发明背景
生物和化学武器的扩散加强了对能够长时间监测环境的小型、快速、灵敏的生物战剂探测器的需要。在战场条件下,有用的探测器在探测到特定生物或化学战剂时可以迅速给士兵发出警报,从而可以快速实施应对措施。这些探测器也可以用于非军事领域。快速检测患者中的抗生素抗性细菌将会帮助医生选择更有效的治疗方案。连续监测城市饮用水供应可提供对潜在病原体的早期预警,使公共工作部门有更多的时间处理这种对公众的潜在健康威胁。另外,在肉类和禽类检验中使用这样的探测器将比现有的“刺-嗅”法具有显著的改进。通常,在医学(例如兽医学)、农业、环境保护(例如诊断不良建筑物综合征)和食品加工或管理领域的分析和诊断应用中非常需要这样的探测器。

发明内容
本发明提供了当细胞在可能导致细胞存活力降低或丧失的环境温度或非冷藏温度下保存时保持细胞存活力的方法。
在一个实施方案中,细胞保存技术用于发射细胞(在此也被称为CANARY细胞或传感细胞)。在一个具体实施方案中,发射细胞表达目标粒子的一种或多种受体。在一个实施方案中,所述受体是一种抗体。在另外一个实施方案中,所述受体是Fc受体。在一个进一步的实施方案中,发射细胞是B细胞、巨噬细胞或成纤维细胞。
在再进一步的实施方案中,发射细胞表达发射分子。在一个具体实施方案中,发射分子响应细胞内钙的增加而发出光子。在一个进一步的实施方案中,发射分子是水母发光蛋白(aequorin)。
在一个进一步的实施方案中,细胞保存方法包括用一种或多种胱天蛋白酶(caspase)抑制剂、蛋白水解酶抑制剂和/或蛋白体(proteosome)抑制剂的组合保护细胞(如发射细胞)以阻止其凋亡、氧化和/或蛋白质降解。
在另外一个实施方案中,细胞保存方法另外或者可替代地包括增强负责诱导和/或保持静止的基因在细胞中的表达,从而延长细胞保持静止状态的时间。

发明详述
尽管哺乳动物细胞可以在-80℃或更低的温度下冻存,但是众所周知当它们以液体状态在冷藏温度或环境温度下保存时在数周内丧失存活力。此外,以干燥状态长时间保存细胞而同时保持其存活力的技术仍在发展中。改善活细胞干燥保存的几种技术涉及在细胞内或细胞外使用非还原二糖,如海藻糖,但是它们只获得有限的成功[Nat.Biotechnol.,2000,18(2):168-171;FEBS Lett.,2000,487:199-202;J.Opthalmol.,85:610-612(2001);Cryobiology,42(3):207-217(2001);J.Physiol.,558:181-191(2004)]。最近通过诱导静止状态,同时保持一定水平的含水量并且减少可利用的氧和静电,实现了人类细胞系在环境温度下的6周保存[Jack等人,J.Cell.Physiol.,206(2):526-536(2005)]。通过使细胞在被称为球状体的三维结构中生长来诱导静止状态,认为这种静止状态提供了阻止凋亡和其它形式的细胞死亡的保护。
本发明使用一种或多种以下细胞保存技术改进了上述技术,该细胞保存技术包括:(1)与胱天蛋白酶抑制剂(例如,Z-AEVD-FMK、Z-DEVD-FMK、Z-LEED-FMK、Z-LEHD-FMK、Z-WEHD-FMK、Z-VAD-FMK、Z-VDVAD-FMK、Z-YVAD-FMK、Z-VEID-FMK、Z-IETD-FMK、OPH-109、和/或广谱胱天蛋白酶III抑制剂)、抗氧化剂(例如,亚硒酸钠、N-乙酰半胱氨酸、去铁胺、氨基胍、Trolox、苯并硒唑酮(ebselen)、和/或Tempol)、和/或蛋白水解酶抑制剂和/或蛋白体抑制剂(例如,AEBSF、抑酶肽(aprotonin)、苯丁抑制素(bestatin)、钙蛋白酶抑制蛋白(calpeptin)、组织蛋白酶L抑制剂、E-64、亮抑酶肽、和/或胃酶抑素A)组合,提供阻止凋亡、氧化和/或蛋白质降解的进一步保护;(2)增强负责诱导和保持静止的基因的表达,以延长细胞保持静止状态的时间(例如,可以使用本领域技术人员公知的技术通过转录分析来鉴定这些基因)。