Abstract
本发明公开的光合细菌无氮培养基,其每升培养基中含有:乙酸钠10~20g,硫代硫酸钠8~12g、碳酸氢钠1~2g、磷酸二氢钾0.6~1.2g、硫酸亚铁0.03~0.05g、硫酸镁0.2~0.4g、氯化钠0.3~0.5g,余量位蒸馏水。本发明的光合细菌无氮培养基无需氮元素,且含有高含量的乙酸钠,能抑制杂菌的生长。

技术领域
本发明涉及细菌培养基,特别涉及一种光合细菌无氮培养基。
背景技术
光合细菌英文名:Photosynthetic Bacteria Abbr.name:PSB光合细菌(简称PSB)是地球上出现最早、自然界中普遍存在、具有原始光能合成体系的原核生物,是在厌氧条件下进行不放氧光合作用的细菌的总称,是一类没有形成芽孢能力的革兰氏阴性菌,是一类以光作为能源、能在厌氧光照或好氧黑暗条件下利用自然界中的有机物、硫化物、氨等作为供氢体兼碳源进行光合作用的微生物。光合细菌广泛应用于水产养殖、畜禽养殖、种植、污水处理等行业领域中。
目前光合细菌培养基都含有含有N元素,N是细菌的必要营养元素,如NY/527-2002《光合细菌菌剂》附录A配方。
发明内容
本发明所要解决的技术问题在于针对现有光合细菌培养基所存在需特殊的微生物反应器,不仅价格昂贵,产量也难以提高,因此难以推广应用的问题而提供一种能有效防止杂菌污染,又能促进光合细菌快速繁殖的光合细菌无氮培养剂。该光合细菌无氮培养剂具有良好的防止杂菌生长的功能,能在开放的自然环境中培养,不易被杂菌污染,同时能使光合细菌快速生长,在30~45℃经5天的培养光合细菌的浓度可达到5亿/ml以上,(超过农业部的标准)。
本发明所要解决的技术问题可以通过以下技术方案来实现:
光合细菌无氮培养基,其特征在于,每升培养基中含有:乙酸钠10~20g,硫代硫酸钠8~12g、碳酸氢钠1~2g、磷酸二氢钾0.6~1.2g、硫酸亚铁0.03~0.05g、硫酸镁0.2~0.4g、氯化钠0.3~0.5g,余量位蒸馏水。
碳和氮是所有细菌的必须营养元素,缺一不可,且低含量乙酸钠可提供细菌碳源,本发明的光合细菌无氮培养基无需氮元素,且含有高含量的乙酸钠,能抑制杂菌的生长。
具体实施方式
本发明技术方案不局限于下列所举实施方式,还包括各种具体实施方式间的任意组合。
实施例1
1)、光合细菌无氮培养基的配制
每升光合细菌无氮培养基中含有:乙酸钠10g,硫代硫酸钠8g、碳酸氢钠1g、磷酸二氢钾0.6g、硫酸亚铁0.03g、硫酸镁0.2g、氯化钠0.3g,用蒸馏水配制成1000ml。
2)、光合细菌培养
将上述光合细菌无氮培养基的pH值调至7.0~7.5,120℃灭菌30分钟。将光合细菌(市购)按照接种浓度为30%接种于光合细菌无氮培养基中,混合均匀,置于光照下玻璃培养箱中培养,光照条件为1300~1600lux,温度30~45℃。
本实施例1的光合细菌无氮培养基采用不含N元素培养基,利用光合细菌本身的固氮作用,培养光合细菌。
实施例2
1)光合细菌无氮培养基的配制
每升光合细菌无氮培养基中含有:乙酸钠20g,硫代硫酸钠1.2g、碳酸氢钠2g、磷酸二氢钾0.9g、硫酸亚铁0.04g、硫酸镁0.3g、氯化钠0.4g,用蒸馏水配制成1000ml。本实施例光合细菌培养方法与实施例1相同。
实施例3
光合细菌无氮培养基的配制
每升光合细菌无氮培养基中含有:乙酸钠20g,硫代硫酸钠12g、碳酸氢钠2g、磷酸二氢钾1.2g、硫酸亚铁0.05g、硫酸镁0.4g、氯化钠0.5g,用蒸馏水配制成1000ml。本实施例光合细菌培养方法与实施例1相同。

1.光合细菌无氮培养基,其特征在于,每升培养基中含有:乙酸钠10~20g,硫代硫酸钠8~12g、碳酸氢钠1~2g、磷酸二氢钾0.6~1.2g、硫酸亚铁0.03~0.05g、硫酸镁0.2~0.4g、氯化钠0.3~0.5g,余量位蒸馏水。
