背景技术
在当今临床医学领域，心血管系统疾病一直是最易引起人类死亡的病种之一，而冠心病心绞痛是心血管系统疾病中最常见的多发病。该病是一种心绞痛是心肌急剧的暂时的缺血与缺氧所引起的临床症状，患者以中老年人居多，常发生于某种诱因存在时，其特点为阵发性的前胸压榨或疼痛感觉，主要位于胸骨后部，可放射到患者心前区、左上肢、颈咽部或下颌部，严重者甚至危及生命，极大地影响着人们的健康质量。近些年来，随着社会人口年龄结构的日渐老龄化，冠心病心绞痛的发病率逐年上升，而且有发病年轻化的趋势，已成为当今威胁人类健康的主要疾病，因此有效地对之予以防治具有十分重要的意义。
西医治疗一是应用硝酸甘油类扩张冠状血管，但使大多数病人对此类药物的扩张动脉产生毒副作用：如搏动性剧烈头痛、头昏、眩晕，甚至出现体位性低血压，同时，还可使眼内血管扩张而增加眼压，导致青光眼的发生等不宜承受或接受，影响了治疗效果，且容易复发；二是现代西医心血管内科采用冠状动脉造影检查及置入支架治疗，它又有价格昂贵等不足，更不宜被病人及家属所接受，就其所取得的近期、远期疗效而言，结果难以令人满意。
祖国医学认为，心居胸中，心包围扩其外，与小肠互为表里，在体为脉，其经脉下络小肠，舌为心之窍。心主血脉，故为人体生命活动的中心，又主神明，故为情志思维活动之中枢。由于心主血脉，心脏发病则多与血脉运行不畅有关。其属于中医“胸痹”“心痛”范畴。其病机多为本虚标实。根据五脏虚寒，气阴不足及情感异常导致肝郁气滞，气滞血瘀，血瘀不通，不通则痛的原理，认为本病以气阴两虚为主，标实则以心脉瘀阻多见。再加上情志异常，使本病加重。因此，临床治疗当以益气养阴扶其本，活血化瘀通脉治其标，再投以舒肝解郁，宽胸理气，气行则血行，血行则脉通，脉通则心痛自愈。
目前，临床上不断有新的治疗冠心病心绞痛的药物，包括西药和中成药和新的介入性治疗方法出现，但这些均未能达到真正令人满意的效果。开发一种高效低毒、标本兼治的中药，仍是临床的急切需要。
发明内容
本发明的目的是提供一种标本兼治、副作用小、安全方便、价格低廉的中药组合物。
本申请发明人根据前人经验及自己多年的医疗实践，通过对传统中药的研究，从整体看， 本病源于肾阳不足，而肾阳不足是心阳不振之根，也是气血瘀滞、心脉孪急之因。因此在胸痹的治疗当中，既要注意益气活血通络治标，亦要重视补肾培元治本。传统中医学“水火既济、心肾相关”、“治心之所以治肾，而治肾之所以治心。”的这种“心肾相关”的治疗学理论为补肾法治疗心衰、心绞痛提供了理论依据。同时根据“通则不痛”、“久病入络”的理论观点，在心衰合并心绞痛的治疗上要加以活血通络之品，这也是与符合现代医学微观理论相符合的。
在此基础上，本发明人结合辩证论证，多方收集众家之长，寻求最佳治疗方案，从祖国医学宝库中，筛选出益气养阴，活血化瘀，舒肝解郁的治疗冠心病心绞痛的天然中药，按中医理论组方，结合传统中医技术精心配制成本发明的治疗心绞痛的中药组合物，对心绞痛有治标治本的功效。
为实现上述目的，本发明采用的技术方案如下：
一种治疗心绞痛的中药组合物，其特征在于其由下述重量配比原料制备而得，
黄芪20-25份、红花20-25份、川芎20-25份、鹿角15-18份、女贞子12-15份
白芷10-15份、人参10-15份、黄连8-10份、瓜蒌5-7份、麦冬3-5份。

