发明概述

优选实施方案详述

可添加另外的元件,如果出于任何原因希望消除接受基因构建体的细胞,则所述元件充当用于细胞破坏的靶。可表达形式的疱疹病毒胸苷激酶(tk)基因可包含在基因构建体中。药物更昔洛韦可施用至个体并且所述药物将引起任何产生tk的细胞的选择性杀死,由此提供选择性破坏具有基因构建体的细胞的方式。
为了最大化蛋白质制备,可选择调控序列,所述调控序列非常适用于构建体所施用的细胞中的基因表达。而且,可选择在细胞中最高效地转录的密码子。本领域技术人员可制备在细胞中具有功能的DNA构建体。
在一些实施方案中,可提供基因构建体,其中本文所述的蛋白质的编码序列连接至IgE信号肽。在一些实施方案中,本文所述的蛋白质连接至IgE信号肽。
在使用蛋白质的一些实施方案中,例如使用熟知的技术,本领域技术人员可使用熟知的技术来制备和分离本发明的蛋白质。在使用蛋白质的一些实施方案中,例如,本领域技术人员可使用熟知的技术来将编码本发明的蛋白质的DNA分子插入在熟知表达系统中使用的市售表达载体中。例如,市售质粒pSE420(Invitrogen,SanDiego,Calif.)可用于在大肠杆菌中制造蛋白质。市售质粒pYES2(Invitrogen,SanDiego,Calif.)可例如用于在酵母的酿酒酵母菌株中的制备。市售MAXBACTM完整杆状病毒表达系统(Invitrogen,SanDiego,Calif.)可例如用于在昆虫细胞中的制备。市售质粒pcDNAI或pcDNA3(Invitrogen,SanDiego,Calif.)可例如用于在哺乳动物细胞如中国仓鼠卵巢细胞中的制备。本领域技术人员可使用这些商业表达载体和系统或其它载体和系统来通过常规技术和可容易获得的起始材料制备蛋白质。(例如参见Sambrook等,MolecularCloningaLaboratoryManual,第二版ColdSpringHarborPress(1989),其以引用的方式并入本文)。因此,所需蛋白质可在原核和真核系统中制备,从而产生一系列加工形式的蛋白质。
本领域技术人员可使用其它市售表达载体和系统或使用熟知方法和可容易获得的起始材料来制备载体。含有需要的控制序列(如启动子和聚腺苷酸化信号、并且优选增强子)的表达系统对于多种宿主来说是容易获得的和本领域已知的。例如参见Sambrook等,MolecularCloningaLaboratoryManual,第二版ColdSpringHarborPress(1989)。基因构建体包括可操作地连接至启动子的蛋白质编码序列,所述启动子在构建体所转染的细胞系中具有功能。组成型启动子的实例包括来自巨细胞病毒或SV40的启动子。可诱导启动子的实例包括小鼠乳腺白血病病毒或金属硫蛋白启动子。本领域技术人员可由可容易获得的起始材料容易地制备适用于使用编码本发明的蛋白质的DNA转染细胞的基因构建体。使用包含编码蛋白质的DNA的表达载体来转化相容性宿主,然后培养所述宿主并保持在其中发生外源DNA表达的条件下。
制备的蛋白质通过使细胞裂解来从培养物中回收或者从对于本领域技术人员来说适当和已知的培养基中回收。本领域技术人员可使用熟知技术分离使用这类表达系统制备的蛋白质。使用如上所述特异性结合特定的蛋白质的抗体从天然来源纯化蛋白质的方法可同样应用于通过重组DNA方法制备的蛋白质的纯化。
除了通过重组技术来制备蛋白质,还可以使用自动肽合成器来制备分离的基本上纯的蛋白质。这类技术是本领域技术人员熟知的,并且适用于具有DNA-编码的蛋白质制备中未提供的取代的衍生物。
核酸分子可使用若干种熟知的技术中的任一种来递送,所述技术包括DNA注射(也称为DNA疫苗接种)、重组载体,如重组腺病毒、重组腺病毒相关病毒和重组牛痘。
施用途径包括但不限于肌内、鼻内、腹膜内、皮内、皮下、静脉内、动脉内、眼内和口服以及局部、透皮、通过吸入或栓剂施用、或对粘膜组织施用,如通过灌洗阴道、直肠、尿道、口腔和舌下组织施用。优选的施用途径包括肌内、腹膜内、皮内和皮下注射。基因构建体可通过这样的方式来施用,所述方式包括但不限于电穿孔方法和装置、传统注射器、无针注射装置或"微弹轰击基因枪法"。

