Abstract
本发明公开了一种增强胃肠动力、润肠通便、改善便秘的药食两用品种组合物及制备方法,属于食疗产品、保健品及中药制备领域。该药食两用品种组合物是由:沙棘叶、蒲公英、桑叶、薤白、菊苣组成,其提取物重量份数的组成:沙棘叶提取物15-25,蒲公英提取物15-28,桑叶提取物9-16,薤白提取物5-9,菊苣提取物35-50。上述组合物的制备方法是先分别对沙棘叶、蒲公英、桑叶、薤白、菊苣进行低温超声提取,低温浓缩,低温带式干燥,然后将提取物制备成各种口服剂型。本发明组成全为药食两用品种,安全、可长期服用。本发明产品具有增强胃肠动力,润肠通便,改善便秘的作用。

技术领域:
本发明涉及食疗和保健品及中药制备领域,具体涉及一种润肠通便的药食两用组合物及其制备方法。
背景技术:
功能性便秘是临床上较为常见的一种功能性消化系统疾病。流行病学资料显示,欧洲普通人群便秘的平均患病率为17.1%,我国普通人群慢性便秘的患病率为6%。随着饮食结构改变、生活节奏加快和社会心理因素影响,我国慢性便秘的患病率呈逐年上升趋势。最新便秘诊治指南显示,他有两个年龄高峰:30-40岁人群及60-70岁人群,女性比男性多,60岁以上人群慢性便秘患病率高达22%。2013年9月29日搜狐健康网报道:我国超7千万女性患慢性便秘带来的困扰就可清楚看到慢性便秘严重危及着国民的健康。问题在于临床医学诊治,西医学和中医学对慢性便秘都没有标本兼治的方法。一些刺激性泻药,如番泻叶制剂会出现依赖性其用量越来越大,甚至易导致结肠黒变病,增加致癌风险。
目前,采用沙棘叶、蒲公英、桑叶、薤白、菊苣药食两用品种组合物润肠通便,有效干预慢性便秘的食疗产品和保健品及准字药品尚属空白。
发明内容:
本发明的目的在于根据现有技术中的上述不足,提供一种润肠通便、治疗慢性便秘的药食两用品种组合物。
本发明的另一目的是提供上述润肠通便治疗慢性便秘的药食两用品种组合物制备方法。
本发明通过以下技术方案实现上述目的:
本发明药食两用品种组合物是由:沙棘叶、蒲公英、桑叶、薤白、菊苣组成。
本发明润肠通便治疗慢性便秘的药食两用品种组合物,由以下重量份数的药食两用品种提取物组成:沙棘叶提取物12-25,蒲公英提取物15-28,桑叶提取物9-16,薤白提取物5-9,菊苣提取物35-50.
现代生化研究表明,沙棘叶主要含有黄酮类、维生素类、粗纤维、多糖类、氨基酸、蛋白质、有机酸、胡萝卜素、微量元素、叶绿素等营养及活性成分。现代研究发现沙棘叶含维生素B族元素中有能够促进乙酰胆碱合成的成分,从而能够维持胃肠的正常蠕动和消化液分泌,有利于消化和肠内容物的排出。沙棘叶对小白鼠胃排空和家兔离体肠管的影响实验研究证实,沙棘叶提取液可以加快小鼠的胃排空速率,能够增强离体十二指肠的运动张力,对肠道自身神经有明显的作用。蒲公英主要含有维生素、黄酮类、菊糖、甾醇、胆碱、绿原酸等活性成分。《本草新编》中表述蒲公英泻胃火,火退而胃气自生,但其气甚平,既能泻火,又不损土,可以长久服而无碍。现代研究证实,蒲公英提取物中的阿魏酸、齐墩果酸、绿原酸具有促进胃肠动力作用。桑叶在古代有个雅称叫“神仙叶”,桑叶能有效治疗肺火上亢及肺经风热,降肺火入大肠,起到润肠通便的作用。薤白能活血、助胸阳,开心窍,散胸中与大肠气滞而起到润肠通便作用。菊苣提取物中的主要成分为菊粉,菊粉能在人的结肠中发酵,使双歧杆菌增值5-10倍,同时又能抑制潜在有害菌的生长,对便秘、滥用抗生素造成的腹泻有显著的改善作用。菊粉在结肠中发酵后,提高微生物菌群和气体量提高,从而促进肠道蠕动,缩短了粪便在结肠内的停留时间,使移动加快,减少了水分吸收时间,增加了粪便重量,有效预防便秘。本发明五味合用,形成活性成分群,其共同叠加效应起到更有效的增强胃肠动力、润肠通便有效治疗便秘。
