背景技术

发明内容

实施例1:BML-258的合成
根据以下程序和步骤合成如下文所述和使用的化合物BML-258。
BML-258合成程序

细胞和细胞培养
U937人组织细胞白血病和Jurkat急性T细胞白血病细胞获自美国典型培养物保藏中心(Manassas,VA)。除非另有指明,否则细胞培养于补充了L-谷氨酸、青霉素、链霉素和10%胎牛血清的RPMI 1640培养基(Invitrogen,Carlsbad,CA)中,并维持于对数生长期(Dai等人,CancerRes 61:5106-5115,2001)。U937细胞稳定地过表达Bcl-2、Bcl-xL、组成型活性的Akt(Myc-标签化的豆蔻酰化的Akt),获得它们的空载体对应物并在存在合适的选择抗生素的情况下培养,正如所描述的那样(Rahmani等人,2005,见上文)。
白血病母细胞获自知情同意的两位经历常规诊断抽吸术的AML患者,并经过Virginia Commonwealth University的机构审查委员会批准。根据Helsinki声明提供知情同意书。两个患者样品表征如下:
患者#1:FAB亚型M2,无已知的融合或突变蛋白,无已知的染色体异常。
患者#2:FAB亚型M4,无已知的融合或突变蛋白,16号染色体倒置。
通过在Ficoll/Hypaque(比重1.077-1.081;Sigma,St Louis,MO,USA)上在室温以400xg离心而分离样品,在每种情况下包含85%母细胞。使用无菌巴斯德移液管吸出主要含有母细胞的界面层,并重悬于含有10%FBS的培养基中。通过台盼蓝排除法测定,细胞显示出大于95%的存活,按照上文描述进行培养。从健康供体中类似地分离外周血单核白细胞。
RNA干扰
以获自Qiagen(Valencia,CA)的100pmol的靶向SphK1的RNAi寡核苷酸[靶向的序列为GGGCAAGGCCTTGCAGCTC(SEQ IDNO:1)]和非靶向对照siRNA(非特异性随机序列)转染U937细胞。根据生产商的说明书,使用Cell Line Nucleofector Kit V(Amaxa GmbH,Cologne,Germany),以Amaxa Nucleofector(程序V-001)进行转染。
鞘氨醇激酶的表达和活性
HEK 293细胞培养于含有10%胎牛血清的DMEM中,并按照之前的描述(Paugh等人,FEBS Lett 554:189-193,2003),使用LipofectaminePLUS(Invitrogen),以V5-His-pcDNA3.1载体(Invitrogen)、V5-His-标签化的人SphK1或V5-His-标签化的人SphK2进行转染。然后将细胞培养2天,通过冷冻-解冻裂解,使用0.25%Triton X-100[其抑制SphK2(Hait等人,J Biol Chem 280:29462-29469,2005)]中的[y-32P]ATP(10pCi,1mM,含有10mM MgCI2)和鞘氨醇测定SphK1活性。SphK2活性这样测定:在存在1M KCl的情况下,加入鞘氨醇与4mg/ml BSA形成复合物,在这样的条件下,SphK2的活性是最佳的并且SphK1被强烈抑制(Hait等人,J Biol Chem,2005,见上文)。使用氯仿/丙酮/甲醇/乙酸/H2O(10∶4∶3∶2∶1.v/v)作为溶剂,在硅胶G60上通过TLC提取并分离标记的S1P。使用FX Molecular Imager(Bio-Rad,Hercules,CA)定量对应于S1P的放射活性带。SphK比活性表示为形成的pmol S1P/分钟/毫克蛋白。
Western印迹分析

蛋白激酶谱
