Abstract

技术领域
本发明涉及用于检测体液中分析物的方法以及用于实施该方法的敷料(dressing)。
背景技术
为了以定性方式检测体液中分析物的存在,已经进行了许多工作。
几年来,比色法技术已经被应用于医学领域,特别是以满足确定伤口发展情况的需要以及描述伤口渗出物(exudate)中滋生(developing)的病菌(germ)的需要。
源于细菌(bacteria)的分类,革兰氏(Gram)试验是广泛普及的方法,其由于溶液中的染色试验而可鉴别和区分细菌。该方法需要取得体液样品并且然后在体外对其进行革兰氏试验。该试验基于细菌的壁和膜的特性。
其它比色指示剂不仅用来检测细菌本身,而且用来检测在细菌生长期间产生的一种或多种产物。因此,在论文“Spot Indol Test:Evaluation of FourReagents”(Journal of Clinical Microbiology,1982,589-592),J.M.Miller等中描述了用含有柯氏(kovacs)试剂的溶液浸渍的纸。柯氏试剂因检测溶液中所谓的“吲哚阳性”细菌而著名。这些细菌在它们的生长期间产生吲哚,其是色氨酸通过色氨酸酶水解的副产物。柯氏试剂中含有的化合物二甲基氨基-4-苯甲醛与吲哚反应以形成红色化合物。
最近,已经提出了基于各种类型的指示剂的敷料。用于伤口的敷料必须每隔一定时间进行更换,例如,以便维持有利于伤口愈合(healing)的一定湿度和保持伤口及伤口周围部分是清洁的或者根据伤口的愈合阶段来更换所用敷料的类型。
在愈合过程期间,跟踪观察伤口的发展情况是医务工作者日常工作的重要部分,因为,其使得可知晓,特别是,伤口的愈合状况并且提供关于何时必须更换敷料的指示。所关注的伤口例如为褥疮,糖尿病伤口,由溃疡、烧伤、外科手术或移植、皮肤肿瘤或遗传性疾病形成的伤口。伤口通常通过大致寻找视觉和气味线索(cue)进行检查。在伤口内大的细菌载量(load)的情况下,出现作为感染和身体炎症防御反应的特征的许多炎症信号:大量的、浓的、甚至是令人作呕的(nauseous)并且根据所存在的病菌而有颜色的渗出物,具有自发性疼痛的在损伤周围的(perilensional)红斑,和水肿性反应。
文献US-A-2002091347描述这样的敷料:其能够根据所述敷料的水分含量指示何时必须将其更换。这种敷料依次包括外部涂层、防水层、比色指示剂和敷料内衬(衬料,lining)。敷料内衬具有意图接触伤口的层的形式并且是防水的,直到其被水所饱和并且然后变成透水的。水然后流过敷料,并且到达与水接触时变色的显色指示剂。
还已经提出了其目的是检测作为感染的特征的温度升高的其它敷料。这些产品基于如下原理:敷料在皮肤的或者伤口的区域上的施加涉及该区域中的温度升高。在限制(confined)空间中代谢物的堆积,结合由于敷料的存在而引起的在所述区域周围的压力增加,引起了血液的涌入,所述血液的涌入造成温度的升高。文献US-A-5181905公开了例如如下敷料,其含有温度指示剂,所述温度指示剂由含有液晶的温度检测条带(band)构成。
该方法具有对于该温度升高的原因,特别是所述区域中存在的代谢物的性质并且因此对于需要更换所述敷料与否不是非常敏锐的(有识别力的,discriminating)缺点。
文献US-B-7364918提出了含有聚二乙炔组装体(assembly)和受体的比色传感器以及使用该传感器检测分析物的方法。将所述受体选择成特异性地与分析物相互作用。将该受体引入(incorporate)到在聚合之前或之后的共轭的烯-炔聚合物网络中。由此形成的共轭聚合物结构能够根据受体与分析物的相互作用而改变其构象。在共轭聚合物骨架结构上引起的干扰(disturbance)于是涉及颜色的变化。比色传感器能够指示例如,在生物液体中革兰氏阴性和革兰氏阳性细菌的存在。作者还提出将该比色传感器与敷料结合用于检测感染的存在,例如,通过将其以直接或者间接地与伤口接触的层的形式集成到敷料中,但是,根据该文献,传感器总是与渗出物接触。但是,基于聚合物简单的构象变化的传感器可涉及相当数量的假阳性结果。
