Abstract
本发明涉及至少一种单不饱和脂肪酸、其盐和/或其酯,优选地岩芹酸,通过口服途径的美容用途,所述用途为用于改善头发的质量和/或头发的外观,特别是用于预防和/或治疗随年龄而出现的头皮和/或头发的改变。
Description
本发明涉及在包含美容上或药学上可接受的介质的组合物中的至少一种单不饱和脂肪酸,作为头皮和头发的保护试剂(治疗或预防),通过口服施用的用途。
毛囊是由极具特性的区室形成的,其中一些为真皮起源(毛囊鞘(gaine conjonctive)和真皮乳头),另一些具有上皮性质(外上皮鞘、内鞘、毛干和皮脂腺)。由成纤维细胞合成的毛囊鞘首要地是一种由I型和III型胶原以及蛋白聚糖形成的细胞外基质。有毛细血管网横穿其下部三分之一,毛囊鞘经由真皮乳头(真正的细胞外基质聚集体)延伸至毛囊的底部。真皮乳头由负责局部合成前列腺素E2的特殊的成纤维细胞构成。由IV型胶原、1型和5型层粘连蛋白、纤连蛋白和硫酸乙酰肝素蛋白聚糖构成的基底膜,将真皮区室和上皮区室相分开。然而,层粘连蛋白5的表达特性谱在毛球外周中断,由此突出了真皮-表皮接合处的区域化。上皮区室,就其自身来说,可被分隔为四个不同的区域。在毛囊的底部和在真皮乳头周围存在有基质,那里是强烈有丝分裂活性的地点。使用特定的分化程序,该基质产生三个主要的同中心区域:外鞘、内鞘和毛干。在毛囊的上面部分中存在有皮脂腺,其由特殊的角质形成细胞即负责皮脂合成的皮脂腺细胞(sébocyte)形成。外鞘的上面部分的特征还在于存在朗格汉斯细胞(免疫系统的真正卫士)和充满神经肽的梅克尔细胞(其作用尚不被认识)(“Lavierévélée du follicule de cheveu humain”中的综述;Bernard BA,Medecine Sciences Vol 22(2),2006)。
几年来,本申请者已进行了许多临床试验,从而能够确定与年老的头发相关的症状。这些症状特别是纤维外观的变化(纤细的、灰暗的、不直顺的(柔曲的(mou))、无光泽的、花白的头发),即易脱落的、更新变慢的头发。这些表现起因于带发头皮真皮中的胶原网状物的瓦解;起因于糖化产物的出现(1),所述糖化产物使毛囊周围的真皮变硬,从而对居留在此的毛囊以及新头发的植入产生不利影响;以及起因于皮脂合成的减少,这同样也增加了头发的灰暗和无光泽的外观并且促进头发的脱落(2,3)。

一生拥有健康、强健和大量的头发是大多数男人和女人的愿望。有许多用于处理头发的产品可用。然而,至今还没有任何用于口服途径的组合物使得能够对抗随年龄而出现的头皮和/或头发的改变,以及使得能够保护“头发资本”。
本申请者已经证明,单不饱和脂肪酸可以使得能够对抗衰老的头发的改变。
时序性衰老(vieillissement chronologique)是逐步地并且根据个体而或早或迟地显现出来的一种自然现象。然而,头发上的衰老的表现可以被内源性因素(压力、疾病...)或者外源性因素(例如污染、暴露于UV...)所加速。
单不饱和脂肪酸已被描述用于各种应用,例如干燥皮肤的水化(L′Oréal的EP 0 709 084)。
EP 0 116 439(Suntory)公开了,某些脂肪酸具有抑制头皮细菌菌群的5α-还原酶的强活性,并由此可以在用于治疗头皮屑和头皮痒的局部产品中使用。因此,这些脂肪酸的作用针对头皮的细菌菌群。
US 4,097,604(Oxford Hill)指出,各种脂肪酸的盐针对口腔致病微生物而言是有活性的,因而使得能够减少这些细菌对于牙周变性出现的影响。在这种情况下,这些脂肪酸的作用局限于口腔菌群。
EP 0 355 842(Sansho Seiyaku)描述了用于预防由黑色素的过度产生所引起的色素沉着的护理霜剂,所述霜剂可以包含岩芹酸。