通过SelectScreenTM Kinase Profiling(Invitrogen Drug DiscoverySolutions,Madison,WI)测定SK1-I对于多种蛋白激酶活性的效应。简而言之,在不存在或存在5pM SK1-I和浓度为针对各蛋白激酶的Kmapp的ATP的情况下,使用含有构成FRET配对的两个荧光团的肽底物,使用基于荧光共振能量转移(FRET)的激酶测验系统,在384孔板中进行测验。显色试剂包含特异性消化非磷酸化肽的蛋白酶,并且产生荧光信号。分别在445nm和520nm监视香豆素荧光和荧光素FRET信号。香豆素发射仅在未经切割(磷酸化)的底物肽中激发荧光素(通过FRET)。
含有未经磷酸化的肽及激酶(在不存在ATP的情况下)和化学计量的磷酸化肽的反应分别作为0%和100%磷酸化对照。就背景校正原始荧光值。反应终点计算为:香豆素荧光除以荧光素FRET信号的发射比率。然后,根据以100%磷酸化对照获得的比率校正这些比率。
用于细胞凋亡的Annexin V/PI检验法

通过TUNEL测验法检测DNA链的断裂
在冰上以1%(w/v)多聚甲醛将细胞(106)固定15分钟,以PBS洗涤2次,并在冰上在70%乙醇中进行透化处理30分钟。洗涤细胞并重悬于含有末端脱氧核苷酸转移酶和溴代脱氧尿苷(BrdU)的DNA标记溶液中,并在37℃温育1小时:根据生产商的说明书(Sigma)进行。然后在室温在黑暗中以抗-BrdU-荧光素抗体温育细胞30分钟,并使用EXPO32流式细胞仪分析程序(Beckman Coulter)使用CoulterEpics-XL-MCL细胞荧光计进行分析。
鞘脂和代谢物的质谱分析
以冷的PBS彻底洗涤细胞,并通过在2000xg离心10分钟使之沉淀。取出细胞的等份试样进行DNA和蛋白测定。向其余细胞中加入内部标准物(各0.5nmol的C12-SM、C12-Cer、C12-GlcCer、C12-LacCer、C17-鞘氨醇、C17-二氢鞘氨醇、C17-鞘氨醇1-磷酸、C17-二氢鞘氨醇-1-磷酸和C12-Cer-磷酸、Avanti Polar Lipids,Alabaster,AL),通过液体色谱、电喷雾离子化-串联质谱(ESI-MS/MS,4000 QTRAP,AppliedBiosystems)定量提取的脂和各个神经酰胺酰基链的种类:按照之前的描述(Sullards等人,Science STKE 2001:L1,2001)进行。
异种移植肿瘤模型
所有涉及动物的实验都经过VCU IACUC批准。将U937细胞(2x106,悬浮于100μl无菌PBS中)注射进入6周龄CB17 SCID/beige小鼠(Taconic Farms,Germantown,NY)的双侧腰窝的2个位点,并允许生长7天,达到可触及的肿瘤。当肿瘤达到50-100mm3的体积时,将动物随机分配为2个组:连续7天腹膜内注射200μl盐水或SK1-I(20mg/kg)。每天以卡钳测定肿瘤,并使用公式(πx[以毫米为单位的长度]x[以毫米为单位的宽度]2)/6计算肿瘤体积。在实验末尾,杀死动物并取出肿瘤,固定于福尔马林中并包埋于石蜡或冷冻于液氮中。以苏木精-伊红或以针对Ki-67的抗体(Novocastra,Newcastle,UK)染色福尔马林固定的切片。通过免疫组织化学和过氧化物酶缀合的物种特异性二抗检测抗体结合,并以3,3-二氨基联苯胺显色。使石蜡切片脱蜡、复水化并以蛋白酶K处理,然后进行通透化。以荧光素TUNEL标记试剂盒染色冷冻的切片,然后以DAPI对比染色。通过荧光显微镜分析玻片。
统计分析
实验进行至少三次重复,具有一致性结果。对于每个实验,计算来自一式三份样品的数据并表达为平均值±S.D.。通过用于非配对观测的Student’s t检验测定实验条件之间的差异的显著性。
结果