本发明所述润肠通便、治疗便秘的药食两用品种组合物固体制剂的制备方法,步骤如下:
(1)按比例称取各药物。
(2)制备沙棘叶提取物:沙棘叶用10-30倍纯水浸泡1小时以上;40℃以上超声提取20分钟至1小时,提取2次,300目过滤得提取液;低温(50℃以下)负压(-0.097MPa)浓缩至固形物35%以上的浓缩液;低温(50℃以下)负压(-0.097MPa)带式干燥,制得80目以上的沙棘叶提取物。
(3)制备蒲公英提取物:蒲公英用10-30倍纯水浸泡1小时以上;40℃以上超声提取20分钟至1小时,300目过滤得提取液;低温(50℃以下)负压(-0.097MPa)浓缩至固形物35%以上的浓缩液;低温(50℃以下)负压(-0.097MPa)带式干燥,制得80目以上的蒲公英提取物。
(4)制备桑叶提取物:桑叶粉碎为粗粉,用10-30倍纯水浸泡1小时以上;40℃以上超声提取20分钟至1小时,提取2次,300目过滤得提取液;低温(50℃以下)负压(-0.097MPa)浓缩至固形物35%以上的浓缩液;低温(50℃以下)负压(-0.097MPa)带式干燥,制得80目以上的桑叶提取物。
(5)制备薤白提取物:薤白粉碎至20-40目,用10-25倍量的50-80%乙醇浸泡0.5小时以上;40℃以上超声提取20分钟至1小时,300目过滤得乙醇提取液;低温(50℃以下)回收乙醇,得提取液;滤渣加纯水8-15倍,40℃以上超声提取20分钟至1小时,300目过滤得提取液;合并两次提取液,低温(50℃以下)负压(-0.097MPa)浓缩至固形物35%以上的浓缩液;将浓缩液低温(50℃以下)负压(-0.097MPa)带式干燥,制得80目以上的薤白提取物。
(6)制备菊苣提取物:菊苣粉碎至20-40目,用20-30倍的纯水浸泡1小时以上;40℃以上超声提取20分钟至1小时,300目过滤得提取液;低温(50℃以下)负压(-0.097MPa)浓缩至固形物35%以上的浓缩液;将浓缩液低温(50℃以下)负压(-0.097MPa)带式干燥,制得80目以上的菊苣提取物。
(7)组合物产品制备:将沙棘叶提取物、蒲公英提取物、桑叶提取物、薤白提取物、菊苣提取物粉剂混合均匀,或加入食品级辅料制成各种剂型,包括粉剂、颗粒剂、丸剂。
本发明的有益结果:
1、本发明配方全为药食两用品种,确保了产品的安全性,可以长期服用。
2、本发明沙棘叶五味药食两用品种活性成份群的共同作用,起到促进胃肠动力作用,有效润肠通便,改善便秘。
3、本发明制备方法中,各品种的提取、浓缩、干燥的生产工艺过程,其温度都不高于50℃的低温下进行,最大限度保留了生物活性物质,大大提高了产品的生物活性作用。
4、本发明采用超声提取,不仅降低了提取温度,而且大大缩短了提取时间,降低了生产成本。
具体实施方式:
实施例1:
(1)按比例称取各药物:沙棘叶30公斤,蒲公英40公斤,桑叶25公斤,薤白25公斤,菊苣50公斤。
(2)制备沙棘叶提取物:将30公斤沙棘叶用25倍纯水浸泡1小时以上;45℃超声提取40分钟,300目过滤得提取液;滤渣加纯水10倍,45℃超声提取20分钟,300目过滤,合并两次提取液;低温(50℃)负压(-0.097MPa)浓缩至固形物40%以上的浓缩液;低温(50℃)负压(-0.097MPa)带式干燥,制得80目沙棘叶提取物细粉7.8公斤。
(3)制备蒲公英提取物:将40公斤蒲公英用25倍纯水浸泡1小时以上;45℃超声提取30分钟,300目过滤得提取液;低温(50℃)负压(-0.097MPa)浓缩至固形物40%以上浓缩液;低温(50℃)负压(-0.097MPa)带式干燥,制得80目蒲公英提取物细粉10公斤。