EP 1 013 178(Unilever)描述了岩芹酸作为抗炎剂和用于治疗衰老的皮肤征候(皱纹、松弛的皮肤、老年斑)的用途。
EP 888 773(NESTLE)描述了包含岩芹酸的组合物,其用于治疗或预防炎症和/或调节浅表组织中的脂质。然而,既没有描述,也没有暗示岩芹酸用于对抗随年龄而出现的头皮和/或头发的改变的美容用途。
此外,这些文献中没有一个预测了单不饱和脂肪酸对于毛发纤维的质量(qualité),从而对于头发的质量的活性。
因此,本发明涉及至少一种单不饱和脂肪酸、其盐或其酯通过口服途径的用途,尤其是美容用途,所述用途为用于预防和/或治疗随年龄而出现的头皮(带发头皮)和/或头发的改变。
根据一个具体的实施方案,“随年龄而出现的头皮和/或头发的改变”,应被理解为与衰老过程相关的改变,尤其是不由炎症过程介导的改变。特别地,所述改变应被理解为仅与衰老过程(不论是时序性衰老类型还是光衰老类型的)相关的改变。
作为在本发明中未被考虑的炎症过程,尤其可提及由感染、变态反应、自身免疫疾病、暴露于刺激剂所导致的那些。
因此,根据一个具体的实施方案,本发明涉及至少一种单不饱和脂肪酸、其盐和/或其酯,尤其是岩芹酸,通过口服途径的用途,尤其是美容用途,所述用途为用于预防和/或治疗头皮和/或头发的改变,所述头皮和/或头发的改变随年龄而出现,并且不由炎症过程所介导,特别是不由通过暴露于刺激剂而诱导的炎症过程所介导。
应当理解,在本发明的范围内,“通过口服途径的美容用途”涵盖了通过口服途径施用的产品的用途,这些产品为例如食品补充剂或功能性食品的形式,从而在美学或舒适方面在头皮和/或头发的水平上产生效应,或者用于美丽目的,例如为了进行保护、维持处于良好的状态、修饰外观、和尤其是美化。
头皮的改变,特别是随年龄而出现的头皮的改变,尤其为在毛囊周围的组织的纤维化。
事实上,本申请者已经证明,单不饱和脂肪酸导致:
-胶原网状物的重建,其尤其是在补充了芫荽油的个体的皮肤切片上通过在Masson三色染色法中着色而可见到,这是因为,该油的作用是减少胶原酶和弹性蛋白酶的活性(通过酶测定法来测量);
-在真皮和表皮中所发现的糖化蛋白质的减少(羧甲基赖氨酸衍生物的减少);
-应激和炎症的标志物(例如Hsp70和TNF-α,其测量可以通过使用特异性抗体来进行)的减少,而毛囊周围组织的氧化应激导致头发的改变,甚至是其脱落。
它们还防止组织酶(例如胶原酶、弹性蛋白酶和明胶酶)对真皮的改变,并因此导致一种皮肤剖面,特别是健康、没有纤维化、有利于良好质量的头发生长这样的头皮的皮肤剖面。
使用至少一种单不饱和脂肪酸、其盐和/或其酯还使得能够限制头发的脱落和/或增加头发的密度。
根据在给定的带发头皮面积上的头发数目来确定头发的密度。
根据一个实施方案,本发明的目标用途旨在用于对抗衰老的头发的改变。其还旨在保护衰老的头发。
特别地,根据本发明的用途使得能够预防和/或限制纤细的和/或灰暗的和/或易断的和/或柔曲的(mou)头发的形成。
根据本发明的用途还使得能够改善角蛋白纤维的质量,尤其是通过促进光亮的和/或浓密的和/或强健的头发的生长。
在本发明中,“单不饱和脂肪酸”意指包含单一双键的脂肪酸。
更特别地,它涉及长链脂肪酸。适合于本发明的单不饱和脂肪酸为具有12至22个碳原子的单不饱和脂肪酸。
所述单不饱和脂肪酸可以为酸或盐的形式,或者为衍生物(尤其是脂肪酸的酯或酰胺)的形式。
当它们以盐的形式存在时,所述单不饱和脂肪酸更特别地可以是美容上可接受的盐,即无机盐,例如铵盐,或者碱金属(锂、钾、钠)、碱土金属(镁、钙)的盐,或者铝盐。特别地,所述单不饱和脂肪酸以钙盐的形式存在。
所述脂肪酸也可以以酯的形式存在。因而,它们可以与甘油(单酰基、二酰基、或三酰基)、醇(例如甲醇和乙醇)、糖、生育酚、生育三烯酚、固醇或脂肪醇进行酯化。
