Abstract

Description
技术领域
本发明涉及包含天然来源的色氨酸、特别是来自植物的蛋白结合型色氨酸的组合物;制备所述组合物的方法;所述组合物的物理配制品(physical formulations);以及所述组合物用于增强哺乳动物的性欲和性能力的用途。
背景技术
色氨酸是一种必需氨基酸,见于多种天然存在的植物蛋白,并具有多种令人感兴趣的药用价值,包括用于治疗失眠以及辅助用于治疗多种精神疾患。吸收后,血液循环中的色氨酸大约80%结合于血浆白蛋白,其余的20%以游离色氨酸的形式循环,且在适当的情况下,色氨酸可转运至脑中。一旦通过血脑屏障(BBB),色氨酸便可代谢成为5-羟色胺,后者是一种神经递质,与情绪和睡眠调节有关(Boman,1988)。5-羟色胺转而代谢为退黑激素,其是一种在松果体中发现的睡眠相关激素,而松果体是脑部上丘脑中的一种小球果样结构,负责调节人的24小时的昼夜节律。摄入足量的色氨酸本身一向可减少睡眠等待时间(sleep latency),即自“熄灯”到入睡的时间,并通过改善睡眠结构而改善睡眠的总体质量(Boman,1988)。色氨酸代谢为5-羟色胺对一些存在5-羟色胺水平耗竭的情况也十分有帮助,例如焦虑症、抑郁症、强迫症、一些疼痛性疾病、攻击行为以及进食异常。
色氨酸的催眠作用得到了深入研究,其遵循相当平直的剂量反应曲线,在大约1000毫克时达到平台(综述见Schneider-Helmut and Spinweber,1986)。若单独使用,仅250毫克的色氨酸即足以改善轻度失眠者或那些睡眠等待时间长于平均值的人的睡眠(Hartmann and Spinweber,1976;Hartmann 1982)。剂量为1000毫克时可产生更加一致的结果(Schneider-Helmut and Spinweber,.1986),但更高的剂量(2,000-12,000毫克)产生的额外益处是非常有限的,且实际上,最高的剂量(12,000毫克)则伴有睡眠结构混乱,尽管可减少睡眠等待时间(Griffiths等,1972)。
2001年11月29日公开的Hudson,Susan P.和Hudson,Craig J的PCT申请WO01/89319讨论了前述病症。前述WO01/89319揭示了包含含有蛋白结合型色氨酸的至少是部分脱脂的植物原料粉(优选为南瓜籽(squash seed)),以及任选地包含一种碳水化合物原料的组合物,其量足以能够促进体内产生的色氨酸转运通过血脑屏障。也揭示了包含该组合物的膳食添加剂、食品和饮料用于诱导睡眠,用于焦虑症、抑郁症、强迫症、攻击行为、慢性疼痛和进食异常,或用于为有需求的个体提供色氨酸补充。
通常认为提高脑内5-羟色胺水平的药物会降低性欲并降低总体性能力(Bradford 2001)。与性欲降低有关的药物包括抗抑郁剂特别是那些属于选择性5-羟色胺再摄取抑制剂(SSRI)类的抗抑郁剂,以及5-羟色胺前体,色氨酸(Young 1986)。
人摄入大剂量色氨酸,即每天超过6克,可使性欲/功能增强,这在以往罕有报道。
此外,以往报道认为,基本上所有在使用色氨酸时声称性欲或性能力增强的人,均同时在服用其他药物。尚无关于单独使用相对低剂量的色氨酸作为单一用药可增强性欲或性能力的报道。
此外,至少还没有关于结合了色氨酸的蛋白质在增强性欲或性能力方面的作用的报道。
发明内容
我们惊异地发现,前述WO01189319中所述的至少是部分脱脂的粉组合物可增强哺乳动物的性欲和性能力。