SK1-I是SphK1而非SphK2的有力的和选择性抑制剂
目前,没有关于SphK的结构信息以允许使用计算机对接方法来推理设计抑制剂。因此,替代性方法是使用获自抑制剂研究的信息来设计更有力和选择性的抑制剂。之前已经研究了多种化学合成的短链的鞘氨醇和二氢鞘氨醇类似物作为SphK的抑制剂(Edsall等人,Biochemistry37:12892-12898,1998;De Jonghe等人,Bioorg Med Chem Lett9:3175-4180,1999;Johnson等人,J.Pharmacol Exp 309:452-461,2004;和Niiro等人,Bioorg Med Chem 12:45-51,2004)。发现:将烷基链替换为苯环或以氟取代3-羟基会产生有力的SphK抑制剂。此外,具有4,5-反式双键的类似物一般是良好的抑制剂(Johnson等人,J Pharmacol Exp,2004,见上文)。基于这些较早期的发现,我们合成了(2R,3S,4E)-N-甲基-5-(4’-戊基苯基)-2-氨基戊-4-烯-1,3-二醇(图1A)并测试了其对于重组SphK1和SphK2的效应(图1B-E)。这种水溶性的鞘氨醇类似物以剂量依赖性方式强有力地抑制SphK1的活性(图1C),在5μM时有60%-70%的抑制。如之前所报道的(Edsall等人,Biochemistry,1998,见上文;和Kohama等人,J Biol Chem,1998,见上文),N,N-二甲基鞘氨醇(DMS)也抑制SphK1的活性,虽然效力较低。重要的是,DMS也抑制SphK2(Liu等人,J Biol Chem,2000,见上文)和神经酰胺激酶(Sugiura等人,JBiol Chem 277:23294-23300,2002),与其不同的是,我们的化合物不抑制重组SphK2(图1C、E)或神经酰胺激酶(数据未显示)。因此,由于其对于SphK的特异性抑制效应,该化合物在下文中被称作SK1-I。
Lineweaver-Burk分析揭示:SphK1活性被SK1-I竞争性抑制,Ki值为大约10μM(图1D),几乎等于针对鞘氨醇的Km。SK1-I不被SphK1或SphK2磷酸化(数据未显示)。由于DMS和几种其它的泛SphK抑制剂也抑制蛋白激酶C(Igarashi等人,Biochemistry 28:6796-6800,1989)和潜在地抑制其它激酶(De Luca等人,Biofactors 25:43-60,2005;和Gamble等人,Int J Cancer 118:2412-2420,2006),所以,测定SK1-I对于蛋白激酶的效应具有重要意义。使用了蛋白激酶活性筛选,其包括数种不同的重组蛋白激酶,荧光标记的多肽底物和浓度为针对每种激酶的Kmapp的ATP。SK1-I不显著抑制任何蛋白激酶:其中包括PKC家族的两个不同成员PKCα和PKCδ,PKA,Akt1,ERKI,EGFR,CDK2,1KKβ或CamKIIβ(图1F)。因此,SK1-I在已知的SphK抑制剂中是独特的,因为其同工酶选择性,水溶性,并且对于蛋白激酶无效应。
SK1-I有力地抑制人白血病细胞的生长
之前的研究已经证明:泛SphK抑制剂DMS显著诱导U937和Jurkat T细胞的凋亡(Cuvillier等人,Nature,1996,见上文;Jarvis等人,Mol Pharmacol 52:935-947,1997;Edsall等人,见上文;Hamada等人,Biochem Biophys Res Commun 244:745-750,1998;和Cuvillier等人,Blood,2001,见上文)。如图2A所示,浓度低至5μM的SK1-I显著降低培养于存在10%血清情况下的U937细胞的生长,在培养72小时之后是明显的。T-成淋巴细胞Jurkat细胞对于SK1-I更加敏感,因为5μM的浓度抑制了50%的生长,10μM则完全阻止了细胞生长(图2B)。类似地,10μM SK1-I减少其它白血病细胞系的生长,包括原髓细胞性HL-60、Molt-4T-细胞白血病和K-562 CML细胞,在48小时处理内分别抑制50%、70%、90%。
