(2)室内栽培
a、从菌种厂引进优良菌株进行栽培;
b、培养基的配制
培养基配方: 黑米0.5千克、黑豆渣0.2千克、黑小麦0.1千克、菇渣0.5千克,防污剂35克,水1.5千克;其中防污剂由下列重量份的组分混合配制而得:齐墩果酸0.01-0.02、除虫菊酯0.02-0.03、纳米TiO2 0.01-0.02、硫酸铵0.1-0.2、硫酸锌0.01-0.02、磷酸二氢钾0.02-0.03、硫酸镁0.001-0.002、硼酸0.001-0.003、植物源提取物0.002-0.003、赤霉素0.0001-0.0002、沸石粉50-70、生石灰200-300,
所述的植物源提取物的制备方法为:将卷烟下脚料、辣椒根,切碎后用稀硫酸浸泡8-10h后,过滤,滤液浓缩,干燥得所述植物提取物;
配制方法:
选用厚度0.05毫米以上,规格为15厘米×30厘米或17厘米×30厘米,薄厚均匀、无破漏、韧性强的优质聚丙烯塑料袋为栽培袋,
将黑米、黑豆渣、黑小麦、菇渣混合均匀后按每袋50-60克分装入塑料袋里,将1.5升50℃的热水倒入干净盆内,将防污剂放入热水中,再加入小分子核苷酸盐5g,复合维生素制剂3g、红糖2g、硝酸钾0.02-0.03g,搅至完全溶解即成营养液,按料水比 1克:1.5毫升,称取营养液倒入装好在配料的塑料袋内,分装完后用塑料绳将每个塑料袋中部和口部各扎一道;
c、灭菌:灭菌锅内加适量水,把栽培料袋放入灭菌锅,升温至95-102℃,保持1-1.5小时,取出,冷却后移入接种室;
d、接种: 按常规的无菌接种法,在无菌条件下,打开封口材料,先将菌种捣成指头或豆粒大小块状,每袋接入10-12克菌种,均匀分布在料面上,接种完后按原扎口方法稍扎紧袋口,即可移入经杀菌和防虫处理的培养室内培养,把要把室内封闭好,避光;
e、管理:最初将培养室温度保持15-18℃遮光培养,第五天就可给光,给光后8-10天培养基由白转黄,再过2-3天就可见白芽出现,每袋扎6-8个眼,当菌丝生长至培养基1/2-2/3时,可将温度升至20℃,20天菌丝可发满袋 ,3天后,当培养基上部出现菌丝突起时,给予10-15℃的温差刺激,并增加光照刺激转色,7-10天转完色后对培养室消毒20-30分钟,然后解去培养料袋中部扎绳,用手把袋折成直筒状,当子座长至1厘米,及时对栽培袋通气,温度保持18-22℃,空气相对湿度80-90%;
f、采收:当草顶膜时也就达到6-8公分,采收烘干装袋即可。
小分子核苷酸盐是指:5′- 腺苷酸二钠或钾;5′- 鸟苷酸二钠或钾;5′- 肌苷酸二钠或钾;5′- 鸟苷酸二钠或钾;5′- 混合核苷酸的钠盐或钾盐中一种或多种。
小分子核苷酸物质对于整个植物界生产中的产量提高和产品品质改善有着不可替代的作用;核苷、核苷酸、嘧啶及嘌呤等核酸小分子特别是几种核苷酸盐和脱氧核苷酸盐及其配合,不但可以非常有效地调节植物的生长,又可以作为营养源被植物 迅速吸收,增强植株根系活力,提高植株抗逆能力,从而提高作物的产量、改善产品品质。核酸小分子对植物具有增强根系活力,提高根部对磷、钾肥的吸收;提高农作物苹果酸酶和琥珀酸酶的活性,促进植株的同化能力,增强光合作用的强度,对植物生长具有调节和营养作用,使用小分子核苷酸盐北虫草比一般培养在人工培育成活率高、生长周期短。
本发明的有益效果:
本发明提供一种北虫草工厂化人工种植生产技术,它依托现代农业科技的基础,运用现代工业技术手段,生成了一种复合的生产体系,工厂化生产通过人工控制的环境设施系统,克服自然环境的限制因素,营造一个适应采用高密度、立体化栽培环境,合理增加投入,提高产出水平,实现高投入、高产出、高效益的发展路子。 栽培的成功率高达90%以上, 栽培周期短,车间利用率高、产量高,生物学效率可达32%。
具体实施方式
一种北虫草工厂化人工栽培方法,包括如下内容:
(1)、栽培设施
a、盖栽培房首先要盖栽培房 要求放高应在4.0-4.5m ,长7-8m ,宽5-6m ,栽培房分为实验室、菌种室、灭菌室、接种室、发菌室、出草室、配料间、成品库、原料库,灭菌室、接种室、发菌室和出草室之间设有内隔门相连,每两个室之间有缓冲室,房内外地面及周围必须用水泥砖石铺设,整个建筑设施应结构牢固应结构牢固,具有良好的通风、光照条件,各个房间都应保证水电供应, 并配置控温控湿设备, 房顶每平方米装一盏40 w 或60 W 的日光灯;
b、搭建栽培床架

其中,植物源提取物的制备方法为:将卷烟下脚料、辣椒根切碎后,以任意比例混合,再用稀硫酸浸泡9h后,过滤,滤液浓缩,干燥得植物提取物;
栽培基的配制方法:
选用厚度0.05毫米以上,规格为15厘米×30厘米或17厘米×30厘米,薄厚均匀、无破漏、韧性强的优质聚丙烯塑料袋为栽培袋,
将黑米、黑豆渣、黑小麦、菇渣混合均匀后按每袋50-60克分装入塑料袋里,将1.5升50℃的热水倒入干净盆内,将防污剂放入热水中,再加入小分子核苷酸盐5g,复合维生素制剂3g、红糖2g、硝酸钾0.025g,搅至完全溶解即成营养液,按料水比 1克:1.5毫升,称取营养液倒入装好在配料的塑料袋内,分装完后用塑料绳将每个塑料袋中部和口部各扎一道;
c、灭菌:灭菌锅内加适量水,把栽培料袋放入灭菌锅,升温至95-102℃,保持1-1.5小时,取出,冷却后移入接种室;

f、采收:当草长到到6-8cm,就可以采收,采收前半个月不要喷水,边采边烘干,不要积压,烘干时把虫草放在平架上摊平不要太厚,太厚不易烘干,烘干时温度保持80℃,待干后再放在湿草上回潮,回潮到草有点扎手,但是虫草还不碎时就可装袋。

1.一种北虫草工厂化人工栽培方法,其特征在于,包括如下内容:
(1)、栽培设施
a、盖栽培房
首先要盖栽培房 要求房高应在4.0-4.5m,长7-8m,宽5-6m,栽培房分为实验室、菌种室、灭菌室、接种室、发菌室、出草室、配料间、成品库、原料库,灭菌室、接种室、发菌室和出草室之间设有内隔门相连,每两个室之间有缓冲室,房内外地面及周围必须用水泥砖石铺设,整个建筑设施应结构牢固,具有良好的通风、光照条件,各个房间都应保证水电供应,并配置控温控湿设备,房顶每平方米装一盏40W或60W的日光灯;
b、搭建栽培床架

2.根据权利要求1所述的北虫草工厂化人工栽培方法,其特征在于:制作培养基所用的营养液中所述的小分子核苷酸盐是指:5′-腺苷酸二钠或钾;5′-鸟苷酸二钠或钾;5′-肌苷酸二钠或钾;5′-混合核苷酸的钠盐或钾盐中一种或多种。
