Abstract
本发明公开了一种药物组合物,对具有HLA-A24或A3超型的患者治疗HCV相关疾病,所述药物组合物含有肽作为有效成分,所述肽具有以下的序列:YLLPRRGPRL(序列号1)表示的氨基酸序列。本发明还提供一种组合物,用于检查具有HLA-A24或A3超型限制性细胞毒性T细胞的诱导,所述组合物含有肽为有效成分,所述肽具有以序列号1表示的氨基酸序列。
Description
技术领域
本发明涉及治疗与C型肝炎病毒相关的疾病。
背景技术
C型肝炎病毒(HCV)是属于黄病毒科的单链RNA病毒,估计世界人口的约3%是C型肝炎病毒(HCV)血清反应阳性(World HealthOrganization.Hepatitis C:global prevalence.Wkly.Epidemiol.Rec.1997,72(46):341-344)。HCV感染的急性期经常是无症状的,但是血清反应阳性的个体约70%引发慢性感染,感染的患者易罹患进行性肝脏疾病,例如慢性肝炎、肝硬化及肝细胞癌(Alter HJ,SeeffLB.,Semin LiverDis.2000;20(1):17-35;Shimotohno,K.,Semin.Cancer Biol.2000;10:233-240)。目前,HCV感染的治疗方法是与利巴韦林组合得到的干扰素(IFN)-α,最近开始使用聚乙二醇修饰型IFN-α。但是,只在约50%受治疗的患者中可见持续性应答(Manns MP,et al,Lancet.2001;358:958-965)。更重要的是,抗病毒疗法无法在大部分发展中国家使用,而大部分感染者居住在发展中国家。因此需要开发出新的治疗方法。
HCV感染后的肝细胞损伤的病因机制还未充分阐明,但是大量证据显示,病毒特异性细胞毒性T淋巴细胞(CTL)可能在HCV感染后的肝脏损伤中发挥重要的作用(Chang KM,et al.,Springer SeminImmunopathol.1997;19:57-68)。另一方面,认为CTL对于感染期间控制病毒扩散、排除病毒是有效的(Kurokohchi K,et al.,J Virol.1996;70:232-240)。因此,利用疫苗诱导CTL是控制伴随HCV感染的疾病的有希望的方案。并且,由于成本低以及保存容易,所以希望开发出诱导CTL的肽疫苗。
本发明人等首先发现,从大多数HCV感染者中检测出了与来源于HCV的肽具有反应性的IgG,以及能够由具有与上述肽对应的HLA型的HCV患者的外周血单核细胞(PBMC)诱导肽特异性CTL。进而发现通过给与来源于HCV的肽,在大量HLA-A2的病例中确认肽引起的免疫激活作用和病毒量降低(Takao Y et al.,Microbiol.Immunol.,48(7),507-517,2004(非专利文献1);WO2005/028503(专利文献1);日本专利申请2005-310203(专利文献2))。
但是,由于还存在无法对上述肽诱导CTL的病例,所以在该技术领域中,要求能适用于上述患者的其他肽。特别是由于HLA-A24具有次于HLA-A2的高频率,所以需要能适用于A24的患者的肽。另外,HLA-A11、HLA-A31和HLA-A33以及HLA-A0301和HLA-A6801,具有类似的抗原结合基序(binding motif),因此,被分类为HLA-A3超型(Takedatsu H,et al.,Clin Cancer Res 2004;10:1112-1120:非专利文献2)。至今为止,还没有关于来源于HCV的HLA-A3超型(supertype)限制性抗原肽的报告。

发明内容
本发明目的在于提供一种HCV相关疾病的治疗方法,该方法可以适用于HLA(人白细胞抗原;Human Leukocyte Antigen)-A24及A3超型HCV感染者。
