Abstract

技术领域
本发明涉及一种I型胶原改性的多孔钛涂层及其制备方法,具体涉及一种采用共价键接枝方式在等离子体喷涂钛涂层表面固定有I型胶原并具有良好生物学性能的I型胶原改性的多孔钛涂层及其制备方法,属于医用生物涂层技术领域。
背景技术
钛和钛合金等是临床上常用的骨植入材料。利用等离子体喷涂方法在其表面制备的多孔钛涂层,是改善其与骨组织结合的常用方法。多孔结构能够增加骨和植入体表面之间的机械嵌合作用,使植入体与骨组织间的结合强度明显提高。等离子体喷涂钛涂层的人工关节等硬组织植入体已获临床应用。然而,钛涂层属于生物惰性涂层,不具生物活性,对骨组织的生长并没有促进诱导作用,其临床使用效果仍有待改善。
为了赋予钛涂层生物活性,改善临床使用效果,人们采用多种方法对其进行表面改性。利用等离子喷涂、表面矿化等技术在多孔钛涂层表面沉积生物活性陶瓷涂层,可以有效地改善钛涂层的生物活性。例如中国专利CN101342387A公开一种钛涂层表面快速生成磷灰石的活化方法。但生物活性陶瓷涂层在体内容易降解,影响涂层的长期稳定性。化学处理也可以改善钛涂层的生物活性,但与生物活性陶瓷涂层如羟基磷灰石涂层相比,仍存在较大差距。
生物化学方法是一类新型的生物材料表面改性方法,其目的是将细胞外基质成分(如胶原)、酶或者多肽固定在生物材料上,构建仿生中间层,以引发特殊细胞和组织的响应。I型胶原是细胞外基质主要组成部分,影响着细胞粘附、迁移、增殖和分化。通过物理吸附,在钛涂层表面固定I型胶原,能够提高钛涂层的生物学性能和细胞相容性(StefanRammelt,et al.Journal of Orthopaedic Research.2004(22):1025-1034.)。例如,中国专利CN102145194A公开一种将具有多孔磷酸钙涂层的金属基材浸入胶原溶液后风干以制得多孔磷酸钙-胶原复合涂层的方法;中国专利CN101984144A公开一种采用电化学共沉积法在钛基板上以此组装磷酸钙层和胶原层的方法。但物理吸附固定的胶原有限,并且不稳定,容易脱附。
发明内容
本发明的目的是提供一种I型胶原改性的多孔钛涂层及其制备方法。在上述现有技术的基础上,本发明人意识到通过化学方法将I型胶原共价键接枝在多孔钛涂层表面,不但能够获得稳定的仿生中间层,而且能够提高所固定I型胶原的量以及固定的稳定性,从而获得相对于物理吸附方法生物学性能更好的改性多孔钛涂层。
在此,本发明提供一种I型胶原改性的多孔钛涂层,即采用生物化学改性方法,以共价键接枝方式,在多孔钛涂层表面固定有I型胶原的I型胶原改性的多孔钛涂层。
与物理吸附的胶原改性多孔钛涂层相比,本发明提供的共价接枝的I型胶原改性多孔钛涂层所固定的胶原量增加约一倍;固定稳定性更好,在7天后仍保留有90%以上;并且具有更好的生物学性能,可应用于促进诱导骨组织生长的骨植入材料。
另一方面,本发明还提供所述多孔钛涂层的制备方法,包括碱处理、硅烷化处理和共价键固定I型胶原。
较佳地,所述碱处理包括将多孔钛涂层浸入浓度为5~10M的NaOH溶液中在60~80℃下保温6~12h。
较佳地,所述硅烷化处理采用5~10%的氨丙基三乙氧基硅烷为硅烷化剂。
所述硅烷化处理可以采用甲苯为溶剂,加热回流反应6~12h。
较佳地,所述共价键固定I型胶原包括将经硅烷化处理的多孔钛涂层浸泡在I型胶原/乙酸溶液中并采用1-(3-二甲氨基丙基)-3-乙基碳二亚胺和N-羟基琥珀酰亚胺为羧基活化剂。
更佳地,所述共价键固定I型胶原可以是在浓度为1mg/mL I型胶原/5mM乙酸溶液中,滴加1-(3-二甲氨基丙基)-3-乙基碳二亚胺和N-羟基琥珀酰亚胺以使其在反应体系中的浓度分别为2~3.5mg/mL和0.25~0.875mg/mL。
所述共价键固定I型胶原的反应时间优选为4~10h。
较佳地,在所述硅烷化处理之前还包括去离子水陈化处理。
所述去离子水陈化可以是将经碱处理的多孔钛涂层在去离子水中40~80℃下保温3~7天。
