发明详述
本发明提供了生成CD8+T细胞的方法和材料,所述CD8+T细胞能够识别表达HER2/neu多肽的癌细胞。例如,本发明提供了在体内或体外使用由SEQ IDNO:l中所列氨基酸序列组成的多肽生成CD8+T细胞的方法和材料,所述CD8+T细胞能够识别并裂解表达HER2/neu多肽的癌细胞。在一些情况下,本发明提供了SLAFLPESFD多肽和包含SLAFLPESFD多肽的疫苗组合物以及使用SLAFLPESFD多肽或包含SLAFLPESFD多肽的疫苗组合物生成CD8+T细胞的方法,所述CD8+T细胞能够识别表达HER2/neu多肽的癌细胞。
在一些情况下,本发明所提供的多肽(例如由SEQ ID NO:1所列氨基酸序列组成的多肽)可与树突细胞联用以治疗癌症。例如,可使用与SLAFLPESFD多肽接触的树突细胞治疗癌症。
本发明提供的SLAFLPESFD多肽可以是基本纯的。对于多肽而言,术语“基本纯”表示多肽已与其天然状态下伴随的细胞组分分离。例如,通过合成制备的多肽可以是基本纯的多肽。通常,当本发明提供的多肽是至少60重量百分比(例如,65%、70%、75%、80%、90%、95%或99%)、不含蛋白质和天然产生的在天然状态下相关的有机分子时,可以认为该多肽是基本纯的。一般而言,基本纯的多肽可在非还原性聚丙烯酰胺凝胶上产生单一主条带。
可使用多种方法制备本发明提供的SLAFLPESFD多肽。由于其相对短的尺寸,SLAFLPESFD多肽可根据已知方法在溶液中或固相自动合成器上合成。参见例如Stewart和Young,Solid Phase Polypeptide Synthesis(《固相多肽合成》),第2版,皮尔斯化学公司(Pierce Chemical Co.)(1984);Tam等,J.Am.Chem.Soc,105:6442(1983);Merrifield,The Polypeptides(《多肽》),Gross和Meienhofer编,学术出版社(academic Press),纽约,第1-284页(1979)。在一些情况下,本发明提供的多肽(例如SLAFLPESFD多肽)可使用氨基化合物(如NH2)或游离酸(如COOH)C末端合成,两者均能够结合HLA-A2。

在一些情况下,本发明提供的多肽可与CD8+T细胞群体孵育以生成CD8+T细胞的活化库,所述CD8+T细胞能够识别p373-382或HER2/neu多肽。例如,本发明提供的一种或多种多肽(例如SLAFLPESFD多肽)可以离体方式使用以制备可用于治疗癌症的抗原特异性CD8+T细胞。在一些情况下,本发明提供的多肽可用于生成能够单独使用的活化HER2/neu多肽特异性CD8+T细胞库,或与单克隆抗体疗法、CTL疗法或者单克隆抗体疗法和CTL疗法联用以治疗癌症。例如,抗HER-2/neu单克隆抗体疗法可与使用p373-382多肽生成的CD8+T细胞的输注联用以治疗癌症(例如乳腺癌)。在一些情况下,贺赛汀(曲妥珠单抗)疗法可与使用p373-382多肽生成的CD8+T细胞的输注联用以治疗癌症(例如乳腺癌)。
本发明还提供了包含免疫原性有效量的一种或多种本发明提供的多肽的疫苗组合物。本发明提供的疫苗组合物可用作预防性或治疗性疫苗。可使用任意合适的技术给予和制备本发明提供的疫苗组合物,所述技术包括但不限于他处所述的那些技术(参见例如,美国专利申请公开号2010-0310640的[0132]-[0173])。
在一些情况下,本发明提供的疫苗组合物可包含GM-CSF(例如沙格司亭)、林塔托利(例如)、IL-2、IL-12、佐剂或其组合。例如,本发明提供的疫苗组合物可包括GM-CSF和佐剂。佐剂的示例包括但不限于CpG寡核苷酸、单磷酰脂质A和蒙塔尼ISA-51。在一些情况下,所述佐剂可以是油和水的混合物(如蒙塔尼ISA-51)。