本发明提供一种药物组合物,用于治疗具有HLA-A24或A3超型的患者的HCV相关疾病,所述药物组合物含有肽作为有效成分,所述肽具有用以下的序列:YLLPRRGPRL(序列号1)表示的氨基酸序列。本发明还提供一种组合物,用于检查具有HLA-A24或3超型的患者中的HLA-A24或A3超型限制性细胞毒性T细胞的诱导,所述组合物含有肽作为有效成分,所述肽具有用序列号1表示的氨基酸序列。
本说明书中使用的“HCV相关疾病”表示感染C型肝炎病毒而引起的疾病,例如,包括急性及慢性肝炎、肝硬化及肝细胞癌。HCV相关疾病的进展典型地为由慢性肝炎向肝硬化进展、以及肝硬化向肝癌进展。
本发明中的肽(YLLPRRGPRL(序列号1))来源于HCV核蛋白质序列,在本说明书中也被称为“肽C35-44”。报道了该肽能由HLA-A2阳性者外周血强诱导对排出病毒有效的细胞毒性T细胞(CTL)。本发明中,令人惊奇的是,尽管用现有的方法来计算该肽C35-44的解离常数时,显示与HLA-A24或A11具有极低的结合活性,但是本发明人等发现也能从HLA-A24或A3超型阳性患者中诱导CTL。以HLA-A2阳性者为对象的疫苗仅覆盖约40%日本患者,但是,根据已阐明了能适用于HLA-A24阳性者及A3超型阳性者的本发明,能以大致100%的日本患者作为肽疫苗疗法的对象。因此,即使在患者的HLA型或患者血清中的抗体效价不明确的情况下,也能使用本发明的肽。进而,由于肽C35-44也广泛存在于HCV-1b或HCV-2a等各种基因型的HCV中,所以能适用于大多数HCV感染者。

具体实施方式
本发明提供一种含有来源于免疫原性HCV的肽作为有效成分的疫苗,可以用于治疗感染了HCV的具有HLA-A24、HLA-A11、HLA-A31及HLA-A33的患者。如以下的实施例所示,本发明中,发现肽C35-44由HLA-A24阳性HCV感染者外周血单核细胞(PBMC)诱导了肽特异性CTL。进一步通过与MHC分子的结合试验确认该肽C35-44与HLA-A24结合。另外,具有HCV相关疾病且具有HLA-A11、HLA-A31或HLA-A33的HCV感染血浆中检测出了对肽C35-44具有反应性的IgG抗体。该肽由HLA-A31及A33阳性HCV感染者的PBMC诱导肽特异性CTL。
本发明的来源于C型肝炎病毒的肽(HCV肽)含有来源于C型肝炎病毒的蛋白质的部分氨基酸序列,可以通过C型肝炎病毒感染者的血中抗体来识别。优选本发明的来源于C型肝炎病毒的肽可以诱导CTL,由此被诱导的CTL以HCV感染细胞为目标,攻击该细胞。
本发明的肽只要具有发挥所希望作用效果的程度的上述氨基酸序列即可,还可以含有附加的序列。对本领域技术人员来说,理所当然,为了发挥作为抗原肽所希望的作用效果,其N末端侧及C末端侧可以允许分别附加各种程度的序列。因此,例如本发明的肽的N末端侧及/或C末端侧上可以附加对促进免疫致敏有用的氨基酸序列或容易制剂化的氨基酸序列或便于作为融合蛋白质表达肽的氨基酸序列或便于制备及纯化肽的氨基酸序列等。另外,本发明的肽只要不丧失与抗体结合的特异性,就可以被化学修饰,也可以附加聚合物或糖链。
可以通过通常的化学合成法、蛋白质分子的酶分解法、使用转化得到的宿主来表达编码目标氨基酸序列的碱基序列的基因重组技术等来制备本发明的HCV肽。
用化学合成法制备目标肽时,可以通过通常的肽化学中公知的惯用手法来制备,例如,可以使用肽合成机,通过固相合成法来合成。可以通过蛋白质化学中通常使用的纯化方法例如盐析法、超滤法、反相色谱法、离子交换色谱法、亲和色谱法等来纯化由此得到的粗肽。
另一方面,用基因重组技术来生产所希望的肽时,例如可以通过将编码合成或克隆得到的目标氨基酸序列的DNA片段重组到适当的表达载体,使用该表达载体转化微生物或动物细胞,培养所得的转化体,可以得到所希望的肽。作为能使用的表达载体,可以使用本技术领域中公知的质粒、病毒载体等。
在该肽生产技术中,作为使用了表达载体的宿主细胞的转化方法,可以使用其公知的方法,例如氯化钙法、磷酸钙共沉淀法、DEAE葡聚糖法、脂转染(lipofectin)法、电穿孔法等,可以基于所使用的宿主细胞来适当选择。可以通过上述纯化方法对从培养的培养基中回收的细胞提取液或培养上清进行纯化。
