抗原在刺激免疫反应中的效力可以通过施用佐剂增强。佐剂可通过不同机制发挥功能,包括(1)在机体内捕获抗原引起缓慢释放,(2)将免疫系统的细胞吸引至注射位点,(3)刺激免疫系统的细胞增殖并被激活,以及(4)改进接受者体内抗原扩散。通常使用的佐剂包括铝盐、油包水乳液和水包油乳液、矿物盐和其他可作为刺激性危险信号的化合物。在一些情况下,聚阳离子例如二乙氨乙基葡聚糖(DEAE葡聚糖)也可作为佐剂发生效力。佐剂可以作为添加剂包含在疫苗中或可以分别施用。
许多疫苗组合物是油包水或水包油乳液。具体地,油包水组合物是有效的,原因在于这种组合物在注射位点存在时间延长,这引起抗原在免疫位点的缓慢释放。但是,一旦体内注射,油包水乳液可变得不稳定,引起组合物水相和油相分离。这引起抗原和其他组分的过早成熟或加速释放。
发明概述
一方面,本发明提供组合物,其包含:抗原、两性化合物和疏水载体,其中所述组合物基本上无水。

发明详述
本发明提供包含抗原、两性化合物和疏水载体,主要由抗原、两性化合物和疏水载体组成,或由抗原、两性化合物和疏水载体组成的组合物;其中所述组合物基本上无水。
抗原
本发明的组合物包含一种或更多种抗原。如本文所用,术语“抗原”是指可特异性地结合抗体或T细胞受体的物质。
可用于本发明的组合物的抗原包括但不限于:多肽、微生物或其部分,例如活的、减毒的、灭活的或杀死的细菌、病毒或原生动物或其部分。
如本文和权利要求书中所用,术语“抗原”还包括多核苷酸,其编码发挥抗原功能的多肽。基于核酸的接种策略是已知的,其中将含有多核苷酸的疫苗组合物施用至对象。多核苷酸编码的抗原多肽在对象中表达,使得抗原多肽最终存在于对象中,如同疫苗组合物自身已含有多肽。为了本发明的目的,术语“抗原”,如上下文所示,包括编码发挥抗原功能的多肽的这类多核苷酸。

抗原可以可选地为原生动物来源,例如恶性疟原虫(Plasmodiumfalciparum),其引起疟疾。

如本文所用,术语“多核苷酸”包括任何长度(例如9、12、18、24、30、60、150、300、600、1500或更多个核苷酸)或链数(例如单链或双链)的核苷酸链。多核苷酸可以是DNA(例如基因组DNA或cDNA)或RNA(例如mRNA)或其组合。它们可以是自然发生的或合成的(例如化学合成的)。可预期的是,多核苷酸可含有在核苷酸链中一个或更多个含氮碱基、戊糖或磷酸基的修饰。这些修饰是本领域已知的并可以用于例如提高多核苷酸稳定性的目的。
抗原浓度可以高至有效刺激免疫反应所需的浓度,其中限制抗原的数量,因为抗原不应从组合物中沉淀出来,并且抗原必须可重悬于疏水载体中。进一步,抗原浓度根据抗原类型和组合物中其他组分的量而变化。本领域普通技术人员可容易测定具体应用中所需的抗原数量。例如,对于肽抗原,可以使用约0.01至约5mg/ml(基于组合物总体积),优选的范围是不小于0.1且不大于1.0mg/ml。对于其他抗原,例如重组蛋白,浓度可以是在约0.01至约0.5mg/ml的范围内,优选的范围是不小于0.01且不大于0.5mg/ml。
两性化合物
本发明的组合物包含一种或更多种两性化合物。“两性化合物”是具有亲水和疏水部分或特征的化合物。两性化合物的疏水部分通常是大的烃类部分,例如形式为CH3(CH2)n的长链,其中n>4。两性化合物的亲水部分通常为带电基团或极性不带电基团。带电基团包括阴离子和阳离子基团。阴离子带电基团的实例包括下列(其中分子的疏水部分由“R”表示):羧酸根:RCO2 -;硫酸根:RSO4 -;磺酸根:RSO3 -;以及磷酸根(磷脂中的带电官能性)。阳离子带电基团包括例如胺:RNH3 +(“R”再次表示分子的疏水部分)。不带电的极性基团包括例如具有大的R基团的醇,例如二酰基甘油(DAG)。两性化合物可具有几个疏水部分、几个亲水部分或几个疏水部分和亲水部分。蛋白质和一些嵌段共聚合物是实例。类固醇、胆固醇、脂肪酸、胆酸和皂角甙也是可用于本发明实践的两性化合物。
本发明的组合物可含有单一两性化合物或两性化合物混合物。在一些实施方式中,两性化合物(一种或多种)是磷脂或磷脂混合物。
广泛定义地,“磷脂”是水解产生磷酸、醇、脂肪酸和含氮碱基的一组脂质化合物的成员。可用于本发明实践的磷脂包括磷酸甘油酯,其是磷脂,其中两个脂肪酰基侧链被酯化至甘油分子三个羟基中的两个。甘油分子的第三个羟基用磷酸酯化。磷酸基团通常还被酯化至亲水化合物上的羟基,例如乙醇胺、丝氨酸、胆碱或甘油。磷脂,其是磷酸甘油酯,包括例如磷脂酰乙醇胺、磷脂酰丝氨酸、磷脂酰胆碱、二油酰脂酰胆碱(“DOPC”)、磷脂酰肌醇和二磷脂酰甘油。另一常用磷脂是鞘磷脂。鞘磷脂含有鞘氨醇,一种具有长不饱和烃链的氨基醇。脂肪酰基侧链通过酰胺键连接到鞘氨醇的氨基,以形成神经酰胺。鞘氨醇的羟基被酯化至磷酸胆碱。与磷酸甘油酯类似,鞘磷脂是两性的。所有这些和其他磷脂可用于本发明实践。在一些实施方式中,使用具有4至24之间碳链长度的磷脂。也可以使用卵磷脂,其是通常源自鸡蛋或羊毛的磷脂的天然混合物。磷脂可以购自Avantilipids(Alabastar,AL,USA)以及lipoidLLC(Newark,NJ,USA)。