进行了其它工作以检测(更主要地是检测)取自个体(个人,individual)的体液例如阴道液、血液或尿液中分析物的存在。例如,可列举文献US-A-5910447、US-A-4548906和US-A-2007258860,其提出了用于检测气体形式的氨或胺的检测器和方法。
还可列举文献US-A-5899856,其描述了用于对来自个体排出的汗液中的乙醇或乙醇代谢物进行检测的方法。该方法具体包括:由于放置于皮肤上用于吸收所排出的乙醇的皮肤贴片(dermal patch)而收集乙醇,然后解吸提取所述皮肤贴片中含有的乙醇或代谢物,之后分析所提取的液体。
然而,所提出的解决方案不适合于直接对个体进行实时分析。
发明内容
本发明的目的在于提出用于检测体液中分析物的存在、给出可靠且准确的定性信息、并且能够在实验室外实施的方法。
本发明的目的还为提出用于实施该方法的敷料,其便宜并且对于体液中分析物的存在给出可靠、准确且快速的指示。
根据本发明,该目的通过根据所附权利要求的检测方法以及用于实施该方法的敷料而达到。
特别地,该目的由于如下而达到:将检测器(detector)置于含有体液的个体或动物皮肤区域上方;然后仅以气体形式检测由分析物的代谢或降解产生的至少一种代谢物,所述检测通过将检测器放置成与气态代谢物接触而进行,所述气态代谢物来自于至少一部分所述代谢物的蒸发并且所述接触产生光学上能检测的信号;和在体液与检测器之间没有接触的情况下进行检测。

具体实施方式
根据一个具体实施方式,用于检测体液中分析物的方法包括:将检测器置于含有体液的个体或动物皮肤区域上方,然后检测由分析物的代谢或降解产生的、仅气相形式的至少一种代谢物。该检测通过使置于含有体液的皮肤区域上方的检测器与气态代谢物接触而进行。所检测的气态代谢物来自于至少一部分所述代谢物的蒸发。检测器与气态代谢物的接触产生光学上能检测的信号。在体液与检测器之间没有接触的情况下进行检测。实际上,体液有可能含有许多这样的分析物:如果检测器与体液接触,则所述分析物可与检测器相互作用。这可产生假阳性结果。因此,可发现,避免体液与检测器之间任何接触是有益的。而且,没有接触避免了检测器中含有的物质对体液的任何污染。
所述体液可为例如汗液、血液、眼泪、阴道分泌物、淋巴、唾液、尿液、或者个体或动物皮肤伤口的渗出物。特别地,所述体液存在于个体或动物皮肤伤口的渗出物中。
术语“分析物”包括病原性或非病原性生物体(organism)、酶底物(enzymatic substrate)、抗体、抗原、蛋白质、肽、细菌、病毒、原生动物、酶、氨基酸、脂质、糖、激素、寄生物或真菌或者体液成分,而没有任何限制。还可加入小分子例如脲,术语“小分子”是指其分子量低于500g/mol的分子。
所述分析物有利地为细菌。作为指示并且以非限制性方式,列举葡萄球菌型的革兰氏阳性细菌例如金黄色葡萄球菌(Staphylococcus aureus)或葡萄球菌型的革兰氏阴性细菌例如铜绿色假单胞菌(绿脓杆菌,Pseudomonasaeruginosa),以及埃希杆菌型的所谓的“吲哚阳性”细菌例如大肠杆菌。
公知一些科(family)的细菌能够在其生长期间产生代谢物。这些代谢物中的一些是挥发性的(即具有蒸发的趋势),例如在与空气接触时。因此,在文章“Detection of volatile metabolites produced by bacterial growth in bloodculture media by selected ion flow tube mass spectrometry(SIFT-M S)”,(Microbiol.Methods,2006,65,361-365),Allardyce r.a.等人中已经显示在金黄色葡萄球菌(golden staphilococca)的代谢期间产生代谢物,特别是例如如下的代谢物:乙醇、丙酮、乙醛、氨、乙酸、二甲基硫醚、甲硫醇、硫化氢、吲哚、氨基苯乙酮、三甲基胺和己醛。同样地,已经显示,铜绿色假单胞菌排放二甲基硫醚、甲硫醇和硫化氢。