本发明的肽作为用于治疗具有HCV相关疾病的患者的药物组合物即肽疫苗是有用的。通过将如上所述制造的肽与医药上允许的辅助剂及/或载体适当混合来调制由来源于C型肝炎病毒的肽组成的疫苗。作为辅助剂,可以使用能增强免疫应答的佐剂,例如Floint的不完全佐剂、氢氧化铝凝胶等。另外,作为载体,例如可以使用磷酸缓冲生理盐水(PBS)、蒸馏水等稀释剂、生理盐水等。
作为肽疫苗的使用形态,例如可以通过口服或静脉给药或皮下给药等经皮途径给药。作为其剂型,例如可以举出片剂、颗粒剂、软胶囊剂、硬胶囊剂、液体制剂、油剂、乳化剂等。作为上述药物组合物的给药量,可根据给药患者的症状等适当改变,但通常对于成人每1日的肽量优选为0.1-10mg,给药间隔优选为每数日至数月给与1次。另外,可以并用肽疫苗和干扰素或利巴韦林等抗病毒剂。
给与本发明的来源于C型肝炎病毒的肽前,可以测定存在于患者血液中的抗HCV抗体的肽反应性或CTL的存在。此处,所谓“抗HCV抗体的肽反应性”是指应给与的肽被存在于患者血液中的抗HCV抗体识别。
通过用ELISA法等常用方法监测罹患HCV相关疾病的受试者的血中抗体效价,即对肽具有反应性的抗体的血中浓度,从而可以确认HCV相关疾病的治疗效果。另外,可以通过RT-PCR等常用方法来测定HCVRNA的量,从而可以监测病毒量。
本发明的肽作为用于诊断具有HCV相关疾病的患者、或预测疾病进展的组合物也是有用的。可以使用本发明的肽,使用该技术领域中熟知的抗原抗体反应,测定受试者血液中的抗体效价。例如,使用典型的ELISA法时,可以如下所示进行测定。使作为抗原的肽结合在96孔板等惯用的ELISA板中,为了防止非特异性吸附,适当将板进行封闭(blocking)处理。然后,适当稀释由受试者的血液调制得到的血清,加入板的各孔中,反应规定时间。洗涤板,除去未结合成分后,加入能与人抗体结合的抗体(例如兔抗人抗体)。希望检测出IgG时,可以使用γ链特异性抗人IgG。反应规定时间后,洗涤板,加入能检测的被标记的抗体(例如抗兔IgG)。可以使用酶、荧光色素、化学发光物质、生物素、放射线化合物等本领域技术人员熟知的方法进行标记。使板反应规定时间后,加入适当的基质,测定基质的减少或生成物的增加或测定荧光、发光、放射活性来检测标记。由此,可以测定对应于受试者血清中特定肽的抗体量。进而,可以通过监测抗体的量来预测HCV相关疾病的进展。
通过上述抗原抗体反应检测抗体的方法中,作为灵敏度高的方法之一,有xMAP技术。该技术是利用Luminex公司开发的利用荧光微球有序排列体系(Micro Beads Array System)进行的流式细胞仪(flow metry)测定法,使结合肽的微球与血清接触,然后,使其结合荧光标记第二抗体,通过流式细胞仪测定荧光强度。
另外,在医院的检查室等中测定抗体效价时,可以使用免疫色谱法。该方法为,预先制作在试纸上使抗原(或抗体)呈线状分布的部分,使检体中的抗体和被着色粒子标记的抗原的复合体在试纸上移动时,根据有无被抗原(或抗体)集中捕获而出现的带有颜色的线进行定性分析。使用该方法,能用简便的设备在短时间(20~30分钟以内)内得到结果。
具有HLA-A24的HCV感染者中,HLA-A24限制性细胞毒性T细胞是否被诱导可以如下所述检查,即,调制HCV感染者的外周血单核细胞(PBMC),与本发明的肽一起培养后,以IFN-γ产生为指标或用51Cr游离反应研究对脉冲该肽得到的C1R-A24细胞或HLA-A24阳性的HCV感染细胞(株)等的反应性。对HLA-A11、HLA-A31或HLA-A33也同样进行。另外,被诱导的CTL的MHC限制性可以通过用单克隆抗体抑制MHC I类、CD8的试验来确定,肽特异性可以用被对应的肽致敏的被标记目标细胞进行的抑制试验(cold target inhibition试验)来确认。具体而言,可以通过下述实施例1中记载的方法来测定。