乳化剂
本发明的组合物可包含一种或更多种乳化剂。乳化剂可以是纯的乳化剂或乳化剂的混合物。本发明的乳化剂是药学和/或免疫学可接受的。当混合物重悬于疏水载体中时,乳化剂通常有助于稳定两性化合物和抗原的混合物或两性化合物、抗原和佐剂的混合物。
乳化剂可以是两性的,因此,乳化剂可包括较广范围的化合物。在一些实施方式中,乳化剂可以是表面活性剂,例如举例来说,非离子表面活性剂。
可以使用的乳化剂的实例包括聚山梨醇酯,其是源自聚乙二醇化山梨醇的油状液体,以及脱水山梨醇酯。聚山梨醇酯可包括例如脱水山梨醇单油酸酯。常用的乳化剂包括油酸二缩甘露酯(ArlacelTMA)、卵磷脂、TweenTM80以及SpansTM20、80、83和85。乳化剂通常与疏水载体预先混合。
在一些实施方式中,可以使用已含有乳化剂的疏水载体。例如,疏水载体例如MontanideTMISA-51已含有乳化剂油酸二缩甘露酯。在其它实施方式中,疏水载体可以在与两性化合物和抗原组合之前与乳化剂混合。
疏水载体
疏水载体可以是基本上纯的疏水物质或疏水物质的混合物。
可用于如本文所述组合物的疏水物质是药学和/或免疫学可接受的。载体优选地是液体,但某些在环境温度不是液体的疏水物质可以被液化,例如通过加热,并且也可用于本发明。
油或油的混合物是用于本发明的特别合适的载体。油应是药学和/或免疫学可接受的。油可以是可代谢的或不可代谢的,或者可使用可代谢的和不可代谢的油的混合物。
油的优选实例是矿物油(特别是轻或低粘度矿物油)、植物油(例如大豆油)、坚果油(例如花生油)。在一些实施方式中,可以使用低粘度矿物油例如6VR。在一个实施方式中,油是油酸二缩甘露酯的矿物油溶液,其可以ISA51购得。其他油可包括例如MontanideISA700系列(SeppicInc.,France)或MAS-1(MerciaPharmaceuticals)。在存在纯的疏水载体的实施方式中,疏水载体可以在用于本发明的组合物之前与乳化剂混合。
也可使用动物脂肪和人工疏水聚合材料,特别是在环境温度下是液体或可以相对容易地被液化的那些。
液体碳氟化合物是医学上可用的疏水载体,也可用于本发明实践。
其他组分
组合物可进一步包含一种或更多种另外的组分,例如,举例来说,药学可接受的佐剂、赋形剂等等,如本领域已知的:参见,例如,Remington’sPharmaceuticalSciences(Remington’sPharmaceuticalSciences,MackPublishingCompany,Easton,Pa.,USA1985)和1990年出版的TheUnitedStatesPharmacopoeia:TheNationalFormulary(USP24NF19)。
术语“佐剂”是指增强针对抗原的免疫反应的化合物或混合物。佐剂可用作缓慢释放抗原的组织贮库(tissuedepot),并可用作淋巴系统激活剂,其非特异性地增强免疫反应(Hood等,Immunology,2ded.,Benjamin/Cummings:MenloPark,C.A.,1984;参见Wood和Williams,In:Nicholson,WebsterandMay(eds.),TextbookofInfluenza,Chapter23,pp.317-323)。
合适的佐剂包括但不限于:明矾、其他铝化合物、卡介苗(BCG)、不完全弗氏佐剂(IFA)、皂角甙、表面活性物质例如溶血卵磷脂、普流尼克多元醇(pluronicpolyols)、聚阴离子、肽、短小棒状杆菌(Corynebacteriumparvum)、QS-21、弗氏完全佐剂(FCA)、TLR激动剂家族的佐剂例如CpG、polyIC(双链RNA)、falgellin、脂肽、肽聚糖、咪唑并喹啉、单链RNA、脂多糖(LPS)、热休克蛋白(HSP)以及神经酰胺和衍生物例如αGal-cer。合适的佐剂还包括处于它们多肽或DNA编码形式的细胞因子或趋化因子,例如但不限于:GM-CSF、TNF-α、IFN-γ、IL-2、IL-12、IL-15、IL-21。
如上所述,疏水载体,可在一些情况下发挥佐剂的作用。
所用的佐剂量取决于抗原量和佐剂类型。本领域普通技术人员可容易确定具体应用中需要的佐剂量。
组合物也可含有一种或更多种另外的多肽,其可以是短的合成多肽,例如T辅助表位。
组合物的制备
