本发明涉及一种抗氧化胶囊,属于保健品加工技术领域,其主要成分为蓝莓提取物,加入少量的辅料混合调配而成。本发明除蓝莓提取物和少量的辅料外无任何其他添加成分,充分发挥了蓝莓花青素极强的抗氧化功效,可用于抗氧化清除自由基、抗衰老消除亚健康状态、调理机体功能、促进人体新陈代谢、养生等。长期服用无任何副作用,具有很好的保健功效。

技术领域
本发明属于一种医药保健品,不含有任何化学试剂,为纯天然的抗氧化保健品。
背景技术
蓝莓的营养价值远远高于其它水果,常被誉为“世界浆果之王”。它还富含花青素、低糖、低脂肪,抗氧化能力强,它的果实和茎叶在食品、医药、化工等方面具有很高价值。研究证明,蓝莓提取物的主要有效成分花青素是当今人类发现最有效的天然抗氧化剂。花青素的抗氧化性能比维生素E高50倍,比维生素C高200倍。它对人体的生物有效性是百分之百,服用后二十分钟就能在血液中检测到。另有研究表明,与其它的抗氧化剂不同的是,蓝莓花青素能通过血脑屏障,直接保护大脑和神经系统。由于花青素的小分子及特殊的分子结构,其遇到自由基后会给出一个电子中和自由基,有效的清除自由基,且自身不形成能引发链锁反应的有害物质,所以它在人体内具有强大的生物活性。花青素与胶原蛋白有较强的亲和力,能形成一层抗氧化保护膜,保护细胞和组织不被自由基氧化;同时能帮助维生素C及维生素E的吸收利用,增强其抗氧化作用。目前,市面上对蓝莓花青素的利用较低,仅有的几种产品价位较高。因此,本发明以高纯度的天然提取物蓝莓花青素为原料,经现代工艺制成的具有抗氧化功能的保健食品,充分利用蓝莓提取物丰富的营养价值,具有很好的保健功效,弥补市场上此类产品的稀缺。
发明内容
本发明旨在提供一种由含较高剂量花青素的蓝莓提取物为原料制成的抗氧化保健品,深度开发对蓝莓提取物的应用,以满足人们的保健需要。
本发明的技术方案:蓝莓花青素抗氧化保健品按重量份比,由以下成分混合而成:含25%花青素的蓝莓提取物60-94% 、硬脂酸镁1-5% 、淀粉5-35%。
本发明优先推荐的组成为:含25%花青素的蓝莓提取物94%、硬脂酸镁1%、淀粉5%。保健品的剂型不限于此,按上述各药用原料中常用的辅料,还可制成片剂、粉剂、丸剂或其它常规制剂。
本发明采用的蓝莓提取物(花青素)是天然的强抗氧化剂,有相关研究用矢车菊素-3-葡萄糖苷进行大鼠饲喂实验,结果表明此花色苷可以降低血清蛋白和脂质体的过氧化作用,并且对重灌流诱发的局部贫血肝脏的氧化性损伤具有保护作用。酰化花色甙(芍药色素-3-咖啡酸槐糖甙-5-葡萄糖甙)给大鼠灌胃后,发现其机体抗氧化能力显著增强。与其它抗氧化剂不同,花青素有跨越血脑屏障的能力,可以直接保护大脑中枢神经系统。用双盲随机安慰剂对照试验对花青素和安慰剂进行比较,发现持续2周给61-75岁的老年受试者每天服用119mg花青素可以明显降低缺血再灌注损伤诱导的血浆中F2-异前列腺素浓度,而安慰剂则无明显改善作用。同时,服用花青素也降低了尿液中氧化核酸的分泌(如8-羟基脱氧鸟苷、8-羟基鸟苷),但没有降低尿液中F2-异前列腺素的分泌。因此这些数据表明花青素可以提高老年人体内抗氧化能力。
本发明的有益效果:本发明以纯天然蓝莓提取物为主要成分,生物效价高,蓝莓花青素抗氧化功效得到极强发挥,可以用于调理机体功能、促进人体新陈代谢、防止脑部神经老化、抗癌、养生等。服用方便,长期服用无任何副作用,且保健效果显著。

具体实施方式
下面结合附图和实施例对本发明作进一步详细的说明。
本发明实施例中采用的蓝莓提取物按以下方法得到:野生蓝莓清洗后榨汁,用6倍体积百分浓度80%的乙醇加热回流提取3次,每次3h,合并提取液,减压回收乙醇,所得液体过大孔树脂吸附,用一定体积的洗脱液冲洗柱子,收集洗脱液,低温浓缩后,喷雾干燥,即得蓝莓提取物。紫外分光光度法测量其中总花青素的质量百分含量为25%。
实施例 1
一种抗氧化胶囊,按重量份比,由以下成分混合而成,蓝莓提取物(含25%花青素)94%、硬脂酸镁1%、淀粉5%。
其制备方法为:
1)取上述原料取样化验合格后,充分混合,混合粉末过筛50-80目,筛去可能存在的固体杂质。2)将混合物填充胶囊打光。3)胶囊分装:分装用塑料瓶,去其外包装进入缓冲间,紫外线灯照射后,进入内包装工序使用,胶囊分装合格后铝箔封口旋盖。以Co60剂量5GY辐射灭菌。
在美国,花青素的夏季平均日摄取量大约为215mg,冬季则为180mg。花青素因为其毒理数据非常有限而被认可用于食物中,1982年,计算出0-2.5mg/kg体重的日容许摄入量(ADI)(葡萄皮提取的3%花青素),相当于对于一个60-70kg的成人,每天最大摄取量为150-175mg花青素。参考美国ADI值及上述各双盲试验文献中的实验有效剂量,本发明制成的抗氧化保健品的有效成分花青素日服用剂量为150-170mg,折算成使用优先推荐比例配制的抗氧化保健品的日服用剂量为638-723mg。
