①、取16支试管,1支作空白,3支留作未知样品,其余试管分为两组按表中顺序,分别加入样品、水和试剂,即用1.0mg/ml的标准蛋白质溶液给各试管分别加入:0、0.01、0.02、0.04、0.06、0.08、0.1ml,然后用无离子水补充到0.1ml。最后各试管中分别加入5.0ml考马斯亮兰G-250试剂,每加完一管,立即在旋涡混合器上混合。
②、加完试剂2-5分钟后,即可开始用比色皿,在分光光度计上测定各样品在595nm处的光吸收值A595,空白对照为第1号试管,即0.1mlH2O加5.0ml G-250试剂。
③、用标准蛋白质量(mg)为横坐标,用吸光度值A595为纵座标,作图,即得到一条标准曲线。由此标准曲线,根据测出的未知样品的A595值,即可计算出未知样品的蛋白质含量。
④、结果:结果测定表明标准回归方程为:Y=0.0433X+0.0133,R2=0.9993,考马斯亮蓝G-250浓度在0-11.2μg/ml范围内与吸光度呈良好的线性(如图2所示),鹰嘴豆芽多糖-1中蛋白含量为3.25%,鹰嘴豆芽多糖-2中蛋白含量为0.60%,鹰嘴豆芽多糖-3中蛋白含量为1.27%。
实施例5 硫酸基含量测定运用硫酸钡比浊法测定鹰嘴豆芽多糖硫酸基含量。采用可见分光光度法,最大吸收波长为360nm,以K2SO4为标准品,绘制标准曲线,计算多糖中硫酸基含量。
①、称取1.000±0.001g试样于250mL烧杯中,加15mL盐酸(1:1),置电炉上低温加热,溶解完全。将烧杯移入沸水浴中,蒸至糖浆状。加水5mL混匀,冷却后移入100mL容量瓶中,用水稀释至刻度,混匀。分取试液10mL于100mL烧杯中,加1mL盐酸(1:1)、加5mL抗坏血酸,用水稀释至约20mL,混匀。
②、在50mL容量瓶中,加入0.5mL硫酸乙醇溶液、2mL氯化钡,放置1min后(呈晶体液),将烧杯中的试液移入容量瓶中,用水稀释至刻度、混匀。放置5min后,在460nm处,用3cm比色皿以水做参比,测吸光度。
③、标准曲线绘制和样品硫酸基含量计算:分别取SO4 2-0、0.4、0.6、0.8……1.6mg于一组100mL烧杯中,按上述操作,以吸光度A360为纵坐标,SO4 2-含量为横坐标,绘制标准曲线。再根据对应标准的回归方程,计算出该样品中硫酸基含量。
④、结果:用硫酸钡比浊法测定鹰嘴豆芽多糖中硫酸基含量,以K2SO4作标准,根据它们的标准系列回归方程求出样品中的硫酸基含量。结果测定表明回归方程为:Y=0.0682X+0.0809,R2=0.9997,K2SO4浓度在0-2.5μg/ml范围内与吸光度呈良好的线性(如图3所示)。同法计算鹰嘴豆芽多糖-1中硫酸基含量为1.28%,鹰嘴豆芽多糖-2中硫酸基含量为1.58%,鹰嘴豆芽多糖-3中硫酸基含量为1.19%。
实施例6 糖醛酸含量测定
采用咔唑-硫酸比色法,在最大吸收波长530nm处测定吸光度值,以半乳糖醛酸为对照品,绘制标准曲线,计算多糖中糖醛酸含量。
①、标准曲线绘制:精确称取干燥的单糖标准样品10mg,定容于25mL容量瓶中。然后准确量取0,0.1,0.2,0.4,0.6,0.8mL单糖标准溶液于带塞试管中,各管加水至1mL。在冰水浴中向各管加入6mL浓硫酸,摇匀后,改在85℃水浴中保持20min,取出后冷至室温,各管加0.2mL咔唑液,在室温下保持2h,测定吸收度(530nm),以浓度对吸收度作标准曲线。
②、测定各单糖在530nm处的检测响应:分别准确吸取各种单糖溶液(10mg/25mL)0.4mL,补水至1mL用前述方法测得其吸收值。
③、样品中糖醛酸含量的测定:样品中糖醛酸的吸收值为样品吸收值与中性糖吸收值之差,再根据对应标准的回归方程,计算出该样品中糖醛酸含量。
④、按硫酸-咔唑法测定鹰嘴豆芽多糖中糖醛酸含量,己糖醛酸经含四硼酸钠的硫酸作用可进一步与间羟联苯反应,形成有色物质在530nm处有最大吸收。吸收值与该糖的浓度呈良好的线性关系。以半乳糖醛酸作标准,根据它们的标准曲线,计算回归方程求出样品中的糖醛酸含量。测定结果表明回归方程为:Y=0.0683X-0.0052,R2=0.9996,半乳糖醛酸在2.51-25.1μg/ml浓度范围内与吸光度呈良好的线性(如图4所示)。计算鹰嘴豆芽多糖-1中糖醛酸含量为2.01%,鹰嘴豆芽多糖-2中糖醛酸含量为1.02%,鹰嘴豆芽多糖-3中糖醛酸含量为1.66%。
实施例7 鹰嘴豆芽多糖保湿作用
测定吸湿性需在恒温恒湿环境下进行,可以采用密闭容器中放置某种饱和盐溶液,在规定温度下使之保持一定的相对湿度。本实验选用饱和硫酸铵和乙酸钾溶液置于小干燥器中,环境温度为20℃。选用5%甘油为对照,测定了鹰嘴豆芽多糖的吸湿率。将样品干燥至恒质量后称其质量,分别置于相对湿度为50%的干燥器中,放置4h、12h、24h和48h,称其质量变化。
保湿率计算公式:
保湿率(%)=1-(M1-M2)/M1×100%,
其中,M1为放置前样品质量(g);M2为放置后样品质量(g)。
取实施例2所制得的鹰嘴豆芽多糖,加蒸馏水制成0.05mg/ml溶液备用,进行体外保湿作用评价,分别考察8h,12h,24h和48h时水分测定,计算其保湿率,结果见表1。