在悬浮于疏水载体之前将抗原和两性化合物混合。优选地,抗原和两性化合物以使得形成基本均一的混合物的方式组合。这可以通过在组分被组合之前将抗原和/或两性化合物溶解于合适的溶剂来完成。可选地,可将两种实体以它们的干燥形式混合在一起(例如通过研磨)。在这种情况下,抗原和两性化合物“基本均一的混合物”是混合物,其中两性化合物基本均匀地分散在抗原组分中。
两性化合物或两性化合物的混合物以足够量存在于本发明的组合物中以使抗原可重悬在疏水载体中。本发明组合物中两性化合物的数量可以是,例如,每ml组合物约0.1mg至约250mg两性化合物,更优选地每ml组合物约0.1至约120mg两性化合物。
如果抗原和/或两性化合物被溶解,本领域普通技术人员可容易鉴定用于溶解具体两性化合物或抗原的合适的溶剂或溶剂系统(通常地有机溶剂)。在两性化合物是磷脂的情况下,可以使用极性质子溶剂,例如醇(例如叔丁醇、正丁醇、异丙醇、正丙醇、乙醇或甲醇)、水、乙酸或甲酸或氯仿。抗原例如多肽可以用极性非质子溶剂溶解,例如二甲基亚砜(DMSO)、二甲基甲酰胺(DMF)或四氢呋喃(THF)。可以使用其他溶剂,例如非极性溶剂(例如己烷),以及液体CO2。
在一些情况中,相同溶剂可以用于溶解感兴趣的两性分子和抗原。
然后将溶解的抗原和溶解的两性化合物混合。可选地,可以在溶解之前将抗原和两性化合物混合,然后溶解在一起。进一步可选地,仅将两性化合物或抗原的一种溶解,然后添加未溶解的组分。
然后去除溶剂,这可以使用标准技术完成。如果使用易蒸发溶剂,例如乙醇、甲醇或氯仿,可以采用标准蒸发技术,例如旋转蒸发、减压蒸发或冷冻干燥。溶剂,例如水,可以通过例如冻干、冷冻干燥或喷雾干燥去除。不破坏组分完整性的低热干燥也可使用。加热也可用于在使用前辅助抗原/两性化合物混合物重悬。
优选地,溶解的抗原和溶解的两性化合物充分混合,然后如上所述干燥。优选地,干燥的混合物是抗原和两性化合物的基本均一的混合物,其中两性化合物基本均匀地分散在抗原组分中。如果改为抗原和两性化合物的基本均一的混合物通过无溶剂方法例如干研磨形成,当然不需要干燥步骤。
然后将抗原和两性化合物的干燥混合物重悬于疏水载体中以提供制备好的组合物。在一些实施方式中,疏水载体可含有乳化剂,如上详述,其以足以将抗原和两性化合物的干燥混合物重悬在疏水载体中并在疏水载体中维持抗原和两性化合物悬浮的量被提供。例如,乳化剂可以以疏水载体的约5%至约15%重量/重量或重量/体积存在。
如上所述另外的组分,例如佐剂或其他药学可接受的辅剂,可以在制剂过程的任何时期添加。例如,一种或更多种这些另外的组分可以在溶解之前或之后与抗原或两性化合物组合或添加至溶解的混合物中。另外的组分,例如佐剂,可改成添加至抗原和两性化合物的干燥混合物或与抗原和两性化合物的干燥混合物组合,或在抗原和两性化合物的干燥混合物悬浮在疏水载体中之前或之后与疏水载体组合。类似地,如果使用干混合技术,任何另外的组分可以在研磨之前或之后添加。
意想不到地,在无实质量的水存在的情况下,当抗原悬浮在所述疏水载体中时,获得强免疫反应。不预期抗原可位于疏水载体中,除非在本文所述的组合物中配制。在实践中,可能难以获得完全无水的组合物。即,尽管在制剂过程的适当阶段,例如通过蒸发、冷冻干燥或任何其他合适的干燥技术,完全去除水或基本上完全去除水,仍可能保留少量的水。例如,组合物的各个组分可具有结合水,其不能通过例如冷冻干燥或蒸发的方法完全去除,并且某些疏水载体可含有少量溶解在其中的水。当水存在时,例如,以乳状液的形式存在于组合物中时,预期一定量的抗原可被分配至水中。因此,组合物中水的存在降低了悬浮在疏水载体中抗原的量,因此,不希望在最终组合物中存在水。
制备好的组合物是基本上无水的。“基本上无水”意思是悬浮(例如溶解)在水中的抗原相对于组合物中抗原总量的比例(重量/重量)低至足以使悬浮在水中的抗原数量自身不能产生与组合物作为整体提供的免疫反应相当的免疫反应。相反,悬浮在疏水载体中的抗原数量高至足以使得使用该数量(即总量减去存在于剩余水组分中的抗原数量)可以产生等效免疫反应。因此,组合物的功效(即它产生期望的生物反应的能力)应主要归因于悬浮在疏水载体中的抗原。在这点上,组合物的功效为没有抗原存在于剩余水组分中的相同组合物诱导的至少80%、85%、90%或95%那么大。
“基本上无水”的本发明组合物通常含有基于重量/重量小于约10%、9%、8%、7%、6%、5%、4%、3%、2%、1%、0.5%、0.1%、0.05%或0.01%的水。悬浮在剩余水组分中的抗原量期望小于组合物中抗原总量(基于重量)的20%、15%、10%、9%、8%、7%、6%、5%、4%、3%、2%、1%或更小。
通常地,本发明的组合物是足够无水的,使得没有形成肉眼可见的油包水乳液。例如,不期望的油包水乳液的存在可通过组合物的不透明、浑浊或不透光的外观进行检测。相反,本发明的组合物通常具有清晰或透明的外观并且无可见的颗粒物质,例如未悬浮在疏水载体中的沉淀或聚集的抗原。