增强的基因表达可以用本领域公知的任何手段来实现,例如,利用强启动子在细胞中过量表达一种或多种基因;(3)控制调节凋亡的基因的表达,例如,采用遗传学方法,如过量表达凋亡抑制蛋白(IAP)如XIAP;(4)将条件扩展到包括在环境温度和冷藏温度下在液体中长期保存;和/或(5)利用细胞周期抑制剂(例如,放线菌素D、丝裂霉素C、雷帕霉素、美伐他汀、衣霉素和/或渥曼青霉素)诱导保护性静止状态。如本文使用的“长期保存”是指超过大约10年、超过大约1年、超过大约9个月、超过大约6个月、超过大约3个月、超过大约2个月、超过大约1个月、超过大约2周、超过大约1周、或者超过大约72小时的时间段。
此外,如本文所述,本发明提供了在环境温度、冷藏温度或非冷藏温度下以液体、半液体或干燥状态保存细胞如发射细胞的方法。如本文使用的环境温度或非冷藏温度的范围是从大约5℃至大约50℃、从大约10℃至大约40℃、从大约15℃至大约30℃、以及从大约20℃至大约30℃。
在一个具体实施方案中,对发射细胞(在本文中也被称为CANARY细胞或传感细胞)应用一种或多种细胞保存技术。在此处使用时,发射细胞用于检测目标粒子(如生物抗原、可溶性抗原、核酸、毒素、化学物质等)。目标粒子的检测在过去是通过将目标粒子与发射细胞表面上表达的受体直接或间接结合来进行的。直接结合可以通过直接与目标粒子特异性结合的受体例如抗体来进行。目标粒子的间接结合可以通过(例如)与附着(例如结合)到目标粒子上的抗体结合的Fc受体来进行。抗原与受体的结合导致钙浓度升高。发射细胞在它们的细胞溶质中也含有一种或多种发射分子(例如水母发光蛋白),该发射分子响应细胞溶质中钙浓度的升高而发出光子。光子发射可以被探测到,由此检测目标粒子的存在。这在本文中也被称为“CANARY测定”。关于发射细胞、使用发射细胞的光电检测系统和CANARY测定的进一步的信息,参见,例如,美国专利序号11/001,583、美国专利申请No.2004/0121402、美国专利6,087,114和美国专利6,248,542,它们的内容都全部引入本文作为参考。
一种替代方法是遗传修饰耐干燥的极端嗜极微生物,用作CANARY细胞。例如,已知衣藻在其局部生活环境因缺少降水而干燥时形成孢子。据报道这些孢子再水合的存活力超过孢子形成后70年。此外,还已知衣藻具有类似于B细胞的细胞表面受体活化的钙信号转导级联。因此,可以遗传工程改造一种嗜极微生物,使其表达一种或多种受体,如抗体或Fc受体,和发射分子,如水母发光蛋白。当细胞表面上的受体被病原体或抗原交联时,它的表达刺激信号级联,引起细胞内钙浓度升高,而这种升高的细胞内钙离子浓度刺激发射分子发光,可以测量这种发光,并且与目标抗原或病原体的存在相关联。因此,嗜极微生物发射细胞可以在例如环境温度、冷藏温度或非冷藏温度下以脱水状态保存延长的时间(例如长期保存),并且在再水合后保持存活,例如可用于CANARY测定。
实施例
实施例1
衣霉素可提高CANARYB细胞的细胞存活力和活性(见图2)。在CANARY B细胞(表达鼠疫耶尔森氏菌特异性受体)与衣霉素在室温下一起保存后,CANARY B细胞保持对鼠疫耶尔森氏菌的检测灵敏性(见图2)。
此外,用渥曼青霉素处理CANARY细胞也提高了冷藏的CANARY细胞的存活力和抗原检测活性。
实施例2
与不使用渥曼青霉素在室温下保存1、2、3周的细胞相比,用广谱胱天蛋白酶III抑制剂(在图3的条图中被称为"胱天蛋白酶III")处理细胞提高了在室温下保存1、2、3周的细胞的存活力(见图3,条图)。
实施例3
证明了细胞保存保障(Logistics)的改善
已证明CANARY B细胞生物制剂传感器技术是任何已知生物制剂鉴定技术的速度和灵敏性的最佳结合[Rider等人,Science 301:213(2003)]。CANARY可以检测多种基质和形式(包括空气、食物、表面和医学样品)中的病原体。然而,仍然需要对B细胞本身的容易并且便宜的保存和保障。细胞一般冷冻或冷藏保存到准备使用前,对于许多医学和栖息环境来说这是可接受的(尽管不是最佳的),但不是对所有情况都理想的,例如对于向前展开的军事单位来说。另外,大多数使用者希望有一种发射细胞(例如B细胞)试剂盒,其可以在环境温度下在仓库或实验室架子上放置大约6个月至大约10年或者更长时间,并且可以取出、装入传感器中,用于进行检测。这些试剂盒包含,例如,发射细胞和任选的在测定如CANARY测定中使用发射细胞的说明书。