我们惊异地发现,在一个选定的睡眠研究中,摄入富含蛋白结合型色氨酸的高蛋白食品组合物的人类研究对象报道性欲和性能力得到增强。相对于前述现有技术的量(>6克/天)来说,按照所摄入的结合了色氨酸的蛋白质计算,取得这些结果的研究提供的色氨酸本身的量可能低得多(<250毫克)。
因此,本发明的目的之一在于提供一种用于增强哺乳动物的性欲和性能力的色氨酸的植物原料。
因此,本发明在一个方面提供了一种用于增强哺乳动物的性欲和性能力的组合物,其包含一种来自于含蛋白结合型色氨酸的植物原料粉,及其生理学可接受的稀释剂或载体。
更优选地,所述粉至少是部分脱脂的。
现发现,根据WO01/89319,通过浓缩富色氨酸蛋白质原料的蛋白结合型色氨酸成分而天然衍生的富色氨酸组合物,可增强哺乳动物的性欲和性能力。本发明所用的组合物包含天然含有蛋白结合型色氨酸的植物原料,优选地为南瓜籽(squash seed),例如Butternut瓜籽(Butternut squashseed)、Peppercorn瓜籽(Peppercorn squash seed)和南瓜籽。优选地,所述植物原料至少是部分脱脂的,以浓缩蛋白结合型色氨酸的含量。优选地,所述组合物进一步包含碳水化合物原料,例如葡萄糖,其量足以增强色氨酸跨血脑屏障摄取,并克服对BBB转运位点的竞争而进入中枢神经系统(CNS)。该组合物可进一步任选地包含生理学可接受的载体、调味剂、色素和其他营养剂,例如维生素,优选地维生素B3和/或维生素B6。
在一个优选的实施方式中,该组合物包含至少是部分脱脂的南瓜籽(squash seed)(特别是Butternut瓜籽、南瓜籽和Peppercorn瓜籽)、葡萄糖以及维生素B3和B6。
本发明进一步涉及膳食添加剂,其形式例如为片剂、粉剂、混悬液、液体、胶囊或凝胶;食品例如膳食棒、饼干、烘烤品;小吃、糖果、糖棒、饮料和类似的包含本发明的组合物的食品。
前述WO 01/89319揭示了使用二阶导数光谱学,特定的植物原料、且特别是含有相对高水平的蛋白结合型色氨酸的植物种子,并揭示了这些材料可用于在体内提供色氨酸。提供了一种方法,其用于产生可食用的组合物,所述组合物具有高水平的蛋白结合型色氨酸作为色氨酸的天然蛋白原料,其含量较已知的天然原料高。使用芦竹碱的植物通常含有高水平的色氨酸,并可用作本发明的植物原料。起始植物原料含有至少200毫克/100克或至少0.2%的蛋白结合型色氨酸是较理想的,但并非必需。可通过已知的方法测定色氨酸浓度,包括,例如高压液相色谱(HPLC)、二阶导数光谱学或任何其他已知的方法。二阶导数光谱学是定量分析色氨酸水平的优选方法,原因在于其可消除背景吸附。
根据本发明的一个实施方式,可采用一系列步骤去除植物原料中的油脂,以使原料部分脱脂,提高植物原料中的蛋白结合型色氨酸水平。植物原料可以是种子,例如但不限于,Butternut瓜籽、Peppercorn瓜籽、南瓜籽、兵豆籽、葵花籽、亚麻籽、西瓜籽、大蒜芥籽、棉籽、芝麻籽、Canola籽、月见草籽、红花籽、紫花苜蓿(alfalfa)籽、大麦、大豆及其组合。优选地,种子是Butternut瓜籽,这是因为与其他种子相比,Butternut瓜籽被认为其中含有的色氨酸与总蛋白之比是最高的。植物原料也可以是植物的营养部分(vegetative part),例如紫花苜蓿、海藻或大型海藻。尽管优选地将植物原料脱脂以提高蛋白结合型色氨酸水平,但脱脂对于本发明的实施来说并不是必需的。
前述WO01/89319揭示了一种用于生产浓缩的、天然的色氨酸源的方法,包含鉴别植物原料中的天然存在的蛋白结合型色氨酸源;在足以释放其中所含的油脂的条件下压缩植物原料;并至少部分去除其中所含的油脂,以产生部分脱脂的植物原料,其与起始原料相比具有更高的色氨酸源。