如本文所述的组合物可以适于将抗原递送至对象的任何形式加以配制,作为非限制实例,包括适合于口服、经鼻、直肠或肠胃外施用的形式。肠胃外施用包括但不限于静脉内、腹膜内、皮内、皮下、肌肉内、经上皮、肺内、鞘内和局部施用方式。
试剂盒和试剂
本发明任选地作为试剂盒提供给使用者。例如,本发明的试剂盒含有一种或更多种本发明的组合物。试剂盒可进一步包含一种或更多种另外的试剂、包装材料、用于容纳试剂盒组分的容器,以及一套说明书或用户手册,其详述了使用试剂盒组分用于所期望目的的优选方法。
在一个实施方式中,本发明的组合物可以如下形式提供:其中抗原和两性化合物的干燥混合物包装在第一容器中,而疏水载体包装在第二容器中。然后,在施用至对象之前不久,可将抗原和两性化合物的干燥混合物悬浮在疏水载体中。
用途
本发明可用于在期望将抗原递送至对象的任何情况。对象可以是脊椎动物,例如鱼、鸟或哺乳动物,优选地是人类。
在一些实施方式中,本发明的组合物可以施用至对象以提高抗体对抗原的反应。
“抗体”是蛋白质,其包括基本上或部分由免疫球蛋白基因或免疫球蛋白基因片段编码的一个或更多个多肽。已知的免疫球蛋白基因包括κ、λ、α、γ、δ、ε和μ恒定区基因,以及各种免疫球蛋白可变区基因。轻链分为κ或λ。重链分为γ、μ、α、δ或ε,其反过来分别确定免疫球蛋白类型:IgG、IgM、IgA、IgD和IgE。常见的免疫球蛋白(抗体)结构单元包括含有四个多肽的蛋白质。每个抗体结构单位由两对相同的多肽链组成,每一对具有一个“轻”链和一个“重”链。每个链的N-末端定义为可变区,主要负责抗原识别。抗体结构单元(例如IgA和IgM类)也可相互并与另外的多肽链组装成寡聚体形式,例如IgM五聚体与J-链多肽结合。
抗体是称为B淋巴细胞(B细胞)的白细胞亚型的抗原特异性糖蛋白产物。抗原与B细胞表面表达的抗体结合可诱导抗体反应,其包括刺激B细胞被激活,进行有丝分裂以及最终分化成浆细胞,其被特化用于抗原特异性抗体的合成和分泌。
如本文所用,术语“抗体反应”是指在对象机体内,针对抗原引入至对象机体的反应,抗原特异性抗体数量的增加。
评价抗体反应的一个方法是测量与具体抗原具有反应性的抗体的滴度。这可以通过使用本领域已知的各种方法进行,例如获自动物的含抗体的物质的酶联免疫吸附测定(enzymelinkedimmunosorbentassay,ELISA)来进行。例如,结合具体抗原的血清抗体的滴度可以在暴露至抗原之前和之后在对象中测定。在暴露至抗原后,抗原特异性抗体滴度的统计学显著增加表明对象已经产生针对抗原的抗体反应。
在一些实施方式中,本发明的组合物可以施用至对象以提高针对抗原的细胞介导的免疫反应。如本文所用,术语“细胞介导的免疫反应”是指在对象机体内,应答于抗原引入至对象机体,抗原特异性细胞毒性T淋巴细胞、巨噬细胞、天然杀伤细胞或细胞因子数量的增加。
在组织学上,免疫系统分为两个分支:体液免疫,免疫保护功能可在体液中发现(含有抗体的无细胞体液或血清);以及细胞免疫,免疫保护功能与细胞相关。细胞介导的免疫是这样的免疫反应,其涉及巨噬细胞、天然杀伤细胞(NK)、抗原特异性细胞毒性T淋巴细胞的活化以及应答于“非自身”抗原而释放各种细胞因子。细胞免疫是适应性免疫反应的重要组分,并且是在细胞通过它们与抗原呈递细胞例如树突细胞、B淋巴细胞的相互作用以及较小程度地与巨噬细胞的相互作用识别抗原之后,通过各种机制保护机体,例如:
1.激活抗原特异性细胞毒性T淋巴细胞,其能够诱导在细胞表面上展示外来抗原表位的体细胞凋亡,例如病毒感染的细胞、具有细胞内细菌的细胞以及展示肿瘤抗原的癌细胞;
2.激活巨噬细胞和天然杀伤细胞,使得它们破坏细胞内病原体;以及
3.刺激细胞分泌各种细胞因子,其影响涉及适应性免疫反应和先天免疫反应的其他细胞的功能。
细胞介导的免疫在去除病毒感染的细胞中最有效,而且还参与防御真菌、原生动物、癌症和细胞内细菌。它还在移植排斥中发挥主要作用。
接种后细胞介导的免疫反应的检测
由于细胞介导的免疫涉及各种细胞类型的参与并通过不同机制介导,可使用几个方法来证明接种后免疫诱导。这些可广泛地分为检测:i)特异的抗原递呈细胞;ii)特异的效应子细胞及它们的功能,以及iii)可溶介质例如细胞因子的释放。
i)抗原呈递细胞:树突细胞和B细胞(以及较小程度的巨噬细胞)被配置有专门的免疫-刺激受体,其可进行T细胞的增强活化,并且称为专职抗原呈递细胞(antigenpresentingcells,APC)。感染或接种后,在抗原呈递至效应子细胞例如CD4和CD8细胞毒性T细胞的过程中,在这些细胞上这些免疫刺激分子(也称为共刺激分子)被上调。这种共刺激分子(例如CD80、CD86、MHCI类或MHCII类)可通过使用流式细胞术用荧光染料偶联的针对这些分子的抗体连同特异性识别APC的抗体(例如树突细胞的CD11c)一起检测。
