Abstract

本发明涉及天然抗氧化物质,尤其涉及莲雾叶中抗氧化物质的提取方法及其应用。本发明公开的莲雾叶中抗氧化物质的提取方法,是将莲雾叶剪碎,加入乙醇溶液,回流提取,合并滤液并浓缩至无醇味,制得浸膏,所得浸膏用水溶解后,依次用石油醚、乙酸乙酯、正丁醇萃取,分别收集石油醚有机相、乙酸乙酯有机相、正丁醇有机相、水相,将收集到的有机相和水相分别回收溶剂后即可制备得抗氧化物。本发明的制备方法工艺简单、容易操作、成本低廉;所制备的莲雾叶抗氧化物稳定性强、保质期长。

莲雾叶中抗氧化物质的提取方法及其应用
技术领域
本发明涉及天然抗氧化物质,尤其涉及莲雾叶中抗氧化物质的提取方法及其应用。
背景技术
抗氧化剂是存在于人体或食物中、与可氧化化底物的浓度相比具有较低浓度浓度并能显著延缓或者防止底物氧化的物质。抗氧化剂的作用在于,通过抑制和延缓氧化以及延长食品的保质期并减少浪费和营养损失。在食品中,抗氧化剂或作为内源组分被添加到食品中,通过控制氧化及其有害产物改善产品质量。抗氧化剂保护食品免受氧化的机理是,通过提供电子或氢离子,或者灭火金属离子和纯态氧来消除自由基。理想的用于食品的抗氧化剂要安全,不会影响色泽、气味或者味道,在低浓度下有效,易于整合到食物中,在苛刻的加工条件下不失活,在成品中稳定,并且成本低廉。
长期以来,食品工业主要使用人工合成的抗氧化剂主要为二叔丁基羟基甲苯(BHT)、叔丁基羟基茴香醚(BHA)等化学合成抗氧化剂。但愈来愈多的研究结果表明,人工合成的抗氧化剂具有一定的毒副作用,人们越来越倾向于用天然抗氧化剂取代合成抗氧化剂。因此,研发开发高效、安全的天然抗氧化剂,成为当今食品科学研究的热点。
发明内容
本发明的目的在于提供一种天然抗氧化物质的提取方法及其应用,旨在解决现有技术中使用人工合成的抗氧化剂所带来的问题。
为了实现上述目的,本发明采用了以下的技术方案实现:
莲雾叶中抗氧化物质的提取方法,所述方法包括以下步骤:
1)将莲雾叶剪碎,加入8~10倍质量的乙醇溶液,回流提取2~3小时,重复提取3~4次,过滤,合并滤液;
2)将步骤1)所得滤液浓缩至无醇味,制得浸膏;
3)将步骤2)所得浸膏用水溶解,依次用石油醚、乙酸乙酯、正丁醇萃取,分别收集石油醚有机相、乙酸乙酯有机相、正丁醇有机相、水相,将收集到的有机相和水相分别回收溶剂后即可制备得抗氧化物。
优选的是:步骤1)中所述乙醇溶液中乙醇体积百分数为65~85%。
优选的是:步骤1)中所述回流提取的温度为70~85℃。
优选的是:步骤3)中所述浸膏与水成1:1体积比;所述浸膏与石油醚成1:2体积比;所述浸膏与乙酸乙酯成1:4体积比;所述浸膏与正丁醇成1:3体积比。
上述提取方法获得的莲雾叶抗氧化物可作为添加剂在食品、化妆品、保健品中的应用。
本发明的有益效果如下:
本发明的制备方法工艺简单、容易操作、成本低廉;所制备的莲雾叶抗氧化物稳定性强、保质期长。从莲雾叶提取的高浓度的抗氧化物可用于开发高含量的新型天然莲雾叶抗氧化剂,它可以作为添加剂在食品、化妆品、保健品实现其应用,进而实现了莲雾高附加值的综合利用。

下面结合具体实施例,对本发明作进一步详细的阐述,但本发明的实施方式并不局限于实施例表示的范围。这些实施例仅用于说明本发明,而非用于限制本发明的范围。此外,在阅读本发明的内容后,本领域的技术人员可以对本发明作各种修改,这些等价变化同样落于本发明所附权利要求书所限定的范围。
实施例1
取1千克莲雾叶洗净、沥干、剪碎,加入8千克体积分数为65%的乙醇溶液,于70℃下回流3小时,重复提取4次,合并滤液,58℃减压浓缩至无醇味,制得浸膏。取1L浸膏用1L水分散,依次用2L石油醚、4L乙酸乙酯和3L正丁醇室温下萃取,分别收集石油醚有机相、乙酸乙酯有机相、正丁醇有机相、水相。将所得石油醚有机相、乙酸乙酯有机相、正丁醇有机相、水相分别回收溶剂后即可制备得抗氧化活性极强的抗氧化物。所获得的莲雾叶抗氧化物在食品、化妆品、保健品中可作为添加剂加以使用。
实施例2
取2千克莲雾叶洗净、沥干、剪碎,加入18千克体积分数为70%的乙醇溶液,于75℃下回流2小时,重复提取3次,合并滤液,59℃减压浓缩至无醇味,制得浸膏。取2L浸膏用2L水分散,依次用4L石油醚、8L乙酸乙酯和6L正丁醇室温下萃取,分别收集石油醚有机相、乙酸乙酯有机相、正丁醇有机相、水相。将所得石油醚有机相、乙酸乙酯有机相、正丁醇有机相、水相分别回收溶剂后即可制备得抗氧化活性极强的抗氧化物。所获得的莲雾叶抗氧化物在食品、化妆品、保健品中可作为添加剂加以使用。
实施例3
取3千克莲雾叶洗净、沥干、剪碎,加入30千克体积分数为85%的乙醇溶液,于85℃下回流2小时,重复提取3次,合并滤液,60℃减压浓缩至无醇味,制得浸膏。取3L浸膏用3L水分散,依次用6L石油醚、12L乙酸乙酯和9L正丁醇室温下萃取,分别收集石油醚有机相、乙酸乙酯有机相、正丁醇有机相、水相。将所得石油醚有机相、乙酸乙酯有机相、正丁醇有机相、水相分别回收溶剂后即可制备得抗氧化活性极强的抗氧化物。所获得的莲雾叶抗氧化物在食品、化妆品、保健品中可作为添加剂加以使用。

Claims (5)

1.莲雾叶中抗氧化物质的提取方法,其特征在于:所述方法包括以下步骤:
1)将莲雾叶剪碎,加入8~10倍质量的乙醇溶液,回流提取2~3小时,重复提取3~4次,过滤,合并滤液;
2)将步骤1)所得滤液浓缩至无醇味,制得浸膏;
3)将步骤2)所得浸膏用水溶解,依次用石油醚、乙酸乙酯、正丁醇萃取,分别收集石油醚有机相、乙酸乙酯有机相、正丁醇有机相、水相,将收集到的有机相和水相分别回收溶剂后即可制备得抗氧化物。
2.根据权利要求1所述的提取方法,其特征在于:步骤1)中所述乙醇溶液中乙醇体积百分数为65~85%。
3.根据权利要求1所述的提取方法,其特征在于:步骤1)中所述回流提取的温度为70~85℃。
4.根据权利要求1所述的提取方法,其特征在于:步骤3)中所述浸膏与水成1:1体积比;所述浸膏与石油醚成1:2体积比;所述浸膏与乙酸乙酯成1:4体积比;所述浸膏与正丁醇成1:3体积比。
5.权利要求1所述提取方法获得的莲雾叶抗氧化物可作为添加剂在食品、化妆品、保健品中的应用。