对于种子,不需要在加工前去除其种子的皮或壳以暴露种籽。在一个实施方式中,优选地首先将种子通过一连串平滑滚筒以产生一薄片,该过程称为压片。这一步骤使得油脂细胞至少部分破裂,并增加用于进一步处理的种子的表面积。
随后对压成薄片的种子进行热处理,该过程称为烹调(cooking)或调理(conditioning),以进一步破裂油脂细胞并增加油脂的粘性以便随后脱脂。调理步骤可通过例如微波炉、烤箱或间接蒸气而进行。调理步骤的温度应该足以使得油脂细胞破裂,并提高油脂的粘性,但不会对植物原料中含有的蛋白质造成有害的破坏。优选地,温度为大约40℃至大约50℃。调理步骤进行的时间足以达到目标温度。
冷却前,对加热的种子薄片进行机械压榨以至少部分去除其中含有的油脂。可使用任何已知的机械压具或压榨机,例如Gusta Lab Press:Thedegree of defatting depends in part on the flaking and cooking stepsperformed,temperature and oil viscosity and the pressure exerted on theseed。通常,优选地去除大约2/3至大约3/4的油脂。
对压榨的植物材料的进一步处理取决于最终的使用者。例如,可使用任何常规手段磨碎植物材料,例如但不限于,圆盘式粉碎机、锤式粉碎机或针式粉碎机。所选的粉碎机部分取决于所需的产品密度,例如,针式粉碎机产生的产品具有面粉样密度,而圆盘式粉碎机或锤式粉碎机产生的产品具有颗粒的密度。
上述方法产生了一种蛋白结合型色氨酸的天然原料,其具有的色氨酸含量高于原植物原料。优选地,所述材料应该提供至少0.2%重量的色氨酸。随后可将所得到的、至少部分脱脂的种子粉掺入用于增强性欲和性能力的组合物。
除了部分脱脂的粉,用于本发明的组合物还优选地包含一种具有高血糖指数(glycemic index)的碳水化合物原料,优选地为葡萄糖,不过也可使用蔗糖以及其他可降解为葡萄糖的糖。不限于理论,据认为碳水化合物原料可促进色氨酸本身通过血脑屏障而摄取,然后可代谢为5-羟色胺。人类具有一种屏障,可使得脑在功能上不受肌体其他部位的环境的影响,以便保护敏感的CNS。该屏障由对物质扩散进入脑部起限制作用的血脑接触面处的脑内皮细胞之间的无数的紧密连接形成(Saunders etal.,1991)。由紧密连接产生的多重扩散是一连串进出脑部的转运机制,其调节脑的内部环境,与此有关的大量分子包括电介质、葡萄糖、维生素和氨基酸。其他一些大的中性氨基酸(LNAA)也使用色氨酸的转运机制(Lajtha,1974;Betz and Goldstein,1978)。据认为,对这些转运位点的竞争导致了含大量高蛋白的膳食不能诱导催眠效果,尽管其中含有足量的色氨酸(Moller,1983)。相反,在同一个研究中,含相对小量色氨酸的高碳水化合物膳食缺失诱导了适度的催眠效果。对这种明显的矛盾的解释是,在血清胰岛素水平相对高时,竞争性LNAA分流到肝脏和肌肉组织(Fernstrom and Wurtman,1971)。而色氨酸不会以这种方式分流,因此,任何游离的色氨酸均获得了对通过BBB的转运位点的胰岛素诱导的竞争优势。