本发明的药物组合物其制备方法为：取上述重量份数的原料药(配比单位：克)混合均匀，水煎服，每日一剂，分早晚两次服用，一个月为一个疗程。
具体操作为：
1)称取各原料药，备用；
2)将步骤1)中的原料药用水浸泡(水漫过药面3cm)30分钟后，武火煮沸，文火煎30分钟，冷却、过滤后得到滤液和药渣；
3)将步骤2)中所得到的药渣加水煎煮，加水量以没过药渣面2cm，武火煮沸，文火煎20分钟，冷却、过滤后得到滤液和滤渣，
4)合并两次滤液即得所述治疗心绞痛的中药药物组合物。
本发明药物有理气宽胸、活血止痛之功效，用于心绞痛，症见心胸刺痛、绞痛频作、痛处固定，或心胸满闷如有物压者。
本发明药物可以加一种或多种药学上可接受的载体混合制成任何一种临床上或药学上可接受的剂型，优选口服制剂。以口服给药的方式施用于需要这种治疗的患者时，可将其制成常规的固体制剂，如片剂、胶囊、软胶囊等。
本发明的实验证明，本发明提供了一种能有效治疗冠心病慢性心绞痛疾病的中药组合物为及其制备方法，并严格控制了处方中主要药效物质的含量和比例，进行动物实验，发现本药物具有明显改善心梗后心肌重塑引起的心功能参数、血流动力学及心室组织结构的改变，可有效阻止缺血性心肌重塑引起的心功能恶化；明显缓解心绞痛和抗心肌缺血的作用，其产品的研发为医务工作者和患者提供一个更好的选择，使心绞痛的治疗药物更加完善与合理。

受试药物，实施例1制备的药物；阳性对照药：心得安，由天津力生制药股份有限公司提供；盐酸异丙肾上腺素，由上海禾丰制药有限公司提供。
三、实验方法：
小鼠40只，随机分为4组，每组10只，正常组和模型组灌胃给予生理盐水，实验组给与实施例制备的药物组合物，阳性对照组给与心得安20mg/kg。灌胃1h后，正常组皮下注射等容量生理盐水，其余各组皮下注射盐酸异丙肾上腺素20mg/kg，15min后放入装有15g钠石灰的250mL广口瓶中，立即密闭瓶口，记录缺氧状态下小鼠存活时间。并按照下述公式计算保护率：
所得到的实验结果列为：
正常组存活时间为58.254±11.52
模型组存活时间为22.25±7.65
实验组存活时间为51.06±15.27
对照组存活时间为49.01±5.75
与模型组比较：P＜0.01
结果表明，本发明的治疗心绞痛的中药药物能够显著延长异丙肾上腺素所致的心肌缺氧小鼠的存活时间。
镇痛实验
取雌性小鼠30只，随机分为空白对照组(蒸馏水，20ml/kg)，阳性对照组阿斯匹林，本发明制剂组，每组动物数10只。各组小鼠灌胃给药后30min，每鼠腹腔注射0.65％冰醋酸，每只0.2ml，观察注射冰醋酸后15min内各鼠平均扭体次数。
结果如下：

实施例2
临床资料
选择确诊为冠心病心绞痛患者85例，随机分为两组。治疗组43例，男22例，女21例，年龄50～75岁，平均60.43±5.25岁，病程半年～14年。对照组42例，男20例，女22例，年龄49～75岁，平均56.45±5.37岁，病程5个月～15年，两组病例治疗前在性别、年龄、病程等方面无显著差异(P＞0.05)，具有可比性。
诊断标准
西医诊断标准
参照中华医学会心血管病分会《不稳定性心绞痛诊断和治疗建议》。
中医诊断与辨证标准