类似于以SK1-I进行的处理的效应并与其它白血病细胞系的研究(Bonhoure等人,见上文;and Baran等人,见上文)一致,使用靶向独特序列的siRNA下调SphK1的表达(SphK1蛋白和mRNA水平下降超过60%,见图2C),显著降低了培养于存在2%或10%血清情况下的U937细胞的生长速率(图2C)。综合起来,这些发现是一致性的,注意到:通过药理学或遗传学方法特异性抑制SphK1显著抑制了人骨髓性和淋巴性白血病细胞的生长。
SK1-I诱导人白血病细胞的凋亡
使用DMS抑制SphK或SphK1表达的下调与很多细胞类型中的细胞凋亡的诱导相关,包括人白血病细胞(Cuvillier等人,Nature,1996,见上文;Jarvis等人,1998,见上文,Jendiroba等人,2002,见上文;和Bonhoure等人,见上文)。因此,我们接下来测定了SK1-I对于U937细胞的凋亡的效应:使用了流式细胞术监视在外细胞膜上表达磷脂酰丝氨酸的细胞,通过annexin V染色和PI作为细胞膜通透性的度量。在暴露于SK1-I之后,有时间和浓度依赖性的U937细胞凋亡的增加(图2D)。如图2E所示,大多数细胞是早期凋亡的,非常低的百分率的细胞是坏死的(仅PI阳性)。这些结果与DNA链断裂的发生密切相关,如通过TUNEL测验法所测(图2F)。此外,类似于SphK1的下调(当细胞培养于存在低浓度的血清的情况下时,其更有效地抑制细胞生长,见图2C),当血清浓度降低时,U937细胞对于SK1-I诱导的凋亡更加敏感(图2G)。
SK1-I诱导的细胞死亡中的胱天蛋白酶激活和Bcl-2切割的功能性作用
之前已经证明:在HL-60细胞中以siRNA下调SphK1(Bonhoure等人,见上文)或在Jurkat细胞中以DMS抑制SphK(Cuvillier等人,JBiol Chem 275:15691-15700,2000)足以激发执行者胱天蛋白酶-3的激活以及PARP的切割(细胞凋亡的标志)。类似地,伴随着细胞凋亡的诱导,以SK1-I处理U937细胞增加了胱天蛋白酶-3和胱天蛋白酶-9的激活并诱导了聚ADP核糖聚合酶(PARP)的切割(图3A、B)。此外,暴露于SK1-I16-24小时导致Bcl-2的切割(图3B),Bcl-2为阻止线粒体功能紊乱的抗凋亡蛋白。另一方面,Mcl-1(在恶性造血细胞的存活中具有重要作用的抗凋亡蛋白,见Moulding等人,Blood 96:1756-1763,2000)的水平未显著改变(图3A、B)。接下来,我们研究了胱天蛋白酶激活和Bcl-2切割在SK1-I诱导的致死中的功能性作用。以泛胱天蛋白酶抑制剂ZVAD和BOC预处理U937细胞显著减弱了SK1-I诱导的细胞凋亡以及由DNA损伤剂依托泊甙诱导的细胞凋亡(图3C)。此外,Bcl-2的离原位表达完全免除了SK1-I诱导的致死,并且Bcl-xL的表达使细胞死亡减少60%(图3D)。综合起来,这些发现表明:SK1-I引起的致死主要是通过内部线粒体途径介导的,其为Bcl-2所对抗的事件。
