漆酶最早是由日本学者Yoshida于1883年在漆树(Rhus vennicifen)的汁液中发现的,后来由Bertraucl第一次命名为“漆酶(Lacease)”。此后随着研究的深入,发现漆酶广泛存在于多种植物,如马铃薯、毒莴苣、香蕉、桃子、咖啡豆、苜蓿花等,和部分细菌,如节杆菌(Arthrobacter)和链霉菌(Streptomycete)等,以及各种真菌,如担子菌(Basidiomycetes)、子囊菌(Ascomycete)和半知菌(Adelomycete)。此外,一些动物肾脏和血清中也有漆酶的发现。按其来源主要分为漆树漆酶(Rhus Laccase)和真菌漆酶(Fungal Laccase)两大类,其中真菌中的白腐菌(White-rot fungi)是漆酶的主要生产者。
尽管漆酶在自然界中广泛分布,但真正具有较高漆酶产量的菌株并不多,其漆酶产量还不能满足工业生产所需的酶量,且发酵周期较长(7~14天)。当前用于漆酶生产的菌株主要有:黄孢原毛平革菌(Phanerochaete chrysosporium)、变色栓菌(Trametes versicolor)、彩绒革盖菌(Coriolus hisutus)、杂色云芝(Coriolus versicolor)、血红密孔菌(Pycnoporus sanguineus)以及糙皮侧耳(Pleurotus ostreatus)等,但目前未见竹黄菌(Shiraia bambusicola)产漆酶的研究报道。
竹黄是一种药用真菌,具有很高药用价值和保健功效。竹黄发酵生产漆酶具有发酵周期短和酶活力高的优势,同时发酵过程还可产生高活性的超氧化物歧化酶(SOD)和苝醌色素,开发前景广阔。
发明内容
本发明的目的是,利用竹黄菌株(Shiraiasp.) SUPER-H168,保藏号为CCTCC NO:M 207104,已在ZL200710132510.4公开,通过液态发酵法生产漆酶,通过调整各种培养基成份提高漆酶产量。
本发明的技术方案:一种竹黄菌发酵生产漆酶的方法,
出发菌株为:竹黄菌株(Shiraiasp.) SUPER-H168,保藏号为CCTCC NO:M 207104;
液态发酵培养基组成为:碳源5-50 g/L,氮源5-50 g/L,K2HPO4 2-4 g/L,KH2PO4 2-4 g/L,马铃薯汁200 g/L,用纯水配制;
液态发酵条件为:在发酵罐内装入发酵罐体积60%-70%(v/v)的液态发酵培养基,灭菌冷却后接入竹黄菌,接种量为5%-10%,种龄为10-48小时,起始pH和发酵过程pH均为自然,发酵温度为24-32℃,发酵周期为2-5天;在发酵过程中可以添加表面活性剂,以提高漆酶的产量。
发酵过程中添加的表面活性剂主要有:Tween 20、Tween 40、Tween 60、Tween 80、Triton X-100、TritonX-114、TritonX-116、Triton X-405、TX-4、TX-6、TX-9、TX-10、AEO-3、AEO-7、AEO-9、一乙醇胺、二乙醇胺、三乙醇胺。最终发酵液中漆酶活力最高可达500 U/mL。
所述的竹黄菌菌株,能够同时发酵产漆酶、超氧化物歧化酶SOD和苝醌色素。
所述的竹黄菌发酵生产漆酶的方法,可利用的产漆酶碳源为葡萄糖、果糖、可溶淀粉、麦芽糖、木糖、蔗糖、乳糖、或半乳糖。
所述的竹黄菌发酵生产漆酶的方法,可利用的产漆酶的氮源为无机铵盐、无机硝酸盐、蛋白胨、酵母膏、尿素、玉米浆、或豆饼粉。
所述的竹黄菌发酵生产漆酶的方法,添加Cu2+、Fe2+、Fe3+、Al3+、Mg2+、Mn2+、Na+、K+可以提高竹黄菌的漆酶产量。
最终发酵液中漆酶活力最高可达500 U/mL。在漆酶发酵过程中,菌体还会生成苝醌色素,产量最高可达7 g/L左右,其中竹红菌甲素占90%左右,竹红菌乙素占5%左右。同时发酵过程还能产生SOD酶,酶活达100-200 U/mL。
漆酶酶活测定是以2,6-二甲氧基酚(DMP)为底物,以每分钟氧化1 μmol的DMP(ε=49.6 L/(mmol·cm) 所需要的酶量定义为1个酶活力单位,测定方法和活力定义详见:D Litthauer, M J Van Vuuren, A Van Tonder, F。W. Wolfaardt (2007) Purification and kinetics of a thermostable laccase from Pycnoporus sanguineus. Enzyme Microb Technol, 2007, 40(4): 563~568.
SOD酶活的测定方法是基于SOD抑制氮蓝四唑(NBT)在光下的还原作用。SOD活力测定方法和SOD活力定义详见:Chin-Wen Lin, Jeng-Huh Yang and Lieh-Chi Su,(1997) The extraction and properties of superoxide dismutase from porcine blood,Meat Science,46(3):303-312.