另外,还已知所谓的“吲哚阳性”细菌例如大肠杆菌、嗜水气单胞菌和一些柠檬酸杆菌在它们的生长期间产生吲哚,吲哚也是挥发性产物。
在细菌的代谢期间产生的这些挥发性代谢物的排放可构成细菌真正的指纹(fingerprint)或者特征(signature)。对于挥发性代谢物,其理解为有可能变成气态的任何代谢物。
体液中存在的其它分析物还可例如通过氧化或者在细菌或酶的作用下被降解,然后释放一种或多种挥发性代谢物。作为一个实例,可列举存在于汗液中并且可分解为氨和二氧化碳的脲。
由分析物的代谢或者降解产生的挥发性代谢物有利地为挥发性有机化合物或“VOC”。
因此,通过检测由分析物的代谢或者降解产生的气态代谢物,如果该代谢物或者代谢物的组合是所述分析物所特有的,则可检测所述分析物的存在。通过使用特定的检测器,还可检测同族化合物(例如,乙醇和甲醇或腐胺和尸胺或H2S和CH3SH)的代谢物。
根据一个具体实施方式,为了有利于检测,所述检测方法的第一个步骤在于通过将包括检测器的限制元件(element of confinment)施加在含有体液的个体或动物皮肤区域上方而形成限制空间。所述检测器优选地置于所述含有体液的个体或动物皮肤区域上方。
对于所述限制空间,其理解为通过所述限制元件以及所述个体或动物皮肤区域所界定的空间。另一方面,该术语不一定意味着在所述限制元件和体液之间存在空闲(未占,empty)空间。所述限制元件可与体液接触:由此,体液不与检测器接触。
所述限制元件对于某些气体物质可为至少部分地能透过的。
在将所述限制元件施加到皮肤之上后,由分析物的代谢或降解产生的代谢物的至少一部分逐渐蒸发。未蒸发的代谢物的部分以溶解的形式留在体液中。气体形式的代谢物的浓度相对于液体形式的相同代谢物的浓度的增加取决于若干参数,特别是,取决于代谢物的性质、所述限制元件的尺寸以及体液的量。该浓度取决于表示液相中物质的浓度对气相中该相同物质的浓度之比的亨利定律。
气体形式的代谢物渗透到所述限制空间中直到与检测器接触。
检测器与气态代谢物之间的接触产生光学上能检测的信号。对于光学上能检测的,其理解为通过光学设施(means)可直接地或间接地观察或读取的任何可感知的(perceptible)信号。在接触期间,检测器与气态代谢物一起相互作用。该相互作用造成检测器的光学性质的改变。
检测器有利地对于一种或多种待分析的代谢物是特异性的,即其在与以气体形式存在的待分析的代谢物接触时产生一个或多个光学上能检测的信号。检测器有利地产生与所接触的代谢物的量成比例的信号以进行定性和定量分析。
检测器优选地在生色团(发色团,chromogene)检测器或荧光团(fluorogene)检测器之间选择。由气态代谢物与检测器的接触引起的相互作用导致发射光谱或吸收光谱的改变。在气态代谢物和检测器之间的共价型反应是优先的,因为其使得能够产生至少一个在该时间内稳定的共价键,并且其避免了在该时间期间信息的任何损失。同样地,在气态代谢物和检测器之间的共价型相互作用与在产生弱键例如范德华型弱键的情况下相比可导致光谱更显著的(important)改变,从而改善检测器的灵敏度。
在生色团检测器的情况下,气态代谢物与检测器的接触产生检测器的至少一部分的颜色的变化。生色团检测器具有肉眼直接读出体液中是否存在分析物的优点。
在荧光团检测器的情况下,气态代谢物与检测器的接触产生检测器的至少一部分的荧光发射。在这种情况下,用于激发荧光的设施特别是合适的光源例如UV灯对于检测荧光的辐射而言可为必需的。
根据一个具体实施方式,检测器包括载体以及对于代谢物敏感的探针。对于探针,其理解为对于目标物(target)具有连接亲合力的任何分子或物质。探针是具体地造成检测器例如上述的检测器的生色团或荧光团性质的原因。
探针具有一个或多个与构成目标物的代谢物相互作用的官能团(官能,function),使得一旦结合至代谢物,探针就产生光学上能检测的信号。探针优选地以不可逆且选择性的方式与至少一种代谢物进行反应,优选地通过共价键。探针与代谢物的反应有利地形成吸收性化合物或荧光化合物。荧光团检测器通常具有更大灵敏性的优点。