在申请人的本发明之前,CANARY细胞试剂的搁置保存期为室温下2天和4℃下2周。细胞可以在-80℃或更低的温度下无限期地冷冻保存,但是这需要液氮或特殊的冰箱,不是所有的实验室都能够拥有。本文描述了可提高在室温(例如环境温度)和/或4℃下长期保存的细胞存活力和活性的多种处理、添加剂和基因过量表达的实例。
方法与材料
研究的化学添加剂是:胱天蛋白酶抑制剂AEVD、DEVD、LEED、LEHD(在50μM时具有毒性)、WEHD(在50μM时具有毒性)、VAD、VDVAD、YVAD、VEID、IETD(R&D Systems)、OPH-109(MPBiomedicals)、胱天蛋白酶抑制剂III(Calbiochem);蛋白水解酶抑制剂AEBSF、抑酶肽、苯丁抑制素、钙蛋白酶抑制蛋白、组织蛋白酶L抑制剂、E-64、亮抑酶肽、胃酶抑素A(Sigma);细胞周期抑制剂雷帕霉素(LC Labs)、放线菌素D、丝裂霉素C、美伐他汀、衣霉素、渥曼青霉素(Sigma);抗氧化剂亚硒酸钠、N-乙酰半胱氨酸、去铁胺、氨基胍、Trolox、苯并硒唑酮、Tempol(Sigma)、MnTBAP(A.G.Scientific)。
为了在4℃下保存,将细胞以2.5-3×105细胞/mL的密度在1-3μM渥曼青霉素中37℃孵育24小时,然后以5×105细胞/mL的浓度在2%DMSO中37℃孵育24小时。然后将细胞在含有50μM腔肠素(coelenterazine)(Molecular Probes,Eugene,OR)的测定培养基[不依赖于CO2的培养基,10%胎牛血清,50μg/ml链霉素,50U/ml青霉素和250ng/mL两性霉素B(Life Technologies)]中在暗处室温孵育2小时。然后将细胞洗涤两次,以5×105细胞/mL的终浓度重悬浮在添加有下列成分的测定培养基中:100μM Z-LEED-FMK,50μg/mL N-乙酰半胱氨酸,150ng/mL亚硒酸钠,15μM去铁胺(新鲜制备的贮存液),1.5mM氨基胍(新鲜制备的贮存液),使其在室温下旋转过夜。
为了在室温下保存,将细胞以2.5-3×105细胞/mL的密度在0.1μg/mL衣霉素中37℃孵育24小时,然后以5×105细胞/mL的浓度在2%DMSO中37℃孵育24小时。然后将细胞在含有50μM腔肠素的测定培养基中在暗处室温孵育2小时,洗涤两次,以5×105细胞/mL的终浓度重悬浮在含有100μM胱天蛋白酶抑制剂III的测定培养基中。
使用Magic Vac密封机将细胞管真空密封在食物保存袋中,并且通过将细胞保存在空间没有充满的15mL和50mL管中提供顶部空间。用抗CD40刺激的细胞以2×105细胞/mL的浓度与抗体(BDBiosciences)在37℃下孵育24小时,然后如上所述用2% DMSO处理并且在腔肠素中孵育。热休克条件为42℃下2-5小时。通过用驱动荧火虫萤光素酶和花虫萤光素酶质粒(作为转染率的对照)表达的Hsp70-反应性启动子转染,测定热休克反应。通过电穿孔法转染细胞,使其恢复24-48小时,进行热休克,次日用双萤光素酶报道测定法(DualLuciferase Reporter Assay)(Promega)进行测定。
在500μg/mL潮霉素(Invitrogen)中筛选用卤虫p26和pcDNA3.1质粒中的artemin转染的细胞。通过RT-PCR克隆鼠GADD45β,将其插入pEF6/V5/His-TOPO(Invitrogen)中。在5μg/mL杀稻瘟素(Blasticidin)(Invitrogen)中筛选用该质粒转染的细胞。通过PCR向人Bcl-XL基因(Invivogen)中添加限制性酶切位点,将其插入pcDNA3.1(Invitrogen)的NheI和XhoI位点中,并且证实其序列。在保存期间虽过量表达但不提供保护的其它基因包括卤虫p22和p21(两种形式,一种在氨基酸位点62处具有G,另一种具有E)、BiP、XIAP、杆状病毒p35、和显示组成型活性的HSF1的缺失突变体(氨基酸221-315)[Voellmy,Methods35:199(2005)]。
