背景技术
微生物的宿主细胞用于生产纤维素酶。原料成本最大的部分是葡萄糖(碳源)和纯的纤维素(诱导剂和碳源)。加入纯化的纤维素作为诱导剂来刺激纤维素酶产生在本领域是熟知的。纯化的纤维素可从商业供应商获得,但昂贵。所以,有必要提供一种容易获得并且便宜的诱导剂来代替目前使用的昂贵的纯的纤维素。
发明内容
本发明的目的是提供一种在宿主细胞中产生纤维素酶的方法,该方法使用一种容易获得并且便宜的诱导剂替代昂贵的纯的纤维素或类似的诱导剂。
根据第一个方面,本发明涉及一种在宿主细胞中产生纤维素酶的方法,包括在有益于纤维素酶产生的条件下,培养能够产生纤维素酶的所述宿主细胞,其中加入预处理的木质纤维素材料以诱导纤维素酶产生。宿主细胞可以是重组的或野生型宿主细胞,将在下面作进一步地描述。
本发明还涉及预处理的木质纤维素材料作为在宿主细胞中产生纤维素酶的诱导剂和/或碳源的用途。

发明详述
本发明的目的是提供使用容易获得并且便宜的诱导剂在宿主细胞中产生纤维素酶的方法,所述诱导剂材料可替代昂贵的纤维素酶诱导剂,例如,目前使用的纯的纤维素。
用于纤维素酶生产的标准原料是葡萄糖原料连同悬浮的纯纤维素。
本发明人发现目前用于在宿主细胞中生产纤维素酶的纯的纤维素,可以用预处理的木质纤维素材料(pre-treated ligno-cellulosic material)代替,例如,尤其是预处理的玉米秸(PCS)。在使用前,优选将预处理的木质纤维素材料进行解毒(detoxify),例如,通过洗涤,例如,通过在水中重复浸渍、离子交换、反萃取(stripping)等。至少部分地进行解毒以去除抑制宿主细胞性能的化合物。本发明的一个优点在于因为使用容易获得且比纯的纤维素更廉价的诱导剂而降低了生产成本。
生产酶的方法
在真菌来源(例如丝状真菌(filamentous fungi))或细菌来源的宿主细胞中生产纤维素酶是本领域熟知的。除了用预处理的木质纤维素材料替代诱导剂(如纯的纤维素)之外,本发明的方法可为熟知的方法。
能够产生纤维素酶的宿主细胞以特定的生长速率在精确的培养条件下生长。当把宿主细胞培养物引入发酵培养基时,该接种的培养物经过多个阶段。最初不生长。这个阶段称为迟滞期并可认为是一个适应期。在下一个被称为“指数期”的阶段,宿主细胞培养物的生长速率逐渐增加。在最大生长期之后,生长速率停止然后培养物进入稳定期。在又一段时间之后,培养物进入死亡期并且活细胞的数量下降。生长期中纤维素酶的表达取决于纤维素酶和宿主细胞。在一个实施方案中,纤维素酶可以在指数期进行表达。在另一个实施方案中,在指数期和稳定期之间的过渡期(transient phase)产生纤维素酶。在一个实施方案中,纤维素酶也可以在稳定期和/或恰在孢子形成(sporulation)前进行表达。根据本发明的纤维素酶也可以在一个和/或多个上述阶段中产生。
换句话说,根据本发明,在合适的培养基中和允许表达(优选分泌和任意地回收)纤维素酶的条件下培养宿主细胞。在发酵培养基中进行培养,该培养基包含至少一种碳源和预处理的木质纤维素材料作为诱导剂。根据优选的实施例,诱导剂是洗涤的、预处理的木质纤维素植物材料。纤维素酶生产步骤是本领域熟知的。在本发明的下文中,纤维素酶优选是通过宿主细胞分泌到发酵培养基中的细胞外的纤维素酶。可替换地,纤维素酶是细胞内的。发酵后,可以使用本领域中熟知的方法任选地回收纤维素酶。例如,可以使用常规方法从发酵培养基中回收细胞外纤维素酶,所述常规方法包括,但不限于,离心、过滤、提取、喷雾干燥、蒸发或沉淀。细胞内纤维素酶的回收方法也是本领域熟知的。