由于SphK1是促细胞凋亡性神经酰胺和抗细胞凋亡性S1P之间的平衡的重要调节者(Olivera等人,见上文;和Bonhoure等人,见上文),所以通过高效液相色谱ESI-MS/MS(Sullards等人,见上文)测定了SK1-I对于这些鞘脂代谢物水平的影响。以SK1-I进行的处理导致总的细胞S1P下降50%(图4A),未改变鞘氨醇或二氢鞘氨醇(sphinganine)的水平,伴随着总的细胞神经酰胺的升高和鞘磷脂的下降(图4A)。U937细胞中最丰富的神经酰胺种类为24∶1(图4A)。以Sk1-I进行的处理分别使C16∶0和C24∶1神经酰胺种类的水平升高3倍和2倍,但是对于其它神经酰胺种类无显著影响(图4A)。
为了确认SK1-I的细胞凋亡效应是由于其抑制SphK1的能力造成的,进行了反加入S1P的实验。与SK1-I引起的S1P水平的下降相一致,外源S1P的加入以剂量依赖性方式减少了SK1-I诱导的细胞凋亡(图4B)。联合起来,这些发现表明:SK1-I通过抑制SphK1和S1P的产生而诱导人白血病细胞的凋亡,并且伴随着神经酰胺的升高。
SK1-I诱导的细胞凋亡与ERK1/2和Ark生存信号的失活相关
众多证据表明:有丝分裂原激活的蛋白激酶(ERK1/2,JNK和p38MAPK)和Akt在白血病细胞命运中具有重要作用(Steelman等人,Leukemia 18:189-218,2004)。以SK1-I处理U937细胞引起ERK1/2和Akt的磷酸化的迅速且显著的减少(图5A-C)。在存在低浓度血清的情况下,生存信号的这些失活持续长达2小时(图5A),而在存在10%血清的情况下,p-ERK1/2和p-Akt水平的减弱逐渐克服(图5B),并且依赖于SK1-I的浓度(图5C)。此外,在5分钟的时候观察到p38磷酸化的短暂增加,然后是后来的较不稳健的激活(图5A、B)。另外,在较晚的时间也检测到JNK激活和c-Jun磷酸化(图5A、B)。因为SK1-I显著灭活Akt,所以测定其在SK1-I的致死效应中的作用是有趣的。在U937细胞中过表达组成型活性的豆蔻酰化的Akt显著减弱了浓度低于15μM的SK1-I诱导的细胞凋亡(图5D),这说明Akt的失活可能是促成SK1-I的细胞凋亡效应的因素之一。
原代人AML母细胞对于SK1-I诱导的细胞凋亡高度敏感
为了测定SK1-I对于原代AML样品的有效性,在获自两位具有AML(FAB,M2亚型)的患者的骨髓抽吸物的白血病母细胞中进行了平行研究。使用浓度渐增的SK1-I处理母细胞揭示出相对于U937和Jurkat细胞系而言对于细胞凋亡诱导的增强的敏感性。当暴露于Sk1-I 24小时的时候,两位患者的样品都显示出细胞凋亡的显著增加,使用7.5μMSk1-I观察到40%-50%的细胞凋亡,如annexin V/PI分析所揭示(图6A)。与之前的结果(Rosato等人,Mol Cancer Ther 6:692-702,2007)一致,在不存在处理的情况下,小于10%的母细胞显示出细胞凋亡,这非常类似于正常外周血单核细胞的细胞凋亡。值得注意的是,SK1-I对于正常外周血单核细胞的生存具有不甚显著的效应(图6B)。这结果说明,被报道为过表达SphK1的原代人AML细胞(Sobue等人,Leukemia20:2042-2046,2006),对于SK1-I的易感性比连续培养的白血病细胞系更高,而SK1-I对于正常外周血单核白细胞是相对无影响的。
SK1-I在体内的抗白血病活性
我们接下来测定了SK1-I在免疫缺陷小鼠中的异种移植物中抑制白血病细胞的肿瘤生长的能力,免疫缺陷小鼠是已经被广泛用于促进开发数种新的治疗形式的模型(McCormack等人,Leukemia 19:687-706,2005)。皮下注射进入SCID/beige小鼠的腰窝的U937细胞迅速产生指数生长的肿瘤。当肿瘤达到50-100mm3的体积时,每日向小鼠腹膜内注射盐水或Sk1-I(20mg/kg)。如图7A中可以看出,SK1-I显著减少了肿瘤生长。在7天后,以SK1-I处理的小鼠中的U937肿瘤的平均体积比以盐水处理的小鼠中的肿瘤小50%以上(对照组平均值=747mm3,Sk1-I组平均值=332mm3,p<0.001)。以SK1-I处理的小鼠在尸体解剖时的肿瘤重量也显著较低(图7B)。以SK1-I处理的小鼠未显示出消瘦的迹象,7天之后的体重与对照无显著性差异。