碳水化合物原料的量优选地足以在摄入此组合物的个体中诱导血胰岛素水平升高。色氨酸/LNAA比随着胰岛素水平升高而升高,自15微单位/ml升高至60微单位/ml引起色氨酸/LNAA比升高大约35%。这种水平的升高是足够的,不过,较不明显的升高同样是有益的。优选地,组合物中的葡萄糖的量是大约25克至大约150克,最优选的是75克。色氨酸的量将保持不变,但碳水化合物的升高将使得色氨酸/LNAA比升高。其他碳水化合物原料可包括麦芽糖、蔗糖等等,但优选地不是果糖,这是因为其具有低血糖指数。对于肥胖个体或含有2型糖尿病的个体,由于存在异常的葡萄糖胰岛素反应,因此可能需要更高量的碳水化合物,例如100克。
由于体内血液中的色氨酸80%与蛋白结合,因此仅有小量的游离色氨酸真正与其他LNAA竞争进入脑内。因此,在一般情况下,摄入的植物中的蛋白结合型色氨酸经代谢后迅速储存在“白蛋白库”中,除非摄入的是超生理学量,其对CNS色氨酸的生物利用度几乎没有影响。不过,如果在胰岛素水平升高时可以得到色氨酸,那么游离脂肪酸会竞争“白蛋白库”,并使得已有的蛋白结合型色氨酸转变为游离色氨酸,同时防止新摄入的色氨酸的掺入。因此,当竞争性LNAA的血清水平降低时,两种不同来源的色氨酸,即已有的蛋白结合型色氨酸和新摄入的色氨酸使得游离色氨酸升高。有鉴于此,值得提供一种含有至少一些残留油脂成分的产品以便在植物原料或种子中保留一部分脂肪酸。对于南瓜籽(squash seed),最好在种子粉中保留大约20%残留油脂。对于其他种子,有必要将在部分脱脂的产品中再加入其他脂肪酸,以便提供最佳的脂肪酸平衡。为此,可以添加氢化的油脂(Hydrogenated oils)或其他油脂,例如Canola油、葵花油、红花油、棕榈仁油、玉米油或奶粉。
在一个优选的实施方式中,用于本发明的组合物包含:至少部分脱脂的南瓜籽(squash seed),特别是Butternut瓜籽、南瓜籽、Peppercorn籽及其组合;葡萄糖,其量足以在摄入此组合物的个体中促进来自南瓜籽(squash seed)的色氨酸通过血脑屏障而摄取;以及维生素B3和B6,其量可促进色氨酸的摄取。
在另一个实施方式中,该组合物包含至少部分脱脂的Butternut瓜籽粉,例如大约50克至大约100克,其量足以提供大约250毫克至大约1000毫克的色氨酸,以及大约25克至大约200克的葡萄糖。更加优选的一种组合物包含大约25克至大约50克的脱脂Butternut瓜籽粉,其经过压榨后油脂含量降低了75%,并包含大约75克至大约100克的葡萄糖。任选地,该组合物包含维生素B3和/或维生素B6。维生素B3的量微大约5毫克至大约50毫克;而维生素B6的量微大约0.5毫克至大约50毫克,优选地,维生素B3和B6均为50毫克。
本发明的组合物和膳食添加剂可用于每天口服施用。该组合物被配制为可优选地每日睡前单剂施用。或者,该组合物可被配制为多个部分,以便根据需要增加或减少施用频率。为了使得其大小易于吞咽或者为了提高其在入睡前或任何需要的时候的生物吸收或利用,可以将一个给定剂量分为两个、三个或更多个片剂或胶囊,等等。可将每日的剂量以一片施用、以两片一起施用。如上所述,各种成分的推荐量可作为配制本发明的添加剂的指导。每单位剂量的各种成分的真实剂量取决于施用于对其有需要的个体的单位数。这涉及产品设计且是膳食添加剂配制者所熟知的。
可使用任何药物学可接受形式的维生素、矿物质和其他上述营养成分及其盐来配制用于本发明的组合物。组合物可配制为胶囊、片剂、粉剂、混悬液、凝胶或液体,任选地包含生理学可接受的载体,例如但不限于,水、乳、果汁、淀粉、植物油、盐溶液、羟甲基纤维素、碳水化合物。组合物可配制为粉剂,例如用于与消费型液体例如乳、果汁、水混合,或消费型凝胶或糖浆,用于混入膳食液体或食品。用于本发明的组合物可与其他食品或液体混合,以提供预定量的添加食品,例如作为一种棒。