在一些情况下,本发明提供的疫苗组合物可包括多肽组合。例如,本发明提供的疫苗组合物可包括SLAFLPESFD多肽和/或表1所列一种或多种其他多肽以及美国专利申请公开号2010-0310640、Karyampudi等参考文献(Clin.Cancer Res.,16(3):825-34(2010))、Holmes等参考文献(J.Clin.Oncol.,26(20):3426-33(2008))、Gritzapis等参考文献(Vaccine,28(l):162-70(2009))、Perez等参考文献(Cancer Immunol.Immunother.,50(11):615-24(2002))、Knutson等参考文献(J.Clin.Invest.,107(4):477-84(2001))或Salazar等参考文献(Clin.Cancer Res.,9(15):5559-65(2003))中所列一种或多种多肽。
可使用任何合适的方法以向哺乳动物(例如人)给予本发明提供的疫苗组合物。例如,本发明提供的疫苗组合物或多肽可以100至10,000毫克的剂量单独给予或与其他多肽联合给予,每个月通过皮内或皮下途径给予并持续总共四至十二个月(例如4、5、6、7、8、9、10、11或12个月)。
本发明提供的方法和材料可用于治疗任何类型的表达HER2/neu多肽的癌症。例如,本发明提供的方法和材料可用于治疗乳腺癌、卵巢癌、结肠癌、食道癌或肺癌。
以下实施例进一步描述了本发明,这些实施例并不限制权利要求书所述的本发明范围。
实施例
实施例1-HER2/neu多肽的有效I型MHC分子表位的鉴定
HER2/neu是一种在多种癌症中过表达的多肽并且是一种治疗靶标,尤其在乳腺癌中。例如,NeuVax是一种早前开发的疫苗,其包括HER-2/neu衍生的多肽p369-377(氨基酸序列:KIFGSLAFL(SEQ ID NO:2),也称作E75),该多肽衍生自HER-2/neu的胞外结构域并与GM-CSF混合。该疫苗旨在刺激针对E75的免疫反应,从而生成能够识别并杀死患者体内癌细胞的T细胞,该T细胞可能在与MHC I型分子相关的其细胞表面展示相同肽段。
进行了以下实验以确定E75是否由HER-2/neu或HER-2/neu片段通过被称作蛋白酶体和免疫蛋白酶体的多亚基酶加工而产生,这是将多肽加载至MHC I型分子上所必需的。为确定这一问题,合成了19聚体多肽FAGCKKIFGSLAFLPESFD(SEQ ID NO:3)。该多肽与HER-2/neu氨基酸364-382匹配,且其完全包含E75(下划线)。随后使用纯化的20S蛋白酶体和免疫蛋白酶体切割19聚体。虽然E75被报道按蛋白酶体算法所确定的方式切割,但本文提供的体外数据并未发现该多肽由较长的HER-2/neu多肽加工生成(图1A和1B)。然而,发现所述19聚体始终被加工成数个其他较短的多肽(图1A和表1)。使用数学算法发现,这些较短的多肽的评分均无法达到E75的高度(表1)。
通过免疫蛋白酶体和蛋白酶体,来自HER-2/neu的19聚体序列被加工成较小的多肽片段,且预测这些片段可结合HLA-A*0201。符号(+)表示该多肽在体外试验中由相应的酶生成。符号(-)表示在体外试验中无法在包含相应酶的样品中检测到肽。使用SYFPEITHI服务器预测结合HLA-A*0201的九聚体和十聚体多肽。使用Netchop 3.1服务器上的20S和C端3.0预测法和用于蛋白酶体和免疫蛋白酶体酶的带有模型1、2和3的蛋白酶体切割预测服务器(Proteasome Cleavage Prediction Server)预测是否可通过酶将较大的19聚体序列加工生成较小的多肽(与体外数据无关)。NA(不适用)表示该多肽是缺失产物,是起始材料并且因此不会在试验中生成,或者太大以至于无法预测对HLA-A*0201的结合。图1A中数字和星号表示肽标记。
观察到的经加工的多肽之一(p373-382)显示带有19聚体的末端10个氨基酸。为确定该多肽是否可由较大的肽加工生成,合成了包含p373-382的23聚体并使用蛋白酶体处理。如图1C所示,p373-382确实释放自所述23聚体。