至少在本发明的上下文中,可互换的术语“培养(cultivation)”和“发酵(fermentation)”是指使用包含一种或多种宿主细胞的大量培养(mass culture)产生纤维素酶的任意方法。本发明尤其适用于工业规模生产,例如,具有至少50升,优选至少100升,更优选至少500升,甚至更优选至少1000升,尤其是至少5000升的培养基。
本发明的方法可作为分批、补料分批、重复补料分批或连续方法来进行。
本发明的方法可以在需氧或厌氧的条件下实施。一些酶通过深层培养(submerged cultivation)产生,一些通过表面培养(surface cultivation)产生。根据本发明优选深层培养。
因此,根据第一个方面,本发明涉及在宿主细胞中产生纤维素酶的方法,其包括在有益于纤维素酶产生的条件下,培养所述能够产生纤维素酶的宿主细胞,其中加入预处理的木质纤维素材料来诱导纤维素酶产生。
底物
本发明的方法中使用的底物可以是本领域中任何底物。合适的底物可以从商业供应商获得或根据公布的组成(例如,美国典型培养物保藏中心(theAmerican Type Culture Collection)的目录)制备。
通常用于纤维素酶生产的碳源底物包括葡萄糖或类似的糖。可加入氮源底物、生长刺激剂等来改进培养和纤维素酶生产。氮源包括氨(NH4Cl)和肽。可以使用蛋白酶,例如,来消化蛋白质以产生游离氨基氮(FAN)。这些游离氨基酸可以作为宿主细胞的营养物起作用,从而促进生长和纤维素酶产生。优选用于生长的发酵刺激物包括维生素和矿物质。维生素的例子包括多种维生素、生物素、泛酸/泛酸盐(pantothenate)、烟酸、烟酸肌醇(meso-inositol)、硫胺素、吡哆醇(pyridoxine)、对-氨基苯甲酸、叶酸、核黄素和维生素A,B,C,D,和E。矿物质的例子包括矿物质和矿物质盐,其能够提供包含P,K,Mg,S,Ca,Fe,Zn,Mn和Cu的营养物。
根据本发明,用预处理的木质纤维素材料,优选解毒的(例如水洗的)预处理的木质纤维素材料代替在纤维素酶的生产过程中通常用作诱导剂(和碳源)的纯的纤维素。
根据本发明,可将预处理的木质纤维素材料和碳源一起加入到培养基中,但是也可以与碳源分开加入。根据本发明,可将预处理的木质纤维素材料在接种宿主细胞培养物之前,接种的同时或接种之后加入到培养基中,加入量对应于通常使用的纯的纤维素的量。这意味着预处理的木质纤维素材料优选的加入量等于通常使用的纯的纤维素的量。在本发明的纤维素酶产生过程中,本领域普通技术人员能够容易地确定何时加入以及加入的预处理的木质纤维素的量。在培养的时间间隔(time span)中,预处理的木质纤维素材料优选的加入量对应于通常使用的纯的纤维素的量。在优选的实施方案中,预处理的木质纤维素材料的量(对应于纯的纤维素的量)与碳源(例如葡萄糖)量的比例范围是大约1∶10至2∶1,优选大约1∶5至1∶1。
如上所述,预处理的木质纤维素材料的使用方式与通常用于熟知的纤维素酶产生方法的纯的纤维素的使用方式相同。
举例说明,当使用木霉属(Trichoderma)的菌株,例如里氏木霉(Trichoderma reesei)作为宿主细胞产生纤维素酶时,将碳源底物水平保持在低水平,例如,低于1g碳源底物/L,如低于1g葡萄糖/L。本发明方法可持续的时间与相应的传统方法的相同,例如在3-10天。木霉属发酵(包括里氏木霉发酵)一般持续5-9天。
木质纤维素材料