该组合物可制成各种形式,例如烘烤品如饼干、核仁巧克力饼、牛奶巧克力软糖、蛋糕、面包、软饼、脆饼、布丁、蜜饯即糖果、小吃食品如咸饼干、薯片、膳食饮料、冰淇淋、冰冻蜜饯以及一些新鲜事物,或非烘烤的挤压产品如棒。优选的形式是非烘烤的挤压营养棒。
组合物还可含有其他成分如一种其他的维生素、矿物质、抗氧化剂、纤维和膳食添加剂或其组合。选择这些成分中的一或多种涉及配制品设计、消费者和终端用户的喜好。添加至用于本发明的组合物中的这些成分的量是本领域人员容易得知的,这方面的指导可参考美国成人RDA剂量。可以添加的维生素和矿物质包括,但不限于,磷酸钙或乙酸钙,三价;磷酸钾,二价;硫酸镁或氧化镁;盐(氯化钠);氯化钾或乙酸钾;抗坏血酸;正磷酸铁;烟酰胺;硫酸锌或氧化锌;泛酸钙;葡糖酸铜;核黄素;β-胡萝卜素;盐酸吡哆醇;硝酸硫胺素;叶酸;生物素;氯化铬或吡啶铬;碘化钾;硒酸钠;钼酸钠;维生素K1;维生素D3;维生素B12;亚硒酸钠;硫酸铜;维生素A;维生素E;维生素B6及其盐酸盐;维生素C;肌醇;维生素B12;碘化钾。
调味剂、色素、香料、坚果等等也可掺入到产品中。调味剂的形式可以是香精、挥发性油、巧克力调味剂、花生酱调味剂、碎饼屑、脆米、香草或任何商品化的调味剂。合适的调味剂的实例包括但不限于纯茴香精、人造香蕉精、人造樱桃精、巧克力精、纯柠檬汁、纯橙汁、纯薄荷精、人造波萝精、人造朗姆酒、人造草莓精、或纯香草精;或挥发性油例如香油、月桂油、香柠檬油、雪松木油、樱桃油、胡桃油、肉桂油、丁香油、或薄荷油;花生酱、巧克力调味剂、香草碎饼屑、奶油硬糖或太妃糖。在一个优选的实施方式中,膳食添加剂含有不含咖啡因的(non-caffeinated)可可粉或巧克力、或巧克力代用品如角豆。食品组合物还可以例如酸奶进行涂覆。
可添加乳化剂以稳定终产品。合适的乳化剂包括,但不限于,卵磷脂(例如来自蛋或大豆)和/或单和二甘油酯。其他乳化剂对于本领域人员来说是显然的,且合适的乳化剂的选择部分取决于配方和终产品。
可在组合物中添加防腐剂以延长产品的保存期。优选地,可使用的防腐剂例如为山梨酸钾、山梨酸钠、苯甲酸钾、苯甲酸钠或乙二胺四乙酸二钠钙。
除了上述碳水化合物,组合物可含有人造甜味剂,例如糖类、环己(基)氨基磺酸盐、阿斯巴甜(Aspartamine)、天冬氨酰苯丙氨酸甲酯、丁磺氨K、和/或山梨醇。如果膳食添加剂拟用于体重超重个体或肥胖个体,或易于出现低血糖的患2型糖尿病的个体,则需要添加此类人造甜味剂。
为生产这种食品棒,将液体成分进行烹制;将干燥成分与液体成分在混合器中混合直至成为生面团;将生面团放入挤压器中挤压;将挤压好的生面团切成合适的长度;并将产品冷却。为生产其他食品或饮料,可将包含用于本发明的组合物的成分添加到常规配方中,或者可以之替代常规成分。食品配方领域的技术人员能够设计出适合本发明目标的合适的食品/饮料。