在270V,950μP条件下通过电穿孔(BioRad)转染细胞,并通过有限稀释法进行克隆。使用基因表达测定法(AppliedBiosystems)通过定量RT-PCR分析克隆的表达。选择几个具有中度和高度表达的克隆进行随后的保存实验。通过在添加2% DMSO的生长培养基中以5×105细胞/mL的浓度孵育B细胞,准备B细胞用于发光测定。20-24小时后,将细胞在含有50μM腔肠素(Molecular Probes,Eugene,OR)的测定培养基[不依赖于CO2的培养基,10%胎牛血清,50μg/ml链霉素,50U/ml青霉素和250ng/mL两性霉素B(LifeTechnologies)]中在暗处室温孵育2小时。然后将细胞洗涤两次,以5×105细胞/mL的终浓度重悬浮在1.5mL微量离心管中的测定培养基中,在室温下旋转过夜。准备后,将细胞保存在1.5mL微量离心管中的测定培养基中。
结果
检测在准备测定和/或保存过程中向细胞中加入化学物质提高细胞在4℃或室温下保存时的活性和/或存活力的情况。化学物质选自以下类别:凋亡抑制剂、抗氧化剂、细胞周期抑制剂、和蛋白水解酶抑制剂。完全清单在“方法与材料”部分中给出,见上文。每种添加剂都以不同浓度应用。在保存前以及在保存期间以1周间隔评价存活力并且检测活性。然后将确定为有益的添加剂在进一步的实验中互相组合。
通常,在一种保存温度下有益的处理在另外一种保存温度下不一定具有益处,而且在一种情况下(在4℃下保存的衣霉素处理的细胞)是有害的。当细胞在准备用于CANARY测定之前与渥曼青霉素一起孵育,并且在胱天蛋白酶抑制剂Z-LEED-FMK和抗氧化剂N-乙酰半胱氨酸、亚硒酸钠、去铁胺和/或氨基胍的存在下保存时,发现了在4℃下保存的细胞的存活力和活性提高。该处理使细胞在4℃下保存而不丧失活性的时间从2周延长到3周。当独立应用时,发现渥曼青霉素处理提高了存活力和活性,而在Z-LEED-FMK和抗氧化剂中保存仅提高了活性。类似地,发现当细胞在准备用于测定之前与衣霉素一起孵育,并且在胱天蛋白酶抑制剂III的存在下保存时,在室温下保存的细胞的存活力和活性提高。该处理使细胞在室温下保存而不丧失活性的时间从2天延长到1周。当独立应用时,衣霉素处理和在胱天蛋白酶抑制剂III中保存都提高了存活力和活性。使细胞活性提高的其他处理包括真空密封、利用顶部空间保存细胞、以及用抗CD40抗血清刺激细胞。然而,在这些处理后保存的细胞的应答是不一致的,图6所示的结果代表在一半以上、但不超过75%的实验中发生的结果。
还研究了某些基因的过量表达在保存期间的保护性益处。生长抑制和DNA损伤诱导β(GADD45β)基因在HEK293细胞保存期间显著地转录上调,这些细胞在室温保存6周后仍然存活[Jack等人,J.CellPhysiol.206:526(2006)]。类似地,CANARYB细胞也在准备用于测定后上调GADD45β,并且在整个保存期间保持该水平(见图7)。已经证明来自旧金山湾卤虫的热休克蛋白在哺乳动物细胞中表达时提供对应激的抗性[参见,例如,Ma等人,Cryobiol.51:15(2005);Villeneuve等人,Cell Stress Chaperones 11:71(2006);Collins和Clegg,Cell Biol.Int.28:449(2004)]。GADD45β和卤虫p26和artemin的过量表达提高了在室温下保存的B细胞的活性,使保存时间延长到1周而不丧失活性(见图8)。通常,表达artemin的细胞的活性好于表达p26或过量表达GADD45β的细胞的活性。Bcl-XL(一种抗凋亡蛋白)的过量表达提高了在4℃下保存的细胞的存活力和活性,使最长保存时间从2周延长到4周(参见图9)。在少数实验中,这些细胞在长达6周时间里保持完全活性。当对任意过量表达的细胞系应用上述添加剂和处理时,没有观察到益处。