以下,通过实施例更详细地说明本发明。但是,下述实施例是为了更具体地说明本发明而列举的实施例,本发明并不限于实施例。
实施例1
利用肽C35-44诱导肽特异性CTL以及肽刺激PBMC的细胞毒活性
受试者
以具有HLA-A24或HLA-A3超型的HCV感染者作为受试者。确认上述受试者均未感染人免疫缺陷病毒(HIV)。为了得到外周血单核细胞(PBMC),采集20ml外周血,通过Ficol-conray密度梯度离心分离调制PBMC。所有试样均低温保存直至用于试验。需要说明的是,实施本试验时,获得久留米大学医疗伦理委员会的事先批准,并且也获得了受试者的书面同意。HCV感染者的PBMC中的HLA-A11、HLA-A31及HLA-A33分子的表达是使用以下抗体通过流式细胞仪测定的:抗HLA-A11单克隆抗体(mAb)(Cat#0284HA:One Lambda Inc.,Canoga,CA),抗HLA-A31mAb(Cat#0273HA;One Lambda)及抗HLA-A33mAb(Cat#0612HA;One Lambda)。
细胞株
使用的细胞株C1R-A24是表达HLA-A24的C1R淋巴瘤的亚系(subline)(Dr.M.Takiguchi,Kumamoto University,Japan)。该细胞保存在补充了10%FCS(牛胎儿血清)及500μg/ml的潮霉素B(GibcoBRL,New York,NY,USA)的RPMI-1640培养基中。另外,细胞株C1R-A11、C1R-A31及C1R-A33是为了分别转染HLA-A1101、HLA-A3101及HLA-A3303基因,稳定地表达该抗原而建立的C1R淋巴瘤的亚系。上述亚系中的HLA-A11、HLA-A31及HLA-A33分子的表达已在之前报道(Takedatsu H,et al.,Clinical Cancer Research 2004;10:1112-20)。上述细胞株维持在含有10%FCS的RPMI1640(Invitrogen)中。肽
使用肽C35-44(YLLPRRGPRL(序列号1))及作为阳性对照的具有HLA-A24结合基序的来源于流感病毒(Flu)的肽(RFYIQMCTEL(序列号2))、来源于EBV的肽(TYGPVFMCL(序列号3)),作为阴性对照的来源于HIV的肽(RYLRDQQLL(序列号4))。作为与HLA-A3超型结合的对照肽,使用来源于流感(Flu)病毒的肽(NVKNLYEKVK(序列号5))、来源于艾伯斯坦-巴尔病毒(EBV)的肽(AVFDRKSDAK(序列号6))、来源于酪氨酸酶相关蛋白质2(TRP2)的肽(LLGPGRPYR(序列号7))及来源于HIV的肽(RLRDLLLIVTR(序列号8))。所有肽均从BIOSYNTHESIS(Lewisville,TX,USA)、SynPep(Dublin,CA,USA)或Biologica Co.(Nagoya,Japan)购得,纯度为90%以上。肽以10mg/ml的浓度溶解在二甲亚砜(DMSO)中,保存在-20℃下。
统计学
使用Student t检验进行统计学分析。P<0.05的值为统计学上有意义。肽特异性IgG的检测
首先,使用感染HCV的12名具有A3超型的受试者血清的1:100稀释试样,测定肽特异性IgG的水平。血浆(或血清)中肽特异性IgG的水平根据前面记载的方法通过Luminex(注册商标)法来测定(Komatsu N.etal.,Scand J Clin Lab Invest,64,535-546,(2004))。简单而言,将200μL血浆(1:100稀释)与25μL肽结合彩色编码珠(color code beads)(Luminex Corp.(Austin,TX))一起在96孔过滤器板(MABVN1250;Millipore Corp.,Bedford,MA)中,用板振荡器在室温下孵育2小时。2小时后,将板用0.05%吐温20-PBS洗涤,与100μL生物素化山羊抗人IgG一起用板振荡器在室温下孵育1小时。然后,洗涤板,将100μL链霉亲和素PE(5μg/ml)加入孔中,用板振荡器在室温下孵育30分钟。通过Luminex(注册商标)测定荧光强度(FI),求出肽特异性抗体的水平。使用15名健康供体的血浆作为负对照。截止(cut off)FI值确认为高于健康供体的FI值的平均+2SD。
