实施例4:双位点氨基酸突变对β-葡萄糖苷酶活性的影响
为了进一步研究105位Ser和273位Ile的共同突变是否会有协同效应,从而进一步提高β-葡萄糖苷酶的最适反应温度,进行了双位点氨基酸突变,即105位Ser和273位Ile均突变为Asn,得到S105N-I273N,核苷酸序列和氨基酸序列如SEQ ID No.15-16所示;按照实施例3的方法,对S105N-I273N进行了重组表达、纯化并对活性进行了测定;图2显示S105N-I273N的最适反应温度可以提高到48℃左右,表明105位Ser和273位Ile共同突变具有协同效应。
图3显示双突变体S105N-I273N在48℃时的酶活与野生型在36℃时的酶活相差不大,即,双位点氨基酸的突变没有影响β-葡萄糖苷酶的活性;而且,突变体在48℃保温12小时后,仍然具有80%以上的酶活性,表明其具有良好的热稳定性。
上述结果表明,双突变体S105N-I273N的最适反应温度提高到了48℃,其活性没有降低,并具有良好的热稳定性,可以用于高温条件下纤维素材料的降解以及食品工业等领域。