苝醌色素的产量采用液相色谱法测定,详见:Xiao-Hui Lianga, Yu-Jie Cai, Xiang-Ru Liao, Kang Wu, Lei Wang, Da-Bing Zhang and Qiang Meng (2009). Isolation and identification of a new hypocrellin A-producing strain Shiraia sp SUPER-H168. Microbiological Research 164(1): 9-17.
本发明有如下特点:
(1)用于Laccase发酵的碳源可以是葡萄糖、果糖、可溶淀粉、麦芽糖、木糖、蔗糖、乳糖、半乳糖。氮源可以是无机铵盐、无机硝酸盐、蛋白胨、酵母膏、牛肉膏、尿素、玉米浆、豆饼粉。马铃薯汁既可以补充碳源也可以补充氮源,200 g/L是指称取200g马铃薯,洗净去皮切成小块,加水1000mL煮沸半个小时,纱布过滤得到滤液,所有滤液加入到培养基中,培养基终体积为1000 mL。
(2)竹黄菌发酵漆酶可采用机械搅拌或气升式发酵罐。对于机械搅拌发酵罐,搅拌转速为70-150 r/min,通风量1:0.1-1:0.5 v/v/m。对于气升式发酵罐,通风量为1:0.3 - 1:1.5 v/v/m。 通风量数值与单位说明:1:0.5 v/v/m指1个体积的发酵液每分钟通0.5体积的无菌空气。发酵罐体积为5-500L,结合本领域的基本常识可根据生产情况调整发酵罐大小。
(3)竹黄菌生产漆酶的过程,伴随着如下所示的三种母核为基本结构的苝醌色素的生成,其中主要是竹红菌甲素。同时该菌可产生以此三种母核结构进行基团修饰的多种苝醌类化合物。可在2, 3, 6, 7, 11, 12, 17, 18, 26, 28, 29, 30, 32, 36, 37, 45, 46, 53, 55, 56, 58, 62, 63, 71, 72位置修饰上各种化学基团,如CH3、OCH3、COCH3、CH(OH)CH3、CHO、OH、H等,这些化学基团之间还可以进一步形成更为复杂的结构。随着发酵条件的不同,各苝醌色素的产量和比例会略有不同。
伴随SOD产生的苝醌色素母核
(4)竹黄菌生产漆酶的过程,伴随着SOD酶的生成。
(5)为了提高漆酶的产量,本发明通过添加表面活性剂,以改善竹黄菌细胞膜的通透性。添加时间为发酵开始后的0-40小时,使发酵液中表面活性剂的浓度达0.1%-2% (w/w),随后的发酵不再补加表面活性剂。结果表明所有表面活性剂对于漆酶的产量均有促进作用,其中Tween 80添加量为0.8%时的效果最好。
(6)为了提高漆酶的产量,本发明可通过添加金属离子,以刺激菌体生产漆酶。在发酵开始前添加离子,离子主要有Cu2+、Fe2+、Fe3+、Al3+、Mg2+、Mn2+、Na+、K+,浓度控制在0-4 mmol/L。结果表明这些离子对于漆酶的产量均有一定的促进作用。
本发明的有益效果:目前未见竹黄菌(Shiraia bambusicola)产漆酶的研究报道。竹黄是一种药用真菌,具有很高药用价值和保健功效。竹黄发酵生产漆酶具有发酵周期短和酶活力高的优势,同时发酵过程还可产生高活性的超氧化物歧化酶(SOD)和苝醌色素,开发前景广阔。

具体实施方式
实施例1
培养基组成为:碳源20g/L,马铃薯200g/L,蛋白胨20g/L,NH4NO3 10g/L,玉米浆粉12g/L,K2HPO4 2 g/L, KH2PO4 2 g/L, pH自然,接种量为10%,种龄24小时,发酵温度为 30 ℃,300L 气升式发酵罐,装液量70%,通风量为1:1.5 v/v/m,发酵时间为3天。在此条件进行8批发酵,每批发酵所采用的碳源不同,依次为葡萄糖、果糖、可溶淀粉、麦芽糖、木糖、蔗糖、乳糖、半乳糖,每批碳源浓度为20g/L。漆酶、SOD和苝醌色素的产量如图1所示。
实施例2
培养基组成为:葡萄糖 20g/L,氮源30g/L,K2HPO4 4 g/L, KH2PO4 4 g/L, pH自然,接种量为5%,种龄48小时,发酵温度为 24 ℃,300L 气升式发酵罐,装液量60%,通风量为1:1.5 v/v/m,发酵时间为3天。在此条件进行9批发酵,每批发酵所采用的氮源不同,依次为氯化铵、硝酸铵、硫酸铵、尿素、蛋白胨、酵母膏、牛肉膏、玉米浆、豆饼粉,每批氮源浓度为30g/L。漆酶、SOD和苝醌色素的产量如图2所示。
实施例3