如预期的那样,来自以盐水处理的小鼠的肿瘤被Ki67强烈染色,这表明肿瘤组成为高度增殖性的细胞,极少有凋亡细胞被TUNEL染色(图7C)。与此相反,来自以SK1-I处理的小鼠的肿瘤的免疫组织化学分析揭示有很多凋亡细胞,如细胞核片段化(TUNEL染色)和致密的细胞核(图7C)所测定。SK1-I也显著减少肿瘤中的有丝分裂细胞(图7C)。这些结果表明:SK1-I在体内具有有力的抗白血病活性。
讨论
有充足的证据证明:在很多类型的癌症中,S1P的产生失调,导致异常细胞生长和生存信号传导(Milstien等人,见上文;和RA Sabbadini,Br J Cancer 95:1131-1135,2006)。Sphk1在多种实体肿瘤中过表达(French等人,Cancer Res 63:5962-5969,2003),在AML中也过表达(Sobue等人,见上文)。此外,已经证明bcr/abl(慢性髓性白血病中的常见的遗传异常和至少20%的患有急性淋巴细胞性白血病的患者中的差预后指示)上调SphK1的表达(Li等人,Oncogene 26:7904-7908,2007)。因此,SphK1现在被认为是用于药理学干预(尤其是在白血病细胞中)的潜在靶标,在白血病细胞中,其水平与化疗抗性和放疗抗性相关联(Bonhoure等人,见上文;和Baran等人,见上文)。之前关于SphKi在白血病细胞中的作用的研究着眼于通过特异性siRNA或使用抑制SphK1和SphK2二者和潜在地抑制蛋白激酶的药理学试剂使之下调。本研究描述了第一个有力的且水溶性的SphK1同工酶特异性抑制剂SK1-I的开发。此外,SK1-I不抑制PKC或很多其它蛋白激酶。与多数小分子蛋白激酶抑制剂不同,SK1-I与脂底物是竞争性的,而多数小分子蛋白激酶抑制剂是在非常保守的ATP结合袋处与ATP竞争性的,并且潜在有交叉反应。
Sk1-I在数种白血病细胞系和AML白血病母细胞中有力地诱导细胞凋亡,如通过磷脂酰丝氨酸的外化、增加的DNA链断裂、胱天蛋白酶3和9的激活、PARP和Bcl-2的切割所反映。SK1-I是通过什么机制如此深刻地诱导这些致死效应的呢?这可能是由于数种非互斥的相关作用造成的。SK1-I抑制促生存性S1P的产生,S1P能够在细胞内作用以增强生长和生存,虽然其细胞内靶标仍然未知(Kohno等人,MolCell Biol 26:7211-7223,2006)。普遍接受的是:细胞内产生的S1P可以从细胞中释放(Mitra等人,Proc Natl Acad Sci USA 103:16394-16399,2006),并且通过与生存途径、包括ERK1/2和Akt相关的其细胞表面受体而作用(Spiegel等人,见上文)。在此意义上,SK1-I的减少白血病细胞中的激活的ERK1/2和Akt的能力可能是相关的,因为Raf/MEK/ERK和PI3K/Akt途径在AML中通常是组成型激活的(Steelman等人,见上文;和Nyakern等人,Leukemia 20:230-238,2006)。由于ERK1/2磷酸化SphK1并将其激活(Pitson等人,J Exp Med201:49-54,2005)、导致S1P增加,S1P继而又能够刺激ERK1/2,所以,Sk1-I能够通过抑制SphK1而打断该阳性回馈回路,减少促生长和生存的S1P,同时增加其前体,促细胞凋亡的神经酰胺。因此,SK1-I整合了白血病中的多个分子治疗靶标。