探针可对于具有特定官能团或反应性的挥发性代谢物或者一类代谢物是特异性的。探针与目标物相互作用的能力取决于构成目标物的代谢物。根据待检测的代谢物,本领域技术人员能够确定要使用的已知探针,并且能够根据任何已知的方法实施合成和/或插入到载体中和/或接枝到载体上的步骤,以获得适合的检测器。
作为第一实例,探针可为4-氨基戊-3-烯-2-酮,其是已知的检测在空气中以挥发性有机化合物(VOC)形式存在的轻质醛例如甲醛的化合物。作为实例,4-氨基戊-3-烯-2-酮与甲醛按照以下反应(1)反应:

所得到的化合物3,5-二乙酰基-1,4-二氢二甲基吡啶是荧光性的。
另外,被称为James试剂的化合物4-(二甲基氨基苯甲醛)(记作“DMABA”)以及被称为柯氏试剂的化合物4-二甲基氨基肉桂醛(记作“DMACA”)可分别根据以下反应(2)和(3)检测溶液中的吲哚:

目标物吲哚与DMABA或DMACA型探针之间的相互作用导致azafulivenium chlorides,其是吸收性产物。
作为第二实例,目标物可为化合物2'-氨基苯乙酮,其已知作为铜绿色假单胞菌的特异性的挥发性有机化合物。2'-氨基苯乙酮的检测可通过反应进行。实际上,在催化剂量的乙酸的存在下,化合物2'-氨基苯乙酮与环己酮根据以下反应(4)反应,得到喹啉衍生物。
2'-氨基苯乙酮环己酮9-甲基-1,2,3,4-四氢吖啶
该喹啉衍生物(具体地,9-甲基-1,2,3,4-四氢吖啶)是有色的分子。
该检测还可通过在催化剂量的乙酸的存在下2'-氨基苯乙酮在醛例如苯甲醛上的缩合进行。所获得的反应产物则为根据以下反应(5)的共轭且有色的稳定化的亚胺,即1-(2-(亚苄基氨基)苯基)乙酮。

探针可根据任何已知的方法例如通过简单地俘获(trap)或者吸附到载体的结构中而引入到载体中。探针还可通过共价键或通过实现吸附的弱键例如范德华型键、离子键、氢键、静电键或配位键结合至载体。
根据一个替代方案,探针可存在于载体本身中,特别是当挥发性代谢物有色的时候。其在载体上的吸附于是涉及载体的吸收光谱的改变。
载体有利地为透明的或半透明的以便于检测结果的读取。
载体优选地对于气体形式的代谢物为能透过的。载体有利地是多孔载体以容许代谢物在载体内更好的扩散和促进代谢物与探针的接触。载体的多孔性还使得可控制气态物质在检测器内的扩散,特别是通过根据不同气态代谢物的尺寸确保分类(拣选,sort)来控制气态物质在检测器内的扩散。载体的多孔性是可在所述检测的选择性方面具有作用的参数。
载体有利地是聚合物。对于聚合物,其理解为可溶解的和不可溶解的聚合物以及共聚物。载体可选自有机聚合物。例如,载体可为大孔树脂,例如大孔聚苯乙烯树脂。优选地,聚合物选自已知的非毒性和/或生物相容性的聚合物。
其还可为多孔氧化物,优选非金属氧化物,例如包括二氧化硅。
根据一个具体实施方式,载体是多孔聚合物以容许气体形式的代谢物循环(流动,circulation)通过聚合物骨架(skeleton)。
多孔聚合物的比表面积优选地高于或者等于50m2/g、优选高于或者等于500m2/g,以提高探针与气体形式的代谢物之间的相互作用的概率以及因此改善所述检测的灵敏性。
探针可根据任何已知的方法通过共价键结合至形成载体的聚合物,例如通过聚合物的后官能化或者已经预先官能化的单体的共聚。
根据另一具体实施方式,载体是具有强的使气态化合物增溶溶解(solubilize)的能力的聚合物。因此,除了载体的作用之外,聚合物还可用来吸收和/或储存气态代谢物。因此,与检测器接触的以气体形式的代谢物的至少一部分可在聚合物中增溶溶解并且然后以增溶溶解的形式与探针反应。
聚合物优选地选自任何已知的具有良好的溶解气体的能力的凝胶聚合物,例如强烈水合的聚合物。
聚合物例如选自水凝胶,例如琼脂糖、水凝胶、藻酸盐、透明质酸的凝胶、聚二甲硅氧烷、纤维素、聚乙二醇、聚(乙烯醇)(记为“PVA”)、聚(富马酸丙二醇酯)、聚(α-羟基酯)、聚(原酸酯)、聚酐、聚(磷腈)、聚(富马酸丙二醇酯)、聚(酯酰胺)、聚(富马酸乙二醇酯)、聚乳酸、聚乙醇酸、聚己内酯(记为“PLC”)、以及聚二氧六环酮(记为“PDO”)、聚氨酯和羧甲基纤维素。
