Abstract
La presente invención se relaciona con un método para la producción de microglía a partir de células madre que comprende los pasos de a) inserción dirigida de una secuencia de nucleótidos que codifica para una proteína reguladora transcripcional en un primer sitio de puerto genómico seguro; y b) inserción dirigida en la secuencia codificadora del factor de transcripción PU.1 (SEQ ID NO: 1) en un segundo sitio de puerto genómico seguro, donde el gen se une operativamente a un promotor inducible, el cual es regulado por la proteína reguladora transcripcional; expresión de PU.1 (SEQ ID NO: 2); y cultivar las células madre recibidas de los pasos a) y b) con exposición a al menos un factor de crecimiento o molécula pequeña que imita la señalización durante al menos una etapa del desarrollo embriónico de la microglía o proliferación, diferenciación o polarización de microglía adulta. Además, la presente invención se relaciona con la microglía obtenida por los métodos de la presente invención y varios usos de la misma.