1990年修订的冠心病中医辨证标准进行中医辨证分型。总体上分为血瘀证和非血瘀证。血瘀证候评分参照中国中西医结合学会活血化瘀专业委员会制定的血瘀证的诊断标准。结合冠心病患者的发病特点进行观察，包括心绞痛、舌质紫暗或有瘀斑、口唇及齿龈紫暗、舌下脉络曲张、脉涩或结代等症状和体征并进行评分：(1)心绞痛：轻度(有典型心绞痛发作，发作持续时间数分钟，每周至少发作2-3次，但疼痛不重，有时需含服硝酸甘油)记3分；中度(每天有较典型的心绞痛发作.每次持续数分钟，每次需含服硝酸甘油)记6分；重度(每人有数次较典型的心绞痛发作，持续数分钟，需数次含服硝酸甘油)记10分。(2)舌质紫暗或有瘀斑：轻度(舌质暗红)记3分；中度(舌质暗红，有散在疲斑)记6分；重度(舌质紫暗)记9分。(3)口唇及齿龈暗：轻度(口唇或齿龈颜色暗红)记3分；中度(口唇或齿龈颜色紫暗)记5分；重度(口唇或齿龈颜色紫暗且有瘀斑)记6分。(4)舌下脉络曲张：轻度(舌根部脉络曲张)记8分；中度(舌下脉络曲张超过舌下脉的1/2)记9分；重度(整个舌下脉曲张)记10分。(5)脉涩或结代：脉结代，记8分；脉涩，记10分。上述病例，无论标实还是本虚，皆同时存在多种兼证。
排除标准
①有下列情况之一者不能入选，血压剧烈波动控制不佳者，重度神经官能症、更年期症候群、颈椎病所致胸痛者，重度心肺功能不全，重度心律失常，肝、肾、造血系统等严重原发性疾病，精神病，主动脉夹层及出血性疾病等；②孕妇或哺乳期的妇女；③其他干扰试验的完成或影响对结果解释的主要疾病或因素。
治疗方法：
治疗组(本发明组)服用实施例1制备的中药组合物；
对照组复方丹参滴丸组，天津天士力制药股份有限公司生产，用法每天3次，每次10丸，一日三次，一个月为一疗程。
治疗结果：
疗效标准心绞痛和心电图疗效参考《冠心病心绞痛及心电图疗效评定标准》(中西医结合治疗冠心病心绞痛及心律失常座谈会，1979，上海)
具体治疗结果如下(P＜0.05)
由上表可见：治疗组缓解冠心病心绞痛显效率和总有效率均明显优于对照组，有显著性差异。
实施例3
典型实施例
李某，男，50岁，心前区疼痛已有半年，伴随胸闷、气短，体力活动时疼痛加重。治疗前口服消心痛每次一片，每日2次；欣康每次1片，每日2次；心痛定每次1片，每日2次，无明显效果；经服用本发明治疗心绞痛的中药组合物一个疗程后，疼痛明显减轻，心绞痛、胸闷、睡觉憋醒，只发作一次，经继续服两个疗程(两个月)，诸症大消失，随访无复发。
谢某，女，47岁，自述左胸刺痛，有时痛引背部，苔暗脉弦细，证属心血瘀阻，舌暗苔腻，脉沉细，近半年来疼痛加重且发作频繁，每日发作4～5次，每次均持续2～3分钟，多发生在夜间，一般口含硝酸甘油片。近1个月病情加重为持续性疼痛，尤以夜间为甚，伴随胸闷、出汗、心慌，无法平卧，服用硝酸甘油效果不明显。经服实施例1制备的中药组合物三个疗程痊愈，经追访1年未复发。
本发明采用的上述各中西药互作配伍，能发挥其协同治病作用，所用中药原料各成分之间具有药效相互交织相互促进和协调效能，经临床验证，对心绞痛有很好的治疗效果，本申请人多年的医疗实践中，治愈的病例数目众多，均取得良好的效果，而且成本低廉，减轻患者的负担。
本申请的治疗心绞痛的药物组合物成本低廉，疗效显著，具有广阔的应用前景。
Claims (2)
1.一种治疗心绞痛的中药组合物，其特征在于其由下述重量配比原料制备而得：
黄芪25份、红花20份、川芎20份、鹿角15份、女贞子12份
白芷10份、人参10份、黄连8份、瓜蒌5份、麦冬3份。
2.权利要求1所述中药组合物在制备治疗心绞痛药物中的用途。