由表1可见,鹰嘴豆芽多糖较阳性对照在8h-48h内,均具有保湿效果,尤其是鹰嘴豆芽多糖-2,在48h时,保湿率仍为8.8%,可见其具有较强时间的保湿作用,且较阳性对照(5%甘油)强。
实施例8:含有鹰嘴豆芽多糖的皮肤外用剂
取实施例2中制备得到的鹰嘴豆芽多糖,用于皮肤外用剂的制备。所述皮肤外用剂优选为化妆品组合物,例如化妆水、精华液、乳霜等。所述鹰嘴豆芽多糖在皮肤外用剂中的重量百分比是0.001-20,优选5-20。以下是鹰嘴豆芽多糖在皮肤外用剂中的具体应用的实施例。以下各表中“-”表示无添加。
实施例9:含有鹰嘴豆芽多糖的化妆水含有的组分如表2所示:

取实施例2制备的鹰嘴豆芽多糖,用于制备具有保湿作用的精华液,所述精华液含有的组分如表3所示:

Claims (7)
1.一种鹰嘴豆芽多糖,其特征在于,所述鹰嘴豆芽多糖中总多糖含量为33.85%-40.38%,蛋白含量为0.60%-3.25%,硫酸基含量为1.19%-1.58%,糖醛酸含量为1.02%-2.01%。
2.如权利要求1所述的鹰嘴豆豆芽多糖,其特征在于所述鹰嘴豆芽多糖通过以下方法制备:
A、取鹰嘴豆、洗净、于温水中浸泡至表面起皱后,取出置于豆芽箱中,每隔一段时间淋水一次,直至豆芽长至5厘米,将豆芽和豆瓣分开,取豆芽;
B、取豆芽加乙醇超声提取,过滤,取豆芽残渣;
C、取豆芽残渣加入沸水煮,将每次滤液合并,浓缩,加三氯醋酸脱蛋白,离心,上清液加氢氧化钠中和,流水透析,浓缩,再加入乙醇使体系含醇量达70-85%,沉淀冷冻干燥得鹰嘴豆芽多糖。
3.如权利要求1所述的鹰嘴豆豆芽多糖,其特征在于所述鹰嘴豆芽多糖通过以下方法制备:
A、取鹰嘴豆,洗净,于温水中浸泡6-8小时后,取出置于豆芽箱中,豆芽箱的温度30℃,淋水时间5秒,淋水间隔为180分钟,豆芽长至5-6cm,将豆芽和豆瓣分开,取豆芽,备用;
B、取新鲜鹰嘴豆芽加70%乙醇超声提取2次,每次1小时,过滤,滤液合并,得豆芽残渣备用;
C、取鹰嘴豆芽醇提后残渣加入沸水煮3次,将每次滤液合并,浓缩,加三氯醋酸在4℃下脱蛋白,离心,上清液加NaOH中和,流水透析2d,浓缩,再加入95%乙醇使体系含醇量达75-85%,沉淀冷冻干燥得鹰嘴豆芽多糖。
4.如权利要求2或3所述的鹰嘴豆豆芽多糖,其特征在于C步骤为:
1)、取鹰嘴豆芽醇提后残渣加入沸水煮3次,将每次滤液合并,浓缩,加三氯醋酸在4℃下脱蛋白,离心,上清液加NaOH中和,流水透析2d,浓缩,加入95%乙醇使体系含醇量达75%,沉淀冷冻干燥得鹰嘴豆芽多糖-1;
2)、取鹰嘴豆芽醇提后残渣,加入沸水煮3次,将每次滤液合并,浓缩,加三氯醋酸在4℃下脱蛋白,离心,上清液加NaOH中和,流水透析2d,浓缩,加入95%乙醇使体系含醇量达80%,沉淀冷冻干燥得鹰嘴豆芽多糖-2;
3)、取鹰嘴豆芽醇提后残渣,加入沸水煮3次,将每次滤液合并,浓缩,加三氯醋酸在4℃下脱蛋白,离心,上清液加NaOH中和,流水透析2d,浓缩,加入95%乙醇使体系含醇量达85%,沉淀冷冻干燥得鹰嘴豆芽多糖-3。
5.如权利要求2所述的鹰嘴豆豆芽多糖,其特征在于,鹰嘴豆芽多糖C制备步骤中,所述加入乙醇的体积浓度为95%。
6.一种含有如权利要求1-5任一项所述的鹰嘴豆芽多糖的皮肤外用剂组合物,其特征在于,所述鹰嘴豆芽多糖在皮肤外用剂组合物中的重量百分比为0.001-20。
7.根据权利要求6所述的皮肤外用剂组合物,其特征在于,所述鹰嘴豆芽多糖在皮肤外用剂组合物中的重量百分比为5-20。