(4)制备桑叶提取物:将25公斤桑叶,用25倍纯水浸泡1小时以上;45℃超声提取40分钟,300目过滤得提取液;滤渣加纯水10倍,45℃超声提取20分钟,300目过滤,合并两次提取液;低温(50℃)负压(-0.097MPa)浓缩至固形物40%以上的浓缩液;低温(50℃)负压(-0.097MPa)带式干燥,制得80目桑叶提取物细粉5公斤。
(5)制备薤白提取物:将25公斤薤白粉碎至20-40目,用15倍量的60%乙醇浸泡0.5小时;40℃超声提取0.5小时,300目过滤得乙醇提取液;低温(50℃以下)回收乙醇,得提取液;滤渣加纯水8倍,40℃超声提取20分钟,300目过滤得提取液;合并两次提取液,低温(50℃以下)负压(-0.097MPa)浓缩至固形物40%以上的浓缩液;将浓缩液低温(50℃)负压(-0.097MPa)带式干燥,制得80目的薤白提取物细粉2.5公斤。
(6)制备菊苣提取物:将菊苣50公斤粉碎至20-40目,用30倍的纯水浸泡1小时;45℃超声提取40分钟,300目过滤得提取液;低温(50℃)负压(-0.097MPa)浓缩至固形物40%以上的浓缩液;将浓缩液低温(50℃)负压(-0.097MPa)带式干燥,制得80目菊苣提取物细粉25公斤。
(7)粉剂制备:将7.8公斤沙棘叶提取物粉剂、10公斤蒲公英提取物粉剂、5公斤桑叶提取物粉剂、2.5公斤薤白提取物粉剂、25公斤菊苣提取物粉剂加入高效混合机混制45分钟,将混制均匀的粉料分装包装。每袋5克。其理论产量10000袋。检验、成品。待用。制成润肠通便,改善便秘的药食两用品种组合物粉剂。
(8)服用剂量及方法:
早晚服用,每次1袋。
实施例2:
(1)按比例称取各药物:沙棘叶45公斤,蒲公英60公斤,桑叶37.5公斤,薤白37.5公斤,菊苣75公斤。
(2)制备沙棘叶提取物:将45公斤沙棘叶用25倍纯水浸泡1小时以上;45℃超声提取40分钟,300目过滤得提取液;滤渣加纯水10倍,45℃超声提取20分钟,300目过滤,合并两次提取液;低温(50℃)负压(-0.097MPa)浓缩至固形物40%以上的浓缩液;低温(50℃)负压(-0.097MPa)带式干燥,制得80目沙棘叶提取物细粉11.7公斤。
(3)制备蒲公英提取物:将60公斤蒲公英用25倍纯水浸泡1小时以上;45℃超声提取30分钟,300目过滤得提取液;低温(50℃)负压(-0.097MPa)浓缩至固形物40%以上浓缩液;低温(50℃)负压(-0.097MPa)带式干燥,制得80目蒲公英提取物细粉15公斤。
(4)制备桑叶提取物:将37.5公斤桑叶,用25倍纯水浸泡1小时以上;45℃超声提取40分钟,300目过滤得提取液;滤渣加纯水10倍,45℃超声提取20分钟,300目过滤,合并两次提取液;低温(50℃)负压(-0.097MPa)浓缩至固形物40%以上的浓缩液;低温(50℃)负压(-0.097MPa)带式干燥,制得80目桑叶提取物细粉7.5公斤。
(5)制备薤白提取物:将37.5公斤薤白粉碎至20-40目,用15倍量的60%乙醇浸泡0.5小时;40℃超声提取0.5小时,300目过滤得乙醇提取液;低温(50℃以下)回收乙醇,得提取液;滤渣加纯水8倍,40℃超声提取20分钟,300目过滤得提取液;合并两次提取液,低温(50℃以下)负压(-0.097MPa)浓缩至固形物40%以上的浓缩液;将浓缩液低温(50℃)负压(-0.097MPa)带式干燥,制得80目的薤白提取物细粉3.75公斤。
(6)制备菊苣提取物:将菊苣75公斤粉碎至20-40目,用30倍的纯水浸泡1小时;45℃超声提取40分钟,300目过滤得提取液;低温(50℃)负压(-0.097MPa)浓缩至固形物40%以上的浓缩液;将浓缩液低温(50℃)负压(-0.097MPa)带式干燥,制得80目菊苣提取物细粉37.5公斤。