ii)细胞毒性T细胞:(也称为Tc、杀伤性T细胞或细胞毒性T淋巴细胞(CTL))是T细胞亚群,其诱导病毒(和其他病原体)感染的或表达肿瘤抗原的细胞死亡。这些CTL直接攻击在其表面上携带某些外源或异常分子的其他细胞。这种细胞毒性能力可以使用体外细胞溶解测定(铬释放测定)加以检测。因此,当负载抗原的靶细胞被接种或感染后体内产生的特异性CTL裂解时,适应性细胞免疫的诱导可通过这种细胞毒性T细胞的存在加以证明。
当它们的T细胞受体(TCR)与肽-结合的MHCI类分子强烈地相互作用时,幼稚细胞毒性T细胞被激活。这种亲和性取决于抗原/MHC复合物的类型和方向,并且是这种亲和性保持CTL和感染的细胞结合在一起。一旦激活,CTL进行称为克隆扩增的过程,其中它获得功能性,并快速分裂以产生大量的“防卫”效应子细胞(“armed”-effectorcells)。然后,激活的CTL在整个机体内游走寻找具有独特MHCI类+肽的细胞。这可用于通过在流式细胞术测定中使用肽-MHCI类四聚体在体外鉴定这种CTL。
当暴露至这些感染的或机能紊乱的体细胞中,效应子CTL释放穿孔素(perforin)和粒溶素(granulysin):细胞毒素,其在靶细胞质膜中形成孔,使离子和水流进感染的细胞,并导致它裂解或溶解。CTL释放粒酶(granzyme),一种丝氨酸蛋白酶,其经由孔进入细胞以诱导凋亡(细胞死亡)。这些分子从CTL的释放可以用作接种后成功引入细胞免疫反应的量度。这可以通过酶联免疫吸附测定(ELISA)或酶联免疫斑点测定(ELISPOT)进行,其中CTL可以定量测定。由于CTL还能够产生重要的细胞因子例如IFN-g,产生IFN-g的CD8细胞的定量测量可通过在这些细胞进行ELISPOT和流式细胞术测量细胞内IFN-g来实现。
CD4+“辅助”T细胞:CD4+淋巴细胞或辅助T细胞是免疫反应介质,并在建立和最大化适应性免疫反应的能力方面发挥重要作用。这些细胞不具有细胞毒性或吞噬活性;并且不能杀死感染的细胞或清除病原体,但,本质上通过引导其他细胞进行这些任务,“管理”免疫反应。两个类型的效应子CD4+T辅助细胞反应可以通过专职APC诱导,命名为Th1和Th2,每个被设计为清除不同类型的病原体。
辅助T细胞表达T细胞受体(TCR),其识别结合到II类MHC分子的抗原。幼稚辅助T细胞的活化使其释放细胞因子,其影响许多细胞类型的活性,包括激活它的APC。辅助T细胞需要比细胞毒性T细胞温和得多的活化刺激。辅助T细胞可提供“辅助”激活细胞毒性细胞的额外信号。两个类型的效应子CD4+T辅助细胞反应可以通过专职APC诱导,命名为Th1和Th2,每个被设计为清除不同类型的病原体。与Th1或Th2反应相关的细胞因子的测量将提供成功接种的测量。这可以通过为Th1-细胞因子例如IFN-g、IL-2、IL-12、TNF-a及其他,或Th2-细胞因子例如IL-4、IL-5、IL10等等设计的特异ELISA来实现。
iii)细胞因子的测量:从局部淋巴结的释放提供了成功免疫的良好指示。作为抗原呈递和APC及免疫效应子细胞例如CD4和CD8T细胞成熟的结果,若干细胞因子由淋巴结细胞释放。在抗原存在的情况下,通过体外培养这些LNC,可以通过测量某些重要的细胞因子例如IFN-g、IL-2、IL-12、TNF-a和GM-CSF的释放来检测抗原特异性免疫反应。这可使用培养上清液和重组细胞因子作为标准,通过ELISA进行。
本发明可广泛用于治疗和预防易于通过抗原施用而预防和/或治疗的任何疾病。本发明的代表性应用包括癌症治疗和预防、基因治疗、辅助治疗、传染性疾病治疗和预防、变态/过敏反应治疗和预防、自身免疫疾病治疗和预防、神经退行性疾病治疗以及动脉粥样硬化治疗、药物依赖性治疗和预防、用于疾病治疗和预防的激素控制、用于避孕目的的生物过程控制。
疾病的预防或治疗包括获得有利或期望的结果,包括临床结果。有利或期望的临床结果可包括但不限于:一个或更多症状或状况的减轻或改善、疾病程度的降低、疾病状态的稳定、疾病发展的防止、疾病扩散的防止、疾病进程的延迟或减慢、疾病发作的延迟或减慢、赋予针对疾病引发剂的保护性免疫以及疾病状态的改善或减轻。预防或治疗还可意味着延长患者的存活时间,使其高于缺乏治疗所预计的时间,还可意味着临时地抑制疾病进程,尽管更优选地,它涉及例如通过防止在对象中的感染来防止疾病的发生。
本领域普通技术人员可确定任何具体应用的合适治疗方案、施用途径、剂量等等,以获得期望的结果。可考虑的因素包括例如:抗原性质;要预防或治疗的疾病状态;对象的年龄、身体条件、体重、性别和饮食;以及其他临床因素。