实施例 2
蓝莓花青素对健康果蝇子代生长发育的影响
1 实验材料
1.1实验试剂
按照实施例1的比例称取蓝莓提取物(含25%花青素)。总超氧化物歧化酶(T-SOD)测试盒、过氧化氢(Catalase CAT)测定试剂盒及丙二醛(MDA)测试盒均购于南京建成生物工程研究所。
1.2实验昆虫
野生型黑腹果蝇(Canton-S)品系
2实验方法
在基本培养基中分别添加入 0.01%、0.05%、0.1%、0.3%的蓝莓花青素提取物,每组接种2对未交配的雌雄蝇,产卵3天后去亲本,每组处理10管,管中果蝇为F1代。各处理组的F1代果蝇用同样方法再接种培养得到F2代,以上实验均重复3次。选取0.1%蓝莓花青素处理的8h内孵化的F1代果蝇,雌雄分开,每组 10 只,进行荧光定量 PCR 检测Hsp70、Sod、Cat 的表达量,用试剂盒检测蝇体内总 SOD 和 CAT 的活性及MDA 的含量。
3实验结果
如图1、2所示,通过用0.1%蓝莓花青素处理对果蝇 Hsp70、Sod 及 Cat 基因表达量的影响以及对果蝇 SOD、CAT活性及 MDA 含量的影响的结果可见,蓝莓花青素处理组雌雄果蝇的分子标志物Hsp70基因表达水平和代谢标志物MDA的含量都出现了显著下降,说明果蝇机体氧化应激水平的降低,同时机体内环境也得到了改善。而Sod和Cat基因表达水平的结果可能与花青素具有保护核酸、激活酶活性等功能相关,同时其酶活性的结果,主要是花青素通过果蝇体内的非酶性防御系统直接参与清除自由基等代谢有害物质,从而对机体起保护作用。
上述结果表明,蓝莓花青素能够同时通过果蝇体内的酶性和非酶性防御系统发挥其抗氧化功效,降低果蝇体内的氧化损伤水平。表明该受试样品具有抗氧化功能。
实施例 3
蓝莓花青素对老龄大鼠的抗氧化作用
1实验材料
1.1 实验试剂
按照实施例1的比例称取蓝莓提取物(含25%花青素)、硬脂酸镁和淀粉,混合均匀制成受试样品;丙二醛(MDA)试剂盒、谷胱甘肽过氧化物酶(GSH-PX)试剂盒、血羰基和超氧化物歧化酶(SOD)试剂盒,购自南京建成生物工程研究所。
1.2 实验动物
清洁级雄性 SD老龄大鼠40只(10月龄),由上海斯莱克实验动物有限责任公司提供。
2 实验方法
按卫生部保健食品功能检测与评价技术规范,实验设三个剂量组和一个对照组,以人体推荐量的5倍(大鼠)为其中的一个剂量组,另设两个剂量组,高剂量一般不超过30倍,本发明中的抗氧化保健品推荐的成人日服用剂量设定为0.67g/60kgbw,即0.01g/kgbw,因此,试验设一个老龄对照组和三个剂量组,分别相当于推荐量的5、10、30倍:低剂量组:0.05g/kgbw、中剂量组:0.10g/kgbw、高剂量组:0.30g/kgbw。大鼠适应环境5d后,采尾血测定老龄鼠血清中丙二醛(MDA)的含量。根据其含量将老龄鼠随机分为3个剂量组,即0.05、0.10、0.30g/kgBW和1个老龄对照组。连续给予受试样品30d,灌胃量为10ml/kgBW。实验结束时采血并分离血清,测定MDA含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)的活力。
3实验结果
如图3、4、5、6所示,实验开始时,各剂量组体重、初始MDA水平均无统计学差异;实验结束时,高剂量组血清SOD含量明显高于老龄对照组,MDA水平及血羰基蛋白含量低于老龄对照组,差异有统计学意义,说明老龄大鼠体内脂质过氧化反应主要终产物MDA的含量显著降低,并提高主要抗氧化酶SOD的活性。而各组GSH含量与老龄对照组比较均无统计学意义。根据结果表明该受试样品作为有效的抗氧化剂,具有经济、多效的抗氧化优点。
Claims (4)
1.一种花青素抗氧化保健品,其特征是用下述重量份组分配制而成:含花青素的蓝莓提取物60-94% 、硬脂酸镁1-5% 、淀粉5-35%。
2.按权利要求1所述的花青素抗氧化保健品,其特征在于每份蓝莓提取物中花青素含量达25%。
3.按权利要求1所述的花青素抗氧化保健品,其优先推荐的组成为:含25%花青素的蓝莓提取物94%、硬脂酸镁1%、淀粉5%。
4.按权利要求3所述的花青素抗氧化保健品,其特征是:所含有效成分花青素的日服用剂量为150-170mg,折算成使用优先推荐比例配制的抗氧化保健品的日服用剂量为638-723mg。