凝胶聚合物优选地选自藻酸盐、水凝胶、含有硅油的凝胶聚合物、以及氟化的或有机硅有机凝胶,例如蒽醌-2-羧酸胆甾醇酯的凝胶和聚甲基硅氧烷的凝胶,或者1,3:2,4-二亚苄基-山梨糖醇与八甲基环四硅氧烷的凝胶、或者具有全氟化链的芳二酰胺与全氟三丁基胺的凝胶,在J.Loiseau等的出版物(Tetrahedron,2002,4049-4052)中描述了这样的凝胶。
根据图1-5中所示的优选实施方式,限制元件包括敷料1。特别地,限制元件可为敷料1。体液2优选地由来自皮肤4的伤口3的渗出物组成。将敷料1施加至含有体液2的个体或动物皮肤4的区域之上以形成限制空间5。限制空间5通过敷料1(在图1的顶部)以及被敷料1覆盖的个体或动物皮肤4(在图1的底部)界定。敷料1与皮肤4是一体的(integral)。
如图1-5中所示,容许实施上述检测方法的敷料1包括检测器6,对于个体或动物皮肤4的覆盖面7,以及在覆盖面7与检测器6之间的保护设施(means)8。检测器6如前面所述,特别地,其为生色团或荧光团检测器。
更准确地说,限制空间5通过敷料1的覆盖面7以及被覆盖面7覆盖的皮肤4的表面界定。检测器6位于敷料1之中或之上并且优选地布置于体液2上方以有利于挥发性气体形式的代谢物与检测器6的接触。
保护设施8对于代谢物是惰性的,对于气体形式的代谢物是能透过的且对于体液2是不能透过的。保护设施8的存在使得可避免体液2与检测器6之间的任何接触。保护设施8构成了防止体液2到达检测器6的物理阻隔物(barrier)。保护设施8有利地由对于气体形式的代谢物是能透过的并且对于体液2是不能透过的一种或多种材料的至少一层形成。有利地,保护设施8包括疏水聚合物(例如聚氨酯,该聚合物在性质上是疏水的)的层。
根据示于图1中的第一具体实施方式,敷料1包括第一(primary)敷料9、第二(secondary)敷料10、和位于第一敷料和第二敷料(分别为9和10)的界面处的检测器6。如图1中所示,敷料1可由依次包括第二敷料10、检测器6和第一敷料9(从图1中的顶部至底部)的一组相邻层形成。
第一敷料9通常与渗出物接触或者潜在地与渗出物接触。其可由选自如下的材料组成:机织(woven)或非机织(not woven)织物(textile)例如薄纱(绢网,tulle)、粘胶纱布(viscose gauze)、棉花、针织(knitted)聚酰胺织品(fabric)、聚酯纬纱(weft)、聚合物或凝胶聚合物例如聚氨酯、羧甲基纤维素、藻酸盐、胶原、纱布、乙酸酯、粘胶、包裹在例如纱布块的织物的非机织片中的活性炭。第一敷料9可为可能亲水的机织和/或非机织棉纱布块。机织和非机织棉纱布块构成包层(填絮,wadded)纱布块。
第一敷料9可提供一种或多种下列的功能:泵送体液2,抗菌功能,水合作用,愈合,防臭(anti-odor)。
第二敷料10主要是保证保持功能。第二敷料10可包括任何已知的粘合带(adhesive band)或常规的敷料。第二敷料10有利地是透明的或半透明的以容许直接读取由检测器6发出的信号。因此,在体液2中一种或多种分析物的存在与否的读取可在不将敷料1从皮肤4移除的情况下进行。
敷料通常由这样的第一敷料9和这样的第二敷料10组成,第一敷料9和第二敷料10的材料和功能如前所述。
在该第一实施方式中,检测器属于第一敷料9,保护设施也形成第一敷料9的一部分,防止体液2向检测器6迁移。
在下面的实施方式中,将看到,检测器6也可属于第二敷料10,保护设施则布置于第一敷料9和/或第二敷料10之中。
还将看到,当检测器6为第一敷料9的一部分时,第一敷料9可由相同的材料制成,检测器6被置于该材料的第一疏水部分之中,保护设施用该材料的第二亲水部分制得。
根据一个未示出的替代方案,第二敷料10具有与检测器6相对并且在检测器6上方的透明窗口(在图1的顶部),以使得当将敷料1施加至皮肤4时,能够用肉眼看见检测器6的颜色的变化。