本发明通过下列非限制实施例进一步说明。
实施例1
无病原体的6-8周龄雌性C57BL/6小鼠获自CharlesRiverLaboratories(StConstant,Quebec,Canada)并根据专业指南在滤器控制的空气循环中饲养,任意提供水和食物。
用于该研究的C3细胞系——用于临床前宫颈癌研究的充分描述的小鼠模型——是表达HPV16的C3肿瘤细胞,其源自B6小鼠胚胎细胞(B6mec)并转化了在它的自身启动子控制下的全部HPV16基因组、以及激活的-ras原癌基因。当皮下注射时C3细胞系发展为肿瘤,并且已用于癌症风险研究中以检测C3肿瘤细胞植入前或植入后施用的疫苗的效力。将C3细胞系在添加10%热灭活胎牛血清(Sigma,St.Louis,MO)、2mM1-谷氨酰胺、50mM2-巯基乙醇、青霉素和链霉素的IscoveModifiedDulbecco’sMedium(IMDM;Sigma,St.Louis,MO)中维持。将细胞在37摄氏度/5%CO2中温育。
将含有CTL表位的HPV16E7(H-2Db)肽RAHYNIVTF49-57(SEQIDNO:1)与DaltonChemicalLaboratoriesInc.(Toronto,Ontario,Canada)提供的含有CD4+辅助表位的PADRE融合。下文将这种肽命名为FP并在疫苗中以50微克/100微升剂量用作抗原。FP已用于疫苗研究来预防或清除小鼠中的C3肿瘤。
为了配制本文所述的疫苗,将二油酰磷脂酰胆碱(DOPC)溶解在叔丁醇中。首先将FP溶解在二甲基亚砜中,尽管FP水悬浮液也可使用。然后将FP添加至DOPC/叔丁醇混合物。在指出的情况下,合成的基于脂肽的免疫刺激化合物(佐剂)重悬在水中并添加至DOPC/FP/叔丁醇混合物。通过冷冻干燥去除制剂中存在的溶剂和水来制备抗原的干燥均一混合物(含有或不含有佐剂)。
然后,将干燥混合物悬浮在不完全弗氏佐剂——基于矿物油的模式疏水载体中。
这些制剂的效力可与由在常用的油包水乳液中的抗原(含有或不含有佐剂)组成的制剂比较,乳液例如基于不完全弗氏佐剂的乳液,其主要由在连续的基于矿物油的油载体中的水(含有抗原/佐剂)组成。
为了检测这些无水制剂的效力,在尾基部上方左侧腹向各组小鼠(每组7至10只小鼠)皮下注射五十万个C3细胞。植入后八天,在右侧腹向所有小鼠皮下注射疫苗制剂(每个剂量100微升)。五组小鼠进行如下接种:组1小鼠用作对照小鼠并注射磷酸盐缓冲液以使肿瘤正常发展;用在乳液的水组分中含有FP抗原(每剂量50微克)的标准油包水矿物油基乳液(不完全弗氏佐剂)接种组2小鼠;用如上所述制备的并含有FP抗原(每个剂量50微克)、DOPC(每个剂量12微克)和100微升疏水载体(不完全弗氏佐剂)的均一无水制剂接种组3小鼠;用在乳液的水组分中含有FP抗原(每个剂量50微克)和Pam3Cys佐剂(每个剂量50微克)的标准油包水矿物油基乳液(不完全弗氏佐剂)接种组4小鼠;用如上所述制备的并含有FP抗原(每个剂量50微克)、Pam3Cys佐剂(每个剂量50微克)、DOPC(每个剂量12微克)和100微升疏水载体(不完全弗氏佐剂)的均一无水制剂接种组5小鼠;每周在所有小鼠中监测肿瘤生长,持续42天,以评估接种对肿瘤生长的作用。
组1中移植C3细胞的所有小鼠发展出肿瘤,其中肿瘤大小在肿瘤植入后第42天达到1881立方毫米。由油包水乳液中的FP抗原组成的对照接种控制小鼠肿瘤生长(组2),其中肿瘤在第42天进展为平均大小548立方毫米。组3小鼠,其用疏水载体中的FP抗原无水组合物接种并利用DOPC作为两性载体以确保抗原在油中的均一重悬,在控制肿瘤生长方面明显比组2疫苗更有效。在肿瘤植入后第42天组3小鼠肿瘤平均大小为73立方毫米,8只小鼠中的6只为无肿瘤。在该实例中佐剂Pam3Cys的添加略微提高了组2使用的油包水疫苗制剂的效力(在第42天组4肿瘤平均大小为320立方毫米,相对于组2肿瘤平均大小为548立方毫米)。佐剂Pam3Cys的添加进一步提高无水疫苗制剂的效力(组5疫苗),在第42天肿瘤平均大小为14立方毫米,7只小鼠中的6只为无肿瘤。
这些结果清楚地显示,从疫苗制剂中排除水并使用基于磷脂的两性分子以确保疏水载体中免疫激活化合物均一性,产生增强的靶向免疫反应。
实施例2
无病原体的6-8周龄雌性C57BL/6小鼠获自CharlesRiverLaboratories(StConstant,Quebec,Canada)并根据专业指南在滤器控制的空气循环中饲养,任意提供水和食物。
将含有CTL表位的HPV16E7(H-2Db)肽RAHYNIVTF49-57(SEQIDNO:1)与DaltonChemicalLaboratoriesInc.(Toronto,Ontario,Canada)提供的含有CD4+辅助表位的PADRE融合。这种肽在下文命名为FP并在疫苗中以20微克/100微升剂量用作抗原。