合成了试验中加工的许多多肽并使用标准T2HLA-A2稳定性试验测试其与I型MHC分子HLA-A2的结合。HLA-A2是一种MHC I型分子,普遍存在于约30-40%白种人中。该分子常被用作免疫试验中的靶标,因为其可能使大量乳腺癌患者受益。合成的多肽之一p373-382(SLAFLPESFD)能够牢固地结合HLA-A2分子,结合水平与阳性对照(来自流感病毒(FLU)的多肽)相当(图2)。出乎意料的是,与需要较高浓度的p369-377相比,甚至低水平的p373-382也有结合(图2)。
总之,这些结果显示HER-2/neu多肽p373-382由较长的HER-2/neu多肽加工生成并结合HLA-A2。
进行了ELIspot以确定p373-382表位是否通过癌细胞的细胞机理天然地加工生成,并确定其是否具有结合癌细胞表面上HLA-A2的潜力,此时其可作为已接触抗原并活化的免疫细胞的靶标。如果特定的免疫细胞能够识别癌细胞上的p373-382:HLA-I型复合物,则这些免疫细胞可杀死癌细胞并防止癌症在患者体内发展。由于已显示p373-382多肽在体外通过细胞机理加工生成且能够牢固地结合HLA-A2分子,因此进行了ELIspot以确定是否可以使用p373-382多肽生成CD8+T细胞且这些T细胞是否能够识别HER-2/neu+乳腺癌细胞。
图3A显示使用pFLU多肽(对照)、p369-377和p373-382生成了CD8+T细胞系。如预期的那样,对照FLU T细胞仅识别用FLU多肽脉冲处理的靶细胞。HER-2/neu p369-377多肽生成的T细胞识别用p369-377多肽脉冲处理的靶细胞和用p373-382脉冲处理的靶细胞。p373-382多肽生成的T细胞识别用p373-382多肽或p369-377多肽脉冲处理的靶细胞,表明这两种多肽之间具有交叉反应性,这可能是由于其含有五个相同的氨基酸。
随后,在体外ELISPOT中评估生成的T细胞系以确定其是否能够识别在表面表达有不同HER-2/neu水平的乳腺癌细胞系组。在所有情况下,与p369-377生成的CD8+T细胞和对照FLU CD8+T细胞相比,p373-382生成的CD8+T细胞能够以明显更高的水平识别乳腺癌细胞(图3B)。BT20细胞表达HER-2/neu但不表达HLA-A2,因此用作阴性对照。这些结果表明在HLA-A2存在的情况下乳腺癌细胞在其表面上表达p373-382,且使用p373-382生成的CD8+T细胞能够识别这些癌细胞。
进行了另一个体外试验以测定T细胞对乳腺癌细胞的裂解。同样,与使用p369-377生成的CD8+T细胞相比,使用p373-382多肽生成的CD8+T细胞以明显较高的水平识别并裂解所有测试的乳腺癌细胞系(图3C)。在该试验中,BT20细胞是阴性对照,同样作为阴性对照的还有表达HLA-A2但不表达HER-2/neu的FLO细胞。
最后,为确认p3737-382多肽以HLA限制性方式活化CD 8T细胞,在中和性HLA-A2或HLA-ABC单克隆抗体存在的情况下测试了使用图3中所用三种多肽生成的T细胞系的肽特异性反应性或裂解活性。如图4A-B所示,通过IFN-γ释放的评估,与使用对照同种型匹配的抗体处理的T细胞相比,无论何种抗体的介入均会显著抑制p373-382生成的T细胞的反应性。平行地,还观察到中和性HLA-A2或HLA-ABC单克隆抗体阻断了p373-382生成的T细胞对肿瘤细胞的裂解,分别如图4C-D所示。
以上三段中所述的发现使用由二至三种HLA-A2+供体制备的T细胞重复了二至四次。
使用算法以确定HLA-A2之外其他HLA等位基因结合p373-382或一些其他嵌合多肽的潜力。使用的算法是SYPEITHI和NetMHCpan。该研究的结果暗示p373-382或一些片段能够结合其他HLA类分子(表2)。