结果在12名受试者中7名的血浆中检出对C35-44肽具有反应性的IgG。而该肽对来源于15名健康供体的任一个供体的血浆均没有反应性。受试者中的作为对C35-44肽的反应性的平均IgG的水平在统计学上显著高于各健康供体。
由HCV感染者的PBMC诱导肽特异性CTL
研究肽C35-44是否能由具有HLA-A24、HLA-A11、HLA-A31或HLA-A33的HCV感染者的PBMC诱导肽特异性CTL。将PBMC在体外与来源于HCV的肽或对照肽一起培养后,以IFN-γ产生作为指标,研究对脉冲对应的肽得到的C1R-A24、C1R-A11、C1R-A31或C1R-A33细胞的反应性。
用于检测肽特异性CTL的分析根据前面所报道的方法,可以适当进行一些改变(Hida N et al.,Cancer Immunol Immunother 2002;51:219-28)。简单而言,将HLA-A24阳性的HCV感染者的PBMC(1×105细胞/孔)在U底96孔微培养板(Nunc,Roskilde,Denmark)中,于200μl培养液中与10μl/ml的各肽一起进行培养。试验进行4次。培养液由45%RPMI1640、45%AIM-V培养基(Gibco-BRL,Gaithersburg,MD,USA)、10%FCS、100U/ml的白介素-2(IL-2)及0.1mM MEM非必需氨基酸溶液(Gibco-BRL)组成。每隔三天除去一半培养液,置换为含有肽(20μg/ml)的新培养基。培养至第15天时,将一半培养细胞用由肽C35-44脉冲得到的C1R-A24细胞进行刺激,将剩下的一半细胞用由对照HIV肽脉冲得到的C1R-A24细胞进行刺激。培养18小时后,回收上清液,通过ELISA测定IFN-γ的水平。
结果,使用肽C35-44时,从5名HLA-A24阳性HCV感染者中的3名PBMC诱导了对肽具有反应性的CTL(表1)。表中,数字为IFN-γ的水平(ng/ml)。

下面,研究C35-44肽是否能诱导HLA-A3超型限制性CTL。将HLA-A2及HLA-A3超型阳性的HCV感染者的PBMC在体外用C35-44肽或对照肽刺激,研究对脉冲对应的肽得到的T2(具有HLA-A2)、C1R-A11、C1R-A31、C1R-A33细胞应答产生的IFN-γ。结果,C35-44肽由HLA-A11阳性HCV感染者(0/7)、HLA-A31阳性HCV感染者(1/2)及HLA-A33阳性HCV感染者(1/2)的PBMC诱导了肽反应性CTL。
上述发现表明C35-44肽在具有HLA-A11、HLA-A31或HLA-A33的HCV感染者的PBMC中能诱导肽特异性CTL。
已知HCV的C35-44肽能诱导HLA-A2限制性CTL(Takao Y et al.,Microbiol.Immunol.,48(7),507-517,2004;日本专利申请2005-310203)。上述结果表明该C35-44用现有的方法计算解离常数时,尽管显示与HLA-A24、HLA-A11、HLA-A31及HLA-A33极低的结合活性,但是也能从HLA-A24、HLA-A11、HLA-A31或HLA-A33阳性的HCV感染者中诱导CTL。
肽刺激PBMC的细胞毒活性