疫苗效力通过酶联免疫斑点测定(ELISPOT)评价,其是在从免疫的C57BL/6小鼠收获的脾细胞中抗原特异性细胞免疫反应的离体检测方法。ELISPOT测定可用于评价抗原特异性免疫反应的存在/缺乏,但当用作体内针对靶的疫苗效力的相关性时,它具有局限性。简言之,免疫后第8天,通过在4℃温育过夜将96孔硝酸纤维素板用捕获抗体——纯化的抗小鼠IFN-γ抗体包被,然后用完全培养基封闭。将脾细胞以起始浓度5×105个细胞/孔以100μl体积添加至孔并制备一排连续稀释。将连续稀释的细胞用特异的肽RAHYNIVTF49-57(10μg/ml)刺激。将板在37℃/5%CO2中温育过夜。第二天,将板用检测抗体(生物素化的抗小鼠IFN-γ抗体)在室温温育2小时。通过洗涤去除未结合的检测抗体并添加酶偶联物(Streptavidin-HRP)。室温温育1小时后,通过洗涤去除未结合的酶偶联物并将板用AEC底物溶液染色20分钟。洗涤板,使其风干过夜,并使用放大镜计算染色点。
为了配制本文所述的疫苗,将二油酰磷脂酰胆碱(DOPC)溶解在叔丁醇中。首先将FP溶解在二甲基亚砜中,尽管FP水悬浮液也可使用。然后将FP添加至DOPC/叔丁醇混合物。通过冷冻干燥去除制剂中存在的溶剂来制备抗原和DOPC的干燥均一混合物。
然后,将干燥混合物悬浮在不完全弗氏佐剂——基于矿物油的模式疏水载体中。
这些制剂的效力可与由常用的油包水乳液中的抗原组成的制剂比较,例如基于不完全弗氏佐剂的乳液,其主要由连续的基于矿物油的油载体中的水(含有抗原)组成。制剂的效力还可与由抗原组成的制剂比较,所述抗原直接从抗原/二甲基亚砜储存溶液稀释入典型的油包水乳液,例如基于不完全弗氏佐剂的乳液。
为了检测这种无水制剂的效力,在右侧腹向各组小鼠(每组4只小鼠)皮下注射疫苗制剂(每个剂量100微升)。三组小鼠进行如下接种:组1小鼠用作对照小鼠并用在乳液的水组分中含有FP抗原(每个剂量20微克)的标准油包水矿物油基乳液(不完全弗氏佐剂)接种;用如上所述制备的并含有FP抗原(每个剂量20微克)、DOPC(每个剂量12微克)和100微升疏水载体(不完全弗氏佐剂)的均一无水制剂接种组2小鼠;组3小鼠用作对照小鼠并用缺乏两性载体(DOPC)但含有FP抗原(每个剂量20微克)和100微升疏水载体(不完全弗氏佐剂)的均一无水制剂接种。8天后去除所有小鼠的脾,通过ELISPOT测定检测抗原特异性免疫反应的存在。
组1小鼠产生显著的抗原特异性细胞反应。通过ELISPOT检测的这种反应的存在表明疫苗制剂具有诱导体内针对靶的有效免疫反应的潜能。注射含有FP、两性载体(DOPC)和疏水油载体的无水制剂的组2小鼠也能够诱导免疫反应,表明诱导体内针对靶的有效免疫反应的潜能。用缺乏两性载体(DOPC)但含有FP和疏水油载体的无水制剂注射的组3小鼠诱导显著降低的免疫反应,如ELISPOT所检测,这清楚地表明这种具体制剂具有相当低的免疫原性潜能。这些结果清楚地显示,如果基于磷脂的两性分子存在于制剂中,从疫苗制剂中排除水具有产生增强的靶向免疫反应的潜能。
实施例3
在该实施例中,polyIC双链RNA(Pierce,Milwaukee,USA)用作代表分子,其具有与基因抗原构建体(基于核苷酸的质粒或RNA分子)类似的物理和化学特征。polyIC还用作基于核苷酸的代表佐剂,其可在本发明中与抗原共配制。为了配制1ml终体积的本文所述的疫苗,在40℃的温度将120.0mg二油酰磷脂酰胆碱(DOPC)溶解于480ul叔丁醇,振荡10至15分钟。首先,polyI:C以5mg/ml浓度溶解在水中。然后将80ulpolyI:C(0.4mg)进一步稀释在320ul水中。然后,将polyI:C稀释液添加并混合至在小瓶21中的DOPC/叔丁醇混合物。通过冷冻干燥去除制剂中存在的溶剂和水,制备DOPC/佐剂的干燥均一混合物。只含有polyIC的对照制剂(小瓶26)以如上所述相同方法制备,除了不向丁醇中添加DOPC。
然后,通过添加0.88ml疏水载体至小瓶21,以及1ml疏水载体至小瓶26,将小瓶21和26的干燥内含物悬浮。使用的疏水载体为含有油酸二缩甘露酯和称为ISA51(Seppic,France)的矿物油。通过涡旋大约3分钟(小瓶21)或最少30分钟(小瓶26)将干燥混合物重悬在疏水载体中。在仅含有1ml疏水载体的小瓶(标记ISA51的小瓶)旁,通过肉眼观察比较小瓶21和26。