本发明工艺简单、效率高、可重复性好。本发明所提供的I型胶原改性的多孔钛涂层,具有良好的稳定性和生物学性能,解决了现有技术中所存在的问题,可改善钛涂层的临床效果,为医用生物涂层技术提供新的方法。

具体实施方式
以下结合附图及下述具体实施方式进一步说明本发明,应理解,下述实施方式和/或附图仅用于说明本发明,而非限制本发明。
基于I型胶原良好的生物学性能和细胞相容性,本发明采用生物化学改性方法,以共价键接枝方式,将I型胶原固定在等离子体喷涂多孔钛涂层表面,从而获得生物学性能更好的I型胶原改性的多孔钛涂层。作为示例,其具体工艺如下。
(1)钛涂层的制备:利用等离子体喷涂技术制备钛涂层。
(2)碱处理:将所述钛涂层浸入浓度为5~10M的NaOH溶液中在60~80℃下保温6~12h,取出后可用去离子水超声清洗,超声清洗可以持续约5分钟。这里采用5~10M的NaOH作为碱处理的碱液,但应理解其他合适的碱液也是适用的,例如氢氧化钾。
(3)去离子水陈化:将经碱处理得到的多孔钛涂层浸泡在去离子水中40~80℃下保温3~7天。可以每天换一次水,最后40℃下干燥。
(4)硅烷化:将干燥的多孔钛涂层浸泡在浓度为5~10%的氨丙基三乙氧基硅烷(APS)/甲苯溶液中,溶液加热至沸腾,冷凝回流,反应6~12h,冷却后,分别用酒精、去离子水超声清洗1-2次,真空干燥。这里采用5~10%的氨丙基三乙氧基硅烷(APS)为硅烷化剂,采用甲苯作为溶剂,但应理解其他合适的硅烷化剂、溶剂也是适用。
(5)共价键固定I型胶原:将所述硅烷化多孔钛涂层浸泡在I型胶原/乙酸溶液中(例如浓度为1mg/mL I型胶原/5mM乙酸溶液)中,滴加入1-(3-二甲氨基丙基)-3-乙基碳二亚胺(EDC)水溶液(例如120mg/mL)和N-羟基琥珀酰亚胺(NHS)水溶液(例如30mg/mL),使得EDC最终溶度为2~3.5mg/mL,NHS最终溶度为0.25~0.875mg/mL,反应4~10h后取出,去离子水超声清洗,真空干燥。
下面进一步例举实施例以详细说明本发明。同样应理解,以下实施例只用于对本发明进行进一步说明,不能理解为对本发明保护范围的限制,本领域的技术人员根据本发明的上述内容做出的一些非本质的改进和调整均属于本发明的保护范围。下述示例具体的试剂的量和浓度、反应时间和温度也仅是合适范围中的一个示例,即、本领域技术人员可以通过本文的说明做合适的范围内选择,而并非要限定于下文示例的具体数值。
实施例1
[基于共价键固定I型胶原的改性钛涂层的制备]
利用等离子体喷涂技术制备钛涂层(标记为TC),所制备的钛涂层浸入5M NaOH溶液中,在80℃下保温12h,用去离子水超声清洗后,在去离子水中60℃下陈化7天,一天换一次水,40℃下干燥。碱处理改性钛涂层标记为TC-A。将干燥的样品TC-A浸泡在浓度为10%的APS/甲苯溶液中,溶液加热至沸腾,冷凝回流,反应12h。冷却后用酒精超声清洗1次,去离子水超声清洗2次,真空干燥。硅烷化的钛涂层标记为TC-AA。将样品TC-AA浸泡在浓度为1mg/mL I型胶原/5mM乙酸溶液中。每38mL胶原溶液中滴入120mg/mL EDC水溶液和30mg/mL NHS水溶液各1mL,使得EDC最终溶度为3mg/mL,NHS最终溶度为0.75mg/mL。反应6h后取出,去离子水超声清洗,真空干燥,样品标记为TC-AAC。
对比例1
[用物理吸附方法将I型胶原固定在钛涂层表面]
将实施例1中的中间样品TC-A浸泡在1mg/mL I型胶原/5mM乙酸溶液中,6小时后取出,去离子水超声清洗,真空干燥,样品标记为TC-AC。
图1为钛涂层表面固定I型胶原前后的表面形貌。在(c)、(d)中均能观察到纤维状的I型胶原,表明通过吸附固定方式和共价键固定方式,I型胶原成功地固定在钛涂层表面,得到新型的胶原改性钛涂层。
