Abstract

Se proporciona un mutante de transcetolasa, su gen codificante y aplicación, se refiere al campo técnico de la ingeniería enzimática. El mutante de transcetolasa proporcionada tiene la secuencia que se muestra en SEQ ID NO:16, es una secuencia en que se produce la mutación de aminoácidos. Se proporciona un mutante de la transcetolasa R con alta estereoselectividad, cuya secuencia de aminoácidos es como se muestra en SEQ ID NO:1; el vector de expresión recombinante que contiene el mencionado gen codificante; la célula huésped que contiene el mencionado vector de expresión recombinante; un método para preparar el mutante de la transcetolasa R con alta estereoselectividad; la aplicación del mencionado mutante de la transcetolasa R al formar la R-dihidroxicetona aromática con alta estereoselectividad a través de catalizar el aldehído aromático; la aplicación de su gen codificante al formar la R-dihidroxicetona aromática con alta estereoselectividad a través de catalizar el aldehído aromático. La estereoselectividad de la p-tiamfenilfenildihidroxicetona producida por el mutante EcTK1_YYH mediante la catálisis es del 95,2% de ee(R), con una alta actividad y una alta estereoselectividad.