根据本发明,“木质纤维素材料”包括含有木质纤维素的任何材料。木质纤维素一般存在于下述物质中:植物的茎、叶、壳、皮、和穗轴,或树的叶、枝和木材。木质纤维素材料也可以是,但不限于,草本(herbaceous)材料、农业残余物、林业残余物、市政固体废物、废纸以及纸浆厂和造纸厂残余物。本文中可以理解的是:木质纤维素材料可以是植物细胞壁材料的形式,所述植物细胞壁材料在混合基质中含有木质素、纤维素和半纤维素。
在一个实施方案中,木质纤维素材料是玉米纤维、稻草、松树木材、木屑(wood chip)、白杨木,麦秆(wheat straw),柳枝稷(switch grass),甘蔗渣,纸和纸浆处理废物。在一个优选实施方案中,木质纤维素材料是玉米秸(cornstover)。在另一个优选实施方案中,木质纤维素材料是木本或草本植物。
预处理
根据本发明,木质纤维素材料是经预处理的。术语“预处理”可以用术语“处理”代替。但是,其包括的优选技术是已知用于“预处理”木质纤维素材料的那些技术,如下文进一步描述的。
如上所述,可以使用本领域已知的常规方法来进行处理或预处理,这可以促进纤维素从木质纤维素材料中分离和/或释放。
预处理技术是本领域熟知的,包括物理的、化学的、和生物的预处理,或它们的任意组合。在优选的实施方案中,木质纤维素材料的预处理作为分批或连续过程来进行。
物理预处理技术包括多种类型的研磨/粉碎(粒度的减小),照射,蒸煮/蒸汽喷发(steaming/steam explosion),和水热解(hydrothermolysis)。
粉碎包括干磨、湿磨和振动球磨(vibratory ball milling)。优选地,物理预处理包括使用高压和/或高温(蒸汽喷发)。在本发明的上下文中,高压包括300至600psi,优选400至500psi,例如大约450psi的压力范围。在本发明的上下文中,高温包括大约100至300℃,优选大约140至235℃的温度范围。在具体的实施方案中,在大约450psi的压力和大约235℃的温度条件下进行浸渍。在优选的实施方案中,使用蒸汽枪水解器系统(steam gun hydrolyzersystem)进行物理预处理,该系统采用高压和高温,例如,使用Sunds水解器(Sunds Hydrolyzer)(可由Sunds Defibrator AB(瑞典)获得)。
化学预处理技术包括酸,稀释酸,碱,有机溶剂,石灰,氨,二氧化硫,二氧化碳,pH-控制的水热解,湿氧化,和溶剂处理。
优选地,化学处理方法是酸处理方法,更优选,连续稀释的酸或弱酸(mildacid)处理,例如用硫酸,或另外的有机酸,如乙酸、柠檬酸、酒石酸、琥珀酸,或它们任意的混合物来处理。也可以使用其它酸。至少在本发明的上下文中,弱酸处理是指处理的pH范围为1至5,优选1至3。在具体的实施方案中,酸浓度的范围是0.1至2.0wt%的酸,优选硫酸。酸与木质纤维素材料混合或接触,该混合物在大约160-220℃的温度范围保持数分钟至数秒钟的时间。具体而言,预处理条件可以是如下所述:165-183℃,3-12分钟,0.5-1.4%(w/w)酸浓度,15-25,优选大约20%(w/w)总固体浓度。其它预期的方法在美国专利4,880,473,5,366,558,5,188,673,5,705,369和6,228,177中描述,所述文件全部并入作为参考。
湿氧化技术涉及氧化剂(如亚硫酸盐类氧化剂等)的使用。溶剂处理的例子包括用DMSO(二甲基亚砜)等处理。化学处理方法一般进行大约5至10分钟,但可以进行更短或更长的时间。

纤维素酶