讨论
CANARY提供了其它方法无法匹敌的速度和灵敏性的结合。然而,活细胞试剂通常具有室温下2天和4℃下2周的有限的保存期。试剂(即CANARY细胞)可以在-80℃或更低的温度下冻存,但是这需要液氮或特殊的冰箱。将保存的细胞与新鲜制备的细胞进行比较的实验表明,保存期间活性的丧失不仅是由于存活力的降低,也是由于每个细胞响应抗原的发光量的降低(见图10)。因此申请人在寻找随时间延长保持细胞应答性和存活力的方法。申请人发现,胱天蛋白酶抑制剂和抗氧化剂都是有益的,这可能是因为胱天蛋白酶抑制剂保持存活力而抗氧化剂保持总体细胞健康或水母发光蛋白、腔肠素的辅因子的氧化状态。
HEK293细胞可以在室温下保存长达6周,而具有良好的存活性,静止状态对于它们的存活是关键性的[Jack等人,J.Cell Physiol.206:526(2006)]。为了诱导静止状态,用阻滞细胞周期的化学物质处理CANARY细胞。有意思的是,尽管证明用渥曼青霉素和衣霉素处理细胞具有有益效果,但这不是在抑制细胞生长的浓度上。
在多种应激条件下诱导热休克应答,其代表为保护细胞避免非折叠中间物聚集和沉淀的蛋白质,包括分子伴侣和蛋白水解酶[Winter和Jakob,Crit.Rev.Biochem.Mo l.Biol.39:297(2004);Mosser等人,Mol.CellBiol.17:5317(1997)]。发现卤虫热休克基因p26和artemin的表达保持在室温下保存的CANARY细胞的活性。
GADD45β在细胞周期调节和凋亡中都起作用。它是一种抗凋亡蛋白质,由B细胞的CD40刺激诱导,并且介导Fas-诱导的凋亡的抑制。它也被描述为应激应答基因,GADD45β的表达在用DMSO处理后的CANARY细胞中上调,申请人发现该DMSO处理步骤有助于确保一致的活性。GADD45β不是唯一的在DMSO处理后的CANARY细胞中显示表达变化的DNA损伤相关基因。GADD45β和组蛋白1H1c(Hist1H1c)的表达提高,而淋巴特异性解旋酶(Hells)的表达降低(见图7)。然而,GADD45β的上调在HEK293细胞中更显著,该细胞在保存后也显示更好的存活力。通过诱导该基因的更高表达,我们能够证明在室温下保存的CANARY细胞的活性提高。
尽管未能证明凋亡是引起保存期间活细胞损失的机制,但是有几条证据表明它们可能至少部分相关。第一,存在至少两种可提高保存细胞的存活力和活性的凋亡抑制剂。第二,转录分析揭示有几种凋亡促进因子如Bcl2-样11(Bcl211l)、程序性细胞死亡1配体1(Pdcd11gl)和Bcl2修饰因子(Bmf)上调(见图7)。已经证明Bmf在物理上与抗凋亡家族的成员(Bcl2、Bcl-w、Mcl-1和Bcl-XL)结合,并且认为Bmf抑制所述成员隔离及灭活促凋亡蛋白的能力,从而解除对凋亡途径的抑制。而且,Bmf在哺乳动物细胞系中的过量表达诱导了凋亡,这一效应可以通过过量表达上述抗凋亡蛋白来克服。申请人已经证明过量表达Bcl-XL的细胞在4℃保存后存活力和活性均提高。有意思的是,其他人已经证明p35的表达也逆转了Bmf过量表达的效果,但是当它在CANARY细胞中表达时没有提供明显的益处。
另外一个发现是,尽管在CANARY细胞中过量表达的许多基因没有可观察到的效果,但是XIAP的过量表达完全抑制了细胞对抗原的应答。这不可能是由于意料之外的插入效应,因为它在转染人类和鼠XIAP时,以及在检测的所有克隆中都发生。
这些结果表明,活细胞试剂如发射细胞的保存期可以延长。可抑制凋亡或用作分子伴侣的基因的过量表达提供了阻止细胞死亡的保护,并且也许提供阻止细胞成分降解的保护,并且使CANARY细胞的保存期延长至室温下1周和4℃下至少大约4周。
已经参照本发明的优选实施方案具体说明和描述了本发明,但是本领域技术人员应当理解,在不背离所附权利要求书包括的本发明范围的情况下可以在形式和细节上进行各种变化。