以下是使用电穿孔技术的实施方案的实例,并且在上述专利参考文献中更详细地讨论:电穿孔装置可被配置为递送产生类似于用户输入的预设电流的恒定电流的能量脉冲至哺乳动物的期望组织。电穿孔装置包括电穿孔部件和电极组件或手柄组件。电穿孔部件可包括且并有电穿孔装置的多种元件中的一种或多种,包括:控制器、电流波形产生器、阻抗试验器、波形记录仪、输入元件、状态报告元件、通信端口、记忆部件、电源和电源开关。电穿孔部件可充当电穿孔装置的一个元件,并且其它元件是与所述电穿孔部件通信的单独元件(或部件)。在一些实施方案中,电穿孔部件可充当电穿孔装置的多于一个元件,其可与电穿孔装置的独立于所述电穿孔部件的其它元件通信。使用电穿孔技术递送改进的HCV疫苗不受到现存作为一个机电或机械装置的部分的电穿孔装置的元件的限制,因为所述元件可充当与彼此通信的一个装置或单独元件。电穿孔部件能够递送在期望组织中产生恒定电流的能量脉冲,并且包括反馈机制。电极组件包括在空间布置中具有多个电极的电极阵列,其中电极组件从电穿孔部件接收能量脉冲,并且通过电极将所述能量脉冲递送至期望组织。多个电极中的至少一个在能量脉冲的过程中是中性的,并且测量期望组织中的电阻和将电阻通知电穿孔部件。反馈机制可接收所测量到的电阻,并且可调整由电穿孔部件递送的能量脉冲以保持恒定电流。
在一些实施方案中,多个电极可以分散型式来递送能量脉冲。在一些实施方案中,多个电极可以分散型式通过控制电极在编程序列下递送能量脉冲,并且编程序列由用户输入至电穿孔部件。在一些实施方案中,编程序列包含依序递送的多个脉冲,其中多个脉冲中的各脉冲通过至少两个有效电极(其中有一个中性电极测量电阻)来递送,并且其中多个脉冲的后续脉冲通过至少两个有效电极(其中有一个中性电极测量电阻)中的不同的那个来递送。

在一些实施方案中,核酸分子是结合多核苷酸功能增强子或基因疫苗促进剂的施用而递送至细胞。多核苷酸功能增强子描述于美国序列号5,593,972、5,962,428和1994年1月26日提交的国际申请序列号PCT/US94/00899,其均以引用的方式并入本文。基因疫苗促进剂描述于1994年4月1日提交的美国序列号021,579中,其以引用的方式并入本文。结合核酸分子施用的辅剂可作为与核酸分子的混合物来施用,或者在核酸分子施用的同时、之前或之后单独施用。另外,可用作转染剂和/或复制剂和/或炎症剂并且可与GVF共同施用的其它药剂包括:生长因子、细胞因子和淋巴因子,如a-干扰素、γ-干扰素、GM-CSF、血小板源生长因子(PDGF)、TNF、表皮生长因子(EGF)、IL-1、IL-2、IL-4、IL-6、IL-10、IL-12和IL-15以及成纤维细胞生长因子、表面活性剂如免疫刺激性复合物(ISCOMS)、弗氏不完全佐剂、LPS类似物(包括单磷酰基脂质A(WL))、胞壁肽、醌类似物和囊泡如三十碳六烯和角鲨烯,并且透明质酸也可与基因构建体结合而使用施用。在一些实施方案中,免疫调节蛋白可用作GVF。在一些实施方案中,联合提供核酸分子与PLG以增强递送/摄取。
根据本发明的药物组合物包含约1纳克至约2000微克DNA。在一些优选实施方案中,根据本发明的药物组合物包含约5纳克至约1000微克DNA。在一些优选实施方案中,药物组合物含有约10纳克至约800微克DNA。在一些优选实施方案中,药物组合物含有约0.1至约500微克DNA。在一些优选实施方案中,药物组合物含有约1至约350微克DNA。在一些优选实施方案中,药物组合物含有约25至约250微克DNA。在一些优选实施方案中,药物组合物含有约100至约200微克DNA。
根据本发明的药物组合物根据所使用的施用方式来进行配制。在药物组合物是可注射的药物组合物的情况下,它们是无菌、无热原质和无颗粒的。优选使用等渗制剂。通常,用于等渗性的添加剂可包括氯化钠、右旋糖、甘露糖醇、山梨糖醇和乳糖。在一些情况下,等渗溶液如磷酸盐缓冲盐水是优选的。稳定剂包括明胶和白蛋白。在一些实施方案中,将血管收缩剂添加至制剂中。