作为单不饱和脂肪酸,可提及岩芹酸(或C18 δ-6-顺-十八烯酸)、棕榈油酸(或C16顺-9-十六烯酸)或油酸(C18顺-9-十八烯酸)。
应当理解,通过考虑包含它们的组合物的最终目的,和尤其是组合物的施用方式(口服或通过呼吸道)来选择脂肪酸。“呼吸道”意指上呼吸道(例如鼻前庭、窦、口)。
在单不饱和脂肪酸之中,特别地使用油酸和岩芹酸。岩芹酸十分特别地适合于本发明。
根据本发明的一个变化形式,以分离的形式(即在从它们的原始来源中提取出来之后)来使用单不饱和脂肪酸。
根据另一个变化形式,单不饱和脂肪酸来自植物提取物例如油。
因此,本发明尤其涉及富含单不饱和脂肪酸的油的用途。
尤其可以使用富含岩芹酸的油,所述油更特别地选自具伞形花序植物(ombellifère)的油。
“富含岩芹酸的油”意指包含至少40%的岩芹酸的油。
具伞形花序植物为其花排列成伞形花序的植物,特别地富含岩芹酸的种类为伞形科(Umbelliferae或Apiaceae)和五加科(Araliaceae)。Thapsia属的植物也是岩芹酸的来源(Avato等人,Lipids,2001,36,845)。用于本发明的优选的种类为芫荽(coriandre)、细叶芹(cerfeuil)、胡萝卜(carotte)、芹菜(céleri)、枯茗(cumin)、蒿(carvi)、欧芹(persil)和莳萝(aneth)。根据本发明使用的具伞形花序植物的油可以是这些具伞形花序植物的种子的提取物,例如通过研磨或压榨,然后精炼而获得的油。具伞形花序植物的油的岩芹酸含量根据从中提取出该油的具伞形花序植物的种子而变化。对于相同的具伞形花序植物,岩芹酸的含量也根据具伞形花序植物的原产地和根据较完全或较不完全的提取而变化。
岩芹酸还是血红老鹳草(Gernium sanguneum)种子油的一种丰富的组分(约48%)(Tsevegsuren等人,Lipids,2004,39,571)。
因此,根据一个实施方案,本发明的目标用途是这样的,即以具伞形花序植物或血红老鹳草的油的形式来使用岩芹酸。
根据一个实施方案,根据本发明的组合物是这样的,即单不饱和脂肪酸、其盐和/或其酯的含量为相对于组合物总重量而言0.0001至30重量%。
在根据本发明可使用的用于口服途径的组合物中单不饱和脂肪酸或其衍生物的含量将是这样的,即所述单不饱和脂肪酸、其盐和/或其酯的日剂量为0.5至2500mg/天,尤其是10至1200mg/天,特别地5至500mg/天。
根据本发明的一个具体实施方案,在可用于口服途径的组合物中单不饱和脂肪酸的含量是这样的,即日剂量为例如5至50mg/天,或150至1200mg/天,和尤其是150至600mg/天。
优选地,根据本发明可使用的口服组合物具有可食用的载体。
为了摄取,口服组合物的许多实现形式,尤其是食品补充剂均是可能的。其配制通过用于生产糖衣丸剂、明胶胶囊(gélule)、凝胶剂、乳剂、片剂、胶囊(capsule)的常用方法来进行。特别地,可以将根据本发明的活性物质掺入到任何其他形式的食品补充剂或强化食品,例如压块食品(barres alimentaires)、或者压缩或未压缩的粉状物中。可以将所述粉状物稀释在水、苏打水、乳制品或大豆衍生物中,或者掺入到压块食品中。
尤其适合于作为食品或药物的载体的有:奶、酸乳酪(yaourt)、干酪、发酵乳、基于乳的发酵产品、冰淇淋、基于发酵谷物的产品、基于乳的粉状物、用于儿童和婴儿的配方、糖果类型的食品、巧克力、谷物、用于动物(特别是家畜)的饲料、片剂、明胶胶囊或锭剂(tablette)、干燥形式的口服补充剂。
当然,根据本发明的口服组合物此外还可以包含多种其他活性物质。
这些活性物质可以选自维生素B1、B3、B5、B6、B8、B12、C、D、E或PP,烟酸,类胡萝卜素,多酚和矿物质,氨基酸,儿茶素类,寡聚原花色素,含硫氨基酸,ω-3和ω-6多不饱和脂肪酸,和明胶。
