Claims (21)
1. Способ защиты микробиологически полученного вещества от окисления, включающий
a) трансформацию микроорганизма, способного продуцировать указанное вещество, рекомбинантной нуклеиновой кислотой, содержащей ген каталазы, кодирующий полипептид, функционально связанный с одной или более контрольными последовательностями, которые управляют продуцированием полипептида в хозяине экспрессии, где полипептид представляет собой
i) каталазу, имеющую по меньшей мере 60% идентичность аминокислотной последовательности с SEQ ID NO. 1, или
ii) фрагмент каталазы согласно i), где фрагмент имеет каталазную активность,
или
обеспечение микроорганизма, содержащего в качестве гетерологичного гена, ген каталазы, кодирующий
i) каталазу, имеющую по меньшей мере 60% идентичность аминокислотной последовательности с SEQ ID NO. 1, или
ii) фрагмент каталазы согласно i), где фрагмент имеет каталазную активность,
b) культивирование микроорганизма при условиях, позволяющих экспрессию указанного гена каталазы.
2. Способ ферментации для получения продукта ферментации, включающий стадии
a) трансформации микроорганизма, способного продуцировать указанное вещество, рекомбинантной нуклеиновой кислотой, содержащей ген каталазы, кодирующий полипептид, функционально связанный с одной или более контрольными последовательностями, которые управляют продуцированием полипептида в хозяине экспрессии, где полипептид представляет собой
i) каталазу, имеющую по меньшей мере 60% идентичность аминокислотной последовательности с SEQ ID NO. 1, или
ii) фрагмент каталазы согласно i), где фрагмент имеет каталазную активность, или '
обеспечения микроорганизма, содержащего в качестве гетерологичного гена, ген каталазы, кодирующий
i) каталазу, имеющую по меньшей мере 60% идентичность аминокислотной последовательности с SEQ ID NO. 1, или
ii) фрагмент каталазы согласно i), где фрагмент имеет каталазную активность, и
b) культивирования микроорганизма при условиях, позволяющих экспрессию указанного гена каталазы и также позволяющих продуцирование указанного продукта ферментации.
3. Способ по п. 1 или 2, где полипептид содержит замещение, делецию и/или вставку в одном или более положениях.
4. Способ по п. 1 или 2, где контрольная последовательность представляет собой или содержит индуцируемый промотор, предпочтительно индуцируемый пероксидом водорода промотор.
5. Способ по п. 1 или 2, где нуклеиновая кислота представляет собой конструкцию нуклеиновой кислоты или экспрессионный вектор.
6. Способ по п. 1 или 2, где микробиологически получаемое вещество или продукт ферментации представляет собой белок, предпочтительно фермент, и предпочтительно выбрано из группы, состоящей из протеазы, амилазы, карбогидразы, липазы, целлюлазы, пуллуланазы, кутиназы, пектиназы, маннаназы, арабиназы, галактаназы, ксиланазы, оксидазы, например лакказы, пероксидазы, изомеразы, трансферазы, киназы и фосфатазы, где предпочтительными протеазами являются субтилизин протеаз.