实施例
实施例1
pConNS4B、pConNS5A和pConNS5B的设计和表达
由获得自LosAlamosNationalLaboratoryHCV序列数据库的170种不同的序列产生HCV蛋白NS4B、NS5A和NS5B的HCV基因型1a共有序列。然后对共有构建体进行若干修饰以便增强它们的表达和检测,所述修饰包括在每个构建体的C-末端处和N-末端的HA-标签处添加前导序列IgE。另外,通过密码子和RNA优化使用GeneOptimizerTM(GENEART,Germany)对每个构建体进行进一步修饰,并且将其亚克隆至临床表达载体pVAX中受CMV启动子的控制。最终构建体称为pConNS4B、pConNS5A和pConNS5B(质粒图谱示出于图8A-图8C)。
通过用每个单独的构建体瞬时转染人RD肌细胞来确定每个构建体的蛋白质表达。使用LipofectamineTM(Invitrogen)根据制造商的指导用pConNS4B、pConNS5A和pConNS5B来瞬时转染RD肌细胞。转染之后48小时,将细胞固定并透化。先后使用抗-HA多克隆兔抗体(Invitrogen)和Cy3偶联的山羊抗-兔二级抗体(Invitrogen)来检测每个蛋白质的表达。
使用共聚焦显微镜并且在250倍放大率下观测细胞(图像未示出)。显示所有三种构建体都表达,pConNS4B示出最高数目的转染细胞而pConNS5B示出最低数目的转染细胞。用空白载体pVax进行转染作为对照。
实施例2
用pConNS4B、pConNS5A和pConNS5B免疫C57BL/6小鼠诱导强的细胞免疫应答
一旦确定了构建体的表达,就免疫C57BL/6小鼠以测定构建体的免疫原性。从杰克逊实验室购买6至8周龄的雌性C57BL/6小鼠,并且根据国立卫生研究院和宾夕法尼亚大学实验动物管理和使用委员会(IACUC)的指导维持它们。对于每个单独的构建体将动物分成三个不同的给料组,每组五只动物。用5μg、12.5μg或25μg的pConNS4B、pConNS5A或pConNS5B肌内免疫动物,随后进行电穿孔。
使用CELLECTRATM适应性恒定电流电穿孔装置和电极阵列(InovioPharmaceuticals,Inc.,BlueBell,PA)进行电穿孔。
动物总共接受两次免疫,相隔两周,并且在第二次免疫之后一周被处死。使用IFN-γELISpot测定来确定构建体的免疫原性。
如先前在Yan,J.等,EnhancedcellularimmuneresponseselicitedbyanengineeredHIV-1subtypeBconsensus-basedenvelopeDNAvaccine.MolTher,2007.15(2):第411-21页中所述进行小鼠IFN-γELISpot测定。用与8个氨基酸重叠并且涵盖每个构建体长度的15mer肽池刺激脾细胞。通过Genscript(Piscataway,NJ)合成肽,重悬浮于DMSO中并以2μg/ml/肽的浓度汇集。以每孔200,000个细胞的浓度铺板脾细胞。调整结果并以每1x10^6个脾细胞的斑点形成单位(SFU)的平均数来作图。结果在图1中可见。
如免疫荧光所测定的,构建体的免疫原性与构建体的表达水平密切相关。虽然所有的构建体都具有强的免疫原性,但对pConNS4B的应答是最大的,而对pConNS5B的应答是最小的。最优剂量:pConNS4B为12.5μg(1687±237SFU/10^6个脾细胞);pConNS5A为5μg(1091±111SFU/10^6个脾细胞);并且pConNS5B为12.5μg(736±136SFU/10^6个脾细胞)。
一旦确定了每种构建体的给料,就对由每种构建体所诱导的细胞免疫应答进行更详细的分析。如先前所述对动物进行免疫和分组。处死之后,将脾分离并使用Stomacher装置来单独压碎。脾细胞用40μM细胞滤过器滤过,并用ACK裂解缓冲液(Biosource)处理5分钟以清除RBC。将脾细胞重悬浮于完全培养基(RPMI1640,具有2mM/LL-谷氨酸,补充有10%热灭活FBS、1X抗生素/抗真菌剂、以及55μM/Lβ-巯基乙醇)中。使用血细胞计数器测定细胞数目。