根据本发明,纤维素酶是指能够降解生物质的纤维素分解酶。根据本发明产生的纤维素酶可以是任意来源的,包括细菌或真菌来源。包括化学修饰或蛋白质工程变体。合适的纤维素酶包括来自芽孢杆菌属(genera Bacillus),假单胞菌属(pseudomonas),腐质霉属(Humicola),镰孢属(fusarium),梭孢壳属(Thielavia),枝顶孢霉属(Acremonium),金孢子菌属(Chrysosporium)和木霉属(Trichoderma)的纤维素酶,例如由特异腐质霉(Humicola insolens)、嗜热毁丝霉(Myceliophthora thermophila)、土生梭孢霉(Thielavia terrestris)、尖镰孢(Fusarium oxysporum)、Chrysosporium lucknowense和里氏木霉产生的真菌纤维素酶。
在一个实施方案中,产生的纤维素酶是与宿主细胞同源的纤维素酶复合物。在一个实施方案中,产生的纤维素酶是与木霉属(优选里氏木霉的菌株)的宿主细胞同源的纤维素酶复合物。
在另一个优选的实施方案中,产生的纤维素酶是含有里氏木霉纤维素酶复合物和此外的一种或多种异源共产生的外源酶的纤维素酶制剂。
在另一个实施方案中,产生的纤维素酶是与腐质霉属的菌株,优选特异腐质霉的菌株,尤其是特异腐质霉,DSM 1800同源的纤维素酶复合物。
在另一个优选的实施方案中,产生的纤维素酶是含有特异腐质霉纤维素酶复合物和此外的一种或多种异源共产生的外源酶的纤维素酶制剂。
在一个实施方案中,产生的纤维素酶是与一种金孢子菌属的菌株,优选Chrysosporium lucknowense的菌株同源的纤维素酶复合物。
在另一个优选实施方案中,产生的纤维素酶是含有Chrysosporiumlucknowense纤维素酶复合物和此外的一种或多种异源共产生的外源酶的纤维素酶制剂。
可以理解的是,产生的纤维素酶可以是单一成分纤维素酶,例如,在合适的宿主细胞中重组产生的内切葡聚糖酶、外-纤维二糖水解酶、葡糖水解酶或β-糖苷酶。合适的宿主细胞在下文中进一步描述。
产生的纤维素酶可以是纤维素酶制剂,其中一种或多种同源的纤维素酶成分从天然产生纤维素酶的宿主细胞中缺失或失活。
能够产生纤维素酶的宿主细胞
宿主细胞可以是任何来源。如上所述,纤维素酶与能产生纤维素酶的宿主细胞可以是同源或异源的。
本文使用的术语“重组宿主细胞”,是指携带编码纤维素酶的基因并能表达所述基因以产生纤维素酶的宿主细胞,其中已将纤维素酶编码基因转化、转染、转导等入宿主细胞。使用的转化、转染、转导等技术是本领域熟知的。在一个优选的实施方案中,将基因以一个或多个拷贝整合入重组宿主细胞的基因组。
当纤维素酶是异源的,能产生纤维素酶的重组宿主细胞优选真菌或细菌来源。重组宿主细胞的选择很大程度上依赖于编码纤维素酶的基因和纤维素酶的来源。
本文使用的术语“野生型宿主细胞”,指天然地携带编码纤维素酶的基因并且能表达所述的基因的宿主细胞。当纤维素酶是同源制剂或纤维素酶复合物时,能产生纤维素酶的野生型宿主细胞或它们的突变体优选为真菌或细菌来源。
“它们的突变体”可为野生型宿主细胞,其中已将一个或多个基因缺失或失活,例如,以富集纤维素酶制剂的某种成分。突变体宿主细胞也可以是用一个或多个附加基因转化的野生型宿主细胞,所述一个或多个附加基因编码附加酶或蛋白质,从而将一种或多种附加酶活性或其他活性引入由野生型宿主细胞天然产生的制剂或纤维素酶复合物。附加酶可以具有相同的活性(如纤维素酶活性),但仅仅是另一种酶分子,例如具有不同的性质。为了增加基因表达以产生更多的酶,突变体野生型宿主细胞也可以具有经转化、转染、转导等,优选整合入基因组的附加的同源酶编码基因。