培养基组成为:葡萄糖20g/L,NH4NO310g/L,KH2PO4 4 g/L, 蛋白胨10g/L,表面活性剂0.08g/L,pH自然。接种量为10%,种龄15小时,培养温度为 28 ℃,50L 机械搅拌发酵罐,装液量60%,通风量为1:0.5 v/v/m,搅拌转速为150 r/min,发酵时间为3天。当发酵开始后,发酵液中立即加入表面活性剂。在此条件进行18批发酵,每批发酵所采用的表面活性剂不同,依次为Tween 20、Tween 40、Tween 60、Tween 80、Triton X-100、TritonX-114、TritonX-116、Triton X-405、TX-4、TX-6、TX-9、TX-10、AEO-3、AEO-7、AEO-9、一乙醇胺、二乙醇胺、三乙醇胺,每批表面活性剂浓度为0.08g/L。漆酶、SOD和苝醌色素的产量如图3所示。
实施例4
培养基组成为:葡萄糖20 g/L,K2HPO4 4 g/L, 牛肉膏10g/L,蛋白胨10g/L,玉米浆粉12g/L,某种金属离子2 mmol/L,pH自然,接种量为10%,种龄24小时,发酵温度为 26℃, 5 L机械搅拌发酵罐,装液量70%,通风量为1:0.3 v/v/m,搅拌转速为100 r/min,发酵时间为2天。在此条件进行8批发酵,每批发酵所添加的金属离子不同,依次为Cu2+、Fe2+、Fe3+、Al3+、Mg2+、Mn2+、Na+、K+。漆酶、SOD和苝醌色素的产量如图4所示。
实施例5
培养基组成为:马铃薯200g/L,乳糖20g/L,NH4NO310g/L,Mg2+ 4mmol/L (MgSO4)、蛋白胨10g/L,pH自然。接种量为10%,种龄10小时,培养温度为 30 ℃,500L 机械搅拌发酵罐,装液量60%,通风量为1:0.5 v/v/m,搅拌转速为150 r/min,发酵时间为3天。当发酵开始后,发酵液中立即加入Triton X-100 1.5%。在该条件下,发酵液中漆酶活力为428 U/mL,SOD活力为316 U/mL。苝醌色素产量为0.53 g/L,其中竹红菌甲素占93%,竹红菌乙素占3%。
实施例6
培养基组成为,葡萄糖20g/L,酵母膏10g/L,蛋白胨10g/L,尿素10g/L, pH自然。接种量为10%,种龄48小时,培养温度为 30 ℃,500 L 机械搅拌发酵罐,装液量70%,通风量为1:0.4 v/v/m,搅拌转速为120 r/min,发酵时间为5天。当发酵开始后40小时,发酵液中立即加入Tween 80 0.3%。在该条件下,发酵液中漆酶活力达460 U/mL, SOD活力为380 U/mL。苝醌色素产量为0.92 g/L,其中竹红菌甲素占91%,竹红菌乙素占5%。
Claims (3)
1.一种竹黄菌发酵生产漆酶的方法,其特征在于
出发菌株为:竹黄菌株(Shiraia sp.)SUPER-H168,保藏号为CCTCC NO:M 207104;
液态发酵培养基组成为:碳源5-50 g/L,氮源5-50 g/L,K2HPO4 2-4 g/L,KH2PO4 2-4 g/L,马铃薯汁200 g/L,用纯水配制;
马铃薯汁的配置:称取200g马铃薯,洗净去皮切成小块,加水1000mL煮沸半个小时,纱布过滤得到滤液,所有滤液加入到培养基中,培养基终体积为1000 mL;
液态发酵条件为:在发酵罐内装入发酵罐体积60%-70%(v/v)的液态发酵培养基,灭菌冷却后接入竹黄菌,接种量为5%-10%,种龄为10-48小时,起始pH和发酵过程pH均为自然,发酵温度为24-32℃,发酵周期为2-5天;在发酵过程中添加表面活性剂,以提高漆酶的产量;
所添加的表面活性剂选用:Tween 20、Tween 40、Tween 60、Tween 80、TritonX-100、TritonX-114、TritonX-116、TritonX-405、TX-4、TX-6、TX-9、TX-10、AEO-3、AEO-7、AEO-9、一乙醇胺、二乙醇胺、或三乙醇胺;添加时间为发酵开始后的0-40小时,使发酵液中表面活性剂质量浓度达0.1%-2%,随后的发酵不再补加表面活性剂;
所述碳源选用葡萄糖、果糖、可溶淀粉、麦芽糖、木糖、蔗糖、乳糖、或半乳糖;
所述氮源选用无机铵盐、无机硝酸盐、蛋白胨、酵母膏、尿素、玉米浆、或豆饼粉。
2.根据权利要求1所述的竹黄菌发酵生产漆酶的方法,其特征在于所述的竹黄菌株,能够同时发酵产漆酶、超氧化物歧化酶SOD和苝醌色素。
3.根据权利要求1所述的竹黄菌发酵生产漆酶的方法,其特征在于添加Cu2+、Fe2+、Fe3+、Al3+、Mg2+、Mn2+、Na+、或K+可以提高所述的竹黄菌株的漆酶产量。