通过肉眼观察(图7),小瓶21内含物看起来类似于ISA51,不存在肉眼可见的颗粒物质。这表明在DOPC存在的情况下,核苷酸分子可有效地悬浮在疏水载体中。相反,在缺乏DOP并仅含有核苷酸分子和疏水载体的小瓶26中,含有通过肉眼观察容易检测到的、核苷酸分子在疏水载体中的不均一悬浮液。缺乏DOPC,亲水核苷酸分子不能悬浮在疏水载体中。这清楚地表明,分子例如实际上是两性的DOPC,在缺乏显著量的水的情况下促进疏水载体中基于亲水核苷酸的分子的配制。
实施例4
在该实例中肽S9L(SVYDFFVWL)(SEQIDNO:2)和F21E(FNNFTVSFWLRVPKVSASHLE)(SEQIDNO:3)用作模式抗原。肽由定制肽生产商(Anaspec,SJose,USA)化学合成。为了配制1ml终体积的本文所述的这些本发明的肽,在40℃的温度将120.0mg二油酰磷脂酰胆碱(DOPC)溶解于470ul叔丁醇,振荡10至15分钟。然后,将混合物冷却至室温(22℃-25℃)。首先,将S9L和F21E分别以5mg/ml浓度溶解在二甲基亚砜中。然后依次添加10ulS9L(50ug)和10ulF21E(50ug)至DOPC/叔丁醇混合物,同时涡旋。为达到混合物体积,将400ul水添加混合至小瓶30中的S9L/F21E/DOPC/叔丁醇混合物。通过冷冻干燥去除制剂中存在的溶剂和水,制备DOPC/肽的干燥均一混合物。含有S9L和F21E肽的对照制剂(小瓶35)以如上所述相同方法制备,除了不向丁醇中添加DOPC。
然后,通过添加0.88ml疏水载体至小瓶30,以及1ml疏水载体至小瓶35,将小瓶30和35的干燥内含物悬浮。使用的疏水载体为含有油酸二缩甘露酯和称为ISA51(Seppic,France)的矿物油。通过涡旋大约30秒(小瓶30)或最少30分钟(小瓶35)将干燥混合物重悬在疏水载体中。在仅含有1ml疏水载体的小瓶(标记ISA51的小瓶)旁,通过肉眼观察比较小瓶30和35。
通过肉眼观察(图8),小瓶30内含物看起来类似于ISA51,不存在肉眼可见的颗粒物质。这表明在DOPC存在的情况下,肽可有效地悬浮在疏水载体中。相反,在缺乏DOP并仅含有肽和疏水载体的小瓶35中,含有通过肉眼观察容易检测到的、在疏水载体中的肽聚集体不均一悬浮液。缺乏DOPC,基于肽的抗原不能悬浮在疏水载体中。这清楚地表明,分子例如实际上是两性的DOPC,在缺乏显著量的水的情况下促进疏水载体中基于肽的抗原的配制。

Claims (17)
1.基本上无水的疫苗制剂,其包含抗原和两性化合物的均一混合物,所述均一混合物悬浮在疏水载体中,
其中在悬浮在所述疏水载体中前,通过如下方法制备所述均一混合物:包括将所述两性化合物溶解在有机溶剂中、混合所述抗原与所述溶解的两性化合物、和干燥所述混合物的方法;
其中所述两性化合物是磷脂或磷脂混合物;
其中所述疏水载体包含油酸二缩甘露酯和矿物油;和
其中所述疫苗制剂包含基于所述疫苗制剂的总重量小于10%重量的水。
2.根据权利要求1所述的疫苗制剂,其中按重量计小于20%的所述抗原被悬浮在水中。
3.根据权利要求1所述的疫苗制剂,其中所述磷脂是卵磷脂,或所述磷脂混合物包含卵磷脂。
4.根据权利要求2所述的疫苗制剂,其中所述磷脂是卵磷脂,或所述磷脂混合物包含卵磷脂。
5.根据权利要求1所述的疫苗制剂,其中所述磷脂是二油酰脂酰胆碱(DOPC),或所述磷脂混合物包含DOPC。
6.根据权利要求2所述的疫苗制剂,其中所述磷脂是二油酰脂酰胆碱(DOPC),或所述磷脂混合物包含DOPC。
7.根据权利要求1至6中任一项所述的疫苗制剂,其中所述抗原包括:多肽;编码多肽的多核苷酸;或活的、减毒的、灭活的或杀死的细菌、病毒或原生动物或其部分。
8.根据权利要求7所述的疫苗制剂,其进一步包含佐剂。
9.根据权利要求7所述的疫苗制剂,其中每ml疫苗制剂包含0.1至250mg的所述磷脂或磷脂混合物。
10.根据权利要求7所述的疫苗制剂,其中所述矿物油包含按重量计5%至15%的油酸二缩甘露酯。
11.权利要求7所述的疫苗制剂,其中所述有机溶剂是醇。
12.权利要求11所述的疫苗制剂,其中所述醇是叔丁醇。
13.制备根据权利要求1至12任何一项所述的疫苗制剂的方法,所述方法包括:
(a)将两性化合物溶解在有机溶剂中;
(b)混合所述溶解的两性化合物与抗原;
(c)干燥所述混合物以形成均一干燥的混合物;和
(d)将所述均一干燥的混合物悬浮在疏水载体中,
其中所述两性化合物是磷脂或磷脂混合物;
其中所述疏水载体包含油酸二缩甘露酯和矿物油。
14.根据权利要求13所述的方法,其进一步包括将佐剂添加至所述疫苗制剂。
15.根据权利要求13所述的方法,其中在与所述溶解的两性化合物组合和混合前,将所述抗原溶解。
16.根据权利要求1至12任何一项所述的疫苗制剂在制备用于诱导个体内抗体反应或细胞介导的免疫反应的药物中的应用。
17.根据权利要求16所述的应用,其中所述疫苗制剂被口服、经鼻、直肠或肠胃外施用。
