Abstract
本发明属于功能性食品添加剂和营养保健品技术领域,具体的说是一种海藻多糖的应用。海藻多糖在功能性食品添加剂和营养保健品中用作制备益生元的用途。本发明作为益生元原料的海藻多糖均来源于大型经济红藻,原料来源丰富,生产成本低,制备工艺更简单,并且具有能提高人体免疫力、防治肿瘤等功效,因此其作为益生元具有非常明显的益生作用。

技术领域
本发明属于功能性食品添加剂和营养保健品技术领域,具体的说是一种海藻多糖的应用。
背景技术
随着肠道微生物学研究的发展,肠道微生物菌群的生殖平衡对健康的重要意义也逐渐被人们所认识。肠道中的益生菌如双歧杆菌、乳酸菌等在净化肠道、改善机体营养和维护宿主健康方面起到重要作用。而益生元是不被宿主胃肠道消化,能够选择性地刺激一种或几种有益菌在宿主肠道内生长,进而增进宿主健康的物质。
目前,益生元可分为以下几类:功能性低聚糖类,如低聚果糖、低聚木糖、低聚异麦芽糖等;微藻类如螺旋藻、节旋藻等,还有一些天然植物,包括蔬菜、中草药、野生植物等。其中功能性低聚糖是最主要,也是研究得最多的一类益生元。而以海洋藻类多糖或低聚糖作为益生元进行开发的报道较少,中国专利(CN100372542C)报道以褐藻胶低聚糖作为益生元进行应用,中国专利(CN1593433A)报道以琼胶寡糖作为益生元进行应用,对于海藻多糖作为益生元进行开发还未见报道。
发明内容
本发明的目的在于提供一种海藻多糖的应用。
为实现上述目的,本发明采用的技术方案为:
一种海藻多糖的应用,海藻多糖在功能性食品添加剂和营养保健品中用作制备益生元的用途。
所述海藻多糖中的单糖组成主要为D-半乳糖、L-半乳糖、3,6-内醚半乳糖;硫酸根含量为5-30%之间,平均分子量在2KDa-100KDa。
所述海藻多糖为以大型海洋红藻为原料通过热水浸提的方式所获得。具体是,以大型海洋红藻为原料在30-100℃,浸提0.5-24小时,而后通过醇沉后冻干、烘干或直接喷雾干燥,即得到海藻多糖。
所述大型海洋红藻为麒麟菜、蜈蚣藻、石花菜、琼枝、鹿角沙菜、卡帕藻等海藻中的一种或几种,但不局限于这几种。
本发明所具有的优点:
本发明作为益生元原料的海藻多糖均来源于大型经济红藻,原料来源丰富,生产成本低,制备工艺更简单,并且具有能提高人体免疫力、防治肿瘤等功效,因此其作为益生元具有非常明显的益生作用。
具体实施方式
实施例1
取破碎后的蜈蚣藻(Grateloupiafilicina)20g,加入600g蒸馏水,于90℃水浴中搅拌4小时,依次用100目,200目和300目的筛绢将滤渣滤出,滤液用减压浓缩设备进行浓缩为原体积的1/10,再用浓缩液4倍体积的95%的乙醇醇沉24小时,离心,沉淀冻干,即为蜈蚣藻多糖。
将上述得到的蜈蚣藻多糖配成1%(质量百分比)水溶液,待用。将市售的益生菌(主要为乳酸菌、双歧杆菌等益生菌混合物)用MRS培养基进行活化扩增。取扩增后的菌液置于96孔板中,分别取不同体积的蜈蚣藻多糖溶液,使其最终浓度为0.1%和0.5%,对照为菌液和培养基的混合液。结果表明,蜈蚣藻多糖浓度为0.1%时,益生菌的数量同对照组相比就增加了3倍,并且当蜈蚣藻多糖浓度为0.5%时,益生菌的数量比多糖浓度为0.1%组并没有明显差别。说明蜈蚣藻多糖具有非常好的益生作用。
实施例2
取破碎后的麒麟菜(E.maricatum)20g,加入800g蒸馏水,于70℃水浴中搅拌2小时,依次用100目,200目和300目的筛绢将滤渣滤出,滤液用减压浓缩设备进行浓缩为原体积的1/10,再用浓缩液4倍体积的95%的乙醇醇沉24小时,离心,沉淀冻干,即为麒麟菜多糖。
将上述得到的麒麟菜多糖配成1%(质量百分比)水溶液,待用。将市售的益生菌(主要为乳酸菌、双歧杆菌等益生菌混合物)用MRS培养基进行活化扩增。取扩增后的菌液置于96孔板中,分别取不同体积的麒麟菜多糖溶液,使其最终浓度为0.1%和0.5%,对照为菌液和培养基的混合液。结果表明,麒麟菜多糖浓度为0.1%时,益生菌的数量同对照组相比就增加了3倍,然而当麒麟菜多糖浓度为0.5%时,益生菌的数量比多糖浓度为0.1%组却有所降低。说明麒麟菜多糖具有非常好的益生作用,并且多糖过量时反而抑制益生菌的生长。
实施例3
取破碎后的石花菜(Gelidiumamansii)20g,加入600g蒸馏水,于100℃水浴中搅拌2小时,依次用100目,200目和300目的筛绢将滤渣滤出,滤液用减压浓缩设备进行浓缩为原体积的1/10,再用浓缩液4倍体积的95%的乙醇醇沉24小时,离心,沉淀冻干,即为蜈蚣藻多糖。

1.一种海藻多糖的应用,其特征在于:海藻多糖在功能性食品添加剂和营养保健品中用作益生元的用途;
所述海藻多糖中的单糖组成主要为D-半乳糖、L-半乳糖、3,6-内醚半乳糖;硫酸根含量为5-30%之间,平均分子量在2KDa-100KDa;
以大型海洋红藻为原料在30-100℃,浸提0.5-24小时,而后通过醇沉后冻干、烘干或直接喷雾干燥,即得到海藻多糖;
所述大型海洋红藻为麒麟菜、蜈蚣藻、石花菜中的一种或几种。