使用肽刺激PBMC,通过标准的6小时51Cr-释放分析,研究对预先用对应的肽或HIV肽中的任一个肽脉冲得到的C1R-A24、A11、A31或A33细胞的细胞毒活性。将每孔2000个51Cr标记细胞与效应细胞一起以规定的效应物/目标细胞比在96圆底孔板中培养。根据以下的式子计算特异性51Cr-释放:(试验组c.p.m.-自然释放c.p.m.)。通过在没有效应细胞下进行培养得到的目标细胞的上清液来确定自然释放。通过与1%Triton-X(Wako Pure Chemical Industries,Osaka,Japan)一起培养得到的目标细胞的上清液确定总释放。在几个试验中,培养初始,在孔中加入10μg/ml抗HLA-I类(W6/32:小鼠IgG2a)、抗HLA-DR(L243:小鼠IgG2a)、抗CD4(NU-TH/I:小鼠IgG1)、抗CD8(NU-TS/C:小鼠IgG2a)或抗CD14(H14:小鼠IgG2a)单克隆抗体。
与HCV肽C35-44一起培养的HLA-A24、HLA-A11、HLA-A31或HLA-A33阳性HCV感染者的PBMC虽然对预先负荷有该肽的C1R-A24、A11、A31或A33细胞显示细胞毒活性,但HIV肽未显示细胞毒活性(图1及2)。
进而,对用肽C35-44脉冲得到的C1R-A24、A11、A31或A33细胞的细胞毒活性被HLA-I类(W6/32)或CD8单克隆抗体(mAb)抑制,但未被试验的HLA-II类(DR-1)、CD4或CD14mAb抑制。该事实表示肽特异性细胞毒活性主要通过CD8+T细胞呈现HLA-I类限制性(图1及2)。
以上结果表示,肽C35-44对HLA-A24、HLA-A11、HLA-A31或HLA-A33阳性的HCV相关疾病患者的肽免疫疗法是有用的。
实施例2
C35-44肽与HLA-A24、HLA-A11、HLA-A31和HLA-A33的结合
实验
关于C35-44肽是否与HLA-A24、HLA-A11、HLA-A31及HLA-A33结合,使用与MHC分子的直接结合分析来研究。
细胞株
RMA细胞株是通过来源于C57BL/6(H-2b)的Rauscher virus进行转化得到的淋巴瘤细胞株,RMA-S是该细胞株的TAP缺损突变株(Ljunggren HG,and Karre K:J Exp Med,162,1745,1985)。RMA-S-A*2402/Kb是将HLA-A*2402基因的α1和α2结构域与来源于小鼠H-2Kb分子的α3、编码膜通过及细胞内结构域的A*2402/Kb嵌合体基因移入RMA-S细胞的小鼠淋巴瘤细胞株(Dr.Takasu(Researchlnstitute of DainiPPon Sumitomo Pharma Co.Ltd.,Osaka,Japan)提供)。另一方面,RMA-S-A*0301、-A*1101、-A*3101、-A*3303细胞株是在来源于小鼠的缺损TAP-1的RMA-S细胞株细胞中导入人的HLA-A*0301、-A*1101、-A*3101、A*3303基因而建立的。
试验方法
RMA-S-A*2402/Kb或RMA-S-A*0301、-A*1101、-A*3101、-A*3303细胞株是TAP缺损株,所以不能与肽形成复合体。因此,在通常的培养条件下,MHC分子不能稳定地存在于细胞表面。利用该性质,在本试验中,研究肽C35-44是否与HLA-A24、HLA-A11、HLA-A31及HLA-A33结合。即,在26℃下培养的RMA-S-A*2402/Kb或RMA-S*A0301、*A1101、*A3101、*A3303细胞株(在该条件下MHC分子能存在于细胞膜上)中加入肽C35-44,在37℃下培养,通过流式细胞仪测定细胞表面上的MHC分子消失程度(Townsend A,etal.,Cold Spring Harb Symp Quant Biol,1,299-308,1989)。
具体而言,按照以下程序进行试验。
1.将RMA-S-A*2402/Kb在26℃下培养一夜(18-20小时)
2.用PBS洗涤1次
3.将细胞(2 x 105个)浮游在含有3μg/ml的β2-微球蛋白(CortexBiochem,Sanleandro,CA,USA)和肽(100μg/ml)的OPTI-MEM(LifeTechnologies)中
4.在26℃下培养3小时
5.在37℃下培养3小时
6.用含有3%FSC的PBS洗涤1次
7.与抗HLA-mAb(第1抗体)一起在4℃下培养30分钟