为了测定对于每种构建体的CD8+和CD4+T细胞应答的相对贡献率,细胞内染色脾细胞的IFN-γ,并使用流式细胞术来观测,图2。细胞内细胞因子染色的结果与先前使用IFN-γELISpot测定所见到的那些结果密切相关。对于pConNS4B的应答是最大的,而pConNS5B具有最小的免疫原性。对于pConNS4B和pConNS5A的IFN-γ应答大部分是由CD8+T细胞产生的,但也识别到对每种构建体特异的CD4+T细胞。有趣的是,对于pConNS5B的IFN-γ应答大部分是CD4+T细胞介导的,而仅识别到很少的IFN-γ+CD8+T细胞。对于pConNS4B、pConNS5A和pConNS5B的IFN-γ+CD4+T细胞的平均百分比分别为0.50%±0.11%、0.27%±0.06%和0.32%±0.11%,图3A。对于pConNS4B和pConNS5A的IFN-γ+CD8+T细胞的平均百分比分别为3.29%±1.33%和0.68%±0.22%,图3B。
实施例3
肌内免疫之后在肝内检测到免疫诱导的NS4B-、NS5A-和NS5B-特异性T细胞
如先前在上文实施例1所述免疫小鼠。在最后一次免疫之后一周,将动物处死。处死之后,将肝分离并使用Stomacher机器来单独粉碎。将所得混合物滤过并使用10mlACK裂解缓冲液(Bioscience)处理5分钟以便清除RBC。沉淀混合物并通过使用35%珀可梯度将肝细胞自淋巴细胞分离。将沉淀的淋巴细胞重悬浮于完全培养基中。在或不在肝灌注下进行实验,没有观察到不同。
自每个肝分离T细胞,并用对应于每个单独构建体的重叠肽刺激。免疫诱导的HCV-特异性T细胞通过经由细胞内细胞因子染色和流式细胞术检测到的IFN-γ表达得到识别。单独分析每只动物。有趣的是,在所有免疫小鼠的肝中识别到HCV-特异性T细胞。在用pConNS4B和pConNS5A免疫的小鼠的肝中检测到CD4+和CD8+T细胞应答,而在用pConNS5B免疫的小鼠中仅检测到CD4+T细胞应答。在肝内检测到的优势T细胞应答与在脾内识别到的那些相同。用pConNS4B和pConNS5A免疫的小鼠在肝内具有强的CD8+T细胞应答,而用pConNS5B免疫的小鼠显示出主要的CD4+T细胞应答和很少的CD8+T细胞应答。对于pConNS4B、pConNS5A和pConNS5B的CD4+T细胞应答分别为0.29%±0.07%、0.41%±0.09%和0.41%±0.06%,图4A。对于pConNS4B、pConNS5A和pConNS5B的CD8+T细胞应答分别为3.73%±0.73%、2.28%±0.68%和0.06%±0.02%,图4B。
实施例4
3.4肝细胞肝特异性表达HCV抗原引起增加的IFN-γ产生和转染肝细胞清除
接着我们意图确定NS4B、NS5A或NS5B蛋白的肝特异性表达是否会活化在肝内检测到的HCV-特异性T细胞。为了诱导NS4B、NS5A和NS5B的肝特异性表达,通过如先前在Ahlen,G.等,InvivoclearanceofhepatitisCvirusnonstructural3/4A-expressinghepatocytesbyDNAvaccine-primedcytotoxicTlymphocytes.JInfectDis,2005.192(12):第2112-6页中所述施用pConNS4B、pConNS5A或pConNS5B的尾静脉注射液来转染免疫小鼠的肝细胞。使肝被转染48小时,之后收集肝并且如上文实施例3所述分离肝淋巴细胞。如之前所提及,免疫诱导的HCV-特异性T细胞通过经由细胞内细胞因子染色和流式细胞术检测到的IFN-γ分泌得到识别。
细胞内细胞因子染色