(7)浓缩丸制备:将11.7公斤沙棘叶提取物细粉、15公斤蒲公英提取物细粉、7.5公斤桑叶提取物细粉、3.75公斤薤白提取物细粉、37.5公斤菊苣提取物细粉、7.4公斤淀粉加入高效混合机混制45分钟,将混制均匀的粉料加入搅拌混合机中,边搅拌边喷进适量纯水,将粉料制成软材,再将软材加入浓缩丸制丸机制丸、整丸、低温负压干燥、检丸、包装。每袋5.5克。其理论产量为15000袋。检验、成品。待用。制成润肠通便、改善便秘的药食两用品种组合物浓缩丸。
(8)服用剂量及方法:
每日2次,早晚服用,每次1袋。
本发明产品主要功效成份及含量:
产品:每100g中, 多糖 ≥5g
黄酮(芦丁计) ≥0.8g
人群服用产品效果:
本发明申请人已完成本发明食疗产品投放市场的前期准备工作,并开展了产品人群试食试验工作。本发明粉剂已有便秘患者8人服用,效果明显,服用本品2-3天后,润肠通便效果显现,且无任何不适出现。

1.一种润肠通便的药食两用组合物,其特征在于由以下品种组成:沙棘叶、蒲公英、桑叶、薤白、菊苣,其提取物重量份数的药物组成:沙棘叶提取物12-25,蒲公英提取物15-28,桑叶提取物9-16,薤白提取物5-9,菊苣提取物35-50。
2.根据权利要求1所述润肠通便的药食两用品种组合物粉剂、颗粒剂和丸剂的制备,其特征在于:
(1)制备沙棘叶提取物:沙棘叶用10-30倍纯水浸泡1小时以上;40℃以上超声提取20分钟至1小时,提取2次,300目过滤得提取液;低温(50℃以下)负压(-0.097MPa)浓缩至固形物35%以上的浓缩液;低温(50℃以下)负压(-0.097MPa)带式干燥,制得80目以上的沙棘叶提取物。
(2)制备蒲公英提取物:蒲公英用10-30倍纯水浸泡1小时以上;40℃以上超声提取20分钟至1小时,300目过滤得提取液;低温(50℃以下)负压(-0.097MPa)浓缩至固形物35%以上的浓缩液;低温(50℃以下)负压(-0.097MPa)带式干燥,制得80目以上的蒲公英提取物。
(3)制备桑叶提取物:桑叶粉碎为粗粉,用10-30倍纯水浸泡1小时以上;40℃以上超声提取20分钟至1小时,提取2次,300目过滤得提取液;低温(50℃以下)负压(-0.097MPa)浓缩至固形物35%以上的浓缩液;低温(50℃以下)负压(-0.097MPa)带式干燥,制得80目以上的桑叶提取物。
(4)制备薤白提取物:薤白粉碎至20-40目,用10-25倍量的50-80%乙醇浸泡0.5小时以上;40℃以上超声提取20分钟至1小时,300目过滤得乙醇提取液;低温(50℃以下)回收乙醇,得提取液;滤渣加纯水8-15倍,40℃以上超声提取20分钟至1小时,300目过滤得提取液;合并两次提取液,低温(50℃以下)负压(-0.097MPa)浓缩至固形物35%以上的浓缩液;将浓缩液低温(50℃以下)负压(-0.097MPa)带式干燥,制得80目以上的薤白提取物。
(5)制备菊苣提取物:菊苣粉碎至20-40目,用20-30倍的纯水浸泡1小时以上;40℃以上超声提取20分钟至1小时,300目过滤得提取液;低温(50℃以下)负压(-0.097MPa)浓缩至固形物35%以上的浓缩液;将浓缩液低温(50℃以下)负压(-0.097MPa)带式干燥,制得80目以上的菊苣提取物。
(6)组合物产品制备:将沙棘叶提取物、蒲公英提取物、桑叶提取物、薤白提取物、菊苣提取物粉剂混合均匀,或加入食品级辅料制成各种剂型,包括粉剂、颗粒剂、丸剂。
3.根据权利要求1所述润肠通便的药食两用组合物产品,其特征在于:每100g中,主要功效成份含量,多糖≥5g,黄酮(芦丁计)≥0.8g。