8.用含有3%FSC的PBS洗涤1次
9.与PE(藻红蛋白(Phycoerythrin):与在488nm下有效激发、发出578nm的荧光的藻胆蛋白(Phycobili-Proteins))-兔抗小鼠IgG(第2抗体)一起在4℃下培养30分钟
10.用含有3%FSC的PBS洗涤1次
11.将细胞悬浮在含有1%甲醛的1mlPBS中
12.用EPICS(流式细胞仪,贝克曼库尔特(Beckman Coulter)公司制)测定
结果示于图3。在26℃下培养RMA-S-A*2402/Kb或RMA-S*A0201细胞后,不脉冲肽,在37℃培养时的平均通道(channel)(X-mean)分别为10。在26℃下培养RMA-S-A*2402/Kb细胞后,用0.1~100μM的不同浓度的肽C35-44脉冲后,在37℃下进行培养时,分别随浓度的增加而增加(图3)。在相同条件下,以具有HLA-A2结合基序的EB-A2、或具有HLA-A24结合基序的肽、NS5A-2132、EB-A24作为阳性对照进行测定,确认分别结合。另外,用于阴性对照的肽(HLA-A2中使用C30、NS5A2132及EB-A24,HLA-A24中使用EB-A2及C30)均确认没有结合。
总结以上结果,确认肽C35-44与各个阳性对照相同地依赖于脉冲肽浓度,荧光强度增加,所以肽C35-44能分别与HLA-A2或-A24结合。
同样地研究肽对HLA-A3超型的结合能力。结果,HCV C35-44肽与阳性对照肽相同程度地结合HLA-A3、-A31及-A33,但未与HLA-A11结合(图4)。
实施例3
用C35-44肽从健康人外周血诱导HLA-A11限制性CTL
研究肽C35-44是否由具有HLA-A11的健常人PBMC诱导肽特异性CTL。将PBMC在体外与来源于HCV的肽或对照肽一起培养后,以IFN-γ产生为指标研究对脉冲对应的肽得到的C1R-A11细胞的反应性。
用于CTL诱导的淋巴细胞使用健康人(HD)6例〔HLA-A11/A11、A2/A2、A24/A24、A2/A24、A11/A2及A11/A24〕。根据之前报道的方法检测肽特异性CTL,并进行了一些改变(Hida N et al.,CancerImmunol Immunother 2002;51:219-28)。简单而言,将HLA-A11阳性的健康人PBMC(1×105细胞/孔)在U底96孔微培养板(Nunc,Roskilde,Denmark)中,在200μl培养液中,与10μl/ml各肽一起进行培养。试验进行4次。培养液由45%RPMI1640、45%AIM-V培养基(Gibco-BRL,Gaithersburg,MD,USA)、10%FCS、100U/ml白介素-2(IL-2)及0.1mM MEM非必需氨基酸溶液(Gibco-BRL)组成。每隔3天除去一半培养液,置换为含有肽(20μg/ml)的新培养基。培养至第15天时,将一半培养细胞用由肽C35-44脉冲的C1R-A11细胞进行刺激,剩下的一半细胞用由对照HIV肽脉冲的C1R-A11细胞进行刺激。18小时培养后,回收上清液,用ELISA法测定IFN-γ的水平。
结果示于图5。使用肽C35-44时,由HLA-A11阳性健康人的3名中3名的PBMC诱导了对肽具有反应性的CTL。图中,横轴数字为IFN-γ的水平(ng/ml)。以上结果表明即使对HLA-A11限制性肽C35-44也诱导CTL。
产业上的可利用性
本发明的肽对治疗HCV相关疾病是有用的。

1.一种药物组合物,用于治疗具有HLA-A24或A3超型的患者的HCV相关疾病,所述药物组合物含有肽作为有效成分,所述肽具有用序列号1表示的氨基酸序列,序列号1表示的序列为:YLLPRRGPRL。
2.一种组合物,用于检查具有HLA-A24或A3超型的患者中的HLA-A24或A3超型限制性细胞毒性T细胞的诱导,所述组合物含有肽作为有效成分,所述肽具有用序列号1表示的氨基酸序列,序列号1表示的序列为:YLLPRRGPRL。