根据一个具体的实施方案,这些活性物质可以选自使得能够对抗头皮的衰老和改善头发的外观的那些,特别地,可以使用牛磺酸、维生素B6、半胱氨酸、维生素B1(生物素)、维生素B12(泛醇)和明胶。
此外,口服组合物还可以包含抗氧化剂复合物,包括维生素C和维生素E,以及至少一种类胡萝卜素(尤其是选自β-胡萝卜素、番茄红素、虾青素、玉米黄素和叶黄素的类胡萝卜素),黄酮类化合物(例如儿茶素类),橙皮苷,原花色素(尤其是以黑茶藨子籽油的形式),和花色素苷。
此外,口服组合物还可以包含至少一种益生菌、至少一种益生元或者益生菌的混合物和益生元的混合物。作为益生微生物,尤其可以提及约氏乳杆菌(Lactobacillus johnsonii)或类干酪乳杆菌(Lactobacillus paracasei)。
根据另一个方面,本发明的目标在于用于改善头发的质量或毛发纤维的质量的美容方法,其包括至少一个摄入口服组合物的步骤,所述口服组合物在生理学上可接受的载体中包含至少一种单不饱和脂肪酸和/或其酯和/或其盐。
优选地,所述单不饱和脂肪酸为岩芹酸。
本发明的美容方法可以包括单次施用。根据另一个实施方案,在一天或更多天内,和通常在至少4周,甚至4至15周的延长的时间段内,每天重复所述施用例如2至3次,其中在需要时,可以有一个或多个中断期。
根据另外一个方面,本发明的目标在于至少一种单不饱和脂肪酸、其盐或其酯在制备用于治疗和/或预防头皮和/或头发,特别是衰老的头皮和/或头发的病理学状态的美容或皮肤病学组合物中的用途。
根据另外一个方面,本发明涉及用于治疗和/或预防头皮和/或头发,特别是衰老的头皮和/或头发的病理学状态的单不饱和脂肪酸、其盐或其酯。

实施例10:岩芹酸对于真皮细胞中糖化产物形成的影响的体外研究
在下面的试验中,本申请者证明了,岩芹酸使得能够在真皮成纤维细胞培养物中显著地减少糖化产物的形成。
真皮成纤维细胞根据本领域技术人员熟知的常用技术来进行培养。
以相对的方式,相对于对照(成纤维细胞培养物,未添加产品)而言,通过糖化产物的免疫检测斑点的光密度测定分析(斑点印迹法)来确定糖化产物的定量。
在一种情况下,向成纤维细胞培养物补充作为参照产品的氨基胍,这是因为在该试验的实验条件下,其对于糖化产物的量的显著减少具有作用。如在下面的表1中所显示的那样,检测到相对于对照而言形成42%的糖化产物。因此,相对于对照而言,氨基胍使糖化产物的量减少了58%。因此,该结果使得能够证明该试验的实验条件是有效的。
在另一种情况下,向成纤维细胞培养物补充岩芹酸。如在下面的表1中所显示的那样,检测到相对于对照而言形成73%的糖化产物,从而表明相对于对照而言糖化产物形成的显著减少(27%)。

1.至少一种单不饱和脂肪酸、其盐或其酯通过口服途径的美容用途,所述用途为用于预防和/或治疗随年龄而出现的头皮和/或头发的改变。
2.根据权利要求1的用途,所述用途为用于预防和/或治疗在毛囊周围的组织的纤维化。
3.根据权利要求1或2中任一项的用途,所述用途为用于限制头发的脱落和/或增加头发的密度。
4.根据权利要求1的用途,所述用途为用于保护衰老的头发。
5.根据前一项权利要求的用途,所述用途为用于预防和/或限制纤细的和/或灰暗的和/或易断的和/或柔曲的头发的形成。
6.根据权利要求1的用途,所述用途为用于改善角蛋白纤维的质量。
7.根据前一项权利要求的用途,所述用途为用于促进光亮的和/或浓密的和/或强健的头发的生长。
8.根据前述权利要求中任一项的用途,其中所述单不饱和脂肪酸以分离的形式或者以在植物提取物例如油中的形式进行使用。

15.用于改善头发的质量和毛发纤维的质量的美容方法,其包括摄入口服组合物,所述口服组合物包含至少一种单不饱和脂肪酸和/或其盐和/或其酯。
16.根据权利要求15的方法,其中所述单不饱和脂肪酸为岩芹酸。