将脾细胞以1x10^6个细胞/100μl的浓度重悬浮于完全培养基中并铺在圆底96孔板中。脾细胞用100μl以下各项刺激:1)2μg/mlpConNS4B、pConNS5A或pConNS5B重叠肽;2)1μg/ml葡萄球菌肠毒素B(阳性对照;Sigma-Aldrich,St.Louis,MO)或3)0.1%二甲亚砜(阴性对照),这些都稀释在完全培养基中,补充有GolgiStop和GolgiPlug(BDBioscience)。在37C下刺激脾细胞共计5小时,之后细胞用PBS洗涤三次并染色以测定生存力。在4C下对脾细胞的表面标记物抗-CD4、CD8细胞外染色30分钟。之后将脾细胞透化并使用BDCytofix/Cytoperm溶液试剂盒(BDBioscience)洗涤,并且接着在4C下用抗-IFN-γ和CD3细胞内染色45分钟。染色之后,将脾细胞用1%多聚甲醛固定并储存在4C下直至进行分析。对于每只动物,特定函数报告为肽刺激的组的百分数函数减去0.1%二甲亚砜刺激的组(阴性对照)的百分数函数。
流式细胞术试剂
使用以下直接偶联的抗体:抗小鼠CD3-别藻蓝素花青染料7(APC-Cy7)[克隆145-C11]、抗小鼠CD4-异硫氰酸荧光素(FITC)[克隆H129.19]、抗小鼠CD8-多甲藻素叶绿素蛋白5.5(PerCP5.5)[克隆53-6.7]、抗小鼠IFN-γ-藻红蛋白花青染料7(PE-Cy7)[克隆XMG1.2](全部来自BDBiosciences,SanJose,CA)。根据制造商的方案使用水性活/死可固定的死细胞染色试剂盒(MolecularProbes,Eugene,OR)以识别活细胞。
在LSRII流式细胞仪(BDBiosciences,FranklinLakes,NJ)上收集样品。使用BDCompBeads(BDBiosciences)和单一荧光染料用于补偿。使用用于Mac的8.7.1版本的FlowJo软件(TreeStar,Ashland,OR)分析数据。
尾静脉注射之后,相比于在脾和所得肝中所检测到的HCV-特异性T细胞的百分比,在所有三个免疫组中见到CD4+和CD8+HCV-特异性T细胞的百分比大幅增加,图5。对于用pConNS4B、pConNS5A和pConNS5B免疫的小鼠,CD4+HCV-特异性T细胞的百分比分别为2.27%±0.70%、2.55%±0.70%和1.22%±0.22%,图6A。对于用pConNS4B、pConNS5A和pConNS5B免疫的小鼠,CD8+HCV-特异性T细胞的百分比分别为9.46%±1.53%、6.98%±0.48%和0.477%±0.16%,图6B。如在尾静脉注射之前和之后在肝中的HCV-特异性IFN-γ+T细胞的百分比所测定的,对于CD4+T细胞应答见到最大的成倍增加。pConNS4B、pConNS5A和pConNS5B免疫的小鼠中的肝内CD4+T细胞应答的成倍增加大约分别为8倍、6倍和3倍。虽然在尾静脉注射之前和之后在肝中CD8+T细胞应答仍然是优势应答,但与CD4+T细胞应答相比所见到的CD8+T细胞应答的成倍增加稍微较小。pConNS4B、pConNS5A和pConNS5B免疫的小鼠中的肝内CD8+T细胞应答的成倍增加大约分别为3倍、3倍和8倍。
在评定由每种构建体产生的肝内HCV-特异性IFN-γ应答之后,进行研究以确定免疫是否也产生肝内细胞毒性HCV-特异性T细胞。自每组的每只动物获得一叶肝,并对其染色以用于测定NS4B、NS5A或NS5B的肝细胞表达。通过在与转染的免疫未处理对照组相比时,每只免疫动物在转染之后清除NS4B、NS5A或NS5B表达肝细胞的能力,评定由用每种构建体进行免疫所产生的肝内T细胞应答的细胞毒性。观察每组的此染色的代表性共聚焦图像(图像未示出)。
共聚焦显微术
