本发明涉及异穿心莲内酯在制备抗炎及抗肿瘤药物中的用途。本发明进一步涉及一种含有异穿心莲内酯作为活性成分以及药学上可接受的载体的药物组合物,以及在制备抗炎及抗肿瘤药物中的用途。

含有异穿心莲内酯的药物组合物及其医药用途
技术领域
本发明涉及异穿心莲内酯在制备抗炎以及抗肿瘤药物的用途。本发明进一步涉及一种含有异穿心莲内酯作为活性成分以及药学上可接受的载体的药物组合物,以及在制备抗炎和抗肿瘤药物中的用途。
背景技术
穿心莲为爵床科植物穿心莲Andrographis panniculata(Burm.f.)Nees的干燥地上部分。原产于印度,我国华东、中南及西南等地有栽培。味苦,性寒,归心、肺、大肠、膀胱经。具有清热解毒,凉血消肿之功效。用于感冒发热,咽喉肿痛,口舌生疮,顿咳劳嗽,泻泄痢疾,热淋涩痛,痈肿疮疡,毒蛇咬伤。
1896年,Boorsma从穿心莲中首次分离出一种无色结晶物,1911年经Gorter结构鉴定为二萜内酯类化合物,被命名为andrographolide(1),中文名称穿心莲内酯,又称雄茸内酯,穿心莲乙素,是穿心莲的主要化学成分之一,在我国已作为合成穿琥宁、炎琥宁等解热抗炎药的原料大量生产。迄今为止,有关异穿心莲内酯的药理活性还没有报道,本发明人对此进行深入研究,体内实验表明异穿心莲内酯具有抗炎以及抗肿瘤的用途。
发明内容
本发明公开了异穿心莲内酯在制备抗炎以及抗肿瘤药物的用途。
本发明进一步涉及一种含有异穿心莲内酯作为活性成分以及药学上可接受的载体的药物组合物,以及在制备抗炎以及抗肿瘤药物中的用途。
具体实施方式
1、异穿心莲内酯的制备

10g(28.5mmol)穿心莲内酯和165mL浓盐酸混合,搅拌至完全溶解,密封,放置过夜。反应毕(TLC监测),加入冰冷的40mL水中,搅拌下缓缓滴加饱和碳酸钾水溶液到pH7,过滤,滤液用乙酸乙酯(50mL×4)萃取,无水硫酸钠干燥,过滤,浓缩,抽滤,与第一次得到的滤饼合并,烘干,得目标物5.9g,收率59%。m.p.199.3~200.2℃。IR(cm-1):3415,2991,2933,2884,1760,1746,1647,1441,1393,1371,1345,1211,1081,1021,890,931,890,836。MS(ESI)m/z:351.2(M+1)+,373.3(M+Na)+,389.2(M+Na+1+CH3)+1H-NMR(DMSO-d6,δ):7.50(1H,s,14-H),5.07(1H,d,3-OH),4.94(2H,s,15-H),4.54(1H,t,J=7.7,12H),4.13(1H,brd,19-OH),3.98,3.28(1H,d,J=10.9,19-H),3.25(1H,d,J=7.6,3-H),1.14(3H,s,17-H),1.09(3H,s,20-H),0.96(3H,s,18-H)。
2、异穿心莲内酯的药理活性研究
2.1对蛋清所致大鼠足肿胀的影响
材料:
动物:wistar大鼠,体重180~220g,均为雄性(河南省医学实验动物中心)方法:
大鼠24只,180~220g,均为雄性,随机分为3组  ,每组8只。一组ip生理盐水,二组和三组分别ip试药300mg/kg,各组注射容积均为1mL/100g。给药前用记号笔标记大鼠右踝关节浸水边界,排水容积法测量1次致炎前大鼠右足排水容积。给药后立即于大鼠右后脚掌sc新鲜蛋清0.1mL/只。测量给药后1h,2h,4h,6h时的大鼠足排水容积。实验结果见表1-1。

实验结果表明:异穿心莲内酯给药4h内能非常显著性抑制蛋清所致的大鼠足肿胀,作用明显强于穿心莲内酯。
2.2抗肿瘤
材料:
动物:昆明小鼠,18~22g,均为雄性(河南省医学实验动物中心)
方法:
取昆明小鼠30只,18~22g,均为雄性。取接种8d生长良好的HepA小鼠腹水,用NS 1∶5稀释,以0.2mL/只接种于小鼠右侧腋窝下。将接种后的小鼠按体重分为3组,每组10只。设第1组为正常对照组,腹腔注射NS;2~3为试药组,腹腔注射穿心莲内酯或异穿心莲内酯300mg/kg,注射容积为0.2mL/10g,每日一次,连续10d。停药次日处死,剥离肿瘤,称体重、瘤重,按下式计算试药的抑瘤率。结果见表1-2。

体内抗肿瘤实验结果表明:异穿心莲内酯具有显著的抗肿瘤活性,其作用强度与穿心莲内酯相当。
以上实验结果证明:异穿心莲内酯具有显著的抗炎及抗肿瘤作用。

2、一种抗炎和抗肿瘤的药物组合物,其特征在于:含有权利要求1所示结构的异穿心莲内酯作为活性成分以及药学上可接受的载体。
3、权利要求2的药物组合物在制备抗炎药物中的用途。
4、权利要求2的药物组合物在制备抗肿瘤药物中的用途。