敷料1有利地包括具有与体液2相对的检测面11的检测器6、与体液2相对的覆盖面7、以及用于将检测器6和体液2隔离的保护设施8。
如图1中所示,保护设施8布置于检测面11和覆盖面7之间并且优选地置于检测器6下面且与检测器6相对,以构成防止体液2向检测器6迁移的物理阻隔物。有利地,保护设施8被集成到第一敷料9中(图1)。
如图1中所示,覆盖面7可与体液2接触,特别是当所检测的体液2为来自伤口3的渗出物时。在此情况下,覆盖面7优选地覆盖皮肤4的其中具有伤口3的区域。
检测器6(其优选为生色团或荧光团检测器6)在与由如上所述的体液2中含有的分析物的代谢或者降解产生的气体形式的至少一种代谢物接触时产生光学上能检测的信号。
如图1中所示,检测器6可由根据任何已知的方法(例如如以前描述的,通过在第一敷料9上浸渍含有探针的凝胶聚合物,所述第一敷料9优选为织物)沉积在第一敷料9之上的层组成。作为一个实例,检测器6可通过如下制造:至少将所述探针与所述凝胶聚合物混合,然后将由此得到的混合物沉积到敷料1的第一织物敷料9之上。将第一敷料9用探针/聚合物混合物浸渍,所述探针/聚合物混合物在其中凝固以形成检测器6。
敷料1还可包括相同或不同的、对于一种或多种代谢物是特异性的多种检测器以用于多参数检测、结果的更好灵敏性和更好的再现性。
根据图2中所示的第二具体实施方式,除了将检测器6布置在第二敷料10的表面之上(所述第二敷料10的表面于是构成检测面11)以外,敷料1与第一具体实施方式(图1)相同。在此情况下,第二敷料10必须对于气体形式的代谢物是至少部分地能透过的。
根据图3中所示的第三具体实施方式,除了敷料1由依次包括第二敷料10、检测器6、保护设施8和第二敷料10(在图3中由顶部至底部)的一组相邻层组成以外,敷料1与所述第一实施方式(图1)相同。检测器6布置于构成保护设施8的层之上。检测面11则与保护设施8的至少一部分接触。
根据图4中所示的第四实施方式,除了覆盖面7有利地至少部分地由保护设施8组成以外,敷料1与第一具体实施方式(图1)相同。作为实例,保护设施8由亲水聚合物(例如,通过任何已知手段而被制成亲水的聚氨酯)的外层12以及疏水聚合物(例如聚氨酯,该材料在性质上是疏水的)的内层13形成。外层12意图与个体或动物皮肤4上的体液2接触并且将内层13布置成与检测器6相对。外层12的亲水性质有利于与体液2的相互作用以及其浸渍到敷料1内并且有利地使体液2更接近敷料1中的检测器6。内层13的疏水性质使保护设施8对于体液2是不能透过的并且防止体液2向检测器6迁移。保护设施8例如由用于构成内层13的聚氨酯层构成。聚氨酯层13通过形成防止体液2越过保护层进入第一敷料9的物理阻隔物而保护检测器6,同时允许气态物质的扩散。
根据图5中所示的第五具体实施方式,该实施方式与其它实施方式的区别在于保护设施8有利地通过第一敷料9的内衬厚度形成。该内衬于是具有抵抗体液2在敷料1中的行进(progression)的物理阻隔物的作用并且将检测器6与体液2隔离。对于机织纱布块,内衬经典地由机织纱布的连续层形成。第一敷料9的内衬厚度必须足够大以构成对于体液2在敷料1中渗透(impregnation)至检测器6的障碍。可局部地使所述内衬在检测器6的附近是疏水的。第一敷料9具有有利地1mm-50mm、优选1mm-10mm的厚度。
根据另一实例,第一敷料由聚氨酯构成,其中保护设施由被制成亲水的聚氨酯构成,含有检测器的其它部分由聚氨酯(该材料在性质上是疏水的)构成。
实施例
实施例1:由多孔聚合物以及俘获于聚合物结构中的探针形成的检测器6的制
造
将300mg 4-(二甲基氨基)苯甲醛(DMABA)溶解于10ml甲醇中。搅拌下,将2mL的1M盐酸加入到该溶液中。
在80℃在搅拌下,将1g琼脂糖溶于20mL水中。溶解后,在80℃,将含有DMABA的溶液加入到琼脂糖溶液中。在回到环境温度(约25℃)后,获得发白的(whitish)凝胶形式的检测器6。
实施例2:包含检测器6的敷料1的制备
将以名称URGO Discret销售的敷料1用于吲哚的检测。该敷料1包括含有织物纱布块的第一敷料9以及粘性且透明的第二敷料10。
