实施例1–凝胶制剂优化
制备并检验不同的凝胶制剂以确定用笔型分配器给予的适合性。许多检验的制剂太易流动或太稠而不能与笔式分配器一起使用。选择基于最初的视觉评估被鉴定为具有可能接受的流变性质并用笔式分配器检验的三种制剂用于更详细的评估。制剂A、B和C根据下表制备(成分以总制剂的重量%计)。
比较制剂在预期使用中的适合性,在两个不同笔式分配器类型中检验制剂,一个分配器具有雌鹿脚末端,另一个具有刷子末端。结果在下表中扼要给出:
制剂B的组合物看起来最适于该应用。三种制剂之间的临界差异与它们的流变性质有关,如在下列总结表中所示:
所选凝胶为非牛顿型,在不同水平的力时显示非线性剪切稀化性质。制剂的具体流变性质使用TA Instruments的AR1000流变仪用4 cm 2度椎体几何学测量。粘弹性,例如弹性模量(G’)和损耗模量(G’’),从应变扫描实验获得。对于应变扫描测量,应变从0.1-500%变化时角频率保持在1 Hz。粘度测量从稳态流实验获得,该实验从1000-0.1 s-1改变剪切率进行。数据标绘在Herschel-Bulkley (HB)模型中(剪切应力=屈服应力+粘度* (剪切率)速率指数):

实施例2–用户试验
然后由潜在用户使用笔式分配器系统评估制剂B组合物的性能。该进展的目的是为了使用菌斑显露技术作为可通过更具靶向的递送系统运送的凝胶。更具靶向的递送允许较低的染料剂量,克服了用菌斑-显露牙膏的早期试验中所见的不可接受的脏乱和用户不满。该试验中探索的递送系统为具有两种不同末端—刷子末端和雌鹿脚末端的笔式涂抹器。受试者使用笔式涂抹器将菌斑-显露凝胶施用于其牙齿,检查着色菌斑,然后刷掉显露的凝胶和菌斑。
用户研究的目的是为了评估使用笔式涂抹器施用时凝胶的菌斑显露能力,评估用该凝胶染色软组织或菌斑是否存在任何问题,并观察用户与用于靶向递送的两种笔式涂抹器的相互作用,以揭露他们如何使用涂抹器、用该系统的任何问题以及是否该系统将改变他们的刷牙行为。
本研究为连续的单一(每天一个原型)中心定位试验。产品呈现无标记。应答者间未进行随机化,这是因为雌鹿脚涂抹器在研究(fielding)的第一个星期一不可用。使用菌斑显露凝胶和笔式涂抹器后给每个应答者的牙齿和牙龈拍照。
三十个试验参与者在两个连续的星期一下午访问了试验设施,在该处他们使用菌斑显露原型的一种,然后用1.0 g牙膏刷其牙齿。使用Compusense直接数据输入系统收集该过程的反馈。使用每种笔式涂抹器及菌斑显露凝胶后照片捕获每个人牙齿上的着色菌斑。要求小组成员在每个试验阶段开始前避免刷牙至少24小时。对于在90%置信水平的每个问题,报告平均等级和频率分布。
受试者对他们使用两种笔式涂抹器的体验的一些意见:
-“比预期更好地显示菌斑”
-“易于施用、扩散得很好并且凝胶具有清新的薄荷味道”
-“认为味道和时间将会较糟糕,不预期愉快的经历”。
100%的受试者报告其牙齿上的菌斑被显示,不管其使用刷子末端还是雌鹿脚末端涂抹器。超过80%的受试者发现使用刷子末端或雌鹿脚末端涂抹器后在刷牙期间很容易到非常容易地除去着色菌斑。刷牙后,几乎没有颜色保留在其牙齿、嘴唇或颊上,并且没有颜色保留在其牙龈上。大部分受试者没有察觉到两种涂抹器中的任一种在使用过程中脏乱。少于10%的受试者表达对该产品的不满。数个受试者评论该产品将通过指示其需要刷更多和需要给予问题区域(更大的斑块)更多关注的区域来改变其刷牙习惯,并且还评论该产品对于儿童将是重要的,其诱使他们更长时间和更细心地刷牙。
实际产品使用的反馈证实该产品比现有产品更具实质性优势,并且有效提供高效和有效的菌斑显露。该反馈与先前使用菌斑-显示洁齿剂的被受试者视为脏乱和无吸引力的试验相比非常积极。
Claims (21)