注意到色氨酸添加剂具有潜在的不良反应。低剂量的色氨酸极少引起不良反应,但是有报道在高剂量时或与某些抗抑郁药物联合使用时可造成问题。当与单胺氧化酶抑制剂(MAOI)联合使用时,色氨酸有引起谵妄和神经系统功能紊乱的危险(Thomas and Rubin,1984)。在更高剂量(每天超过12克),最常见的主诉为日间的镇静作用和恶心(Hartmann,1977)。有少数理论上的危险已经在动物模型中得到证明,单在人类尚未证实。大剂量1-色氨酸在动物中造成脂肪生成(Fears and Murrell,1980),但在人类没有观察到这一效应(Sourkes,1983)。类似地,理论上存在一种危险,即一种色氨酸代谢物黄尿酸(Xanthurenic acid)可导致出现糖尿病(Kotake and Murakami,1971)。因此,本发明的一个目的在于将每日施用于个体的蛋白结合型色氨酸的量限制在低于每天大约12克色氨酸a.i.的水平,以避免这些潜在的不良反应。在另一个方面,本发明提供了一种生产用于增强人类性欲和性能力的药物组合物的方法,所述方法包括将上述定义的组合物与一种其生理学可接受的载体混合。

26.3克脱脂南瓜籽(squash seed)
17.2克葡萄糖(“葡萄糖43”)
5.0克焦糖(Caramel)(New World Flavours)
1.5克干苹果片
20.0毫克维生素B3
20.0毫克维生素B6
病例报告
未经研究者直接询问性欲改变情况,三名研究对象自发报告性欲/性能力增强。
每名研究对象在晚间上床休息前大约1小时摄入其食品棒。两名研究对象报告性欲增强,表现为性欲持久,而另一名报告性欲增强,伴有性生活质量改善。
尽管在此描述并举例说明了本发明的优选实施方式,但应该理解本发明并不限于这些特定实施方式。本发明包括所有那些与在此所描述的并举例说明的具体实施方式和特征的在功能上等价的实施方式。
Claims (13)
1、来自于含蛋白结合型色氨酸的植物原料粉在制备一种用于增强哺乳动物的性欲和性能力的组合物中的用途,其中所述粉至少是部分脱脂的并较所述植物原料具有更高的色氨酸源,植物原料选自胡桃南瓜籽、胡椒南瓜籽、南瓜籽、兵豆籽、葵花籽、亚麻籽、西瓜籽、大蒜芥籽、棉籽、芝麻籽、油菜籽、月见草籽、红花籽、紫花苜蓿籽、大麦、大豆及其组合。
2、权利要求1的用途,其中所述植物原料选自胡桃南瓜籽、胡椒南瓜籽或南瓜籽。
3、权利要求1的用途,其中所述组合物进一步包含一种具有高血糖指数的碳水化合物原料。
4、权利要求3的用途,其中所述碳水化合物原料选自葡萄糖、麦芽糖、蔗糖及其组合。
5、权利要求1或2的用途,其中所述组合物进一步包含选自维生素B3、B6及其组合物的维生素,所述维生素的量足以提高色氨酸的跨血脑屏障摄取。
6、权利要求1或2的用途,其中所述组合物的形式为片剂、粉末、混悬液、液体、胶囊或凝胶。
7、权利要求1或2的用途,其中所述组合物的形式为膳食添加剂。
8、权利要求7的用途,其中所述膳食添加剂为棒状。
9、权利要求1或2的用途,其中所述组合物包含含有蛋白结合型色氨酸的至少是部分脱脂的胡桃南瓜籽粉、25毫克至200毫克的葡萄糖及其生理学可接受的稀释剂或载体,所述含蛋白结合型色氨酸的胡桃南瓜籽粉中色氨酸的量为25毫克至1000毫克。
10、权利要求3的用途,其中所述组合物包含含有蛋白结合型色氨酸的至少是部分脱脂的胡桃南瓜籽粉、25毫克至200毫克的葡萄糖及其生理学可接受的稀释剂或载体,所述含蛋白结合型色氨酸的胡桃南瓜籽粉中色氨酸的量为25毫克至1000毫克。
11、权利要求1或2的用途,其中所述组合物包含提供25毫克至50毫克色氨酸的至少是部分脱脂的胡桃南瓜籽粉、75毫克至100毫克的葡萄糖及生理学可接受的稀释剂或载体。
12、权利要求3的用途,其中所述组合物包含提供25毫克至50毫克色氨酸的至少是部分脱脂的胡桃南瓜籽粉、75毫克至100毫克的葡萄糖及生理学可接受的稀释剂或载体。
13、权利要求1或2的用途,其中所述组合物进一步包含5毫克至50毫克维生素B3、0.5毫克至50毫克维生素B6及其组合。