在一个优选的实施方案中,重组或野生型宿主细胞是丝状真菌来源。宿主细胞的例子包括选自下组的任意一种,包括枝顶孢霉属、曲霉属、短梗酶属、烟管菌属、白腐菌属(Ceriporiopsis)、金孢子菌属、鬼伞属、革盖菌属、隐球菌属、担子菌属(Filobasidium)、镰孢属、腐质霉属、梨孢菌属(Magnaporthe)、毛霉属、毁丝霉属、新考玛脂霉属(Neocallimastix)、脉孢菌属、拟青霉属、青霉属、平革菌属(Phanerochaete)、射脉菌属(Phlebia)、瘤胃壶菌属(Piromyces)、侧耳属(Pleurotus)、裂褶菌属、踝节菌属、嗜热子囊菌属(Thermoascus)、梭孢壳属、弯颈霉属(Tolypocladium)、栓菌属(Trametes)或木霉细胞。
在一个更优选的实施方案中,丝状真菌宿主细胞选自下组,包括泡盛曲霉、烟曲霉、臭曲霉、日本曲霉、构巢曲霉、黑曲霉或米曲霉的菌株。在另一个优选的实施方案中,丝状真菌宿主细胞是杆孢状镰孢、禾谷镰孢、库威镰孢、大刀镰孢、禾本科镰孢、禾赤镰孢、异孢镰孢、合欢木镰孢、尖镰孢、多枝镰孢、粉红镰孢、接骨木镰孢、肤色镰孢、拟分枝孢镰孢、硫色镰孢、圆镰孢、拟丝孢镰孢或镶片镰孢菌株的细胞。在另外优选的实施方案中,丝状真菌宿主细胞选自下组:黑刺烟管菌(Bjerkandera adusta)、Ceriporiopsisaneirina、Ceriporiopsis aneirina、Ceriporiopsis caregiea、Ceriporiopsisgilvescens、Ceriporiopsis pannocinta、Ceriporiopsis rivulosa、Ceriporiopsissubrufa、或Ceriporiopsis subvermispora、Chrysosporium lucknowense、灰盖鬼伞(Coprinus cinereus)、毛革盖菌(Coriolus hirsutus)、特异腐质霉、疏棉状腐质霉、米赫毛霉、嗜热毁丝霉、粗糙脉孢菌、产紫青霉、黄孢平革菌(Phanerochaetechrysosporium)、辐射射脉菌(Phlebia radiata)、Pleurotus eryngii、土生梭孢霉、Trametes villosa、Trametes versicolor、哈茨木霉、康宁木霉、长枝木霉、里氏木霉或绿色木霉的菌株。

用途
在第二个方面,本发明涉及预处理的木质纤维素材料作为在宿主细胞中产生纤维素酶的诱导剂的用途。
在第三个方面,本发明涉及预处理的木质纤维素材料在纤维素酶产生过程中作为碳源的用途。

实施例1
预处理的木质纤维素材料(生物质)用于纤维素酶生产的应用
在这个实施例中,里氏木霉在洗涤的生物质固体上生长,该固体通过加热和稀酸预处理得到。预处理的玉米秸(PCS)由国家可再生能源实验室(theNational Renewable Energy Laboratory(NREL,Golden CO))提供,其葡聚糖含量是53.2%(NREL数据)。在桶(bucket)中将1kg PCS悬浮于~20升双重去离子水(double deionized water)中,在PCS沉降后,将水倾析。重复操作直到洗涤水在pH 4.0以上,此时还原糖为0.06g/L以下。通过下述方法测定洗涤的PCS的干重百分比含量:将样品在105℃的烘箱中干燥24小时以上(直到恒重),并将其与湿重相比较。