将10μl的根据实施例1合成的检测器6的液滴在80℃预先加热,然后沉积到敷料1的第一织物敷料9上。在回到环境温度时,使被第一敷料9的织物部分所吸收的检测器6以发白的凝胶的层的形式凝固。
吲哚检测实验
如图6和7中所示,吲哚水溶液以10-1M制备。将1mL置于透明容器14例如孔板(well plate)中。实施例1中得到的敷料1经由其粘合部分粘在孔板14的盖16的内壁15上,使得通过在第一敷料9的表面上浸渍检测器6而形成的层与吲哚溶液相对。检测器6不与该吲哚溶液接触。在环境温度下10分钟之后,检测器6变为粉红色,从而显示出在气体形式的挥发的吲哚与检测器6之间的反应。
实施例3:包含两种检测器6的敷料1的制造
将300mg 4-(二甲基氨基)肉桂醛溶解于10ml甲醇中。在搅拌下,将2mL的1M盐酸加入到该溶液中。在80℃在搅拌下,将1g琼脂糖溶于20mL水中。溶解后,在80℃下,将含有DMACA的溶液加入到琼脂糖溶液中,然后在回到环境温度之前,将30μl的液滴沉积在由URGO销售的名称为URGO Discret的敷料1的第一织物敷料9上。液滴然后被第一敷料9的织物部分所吸收。在回到环境温度之后,得到橙棕色凝胶层形式的检测器6。
使用DMABA进行相同的程序,除了将检测器6沉积在敷料URGO Discret的相同第一敷料9上但是在不同位置处之外。在回到环境温度之后,得到与橙棕色凝胶层(检测器6DMACA/琼脂糖)相邻的发白的凝胶的层的形式的检测器6(检测器6DMABA/琼脂糖)。
细菌的培养
将由LGC Standards公司销售的、记作EC5的、吲哚阳性的菌株大肠杆菌(Escherichia coli)ATCC 11775的菌落在30℃在搅拌下在4mL LB培养基中预培养约15小时。将琼脂LB盒上的菌落悬浮于4mL的LB中(x2管),搅拌(agitate),然后在30℃下静态保持15小时。
对于其它两种菌株进行相同的程序:由LGC Standards公司销售的、记作HA4的、吲哚阴性的蜂房哈夫尼菌(Hafnia alvei)ATCC 13337以及由LGCStandards公司销售的、记作PP6的、吲哚阴性恶臭假单胞菌(Pseudomonasputida)ATCC 12633。
培养基色氨酸/琼脂的制备
通过如下进行液态培养基色氨酸/琼脂的制备:将3.2g/200ml的DEV色氨酸肉汤(Tryptophan Broth)(FLUKA 31406)和3g/200ml的琼脂(FLUKA05039)混合,然后将该混合物在高压釜中在121℃和2.2巴加热20分钟。
培养基在仍然(still)液态时(即,在约60℃-70℃温度)从高压釜移出,并且直接倒入皮氏培养皿(FALCON 353001,Becton Dickinson,直径35x高10mm)中。
在液体中的培养
具有最浓预培养物的管,在550nm处的OD:
对于HA4:OD(1/5x)=0.16,即0.8的有效OD;
对于EC5:OD(1/5x)=0.25,即1.25的有效OD;
对于PP6:OD(1/5x)=0.29,即1.5的有效OD。
对于0.7的OD,在1/50的基础上进行接种,即1/70种菌(接种物,inoculum)(对于4ml LB,60μL预培养物)。在250rpm的搅拌下,培养开始时的温度为37℃。
在两小时的培养后,获得:
HA4:OD (1x)=0.34即1.35.108cfu/ml
EC5:OD (1x)=0.59即1.40.108cfu/ml
PP6:OD (1x)=0.15即约5.107cfu/ml
为了接种DEV色氨酸/琼脂皿,将20μL培养物用接种环铺展(spread),即1-3百万cfu/皿。
吲哚的检测实验
使用菌株EC5、HA4和PP6制备三个皿14。将根据实施例3的敷料1粘在分别含有EC5、HA4和PP6的皮氏培养皿14的盖16的内壁15上,使得检测器6、DMABA/琼脂糖和DMACA/琼脂糖不接触培养基2并且然后将各皮氏培养皿14在37℃的温度下进行培育(incubation)。