由图2所示的钛涂层改性前后的红外光谱可以看出:与TC-AA谱图相比,TC-AAC谱图上的Si-O-Si系列特征吸收峰(1020-1290cm-1)减弱,而氨基系列特征吸收峰(1300-1700cm-1)增强,进一步表明制备出新型胶原改性钛涂层。
[改性钛涂层的胶原固定效果实验]
利用天狼猩红染色法,从定量和固定稳定性两方面来考察胶原固定效果:将样品TC-AC和TC-AAC浸泡在浓度为1mg/mL的天狼猩红-饱和苦味酸溶液中,2小时后取出,去离子水冲洗,真空干燥。用0.1M的NaOH溶液将染料洗脱下来,然后用紫外分光光度计测定染料-NaOH溶液在540nm下的吸光度值,与吸光度值-胶原量标准曲线对照,得出固定胶原的量。将样品TC-AC和TC-AAC各4个浸泡在tris-HCl溶液中,37度下分别孵育1、3、5、7天。用天狼猩红染色法测量样品表面剩余胶原量。
图3所示是I型胶原的固定效果分析。从图3(a)中可以看出:TC-AAC的胶原量可达391μg/cm2,大于TC-AC的191μg/cm2,表明共价键固定方式能够固定较多的胶原。在图3(b)中,TC-AC的胶原在5天内就脱附殆尽,而TC-AAC的胶原7天后仍保留有90%以上,表明共价键固定的胶原稳定性更好。
[改性钛涂层的生物学性能实验]
用人骨髓间充质干细胞(hMSCs)来考察共价键固定I型胶原钛涂层的生物学性能:用MTT法考察hMSCs在材料表面的粘附和增殖行为;用BCA法对hMSCs的碱性磷酸酶(ALP)的活性进行定量测定。
图4为共价键固定I型胶原改性钛涂层的生物学性能实验。从图中可以看出:与TC相比,表面固定了I型胶原的改性钛涂层,能够促进hMSCs的粘附(图4(a))、增殖(图4(b))和碱性磷酸酶的活性表达(图4(c)),具有更好的生物学性能。而与TC-AC相比,TC-AAC具有更好的促进效果。结果表明:在钛涂层上共价键固定I型胶原,能够提高钛涂层的生物学性能。
产业应用性:本发明通过共价键接枝方式,将I型胶原固定在多孔钛涂层表面,获得一种I型胶原改性的多孔钛涂层,具有良好稳定性和生物学性能,不但能够加强钛和钛合金等骨植入材料与骨组织间的结合强度,而且改善多孔钛涂层的临床使用效果,促进诱导骨组织的生长,在医学生物涂层技术领域有广阔的应用前景。
Claims (8)
1.一种I型胶原改性的多孔钛涂层,其特征在于,采用生物化学改性方法,以共价键接枝方式,在多孔钛涂层表面固定有I型胶原,所述I型胶原改性的多孔钛涂层的制备方法包括碱处理、硅烷化处理和共价键固定I型胶原,其中,
所述硅烷化处理采用氨丙基三乙氧基硅烷为硅烷化剂,硅烷化剂溶解于甲苯中,浓度为5-10%,所述硅烷化处理采用甲苯为溶剂,加热回流反应6~12h。
2.一种权利要求1所述的多孔钛涂层的制备方法,其特征在于,包括碱处理、硅烷化处理和共价键固定I型胶原,其中,
所述硅烷化处理采用氨丙基三乙氧基硅烷为硅烷化剂,硅烷化剂溶解于甲苯中,浓度为5-10%,所述硅烷化处理采用甲苯为溶剂,加热回流反应6~12h。
3.根据权利要求2所述的制备方法,其特征在于,所述碱处理包括:将多孔钛涂层浸入浓度为5~10M的NaOH溶液中在60~80℃下保温6~12h。
4.根据权利要求2所述的制备方法,其特征在于,所述共价键固定I型胶原包括将经硅烷化处理的多孔钛涂层浸泡在I型胶原/乙酸溶液中并采用1-(3-二甲氨基丙基)-3-乙基碳二亚胺和N-羟基琥珀酰亚胺为羧基活化剂。
5.根据权利要求4所述的制备方法,其特征在于,滴加1-(3-二甲氨基丙基)-3-乙基碳二亚胺和N-羟基琥珀酰亚胺以使其在反应体系中的浓度分别为2~3.5mg/mL和0.25~0.875mg/mL。
6.根据权利要求4或5所述的制备方法,其特征在于,所述共价键固定I型胶原的反应时间为4~10h。
7.根据权利要求2所述的制备方法,其特征在于,在所述硅烷化处理之前还包括去离子水陈化处理。
8.根据权利要求7所述的制备方法,其特征在于,所述去离子水陈化包括:将经碱处理的多孔钛涂层在去离子水中40~80℃下保温3~7天。