发酵在Applikon 2L玻璃夹套容器中完成,其工作体积是1.8L。通过电子热电偶测定温度并使用循环水浴控制温度。溶解氧和pH都使用购买于Broadley James Corporation的传感器探针测定。ADI 1030控制器允许比例反馈控制(proportional feedback control)以基于pH设定点和死区(deadband)使用酸和碱进料泵调整pH。ADI 1012搅拌控制器用于驱动Applikon P310电动机,从而以1100至1300rpm的速度搅拌培养液。使用无挡板的Rushton辐流式叶轮(radial flow impeller)。用无菌空气流以大约1vvm的速度向培养液通气;空气通过安装在罐底部叶轮之下的喷头(sparger)进入。
使用里氏木霉菌株SMA135-04进行发酵。制备甘油冷冻原液,并将其用作种瓶的接种物。种瓶按下表所示的进行培养。如所示的,一些接种物的体积减少。
木霉属发酵持续大约165小时,此时对罐进行收集。木霉属发酵法使用葡萄糖原料。根据需要,使用Pluronic_L61表面活性剂(BASF)减少起泡。将PCS分批实例(APE-57,APE-59)和纤维素分批发酵(APE-58)进行比较。

将最终发酵液的等分试样在蒸馏去离子(DDI)水中稀释5倍。然后1体积稀释样品与2体积混有5%β-巯基乙醇的SDS样品缓冲液(BioRad)混合,煮沸5分钟。每个样品取15微升上样于8-16%Tris-HCI凝胶(BioRad)上,进行电泳并用Biosafe Coomassie Blue染色(图1)。蛋白质主条带在~70kDa。所有培养液在~10kDa都显示清晰的条带;纤维素分批也显示在<18kDa的条带。
通过酶培养液水解稀酸预处理的玉米秸(PCS)和产生糖的能力来测定酶培养液的活性,产生的糖可通过化学分析还原性末端来检测。PCS由国家可再生能源实验室(NREL,Golden CO)提供,其葡聚糖含量是53.2%(NREL数据)。在桶中将1kg PCS悬浮于~20升双重去离子水中,PCS沉积后,将水倾析。重复操作,直到洗涤水在pH 4.0以上,此时还原糖在0.06g/L以下。该沉积的浆通过100目(Mesh)筛以确保吸取的能力。洗涤的PCS的百分比干重含量通过下述方法测定:将样品在105℃的烘箱中干燥24小时以上(直到恒重)然后与湿重进行比较。
在用平板密封物(plate sealer)(ALPS-300,ABgene)密封的96-深-孔板(Axygen Scientific)中进行PCS水解。PCS浓度是10g/L,含有50mM醋酸盐,pH 5.0。PCS水解在50℃进行,总反应体积为1.0ml,不用额外搅拌。每个反应进行三次重复。如下所述用p-羟基苯甲酸酰肼(PHBAH)试剂分析释放的还原糖。
具体地,吸取0.8ml PCS(12.5g/L)移入96-深-孔板的每个孔中,其中加入0.10ml醋酸钠缓冲液(0.5M,pH 5.0),然后加入0.10ml稀释的酶溶液以开始反应,并给出1.0ml的终反应体积和10g/L的PCS浓度。在水解开始时和采集每个样品的时间点之前,通过倒转深-孔板使反应混合物混合。混合后,将深-孔板在3000rpm离心2分钟(带RTH-250转子的Sorvall RT7),然后去除20微升水解物(上清液),将180微升0.4%NaOH加入96-孔微量反应板中。如果需要,可将终止的溶液进一步稀释至还原糖的合适范围。