在37℃培养3小时后,对于两种检测器6都获得了颜色的变化。仅在含有EC5(吲哚阳性)的皿14中,含有DMACA的检测器6由橙棕色变为绿色以及含有DMABA的检测器6由发白变为粉红色。注意到了所述检测方法用于检测由吲哚阳性型细菌的代谢产生的吲哚的效力。
实施例4:由含有环己酮或苯甲醛的多孔聚合物形成的检测器6的制造
在制丸机(pill machine)中,制备含有2.3ml具有式Si(OC2H5)4)的四乙氧基硅烷(TEOS)、0.6ml水、3.6ml乙醇、0.1ml乙酸和1.2ml的构成分子探针的环己酮或苯甲醛的溶液。
实施例5:含有根据实施例4的检测器6的敷料1的制造
在均化后,将实施例4中的溶液沉积在与实施例2中相同的敷料1的第一敷料9之上。在该溶液在织物块上的吸收之后,将敷料1在70℃加热3小时。在用环境空气干燥后,获得固态多孔单层形式的含有分子探针的检测器6。
2'-氨基苯乙酮的检测
制备0.5M的2'-氨基苯乙酮水溶液。将5ml置于具有盖的瓶子中。将实施例5中得到的敷料1粘在盖的内壁上以使含有检测器6的第一敷料9朝向2'-氨基苯乙酮溶液,但是不使检测器6与2'-氨基苯乙酮溶液接触。
在37℃下72分钟后,如果分子探针是环己酮,则检测器6变黄色,或者如果分子探针是苯甲醛,则检测器6变橙色。在纯粹对比的基础上,根据相同的程序进行三个对照试验。所得结果示于下表中。

注意到,对照试验保持无色,而瓶2和4确实检测出了2'-氨基苯乙酮的存在。
本发明并不限于实施例和具体实施方式,所述实施例和具体实施方式纯粹是说明性的。在所要求的保护的框架内,变化是可能的,特别地,敷料1可以包括多个层特别是亲水聚合物或织物层,特别地确保机械抵抗性(resistance)和/或改进代谢物的检测条件。另外,当体液为汗液时,限制元件可不同于敷料。
根据本发明的检测方法的目的(finality)是指示体液中是否存在一种或多种分析物。所述检测方法具有对于待分析的不同分析物,灵敏并且敏锐检测的优点。所述方法特别地通过是否观察到信号而容许以高的灵敏度和假阳性结果的低风险进行分析物的定性的并且有利地定量的分析。
所述检测方法实施简便,快速并且避免了体液的任何污染。实际上,容许实施该方法的敷料含有不与体液接触的检测器并且因此不改变体液的组成。特别地,施加于伤口的敷料不干扰治疗并且避免了体液的颜色改变或者毒性的任何风险。另外,通过所述检测方法检测到的信号可提供表明需要更换敷料的相关的中间结果。
Claims (16)
1.用于检测体液(2)中分析物的方法,其特征在于,将检测器(6)置于含有体液(2)的个体或动物皮肤(4)区域上方,然后仅以气体形式检测由分析物的代谢或降解产生的至少一种代谢物,所述检测通过使检测器(6)与气态代谢物接触而进行,所述气态代谢物来自至少一部分所述代谢物的蒸发并且所述接触产生光学上能检测的信号,并且所述检测在体液(2)与检测器(6)之间没有接触的情况下进行。
2.根据权利要求1的方法,其特征在于,所述检测器(6)在生色团检测器和荧光团检测器之间选择。
3.根据权利要求1和2任一项的方法,其特征在于,通过将包括检测器(6)的限制元件施加在含有体液(2)的个体或动物皮肤(4)区域之上而形成限制空间(5)。
4.根据权利要求3的方法,其特征在于,限制元件包括敷料(1)。

13.根据权利要求11的方法,其特征在于,聚合物选自藻酸盐、水凝胶、含有硅油的凝胶聚合物、以及氟化凝胶聚合物。
14.用于实施根据权利要求1-13任一项的方法的敷料,其特征在于,其包括:
-在与至少一种气体形式的代谢物接触时产生光学上能检测的信号的生色团或荧光团检测器(6),
-个体或动物皮肤(4)的覆盖面(7),和,
-在覆盖面(7)和检测器(6)之间的保护设施(8),所述设施(8)对于代谢物是惰性的,对于气体形式的代谢物是能透过的并且对于体液(2)是不能透过的。
15.根据权利要求14的敷料,其特征在于,保护设施(8)包括疏水聚合物。
16.根据权利要求14和15任一项的敷料,其特征在于,检测器(6)包括载体以及对于代谢物敏感的探针,并且探针引入到载体中。