长期以来,神经酰胺的产生被指参与人白血病细胞中细胞凋亡的诱导(Jarvis等人,Proc Natl Acad Sci USA 91:73-77,1994),最近证明,细胞凋亡的数种不同信号转导抑制剂的协同作用与神经酰胺产量的显著增加相关联。例如,向人白血病细胞共同施用组蛋白脱乙酰基酶抑制剂和哌立福新(perifosine)导致了Akt和ERK的破坏,神经酰胺和反应性氧产量的显著升高,以及线粒体损伤和细胞凋亡的陡然增加(Rahmani等人,见上文;和Rosato等人,Mol Pharmacol 69:216-225,2006)。神经酰胺可以通过多个途径转导其细胞凋亡作用(Ogretmen等人,见上文)。所鉴定的重要的神经酰胺靶标包括丝氨酸/苏氨酸蛋白磷酸酶PP1和PP2A,它们使Akt以及SR蛋白去磷酸化,SR蛋白是Bcl-2的备选剪接的调节者(Ogretmen等人,见上文)。我们发现:暴露于SK1-I导致Bcl-2的切割,已经证明该应答与线粒体依赖性细胞凋亡相关(Cheng等人,Science 278:1966-1968,1997)。此外,已经确证,Bcl-2的过表达在很多细胞类型、包括急性淋巴母细胞性白血病和AML中阻止神经酰胺的形成并抵抗神经酰胺诱导的细胞凋亡(Zhang等人,Proc Natl Acad SciUSA 93:5325-5328,1996;Amarante-Mendes等人,Blood 91:1700-1705,1998;和Ogretmen等人,见上文)。与此相符,我们发现:Bcl-2的过表达也阻止SK1-I诱导的致死,这强调了内在的线粒体死亡途径的重要性。与该发现一致,最近已经证明S1P通过以MEK/ERK1/2依赖性方式阻断Bax向线粒体的转位而在Jurkat细胞的凋亡过程中发挥其对线粒体事件的细胞保护性效应(Betito等人,biochem Biophys Res Commun340:1273-1277,2006)。最近的研究表明,神经酰胺在内质网上的持续升高协调性激活ER应激应答并灭活抗-凋亡性Akt,从而导致细胞凋亡(Swanton等人,Cancer Cell 11:498-512,2007)。因此,有可能的是,SK1-I诱导的Akt失活不仅通过减少S1P的形成而介导,而且是通过增加神经酰胺而介导。也已经证明,免疫抑制性药物FTY72O(结构上类似于SK1-I)减少Akt的磷酸化,但是,其为SphK的竞争性底物而非竞争性抑制剂(Ng等人,Int J Oncol 30:375-380,2007)。FTY72O在临床试验中对于多发性硬化具有相对小的毒性(Brinkman等人,Pharmacol Ther115:84-105,2007),最近已经建议其作为治疗慢性髓细胞性白血病原始细胞危象和费城染色体阳性急性淋巴细胞性白血病的替代品(Neviani等人,J Clin Invest 117:2408-2421,2007)。
我们发现,SK1-I有力地诱导分离自患有AML的患者的白血病母细胞中的细胞凋亡、但是对于正常的外周血单核白细胞是相对无细胞毒性的,该发现突出显示了其对于白血病细胞的选择性。此外,在异种移植物AML模型中,SK1-I具有明显的单试剂活性,其抑制肿瘤生长、诱导肿瘤中的细胞凋亡,并减少增殖,这与其体外的作用类似。对于肝脏、肾脏和脾的毒性的初步分析未显示出任何值得注意的效应。因此,特异性SphK1抑制剂值得考虑用作白血病中的潜在药理学干预,可单独使用或作为常规或其它已知的靶向试剂的辅佐。
根据以上详述的本发明的说明书和实施例,毫无疑问对于本领域普通技术人员可以暗示很多明显的变体。本领域技术人员将认识到,经计算用于实现相同目的的任意排列可以替换所显示的具体实施方式。本申请和发明旨在涵盖本发明的改编或变体。所有这些变体都完全包括在本发明的范围和精神内,如随附的权利要求中更具体地定义。