用下述的对-羟基苯甲酸酰肼试剂(para-hydroxy benzoic acid hydrazide reagent)(PHBAH,Sigma,4-羟基苄基酰肼(4-hydroxy benzyhydrazide)分析释放的还原糖:在V-底96-孔热电偶套管平板(Costar 6511)中,将50微升PHBAH试剂(1.5%)与100微升样品混合,在平板加热块上95℃保温10分钟,然后在每个孔中加入50微升DDI水,混合,并将100微升转移到另一个平底96-孔板(Costar 9017)中,在410nm读取吸光度。在相同的条件下使用葡萄糖校准曲线来计算还原糖。将纤维素转化为还原糖的百分比转化率计算为:

因子1.11用于修正纤维素水解为葡萄糖过程中的重量增加。
使用PCS,APE-57和APE-59产生的蛋白质水平与采用纤维素的对照发酵相似(图3),而且,培养液与采用纤维素的对照发酵(图2)的纤维素酶活性相似。
Claims (19)
1.一种在宿主细胞中产生纤维素酶的方法,包括在有益于纤维素酶产生的条件下,培养能够产生纤维素酶的所述宿主细胞,其中加入预处理的木质纤维素材料以诱导纤维素酶产生。
2.根据权利要求1的方法,其中木质纤维素材料是植物材料,优选选自下组:玉米秸,玉米纤维,稻草,松木,木屑,白杨木,麦秆,柳枝稷,纸和纸浆处理废料。
3.根据权利要求1或2的方法,其中木质纤维素材料的预处理通过将木质纤维素材料进行物理处理,化学处理,生物处理,或它们的任意组合来进行。
4.根据权利要求1至3任一项的方法,其中将预处理的木质纤维素材料进行粉碎,稀酸蒸汽喷发,蒸汽喷发,湿氧化,或氨水纤维喷发(或AFEX预处理)。
5.根据权利要求1至4任一项的方法,其中宿主细胞是重组宿主细胞或野生型宿主细胞或它们的突变体。
6.根据权利要求1至5任一项的方法,其中宿主细胞是细菌或真菌来源。
7.根据权利要求1至6任一项的方法,其中产生的纤维素酶是纤维素酶复合物或由野生型宿主细胞或其突变体产生的制备物,优选如下的野生型宿主细胞或其突变体:木霉属,优选里氏木霉的菌株,或腐质霉属,优选特异腐质霉的菌株或金孢子菌属,优选Chrysosporium lucknowense的菌株。
8.根据权利要求1至6任一项的方法,其中产生的纤维素酶是单一成分纤维素酶或由重组宿主细胞产生的纤维素酶制备物,优选如下的重组宿主细胞:木霉属,尤其是里氏木霉的菌株,或腐质霉属,优选特异腐质霉的菌株或金孢子菌属,优选Chrysosporium lucknowense的菌株或曲霉属,优选黑曲霉或米曲霉的菌株。
9.根据权利要求1至8任一项的方法,其中纤维素酶生产在至少50升的发酵罐中进行。
10.根据权利要求1至9任一项的方法,其中该方法作为补料分批方法进行。
11.根据权利要求1至10任一项的方法,其中加入碳源,优选葡萄糖。
12.根据权利要求1至11任一项的方法,其中在加入预处理的木质纤维素材料之前,同时,或之后加入碳源。
13.根据权利要求1至12任一项的方法,其中加入氮源。
14.根据权利要求1至13任一项的方法,其中使用的预处理的材料基本由纤维素组成。
15.根据权利要求1至14任一项的方法,其中在培养后回收纤维素酶。
16.根据权利要求1至15任一项的方法,其中预处理的木质纤维素材料在用于纤维素酶生产前进行解毒。
17.根据权利要求1至16任一项的方法,其中预处理的木质纤维素材料在用于纤维素酶生产前进行洗涤。
18.预处理的木质纤维素材料作为纤维素酶生产过程中的诱导剂的用途。