切割肝并将检体固定在2%多聚甲醛中,随后在30%蔗糖中超低温保护过夜。将检体浸没在Tissue-TekOCT(BayerCorporation,Pittsburgh,PA)中并在干冰氮气上在2-甲基丁烷中快速冷冻。对封固在SuperfrostPlus玻璃载片(FisherScientific,Pittsburgh,PA)上的组织切片(6μm)进行染色,并将其保持在80℃下直至使用。在免疫荧光染色之前,将载片带至室温下并在磷酸盐缓冲盐水(PBS)中洗涤三次,每次10分钟,并且用含有10%的其中产生第二试剂的物种的正常血清和0.1%的Triton的PBS阻断。将第一试剂施用于切片并将切片在室温下孵育1小时或在4℃下孵育过夜。切片在PBS中洗涤三次,每次10分钟,必要时在室温下施用第二试剂30分钟。将切片再次在PBS中洗涤三次,每次10分钟。用ProlongGold封固介质(Invitrogen,Carlsbad,CA)封固盖玻片,并将载片保持在4℃下在黑暗中,直至研究和拍照。所有的染色在潮湿环境中进行。所使用的抗体获得自Invitrogen,或者一个制造抗体的竞争对手公司。使用ZeissAxiovert100倒置共聚焦显微镜获得所有图像,并且使用ImageJ软件(NIH,Rockville,MD)对荧光强度进行分析和量化。
每组转染肝细胞的清除率通过由如核DAPI染色的MFI所测量的每个视野内存在的肝细胞的数目标准化的NS4B、NS5A或NS5B表达的平均荧光强度(MFI)来量化,图9。与未处理的对照组相比,对于所有三种构建体在动物免疫组中见到转染肝细胞的数目大量降低。与未处理的对照组相比,用pConNS4B、pConNS5A或pConNS5B免疫的动物的转染肝细胞表达下降大约9倍、3倍和2倍。在每个免疫组中所观察到的清除量与在转染的肝内检测到的HCV-特异性CD8+T细胞应答密切相关。在用pConNS4B免疫的动物中观察到最大的清除量,而在pConNS5B免疫的动物中观察到最小的清除率。
所提供的结果显示,通过全身免疫诱导的HCV-特异性T细胞在不存在同源抗原的肝特异性表达的情况下复原回到肝中,导致大的肝内HCV-特异性T细胞池形成。这些T细胞仍然在肝内充分发挥功能,这表明它们复原至静止肝中可另外充当免疫监视的持续过程的一部分,并且可证明是肝借以防范感染的重要机制。为了支持这一点,响应于HCV抗原的肝特异性表达,此肝定位的HCV特异性T细胞群体能够快速诱导IFN-γ表达并清除转染的肝细胞。由于先前已报告未观察到T细胞浸润直到肝转染72小时之后(Ahlen等,见上文),所以HCV转染的肝细胞的快速清除似乎很可能取决于在转染之前在肝内存在的肝定位的HCV特异性T细胞群体。另外,如在用pConNS5B免疫的动物中所见,即使相对较小百分比的疫苗特异性应答(如IFN-γ产生所测量的)也足以诱导肝内转染肝细胞的大的2倍减少,这表明如在外围所检测的较小百分比的疫苗特异性应答具有在肝内发挥巨大作用的能力。
肝诱导的T细胞耐受可通过全身免疫来破坏,并且有效的肝特异性免疫可通过利用肝在静止条件下复原和隔绝抗原特异性T细胞的能力来实现。肝的这种独特特性可利用来在已经感染有病毒的患者中增强HCV-特异性应答,并且在未处理个体的肝内产生具有在感染第一信号之后快速应答并迁移的能力的HCV-特异性T细胞池。总之,所述发现结果表明,抗原特异性T细胞至肝的复原连同它们的效应子功能在肝内的保留可以在免疫监视的过程中起到重要且以前未被认识到的作用,这可以在未来T细胞基HCV疫苗中加以利用。
Claims (12)

或
c)SEQIDNO:5。
4.如权利要求3所述的核酸分子,其中所述核酸分子缺少IgE前导序列的编码序列,其中所述缺少编码序列是SEQIDNO:7或SEQIDNO:8。

6.一种表达载体,包含如权利要求1至4中任一项所述的核酸分子,其中编码所述一个或多个蛋白质的序列可操作地连接至调控元件。
7.权利要求1至4中任一项所述的核酸分子、权利要求5所述的质粒或权利要求6所述的表达载体在制备用于治疗诊断患有HCV的受试者的药物中的用途。
8.一种蛋白质,其选自由以下各项组成的组:
a)SEQIDNO:2;
b)SEQIDNO:4;
或
c)SEQIDNO:6。
9.如权利要求8所述的蛋白质,其中所述蛋白质缺少由序列SEQIDNO:9组成的IgE前导序列。
10.权利要求8至9中任一项所述的蛋白质在制备用于治疗诊断患有HCV的受试者的药物中的用途。
